Abstract

Pemurnian enzim cathepsin C diperoleh melalui penggunaan colom kromatografi S-Sepharose FF, Sephacryl S-200 HR, Concanavalin A-Agarose, Affi-gel 501 dan DEAE-Sephacel. Dari 160 g ginjal ikan mas diperoleh 0,33 mg enzim cathepsin C, dengan hasil pemurnian 150 kali. Berat molekul cathepsin C adalah 170 kDa yang terdiri dari 5 subunit, dengan berat molekul berkisar antara 10 sampai 20 kDa. Cathepsin C adalah oligomerik protein dan aktif sebagai eksopeptidase (pH 5) maupun endopeptidase (pH 7)

Abstract

This research aimed to examine the effect of baker’s yeast as immunostimulant on growth of nile tilapia and to find out a proper dose of baker’s yeast that promoted optimal growth of fish. Juveniles with an average weight of 31.83 g were obtained from stock available at Aquaculture Study Program of the Faculty of Fisheries and Marine Sciences.  This research was conducted at the experimental ponds located at the Faculty of Fisheries and Marine Sciences for three months. During the experiment, fish was fed with pellet supplemented with baker’s yeast as treatment for four weeks as much as 4%/bw/day, twice daily.  The doses of baker’s yeast consisted of 0, 10, 20, 30, and 40 g/kg pellet. Fish growth including absolute and relative growth was measured at two weeks interval.  Research results showed that  after two weeks of feeding, supplementation of baker’s yeast did not affect the growth of fish but after four weeks of feeding, supplementation of baker’s yeast significantly increased fish growth. Fish fed pellet supplemented with 10 g baker’s yeast displayed the highest growth. As conclusion, supplementation of baker’s yeast in fish food could enchance the growth of fish.   Keywords:  Oreochromis niloticus, immunostimulant, baker’s yeast, growth

Abstract

This research aimed to gather data and information as well as to evaluate the waters condition and water quality suitable for ornamental coral aquaculture, so the characteristic of Arakan waters for ornamental coral aquaculture could be examined.  The research applied Explorative Descriptive method with Purposive Sampling.  Research stations were established at 20 points representing the overall ecosystem of Arakan waters.  Data collected  consisted of physical conditions (bottom substrate, flow rate, turbidity, water depth, salinity, temperature, TSS) and  chemical conditions (pH, DO, nitrate, phosphate). Data were analyzed using Matching Method by matching the data obtained from sea water standard quality table for marine biota in accordance to Kep.Men. LH No. 51/2004 and data from Hutabarat and Evans (1984).  Based on physical and chemical conditions, Arakan waters was suitable for ornamental coral aquaculture or categorized as suitable. However, water temperature at the time of measurement was not proper,  thus monitoring of this parameter for longer period was required. Abstract

Abstract The purpose of this study was to investigate the effect of vitamin C with different doses of the long time ripe gonads, egg hatchability and larval survival of catfish (Clarias sp). This research used Complete Randomized Design (CRD) with four treatments each with three replications. The treatment in this study were different doses of vitamin C namely 0, 600, 1200 and 1800 mg / kg of feed. Data was analyzed usung analysis of variance and continued with the Smallest Real Difference test.     Parent female catfish  used was 1.5 years age with a weight of 800-900 g. Feed used was  pellets supplemented with vitamin C.  Fish was reared in four tanks measuring 2 x 1 x 1 m and fed every morning and evening ad libitum. The results of the study was the addition of vitamin C in the diet significantly influenced the speed of gonad maturation, egg hatchability and larval survival. Treatment with the addition of 1200 mg / kg of feed was the best treatment with gonadal maturation speed achieved on 39.33 days, the highest egg hatchability 83% and survival of larvae was 7.66 days.   Keywords: Clarias gariepinus, Vitamin C, gonadal development, egg hatchability, larval survival.

Abstract

The aimed of the study was to isolate and select the probiotic Lactobacillus from carp intestine potential for use in aquaculture. Carp as a source of probiotics with a size of about 200 g was  taken from the Center for Fish Hatchery and Disease Control (BP3I), Tateli. After aseptic surgery, the intestine was separated and cleaned, weighed as much as 1 g, put in a mortar and crushed until smooth. The scoured results were put into a tube and added 9 ml NaCl to get a ratio of 1:10. The tube that already contains intestinal scouring was then centrifuged at a speed of 1000 RPM for 10 minutes. The centrifugation process was carried out 2 times to get the supernatant that has been cleared of debris. The supernatant was removed and diluted to obtain a concentration of 10-2  to 10-3. The solution was then spread on MRS (De Man Rogosa and Sharpe Agar) media and incubated at 28oC in the Incubator for 24-48 hours. The bacteria that grew in culture fmedia was reisolated several times to get a single pure colony.. The pure colonies were propagated in the same media to be further identified through gram staining, biochemical, proteolytic, amylolytic, cellulolytic, antibiotic resistance, antagonistic, pathogenicity tests. The results of the isolation obtained the characteristics of a colony of milky white color, round and convex with long cells, gram-positive and confirmed based on biochemical tests as Lactobaccilus sp. The bacterium also had proteolytic, amylolytic, cellulolytic, antagonistic, antibiotic-sensitive activity that were characterized by the formation of clear zones in the area around probiotic discs, and did not cause death either as a disease or as a result of poisoning. Thus it can be concluded that Lactobaccilus sp. isolated from carp intestines can be used as probiotics for cultivation.

Abstract

This study was aimed to examine the effect of immunostimulant bakasang (fermented product from fish) and to determine the optimal dose of bakasang in increasing the non-specific immune response of eels. The eels were obtained from Institute of Freshwater Aquaculture (BPBAT) Tatelu. Bakasang as immunostimulant ingredient contained Lactic Acid Bacteria (LAB) with the bacteria concentration of 105 CFU/ml was prepared by diluting with clean water to get the doses desired. The feed was subsequently mixed with the commercial fish feed in powder form and mixed well with water to form paste. This research used Two-ways ANOVA with 4 different types of treatments A (0 ml/kg feed),   B (50 ml/kg feed), C (100 ml/kg feed), D (150 ml/kg feed) and 3 repetitions. The fish were feed with treatments diets for 3 weeks with a dose of 3% body weight per day with a frequency of twice a day (at 08:00 am) and the afternoon (16.00 pm). The parameters measured was Total Leukocyte Count (TLC) which was measured every week (t0, t1, t2, t3). The results indicated that the addition of bakasang on eels’feed gave a significant increased on TLC, and that the highest TLC was found in the dose of 100 ml/kg feed. This shows that the optimal dose of bakasang in increasing the TLC of eels  was 100 ml/kg feed after administration of 3 weeks. Keywords: Eels, Bakasang, Total Leucocytes Count

Abstract

The purpose of this research was to identify the types of epiphyte that attach to seaweed in cultivation area of Tumbak Village, Posumaen District, Southeast Minahasa Regency.  Identifications were done directly in cultivation site owned by farmers and  in Laboratory of Aquaculture Technology, Faculty of Fisheries and Marine Science Sam Ratulangi University.  Observations were conducted three times at two weeks interval.  Weight of seaweed was measured to obtain data of daily growth rate.  In addition, water quality was also measured in situ including dissolved oxygen, pH, temperature and  salinity.  Research result found there were  three species of epiphyte attaching on seaweed K. alvarezii  including Ulva reticulata Forsskal, Dictyota dicotoma (Hudson), and Achanthophora spicifera (J. Agardh).  Seaweed tested was still clean until the end of research period  and only a few of epiphyte was observed.  This condition, supported by good water quality during the research period, resulted in very good growth of seaweed with  a daily growth rate achieved 4.43%. Keywords: seaweed, epiphyte, daily growth rate

Abstract

This study aimed to analyze the growth of sea cucumber gamat (Stichopus hermanii) on different substrates. This research was conducted in the waters of Talengen Bay, Tabukan Tengah District, Sangihe Islands Regency. The implementation time starts from January - February 2020 for 42 days of research. This research consisted of 3 treatments: A (mud substrate), B (sandy mud substrate), and C (sandy substrate and coral splinter), each treatment consisted of 3 replications. In each experimental unit, four sea cucumbers were placed. This research used a net made from tagaho, with an iron frame which is divided into 3 parts. The size of the container was 30 x 30 x 30 cm3. The results showed that the best absolute length of sea cucumber gamat was the in Treatment A (6.17 cm), then Treatment B (4.67 cm), and Treatment C (1.25 cm). The results of the analysis of the daily growth rate:  Treatment A (1.17%) got the best growth rate results compared to Treatment B (0.87%) and Treatment C (0.29%).

Abstract

The aim of this research was to count the total leukocytes and phagocytosis activity of goldfish seeds treated with probiotics. The test fish used were goldfish seeds measuring 5-8 cm. 500 fish will be taken from the Tateli Fish Cultivation and Development Center (BP3I). This study used a completely randomized design (CRD) with 5 treatments, where each treatment had 3 replications. The treatments used were probiotic bacteria from the intestines of goldfish with different concentrations. The treatments tested consisted of: A: without the addition of probiotics, B: addition of probiotics 1x106 cfu / mL, C: addition of probiotics 1x107 cfu / mL, D: addition of probiotics 1x108 cfu / mL, E: addition of probiotics 1x109 cfu / mL. Each concentration of bacteria that has been prepared is then sprayed on the commercial feed evenly using a sprayer and then dried to room temperature. After drying the feed was coated with 2% egg yolk and dried again at room temperature. Fish are stocked in an aquarium measuring 60x40x40cm3 with a density of 20 fish / aquarium. Each aquarium is equipped with an aerator and a water pump for recirculation. The fish were then given feed that had been added with probiotics at the same dose and frequency of administration as in the acclimatization process. The treatment is given for 6 weeks. At the end of the treatment feeding, fish samples were taken from each experimental unit to measure immune parameters, namely total leukocytes and phagocytic activity. Based on the results of research using the probiotic Lactobacillus sp. which originated from the intestines of goldfish and added to the feed had a significant effect on increasing total leukocytes and phagocytosis activity and the optimal concenration was  1x107 - 1x108 cfu / mL with a duration of 6 weeks of administration

Abstract

Kappaphycus alvarezii seaweed is one of the main commodities of aquaculture which is economically important and has been cultivated commercially. In the Kappaphycus alvarezii seaweed cultivation system, the attack of ice-ice is a major obstacle because it can harm the cultivation business. The spread of the disease can occur vertically and horizontally. This study aimed to observe the horizontal transmission rate of ice-ice disease in cultivation activities of K. alvarezii in Maba Waters, East Halmahera Regency. The results showed that the attack of ice-ice caused a decrease in growth and experienced death. The presence of this disease in seaweed cultivation can result in a decrease in seaweed production. This is a major problem faced by K. alvarezii seaweed cultivation managers and requires serious handling with proper management
