Abstract

Sodium nitrite (NaNO2) found in vegetables, drinking water, and cured meats, can damage tissue because it is an oxidant. Plant phytochemicals such as quercetin are antioxidants. This study aimed to determine the potential of red okra pods ethanol extract (ROE) to repair kidney damage in mice (Mus musculus) induced by NaNO2. The red okra pods were extracted three times with saturated ethanol. The experiment used 36 male BALB/c mice aged 6-8 weeks and body weight of about 28 g. There are six research groups, namely, normal control, negative control (exposure to NaNO2 50 mg/kg BW), treatment of exposure to NaNO2 and administration of ROE at doses of 25, 50, 75, and 100 mg/kg BW. Sodium nitrite and ROE were given daily for 23 days by gavage. On day 24, the serum was isolated. Blood urea nitrogen (BUN) and creatinine (Cre) levels are measured to assess kidney function, as well as measuring the oxidant malondialdehyde (MDA) and the antioxidant enzyme of superoxide dismutase (SOD). The kidneys were made histological preparations and analyzed on the proximal convoluted tubule (PCT).  All data were statistically analyzed (α=0.05). This study indicated that the administration of ROE at a 100 mg/kg BW dose is the most optimal in repairing damage to the PCT with increased normal cells and reduced necrosis. Besides, it degraded BUN, Cre, and MDA levels in the serum of mice exposed to NaNO2 compared to the other treatments. All doses of ROE promoted the SOD level. ROE restore kidney tissue, especially on PCT to normal. Kidney damage due to exposure to NaNO2 preservatives can be reduced by administering ROE. ROE prevents kidney damage through an increase in antioxidant enzymes. ROE can be used as a food ingredient as a source of antioxidants, thereby reducing the impact of oxidant compounds.

Abstract

Osteochilus vittatus is a freshwater fish that needs to be developed because it tastes savory and can be used as a biocleaning agent. Research on the effect of supplementation of Spirulina platensis in nilem fish that is maintained in a biofloc system on digestive enzyme activity has never been done before. The objectives of this study were to determine the effect of S. platensis supplementation on Osteochilus vittatus digestive enzyme activity maintained in the biofloc system and to obtain the most optimum level of S. platensis supplementation in improving the  digestive enzymes activity. The study was conducted experimentally with Completely Randomized Design with four treatments and five replications. Osteochilus vittatus were fed with Spirulina platensis level of 0, 2, 4, and 6 g kg-1 for 56 days. Enzyme activity was measured on days 0 and 56 with three kinds of buffer solutions. Amylase and protease activity was analyzed using ANOVA with a confidence level of 95%. The results showed that the highest amylase activity in the liver was 4.764 ± 1.705 U mg-1 protein and proximal intestine was 2.328 ± 0.838 U mg-1 protein. The highest protease activity was found in the liver and proximal intestine with activity of 6.536 ± 0.911 U mg-1 protein and 6.207 ± 2.195 U mg-1protein. Spirulina platensis supplementation level of 6 g kg-1 feed is the most optimum level (p0.05). Cultivation of fish with S. platensis supplementation in feed can increase enzyme activity and the fish maintenance in biofloc systems can improve water quality.

Abstract

Black Soldier fly larvae is an insect capable to convert organic waste into body biomass. The purpose of this research was to determine the productivity, mass balance of the consumption process, and content of protein and fat of prepupa which has a potency as a livestock feed. This research used 6-day-old larvae, they were fed with cassava skin that has been smoothed with a variety of feeding doses: 12.5; 25; 50; 100; and 200 (mg / larvae / day) with ratio of feed and water 2: 1. The results showed that the feeding of 200 mg / larva / day produced the best larval growth with biomass productivity of 1.54 mg / day, residue yielded of 67.1 mg / day. The organic waste was convertion into larvae biomass. In this study, approximately consumption process scale is feed successfully converted to biomass 2.77%, metabolism 6.98%, and 90.24% residue. Protein content is 25.7%. It can be concluded that the larvae are able to convert organic waste into high protein biomass and potentially being used as a livestock feed. This research acts a model to predict mass balance of substrates for feed. The benefit of this research is the science approach used in this study that can be used as a reference by researchers and the community to determine the biomass production of an organism.

Abstract

Peningkatan jumlah kendaraan bermotor berpotensi meningkatkan pencemaran udara terutama di jalan-jalan protokol. Untuk mengurangi semakin tingginya bahan pencemar yang dihasilkan kendaraan bermotor, perlu adanya pohon-pohon yang berfungsi sebagai penyerap dan penjerap bahan pencemar dan debu di udara yang dihasilkan kendaraan bermotor. Tujuan penelitain ini adalah mengetahui peranan tanaman di jalan-jalan protokol Kota Semarang dilihat dari kualitas dan kuantitasnya. Data mengenai jenis tanaman peneduh yang ada di jalan protokol Kota Semarang dihitung dengan metode line intercept. Hasil penelitian menunjukkan bahwa kualitas udara di Kota Semarang sudah cukup mengkhawatirkan, hal ini dipandang dari kadar CO yang relatif tinggi. Hasil penelitian terhadap jenis tanaman yang ada di jalan protokol Kota Semarang, dapat diketahui jenis tanaman yang ditanam di 5 jalan protokol Kota Semarang berjumlah 29 jenis. Jenis maupun jumlah tanaman yang ada pada masing-masing stasiun penelitian tidak berpengaruh positif terhadap kadar bahan pencemar udara yang ada. Simpulan dari penelitian ini ialah kualitas maupun kuantitas tanaman yang ada pada jalan-jalan protokol lokasi penelitian tidak berpengaruh positif terhadap kadar bahan pencemar udara yang ada. Hal ini dikarenakan jenis dan jumlah tanaman pada masing-masing jalan protokol tidak sesuai dengan tanaman peneduh yang mempunyai fungsi sebagai penjerap dan penyerap polutan udara.The increasing number of motor vehicles might potentially increase the air pollution in main roads. To reduce the increasing concentration of pollutants generated by motor vehicles, the trees are planted to absorb the pollutants and the dust in the air. The objective of the research was to understand the role of the plants along the main roads in Semarang City, from point of view of quality and quantity. Data on the kind and density of shedding plants along the main roads of Semarang City was collected using line intercept method. Result showed that the air quality of Semarang City has been worrying in terms of the high concentration of CO. There were 29 kinds of trees planted along 5 main roads in Semarang City. The kind and the number of plants in each station did not influence positively to the concentration of the air pollutants. It was concluded that the quality and the quantity of the plants along the study sites did not positively influence the concentration of the air pollutants. It was presumed that the kind and the number of plants along each ain road in Semarang was not appropriately functioning as the absorbants of the air pollutants.

Abstract

This research aim was to reveal the genetic variability of local corn cultivars from Kisar Island using microsatellite molecular marker. The use of microsatellite markers is based on one reasons that the genetic information is polymorphic, and will be better to identify genetic diversity of plants including corn. Corn cultivars used were obtained from farmer in Kisar Island, and three reference varieties as outgroup were obtained from Institute of Cereals in Maros South Sulawesi. Polymerase chain reaction was conducted following the protocols from Fast Start kit (Qiagen-USA). The data obtained then analyzed with MVSP 3.1A Software for dendogram construction. The results showed that the appearance of DNA bands was vary (monomorphic and polymorphic) for local cultivars as well as reference varieties, with one or two bands on each primer. Based on the dendogram, there are three main clusters with similarity index ranged from 20% -100%. It can be concluded that there is very low similarity and distant kinship of local cultivars. The novelty of this research is knowing kinship relationship between local corn in Kisar Island which is not known yet. This results is expected to provide benefits regarding the breeding program, and for instance, it can be an important information regarding the development of local corn as a source in assembly the superior corn cultivars.

Abstract

Dental caries is a major cause of tooth loss in children and young adults. Dental caries have been linked to the situation of underprivileged families, nutritional imbalance, and poor oral hygiene techniques, including lack of tooth brushing or flossing the teeth, and also have a genetic etiology. Dietary habits and dental hygiene practice can result in high caries in school children. This research aimed to reveal the correlation between dietary habits and dental hygiene practice with dental caries among school children in urban area of Semarang. The subjects of this research are the elementary student 7 – 9 years old enrolled in schools located in at urban area in Semarang in 2016 and the mother of a student who became the study sample. Data were statisically analyzed usingbivariate analysis and multivariate analysis. Based on the research result, it can be concluded that: there was no correlation between total carbohydrate intake, refined carbohydrate intake, fiber intake, dental hygiene practice with dental caries, bottle feeding and duration of bottle feeding were assosiated with dental cariest-score. Abstract

Currently, agriculture has prospective potential to be developed as a tourist attraction, one of which is agricultural tourism with an ecological perspective. The objective of the research was to examine the potential of ecology-based agrotourism as a means for agricultural education in Besur Village, Lamongan Regency. The research applied a quantitative descriptive approach in the form of scoring, weighting, and ranking of the categories being assessed. The research data consisted of primary and secondary data. The data was collected using direct observation and in-depth interviews. The data analysis was carried out using for the feasibility of  agrotourism area. The results showed that the area of Besur Village was classified as suitable (potential) with zone I that had the highest KKA value (2.6) as the object of the agrotourism. Potential ecological aspects obtained analysis 36 species of food producing plants. The highest Importance Value Index (IVI) of plant species was  recorded for Oryza sativa spp. (83.76%) and Zea mays L. (27.11%). Analysis of the physico-chemical properties of soil resulted in moderate or sufficient fertility. Analysis of public perceptions obtained was 4.08 and classified as good for the development of agrotourism in Besur Village. The novelty of this research was to obtain of the potential for agrotourism management in Besur Village through an ecological approach based on the concept of Healthy Plant Management.

Abstract

Staphylococcus aureus is gram-positive bacteria that often infect the skin. S. aureus has been experiencing resistance to several antibiotics. One of the solutions to overcome the resistance is by using garlic that is believed by the society can overcome bacterial infection. The study aimed to find out the influence of the variation in concentration of local single clove garlic essential oil used on the inhibition and damage of morphological structure of S. aureus bacteria. The inhibition test was conducted using disc diffusion method. The experiment groups consisted of  1% DMSO as negative control, vancomycin 30µg/ml as positive control and single clove garlic essential oil (25 mg/ml, 50 mg/ml, 75 mg/ml and100 mg/ml) as treatment groups. The diameter of inhibition zone was measured using calipers. The morphological damage of the bacterial cells can be seen using Scanning Electron Microscopy (SEM) with magnification of 25000x. The result of Kruskal-Wallis test analysis indicated that the extract of local garlic essential oil has inhibitory activities against S. aureus bacteria (P0.05). The damage to the morphological structure of bacterial cells with the administration of 30 µg/ml vancomycin was equal to 100 mg/ml single clove garlic essential oil extract. Single clove garlic essential oil can be used as an alternative treatment for skin infection diseases by inhibiting S. aureus growth.

Abstract

Cassava stalk can be converted into sugar-based product by using microorganism. Unfortunately, lignin act as a barrier of optimal bioconversion. Cassava stalk needs pretreatment process for removing this barrier. The effect of lime pretreatment on microstructure of cassava stalk fibers and the growth of Aspergillus niger FNCC 6114 were observed in this research. The cassava stalks were reduced into 0.147- 0.297 mm size and pretreated with 1 % Ca(OH)2. Lime pretreated and unpretreated cassava stalk was used as solid medium for Aspegillus niger FNCC 6114. The effect of pretreatment method on fibers microstructure of cassava stalk was evaluated through SEM micrograph. The growth and metabolism activities of Aspergillus niger FNCC 6114 were monitored through SEM micrograph of media after fermentation. The other parameters examined were changes in glucosamine, reducing sugar levels, and spores’ quantity. Lime pretreatment altered microstructure of cassava stalk fibers. However, cassava stalk without lime pretreatment gave better growth of Aspergillus niger FNCC 6144 based on metabolism activities parameters. Cassava stalks is suitable as media for Aspergillus niger FNCC 6144 through solid state fermentation. For better growth of Aspergillus niger FNCC 6144 fine-sized cassava stalk should not be lime pretreated. The results of this study  provide  information about the pretreatment of cassava stems which was effective in supporting the growth of Aspergillus niger. Enhancements the utilization of cassava stems by using fungi, for example Aspergillus niger can overcome the accumulation of organic waste that can interfere with environmental sustainability.

Abstract

A research on the use of sucrose and lactose as carrier media to protect Acetobacter xylinum cell subjected to freeze drying has been done. The aim of the research was to know the number of the viable cells from dried culture and to know the concentration of the carrier medium that would give best result. The best result is sucrose at the concentration of 15% that produced 28.2 x 106 viable cells/ ml of rehidrated culture. The rehidrated culture used in the research was Schramm Herstin medium.
