Abstract

Xylo-oligosaccharide derived from corncob hemicellulose has been reported to possess antioxidant activity. In order to assess the effective scavenging of xylo-oligosaccharide, we conducted in vitro studies based on self-made xylo-oligosaccharide with DPPH (2,2diphenyl-1-picrilhydrazil) method. Xylo-oligosaccharide was prepared with enzymatic hydrolysis. The enzyme used for hemicellulose hydrolysis was endo-β-xylanase enzyme from PC-01 isolated bactrerium. PC-01 isolated bacterium used in this study was Pacet hot spring which was isolated from East Java. Endo-β-xylanase enzyme is an extracelluler enzyme. There was about 0.199 U/mL after purification and dialysis process. Hydrolisis product of hemicellulose A and B from corncob were analyzed with TLC (Thin Layer Chromatography) and HPLC (High Performance Liquid Chromatography). This analysis showed that hydrolysis product of hemicellulose B had a lot of xylo-oligosaccharide hydrolysis product of hemicellulose than Xylo-oligosaccharide hydrolysis product of hemicelluloses A. Xylo-olygosaccharide was analyzed as on antioxidant activity. Xylo-oligosaccharide hydrolysis product ofhemicellulose B (IC = 48.96) has higher antioxidant activity than Xylo-oligosaccharide hydrolysis product of hemicellulose A (IC 50 50 = 92.302). The toxicity of xylo-oligosaccharide can be calculated by the value of LC 50 (Lethality concentration). LC of xylooligosaccharide derived from corncob hemicellulose was 400 ppm so that xylo-oligosaccharide has anti tumor activity because xylooligosaccharide has LC 50 < 1000 ppm.

Abstract

Artocarpus dasyphylla with the local name “cempedak utan” is a member of Artocarpus Genus from Moraceae family. It is a rare and endemic plant from east region of Indonesia. The purpose of this study was to determine the antihypercholesterolemic effect of ethyl acetate extract from A. dasyphylla toward the level of total cholesterol, LDL, and HDL of hypercholesterolemic Rattus norvegicus as the prevention effort of atheroschlerosis and CVD. It was due to antioxidant activities of phenolic compounds. The powder of stem bark of Artocarpus dasyphylla was extracted by maceration and partition method. Phenolic total of ethyl acetate extract from stem bark of A. dasyphylla was determined with Folin Ciocalteu reagent, it was 9,86 mg GAE/g of extract. In vivo experiment toward Rattus norvegicus with hypercholesterol diet used randomized post test only control group design. Ethyl acetate extract with the treatment doses 75, 150, 225 mg/kg body weight showed antioxidant activity by decreasing total cholesterol and LDL level to normal level. Paradox result occured to HDL level, the level of HDL decreasing as the increase of dose of sample, but still above the threshold. The best anti-hypercholesterolemic activity was shown by treatment with the dose of ethyl acetate extract from stem bark of A. dasyphylla 150 mg/kg body weight of Rattus norvegicus. Abstract

Gynura procumbens has a potency to produce raw material for pharmaceutical industry. Liquid culture systems have significant effects on multiplication rates of organ, so the propagation technology to increase biomass of this plant or organ using the liquid culture system was necessary. This research was conducted to compare biomass production of adventitious roots of G. procumbens in different liquid culture (shake flask, temporary immersion bioreactor and balloon-type bubble bioreactor). Adventitious roots culture was maintained in Murashige and Skoog liquid medium supplemented with IBA (indole butyric acid) 5 mg/L and different concentrations of sucrose (10, 30, 50 g/L). Result showed that the highest biomass production (fresh weight) was in shake flask that was 3.9-fold higher than initial explant, while in temporary immersion system was 5.12-fold higher than initial explant, and in balloon- type bubble bioreactor was 13.1-fold higher than initial explant. The highest enhancement of adventitious roots was occurred at sucrose supplementation of 50 g/L. Based on this research we found that the best type of liquid culture to increase biomass of G. procumbens adventitious roots was balloon-type bubble bioreactor. Scaling-up of adventitious root culture are necessary to fulfill the need of raw material for pharmaceutical industry and ballon-type bubble bioreactor was the most suitable method to scale up the performance.

Abstract

AbstrakTelah berhasil diisolasi senyawa golongan fenilpropanoid dengan nama “skopoletin” dari kulit umbi ubi jalar (Ipomoea batatas L.). Ekstraksi senyawa dilakukan dengan metode maserasi menggunakan pelarut metanol, dilanjutkan dengan partisi menggunakan n-heksana dan etil asetat. Pemisahan dan pemurnian senyawa dilakukan menggunakan teknik kromatografi kolom gravitasi. Struktur senyawa dianalisis berdasarkan data spektroskopi UV/Vis, 1D, dan 2D-NMR. Kata kunci : fenilpropanoid, skopoletin, Ipomoea batatas L. AbstractIt has been isolated phenylpropanoid group compound named scopoletin from the tuber peel of Ipomoea batatas L. Extraction of this compound was done by maceration method using methanol solvent, followed by partition with n-hexane and ethyl acetate. Separation and purofication of compound was done by gravity column chromatography techniques. Abstract

Abstract Essential oil from Apium graveolens (celery), a species belonged to Apiceae, was isolated, identified, and tested its activity against fungi Pityrosporum ovale, fungi that cause dandruff problem. Essential oil isolation was conducted by steam distillation. The identification with GC-MS showed that the major compound of celery essential oil was 3-isobutylidenphalide. The anti-fungal activity test was performed using pure celery oil and celery oil added with virgin coconut oil (VCO) and paraffin oil that are usually used as components in hair tonic formulation. The bioactivity test showed that celery oil, whether in pure form or in hair tonic formulation exhibited excellent growth inhibition activity of Pityrosporum ovale Keywords : essential oil, celery, Apium graveolens, hair tonic, anti-dandruff, Pityrosporum ovale

Abstract

AbstrakSintesis ZSM-5 dilakukan dari metakaolin terdealuminasi tanpa penambahan templat organik. Sintesis dilakukan melalui pengaturan suhu hidrotermal mulai suhu 120oC, 150o, dan 170oC. Hasil sintesis dikarakterisasi dengan difraktometer sinar-X , spektrometer FTIR, dan adsorpsi/desorpsi nitrogen. Pengaturan suhu hidrotermal pada 120oC selama 24 jam belum menghasilkan ZSM-5, sedangkan sintesis pada suhu 150oC selama 24 jam diperoleh hasil ZSM-5 dengan kristal yang cukup baik. Hidrotermal pada 170o tetap dapat membentuk ZSM-5 dengan pengotor mordenit dan kurang kristalin dibandingkan sintesis pada 150oC. Kata kunci: AbstrakSintesis ZSM-5 dilakukan dari metakaolin terdealuminasi tanpa penambahan templat organik. Abstract

Two phenolic compoundnamely 4-ethoxy-3-methoxyphenol and methyl-3,4-dihydroxybenzoic have been isolated fromDioscorea hispida Dennst. The isolation of phenolic compounds was done by maceration methods using methanol, followed by partition with n-hexane and ethyl acetate. The process of separation and purification used various chromatography techniques including vacuum liquid chromatography, column chromatography, and radial chromatography. The structure of isolated compounds were determined by spectroscopic methods including UV-Vis, IR, 1D and 2D NMR. The ethyl acetate extract was evaluated for DPPH free radical scavenging activity assay. The IC50of ethyl acetate extract was 415 ppm. Abstract

AbstrakTelah berhasil diisolasi senyawa golongan fenil propanoid  dengan nama “skopoletin” dari ekstrak etil asetat  umbi Ubi Jalar (Ipomoea batatas L.). Ekstraksi senyawa fenolik dilakukan dengan metode maserasi menggunakan pelarut metanol, dilanjutkan partisi menggunakan n-heksana dan etilasetat. Pemurnian dilakukan dengan kromatografi kolom gravitasi dan ktomatografi radial menghasilkan senyawa golongan fenil propanoid. Struktur skopoletin diidentifikasi berdasarkan data spektrum UV, 1D dan 2D NMR.Kata kunci : fenil propanoid, skopoletin, dan Ipomoea batatas L. AbstractPhenylpropanoid group compound has been isolated as “scopoletin” from ethylacetate extract of tuber flesh Sweet potatoes (Ipomoea batatas L.). Abstract

Background: Tentena City is one of the tourist centers with rapid economic growth located in Poso Regency because Poso Lake is very famous and able to change the economic level of the community. But there are still 20% of people who live very poor and 60% of the lower middle class and the remaining 20% of the middle to upper middle. Through the youth community Pukat Tentena is expected to foster and encourage millennials to develop vegetable and fruit entrepreneurs using more modern cultivation techniques. Objectives: empowering the youth community in Tentena city, Pamona Puselemba Subdistrict, Poso Regency, Central Sulawesi with entrepreneurial tomato fruit products and melons produced by hyrdoponik drip irrigation system. Methods: This community service activity was carried out in Tentena village, Pamona Puselemba Subdistrict, Poso Regency, Central Sulawesi Province with TENTENA PUKAT Youth Community partners starting from May to September 2021. Results: Mitra has successfully cultivated tomato and melon plants with hydroponic drip irrigation system techniques in green houses until they are obtained salesable results. The average number of tomatoes per stalk is 4.05 and the average fruit diameter is 5.1 cm; While the average weight of melons is 1.2 -1.8 kg. Conclusion: The results of this community service activity provide benefits for partners, namely providing additional knowledge about how to cultivate tomatoes and melons with hydroponic drip irrigation system techniques and increase partner income by selling tomatoes and melons from hydroponic cultivation.

Abstract

Carbon nanodots (CNDs) have widely received great attention as a result of favorable optical, electrical, optoelectrical, biocompatible, and non-toxic properties these nanoparticles possess. However, the exploration of nanoparticle from natural raw material was limited. In present work, the carbon dots were produced from catechin isolated from Uncaria gambir through a simple and facile process. Carbon nanodots were further produced by the pyrolysis process of catechin, which allowed it for carbonization. Owing to its unique properties such as photoluminescence with an emission peak at 500 nm (lex = 380 nm), average size diameter about 5 nm and non-toxic; Cat-CNDs were incredibly potential for staining targeted tumor cells. The staining ability by confocal microscopy observations showed their green fluorescence images which meant that the CNDs easily penetrated HeLa cells via endocytosis. The resulting CNDs which were analyzed using some significant techniques approved that the prepared Cat-CNDs were tremendously dispersible and water-soluble, good colloidal stability, excellent biocompatibility, favorable hydrophilicity, high photostability, and non-toxicity.
