Abstract

Background: Oryza sativa L. is a major food commodity and a source of carbohydrates for most of Indonesia’s population. Based on BPS data, rice production has declined every year. The decline in rice production is caused by attacks by plant pests (OPT). Xanthomonas oryzae is a bacterium that causes bacterial leaf blight (BLB) in rice, which causes yield losses of up to 60-80%. An environmentally friendly alternative for controlling Xanthomonas oryzae is using endophytic bacteria Bacillus sp. and Pseudomonas fluorescens. Methods: The experiment was conducted using two tests: in vitro and in vivo assays. The in vitro assay was arranged in a completely randomized design with 4 treatments and 4 replications, resulting in 16 experimental units. The experimental unit consisted of one Petri dish containing Nutrient Agar (NA) medium. Antagonistic activity was evaluated using the disk diffusion method against Xanthomonas oryzae. The in vivo assay was arranged in a completely randomized factorial design with 10 treatments and 3 replications. Each replication consisted of 6 rice plants grown in a polybag, resulting in a total of 180 plants as experimental units. Treatments consisted of different concentrations of antagonistic bacterial suspensions applied to soil before pathogen inoculation. The pathogen Xanthomonas oryzae was inoculated onto rice plants during the vegetative stage. Results: Bacillus sp. and Pseudomonas fluorescens can suppress Xanthomonas oryzae disease. The results showed that a consortium treatment of these two bacteria produced the highest inhibitory power of 9,125 mm. In vivo testing, the consortium treatment with 20 ml/plant resulted in the lowest disease intensity of 6,06% and an incubation period of 4 days. Conclusions: The consortium treatment of Bacillus sp. and Pseudomonas fluorescens was able to suppress bacterial leaf blight disease compared to other treatments.

Abstract

Background: This study investigated the effects of varying drought stress durations on the growth and accumulation of antioxidant compounds in basil (Ocimum basilicum L.), a plant sensitive to water availability and threatened by prolonged drought due to climate change. Methods: Four treatment groups were established: control (C0: watered daily), C1 (watered every 3 days), C2 (watered every 6 days), and C3 (watered every 9 days). Morphological parameters (root fresh/dry weight, leaf area) and physiological parameters (Relative Water Content/RWC, root-shoot ratio) were measured, along with biochemical parameters (total phenolic content/TPC and total flavonoid content/TFC). Results: Analysis showed that increasing drought duration significantly reduced root fresh weight (C0: 21 g to C3: 3 g), leaf area (C0: 13.2 cm2 to C3: 7.46 cm2), and RWC, which dropped drastically from 71.70 % in C0 to 30.50 % in C3. However, a mild stress level (C1) significantly induced the highest TPC (97.61 ± 2.94 mg GAE/g extract) and TFC (115,97 ± 3.23 mg QE/g extract), surpassing the control (C0: TPC 76.98 ± 3.55 mg GAE/g extract, TFC 85.42 ± 5.08 mg QE/g extract). The concentrations of these compounds then decreased significantly at severe stress levels (C2 and C3), indicating metabolic breakdown. Conclusion: Mild drought stress (C1) is optimal for inducing secondary metabolite accumulation in basil as an adaptive response, while severe stress significantly inhibits growth and reduces physiological capacity.

Abstract

Background: Biology learning at the high school level requires a solid grasp of concepts and strong learning interest for students to understand the material optimally—particularly the Nervous System topic, which is characterized by abstract and complex concepts. Learning interest is a crucial factor influencing student engagement in the learning process and academic achievement. This study aims to analyze the learning interest of Grade XI students regarding the Nervous System topic at HAS Sepakat Private High School and to identify differences in learning interest between classes XI-1 and XI-2. Methods: This study employed a descriptive quantitative approach. The sample consisted of 60 Grade XI Science students at HAS Sepakat Private High School, divided into two classes: XI-1 and XI-2. Data were collected through Likert-scale questionnaires, observation, and documentation. Data analysis involved descriptive statistics (total scores, means, standard deviations, and percentages) and inferential statistics (normality tests, homogeneity tests, One-Way ANOVA, and Welch's test) using SPSS. Results: The results showed that the total learning interest score reached 3,419 out of an ideal score of 4,500 (75.98%), placing it in the high category. However, the indicators for active engagement and curiosity showed relatively lower achievement compared to other indicators. The mean learning interest score for class XI-1 was 38.39 (standard deviation: 1.927), while for class XI-2, it was 37.45 (standard deviation: 4.356). The One-Way ANOVA yielded a significance value of 0.319, and Welch's test yielded a significance value of 0.326. Both values ​​exceeded 0.05, indicating no significant difference in learning interest between students in class XI-1 and class XI-2. Conclusion: The learning interest of Grade XI students regarding the Nervous System topic at HAS Sepakat Private High School falls into the high category. Nevertheless, active engagement and curiosity still need to be enhanced to support a more optimal learning process. Furthermore, no significant difference in learning interest was found between students in classes XI-1 and XI-2.
