Abstract

Background: Botanical literacy is a part of biological literacy, which focuses on botanical aspects. Biologically literate students need to know and understand the characteristics of scientific knowledge, the values of science, and the methods and processes of scientific inquiry. Botanical literacy needs to be empowered in biology learning. This is because a lack of understanding of botany will lead to plant blindness. Methods: This study follows the research and development model of Lee and Owens, which includes the stages of analysis (needs analysis and front-end analysis), design, development, and implementation, and evaluation. The subjects of this research were students of the Department of Biology at the State University of Malang, totaling 62 participants. Data collection instruments were validation sheets, practicality questionnaires, and botanical literacy instruments to assess product effectiveness. Data were analyzed using descriptive analysis and inferential analysis.. Results: The results showed that e-atlas was highly valid, achieving 100% from material experts, 94% from media experts, and 94% from educational practitioners. Based on the results of individual, small-group, and field tests, e-atlas was found to be very practical. Based on the ANOVA test, the significance value is 0.000, which is smaller than α = 0.05 (Sig < 0.05). Conclusions: Based on these data, it is concluded that the e-atlas of Flower Morphology with Local Potential can be used for learning and has a significant effect on students' botanical literacy.

Abstract

Background: Streptomyces spp. are well known for their ability to produce antibiotic compounds with potential applications in plant disease management, but research in mangrove ecosystems tends to be limited. This study aims to explore and characterize Streptomyces isolates from the mangrove rhizosphere based on morphological characteristics and pathogenicity properties to find candidates as biocontrol agents. Methods: Morphological characterization was carried out macroscopically and microscopically, while hypersensitivity tests and soft rot tests were carried out to determine non-pathogenic properties. Results: A total of 24 isolates of Streptomyces sp. were successfully isolated with the morphological characteristics of circular colonies, filamentous and curved colony edges, convex, flat, raised, and umbonate colony elevations, aerial mycelial pigmentation with six color variations, and substrate mycelial pigmentation with six color variations. Microscopically, two types of spore chains were obtained, namely open spiral and closed spiral. All isolates were nonpathogenic based on the negative results of the hypersensitivity test and soft rot test. Conclusion: The mangrove rhizosphere provides a promising and morphologically diverse source of non-pathogenic Streptomyces isolates. Identification in this study was limited to morphological characterization; therefore, molecular analysis is needed to confirm the isolate species. These findings provide preliminary information as a basis for selecting isolates for further biological control studies.

Abstract

Background: This study was conducted due to the need for innovative biology learning models that can enhance students’ critical thinking and collaboration skills, which remain insufficiently developed in conventional classroom learning. Methods: This research is a quasi-experimental with pretest-posttest control group design. Three XI classes at SMA Negeri 8 Makassar were used as samples. Experimental class 1 applied blended learning with problem-based flipped classroom type, experimental class 2 applied blended learning with traditional flipped classroom type, while one control class followed conventional learning. Critical thinking skills were measured using essay tests based on FRISCO indicators, and collaboration skills were measured through observation sheets. The data were analyzed with prerequisite tests (normality and homogeneity), which showed a significance value <0.05. Furthermore, ANCOVA test was conducted and continued with LSD test. Results: The results of the analysis showed that blended learning with a flipped classroom approach had a significant effect on improving students' critical thinking and collaboration skills. ANCOVA revealed a significant effect of learning model on critical thinking skills (F(2,84)=6.353, p=0.003) and collaboration skills (F(2,84)=7.197, p=0.001). In addition, the pretest covariate also significantly influenced critical thinking (F(1,84)=53.869, p<0.001) and collaboration skills (F(1,84)=372.999, p<0.001). LSD test results further showed significant differences among the experimental and control groups. Conclusions: The findings of this quasi-experimental study indicate that blended learning with a flipped classroom approach was more effective in enhancing students’ critical thinking and collaboration skills than conventional learning.

Abstract

Background: C-Reactive Protein (CRP) is an important biomarker for infection and inflammation; however, quantitative assessment using ECLIA is costly and requires adequate laboratory facilities and trained personnel. This study aims to optimize antibody concentration and conjugate volume, and to evaluate the accuracy of the detection range of an LFIA-based CRP rapid test at two clinical cut-offs (10 and 40 mg/L). Methods: Experimental research was conducted by optimizing antibody concentrations on the T1 and T2 lines, optimizing CGC volume, and testing sensitivity, specificity, and the correlation between color intensity and Image. Results: The optimal antibody concentrations were obtained at T1 of 0.075 mg/mL and T2 of 0.025 mg/mL with a CGC volume of 2 µL. Sensitivity and specificity were 95.7% and 100% at T1, and 83.3% and 95.5% at T2. Conclusions: The developed CRP rapid test has adequate diagnostic performance as a semi-quantitative screening tool in primary healthcare facilities.

Abstract

Background: Scientific literacy is a crucial competence for students in facing the rapid development of science and technology in the 21st century. This study aimed to examine the effect of a science literacy–based learning model on improving senior high school students’ scientific literacy in biotechnology, using a quasi-experimental, non-equivalent control group design. Methods: This study employed a quantitative approach using a quasi-experimental non-equivalent control group design. The research involved two intact classes: an experimental class that received instruction using the Science Literacy-Based Learning model and a control class that was taught using the conventional discovery learning model. Results: The study involved 70 tenth-grade students, consisting of an experimental class of 35 taught using the science literacy–based learning model and a control class of 35 taught using discovery learning. Students’ improvement in scientific literacy was measured using the N-Gain index, and group differences were analyzed using an independent-samples t-test. The results showed that the experimental class achieved a high N-Gain score of 0.73, whereas the control class achieved a moderate N-Gain score of 0.60. The t-test indicated a significant difference between the two groups (p < 0.05). Conclusion: These findings demonstrate that the science literacy–based learning model has a significant and positive effect on students’ scientific literacy skills in biotechnology.

Abstract

Background: Free radical exposure poses significant threats to public health by damaging cells and causing degenerative diseases like cancer and premature aging. Antioxidants are vital in neutralizing these radicals. Jatropha curcas L. leaves contain bioactive compounds with promising antioxidant and wound healing properties, warranting evaluation in a practical gel form. Methods: This study used a true experimental design with a posttest-only control group; quercetin was the control, while the treatment groups received different gel formulations of Jatropha curcas L. leaf extract. Results: The pure extract had very strong antioxidant activity (IC50 0.32 ppm). Gel formulations (Formula 2 and Formula 3) also exhibited strong antioxidant activity, with IC50 values of 30.90 ppm and 35.85 ppm, respectively; Formula 2 performed better. Quercetin showed strong activity at 1.22 ppm. One-way ANOVA (p < 0.01) confirmed significant differences between groups. Conclusions: The Jatropha curcas L. leaf extract gel has significant antioxidant potential and is effective in preventing oxidative damage, supporting its use in safe, gel-based herbal skin products.

Abstract

Background: Write The coconut crab (Birgus latro Linnaeus, 1767) is the largest terrestrial crustacean and is currently classified as Vulnerable by the IUCN. Although its distribution in Central Sulawesi has been documented, information on population size and habitat preferences remains limited for many islands within its range. This study aimed to estimate the population size of B. latro on Bakalan Island, Banggai Archipelago, Indonesia and to identify its preferred habitats. Methods: Field surveys were conducted from November to December 2023 at five sampling stations on Bakalan Island. Coconut crabs were surveyed at night using baited plots established in representative habitat types, including rocky coastal areas, tree-root zones, and coconut plantations. Results: A total of 42 individuals were recorded during the survey. The population size of Birgus latro on Bakalan Island is estimated at 46,950 individuals, corresponding to a population density of 0.00112 individuals/m². The species was most frequently encountered in habitats characterized by rock crevices, tree-root systems, coconut trees (Cocos nucifera), coastal pandanus (Pandanus tectorius), Terminalia catappa, and shrub vegetation. Environmental conditions across sampling stations ranged from 26–31.1°C. Conclusions: Bakalan Island supports an important population of B. latro associated with a variety of coastal habitats that provide shelter and food resources. Given the species’ Vulnerable conservation status and ongoing habitat disturbance on small islands, the protection of key habitats is essential for the long-term conservation of coconut crab populations in the Banggai Archipelago.

Abstract

Background: The implementation of deep learning in junior high school science is often constrained by limited learning media that can provide immediate and meaningful feedback to facilitate student reflection. Objective: This study aims to develop Real-Time Feedback Responsive Learning (RT-FReL), an interactive medium designed to provide immediate feedback and facilitate reflective deep learning in Junior High School Science. Method: The study employed the Research and Development (R&D) method using the ADDIE model. Data were collected through expert validation sheets, pretest-posttest instruments (measuring conceptual mastery as an indicator of deep understanding), student response questionnaires, and reflective journals. The subjects were Grade IX students at SMPN 1 Ciamis, involving two different groups for a small-scale trial (N=15) and a subsequent large-scale field trial (N=30). Results: Expert validation showed that RT-FReL was very feasible, with an average score of 90.23%. The effectiveness test indicated improvement in student understanding, with N-Gain scores of 0.60 in the small-scale trial and 0.62 in the large-scale trial, both categorized as moderate. Additionally, 93% of students perceived that the real-time feedback feature motivated them to reflect on their learning process. Conclusions: RT-FReL was judged very feasible by experts and showed promising results in improving students’ understanding and encouraging reflective responses during learning. The media can be used as a technology-based alternative to facilitate more responsive science learning in junior high school.

Abstract

Background: Anthracnose caused by Colletotrichum sp. is one of the major diseases limiting red chili production. Trichoderma sp. is a promising biological control agent, but its large-scale application requires suitable propagation media. Methods: This study aimed to evaluate alternative media for the mass propagation of Trichoderma sp. and the suppression of anthracnose through in vitro and in vivo experiments using a Completely Randomized Design (CRD). The in vitro assay involved an antagonism test using five treatments with four replications, measuring the inhibitory effect via the dual culture method. The in vivo assay assessed the incubation period and disease intensity using five treatments with four replications each, yielding 20 experimental units. Each experimental unit consisted of five plant samples, making a total of 100 plants. The collected data were analyzed using ANOVA and the Honestly Significant Difference (HSD) test at a 5% significance level. Results: Alternative media affected the growth and effectiveness of Trichoderma sp. In the in vitro test, inhibition against Colletotrichum sp. ranged from 30.67% to 52.58% at 5 days after inoculation. In the in vivo test, disease intensity at 84 days after planting ranged from 6.25% to 22.50% in treated plants, compared with 28.33% in the control. Corn-rice and rice-bran media showed the most favorable overall performance in supporting propagation and disease suppression. Conclusions: Alternative media can support the propagation of Trichoderma sp. and maintain its antagonistic potential against anthracnose in red chili. Corn-rice and rice-bran media showed the best overall potential among the tested media.

Abstract

Background: Bacterial leaf blight is one of the major diseases in rice caused by Xanthomonas oryzae pv. oryzae, which can significantly reduce rice productivity. The use of chemical bactericides may cause environmental problems; therefore, biological control using antagonistic bacteria such as Bacillus spp. is considered a sustainable alternative. This study aimed to evaluate the inhibitory potential of Bacillus mycoides Ba-11 and Bacillus sp. Bcz-20 against Xanthomonas sp. under in vitro conditions. Method: The experiment was conducted using a completely randomized design with three treatments: Bacillus mycoides Ba-11, Bacillus sp. Bcz-20, and a control (sterile distilled water), each with nine replications. Antagonistic activity was tested using the well diffusion method on nutrient agar. The diameter of inhibition zones was measured after 24–48 hours of incubation and analyzed using the Dunn test at a 5% significance level. Result: Both Bacillus mycoides Ba-11 and Bacillus sp. Bcz-20 formed inhibition zones categorized as moderate to strong, indicating their ability to suppress the growth of Xanthomonas sp. Although Bacillus sp. Bcz-20 produced a larger inhibition zone, statistical analysis showed no significant difference between the two isolates. Conclusion: Both Bacillus mycoides Ba-11 and Bacillus sp. Bcz-20 have comparable and significant potential as biological control agents against Xanthomonas sp. through antibiosis mechanisms.
