Abstract

Tithonia diversifolia can be used as an antidiabetic, so it is necessary to study the safety of its use, especially the side effects on the liver and kidneys. This study aimed to determine the effect of using T. diversifolia leaf extract through histomorphometry observations of the liver and kidneys of hyperglycemic albino rats. The study design used a completely randomized design (CRD). This study used 20 male albino rats which were divided into five treatment groups, there were T0 (rat normal/control), T1 (hyperglycemic rat without T. diversifolia leaf extract), T2 (hyperglycemic rat administered with Glibenclamide 10 mg/kg BW), T3 (hyperglycemic rat administered with T. diversifolia leaf extract 150 mg/kg BW), T4 (hyperglycemic rat administered with T. diversifolia leaf extract 300 mg/kg BW). Every treatment was repeated four times. The damage of hepatocyte and the glomerular cell was observed through histological structure observation by histomorphometry method using a photomicrography microscope (Olympus BX51). The results indicate that there were significant differences (P < 0.05) in the variable of hepatocytes diameter and there was no significant different (P > 0.05) result on glomerular diameter, as well as kidney and liver weight. It was concluded that T. diversifolia leaf extract of 150 mg/kg BW and 300 mg/kg BW are safe to be used as antidiabetic. It does not cause any side effects on the liver and kidneys of hyperglycemic albino rats. Thereby T. diversifolia leaf extract can be further tested as preparation of biopharmaca which can be used as herbal medicines for diabetics.

Abstract

Traditionally, the genus Philautus Gistel, 1848 contains small Rhacoporid frogs that lack vomerine teeth and a direct development tadpole stage. In Java, this genus consists of 3 species, including Philautus aurifasciatus, Philautus jacobsoni, and Philautus pallidipes. It is known that Philautus aurifasciatus and Philautus jacobsoni are sympatric species from Mount Ungaran and are morphologically difficult to distinguish. Therefore, information is needed through the phenetic characters of each population in the area to identify differences in their morphology. This study aimed to analyze the morphological characteristics of  Bush Frog Philautus spp. on Mount Ungaran in terms of meristic character through a direct field survey and comparison methods. Furthermore, it was conducted by observing 32 specimens of Philautus spp. from Mount Ungaran and compared with the Philautus aurifasciatus from Mount Gede Pangrango with 18 meristic characters. The results showed that Philautus spp. is similar to Philautus aurifasciatus in terms of their meristic characters. The results of the meristic analysis showed that Philautus spp. has a variety of dorsal colors from brown, purple, green, and gray with two types of patterns including patterned groups (resembling letters H, X, and V) and abstract. Furthermore, the throat, stomach, and lower thighs are granular with three snout shapes (protruding, round, and sloping). It has a loreal oblique tympanum and canthus rostralis. The forelimbs are not webbed while the hindlimbs are half webbed, and the inner metatarsal is an elongated oval in shape. Meristic analysis provides a basis for strengthening the taxonomic status of Philautus spp on Mount Ungaran, as well as information the distribution in Mount Ungaran.

Abstract

Antiobesity medication is available as therapeutic compounds that can reduce fat digestion by the inhibition of pancreatic lipase. Actinobacteria have the potency as source of bioactive compounds with various biological function including as pancreatic lipase inhibitor.  However, the potency of endophytic actinobacteria from Rhododendron spp. as source of pancreatic lipase inhibitor producer has not been reported yet. The aim of this study was to examine the potential of pancreatic lipase inhibitory activity of 23 endophytic actinobacteria from Rhododendron spp.; to characterize their colony based on morphology and molecular analysis. Screening test of pancreatic lipase inhibitor was conducted using the supernatant of endophytic actinobacteria, lipase pancreatic porcine (L3126) and p-nitrophenyl butyrate. The supernatant of selected isolates was extracted using ethyl acetate. The result showed that various inhibitory activities ranging between 0.00 until 91.69%. There were 11 out of 23 isolates that have potential as pancreatic inhibitor. Amongst them, the extract of four selected isolates, i.e. RZP 1.3, RSSB 3.2, RSS 2.1, and RJB F3.2 demonstrated inhibitory percentage of more than 80%. The RJB F3.2 extract showed to have IC50 value by 431.48 µg mL-1 compared to control, i.e. Xenical (89.07 µg mL-1). Phytochemical analysis exhibited that the extract of the selected isolates contained alkaloid which may function as pancreatic lipase inhibitor. Based on the morphological character, the selected isolates have various morphological colonies and 16S rRNA gene sequence revealed the sequence homology to Streptomyces spp. The data clearly indicate that endophytic actinobacteria from Rhododendron spp. have potency as pancreatic lipase inhibitor producer and further studies could be explored for the development of antiobesity agent.

Abstract

Begonia is an ornamental plant that has a high level of morphological diversity. Begonia propagation is mostly done vegetatively and very rarely to propagate sexually. Therefore, this study was conducted to determine the ability of six types of Begonia seeds to germinate. Six types of Begonia, namely B. x albopicta W. Bull 'Coral Rubra', B. gambutensis Ardi & D.C.Thomas, B. multistaminea Burt-Utley, B. nelumbifolia Schltdl. & Cham., B. ulmifolia Wild. and B. varipeltata D.C.Thomas. were grown in a petri dish and placed in a growth chamber with a temperature of 23.6 ℃, humidity of 74.5%, and a light intensity of 3,431 lux. The results showed that the different types of Begonia showed significant differences in all the parameters tested. B. nelumbifolia required 7.89 ± 0.18 days to germinate and is the fastest time than other types. The highest percentage of seed germination was also obtained by this species, amounting to 63.22%. In addition, B. nelumbifolia showed the best response to the germination rate coefficient, germination rate index, and germination index. This study revealed complete information about the germination of six types of Begonia. In addition, the ability to produced many seeds and the ease of germination made this plant have the potential to be cultivated through seeds. Furthermore, the information can be used as a basis for seed storage as an ex-situ conservation method to maintain genetic diversity.

Abstract

Azadirachta indica A. Juss (Neem) is a medicinal plant that contains antioxidants, antihyperlipidemia, and has antidiabetic activity. This research aimed to examine the effect of neem leaf ethanolic extract on blood glucose levels of white rats given high-fat rations. The male white rats (Rattus norvegicus L.) with two months of age were used. The treatment of test animals consisted of six treatment groups and four replications, namely: the control group (P0); given high fat rations + duck egg yolk 2.5 ml/200gBW (P1); given high-fat rations + 8 mg/200gBW simvastatin (P2); and P1+ the dose of neem leaf ethanolic extract of 75; 100; and 125 mg/200gBW (P3; P4; and P5). Blood-glucose levels were analyzed by colorimetric enzymatic method using Roche Diagnostics Cobas C reagents kits. Blood-glucose level was determined using Roche / Hitachi cobas c systems automatically. Homogeneous and normally distributed data of blood-glucose levels, body weight and feed consumption were analyzed statistically using ANOVA followed by Duncan's test with a significance level of 5% using SPSS 16.0 software. The results indicated that giving the neem leaf ethanolic extract of could reduce the blood glucose levels. In this study, it was concluded that the neem leaf ethanolic extract of could potentially be used as an alternative to cure diabetes mellitus.

Abstract

Phycocyanin from Spirulina platensis extract has anticancer activity against various types of cancer cell cultures. However study about its effect on colon cancer cell lines, especially the WiDr, has not been reported before. This study aimed to reveal the anticancer activity of phycocyanin from Spirulina platensis extract on WiDr cells. The research was an in vitro experimental study, with the investigation on cytotoxicity also antiproliferation as the anticancer parameters. Both cytotoxicity and antiproliferation test was conducted through MTT assay to observe the visualization and inhibition of proliferation of different concentrations of phycocyanin in several incubation times on the WiDr colon cancer cell line. The obtained data were then processed statistically with the Two Way ANOVA test at a significance value of p 0.05 and followed with the Post Hoc test since there were significant differences. Based on the results, it could be postulated that phycocyanin extracted from freshwater Spirulina platensis was classified as non-toxic (IC50 of 855 µg/ml). Consequently, it is less potential to be used as the treatment for colon cancer. However, phycocyanin could inhibit the proliferation of the WiDr cell for approximately 47.4%, specifically at the concentration of 1710 µg/ml for 72 hours. It could be concluded that freshwater phycocyanin is less effective as an anticancer substance. The benefit of this study is to provide the new scientific evidence of the contrary results of freshwater phycocyanin activity from Spirulina platensis as an anticancer agent of colon cancer.

Abstract

Gold in nature is covered by rocks which contain sulfide minerals such as pyrite, chalcopyrite, arsenopyrite, and others sulfide minerals. Chemolithotrophic bacteria have the ability to oxidize the sulfur compounds and can be used in the process of releasing gold from carrier rocks which contain sulfide minerals. This research aimed to explore and identify the chemolithotrophic bacteria from gold mining areas as well as determine their potential for sulfur oxidation. The methods used in this study were exploring the potential of bacteria in sulfur oxidation and describing the variety of bacteria that were isolated from gold mining areas by 16s rRNA identification. The results showed that there were six isolates from isolation with Starkey solid medium, i.e. Bl-1, B1-2, B1-3, B1-4, B1-5 and B1-6 that were similar to Paenibacillus sp., Enterobacter ludwigiis train E8-13, uncultured Burkholderia sp., Uncultured bacterium clone N4.5, Bacillus subtilis strain CICC 10023, and Bacterium enrichment culture clone 02 respectively. The B1-3 isolate showed the highest increase of sulfate compound in the medium (8.04 % at 649.55 ppm). This indigenous bacteria will be able to be used to release gold from rock which contains sulfide minerals and reduce the use of hazardous chemicals commonly used in gold mining.

Abstract

AbstrakPembelajaran melibatkan interaksi dosen, mahasiswa, dan sumber belajar. Interaksi antara ketiganya membutuhkan strategi pembelajaran. Penelitian ini bertujuan menghasilkan strategi pembelajaran biologi di Jurusan Biologi Unnes yang telah berkomitmen mengaplikasikan kurikulum berbasis kompetensi dan konservasi dengan ciri pendekatan jelajah alam sekitar (JAS). Metodologi penelitian menggunakan pendekatan kualitatif. Informannya dosen dan mahasiswa yang mengajar dan belajar biologi umum, mikrobiologi, dan taksonomi hewan. Data yang diperoleh berupa deskripsi kegiatan belajar mengajar dan lesson study.  Data-data dianalisis kemudian dinarasikan. Hasil penelitian menunjukkan pembelajaran biologi dengan pendekatan berpusat pada siswa yang dikhususkan pada kegiatan penjelajahan alam sekitar dapat dilakukan menggunakan strategi yang berbasis pada cara belajar siswa aktif dan kooperatif. Strategi-strategi tersebut adalah pembelajaran kontekstual, pembelajaran partisipatif, dan pembelajaran inkuiri. AbstractLearning involves the interaction of lecturer, students, and learning resources. The interaction between them requires a learning strategy. This research aims to generate learning strategies of biology in the Department of Biology Unnes which has committed to apply the competence- and conservation-based curriculum of features the environmental exploration (JAS) approach. The research methodology uses a qualitative approach. The informants are the faculty members and the students who, respectively, teach and learn biology, microbiology, and animal taxonomy subjects. The data obtained were in the form of description of learning activities and lesson studies. The data were then analyzed narratively. The results showed that the student-centered learning of environmental exploration may be conducted using active- and cooperative-based strategies. Examples of these strategies are contextual learning, participatory learning, and inquiry learning.

Abstract

Gynura procumbens has a potency to produce raw material for pharmaceutical industry. Liquid culture systems have significant effects on multiplication rates of organ, so the propagation technology to increase biomass of this plant or organ using the liquid culture system was necessary. This research was conducted to compare biomass production of adventitious roots of G. procumbens in different liquid culture (shake flask, temporary immersion bioreactor and balloon-type bubble bioreactor). Adventitious roots culture was maintained in Murashige and Skoog liquid medium supplemented with IBA (indole butyric acid) 5 mg/L and different concentrations of sucrose (10, 30, 50 g/L). Result showed that the highest biomass production (fresh weight) was in shake flask that was 3.9-fold higher than initial explant, while in temporary immersion system was 5.12-fold higher than initial explant, and in balloon- type bubble bioreactor was 13.1-fold higher than initial explant. The highest enhancement of adventitious roots was occurred at sucrose supplementation of 50 g/L. Based on this research we found that the best type of liquid culture to increase biomass of G. procumbens adventitious roots was balloon-type bubble bioreactor. Scaling-up of adventitious root culture are necessary to fulfill the need of raw material for pharmaceutical industry and ballon-type bubble bioreactor was the most suitable method to scale up the performance.
