Abstract

Improving learning quality in 21st can not be separated from contextual learning and student-centered learning paradigm. The contextual lesson study program conducted in learning-forest prototype to build a learning community. The objectives of this research were to improve learning activities of postgraduate students in Biology Education department and to build a learning community. The implementation of lesson study was conducted in the Biology Learning Innovation subject for postgraduate students while practicing to observe open lesson in the undergraduate students which used learning forest-prototype. The postgraduate students took roles as planner, observer, and reflector in the plan, do (open lesson), and see (reflection) activities. The implementation was done in three cycles in even semester of academic year 2015/2016. Students learned collaboratively and contextually. The postgraduate students’ learning activities were observed by six observers from lecturer colleagues. The research results showed that the students were able to implement planning, open lesson, and reflection properly. The average of student’s learning activity grade was 91.11% from all of students, with the grade averages for planning, open lesson, and reflection activities were 88.89%, 93.33%, and 91.11% respectively. The implementation of this lesson study in the learning forest-prototype can be done in other relevant subjects to strengthen learning activities.

Abstract

Papua has so many wide lowland areas that cover high diversity in plant from life-forms.  In contrast, there is a lack of information concerning species diversity.  The objectives of the research were to describe species of plant life-forms and described conspecific associations between small individuals and large individuals in natural tropical rainforest, south Papua.  Then, 46 nested plots were placed systematically in natural tropical rainforest of Boven Digoel Regency, Papua.  Density, frequency, dominance and importance value index (IVI) were analyzed to describe plant diversity, while detrended correspondence analysis (DCA) was to describe conspecific association between small individuals and large individuals.  Results found 2040 individuals in this forest consisting 194 plant species.  Those are from pteridophytes, angiosperms and gymnosperms.  The plant life-forms are herbs, orchids, palms, pandans, shrubs, rattans, vines and trees in which of the plant life-forms, trees are the most abundant according to number of species.  In the dominant plant species, two types of conspecific association occurred between small individuals and large individuals.  Analysis of DCA revealed that three are large plant species grow along with their small individuals.  Another type is that small individuals grow far from their large individuals.

Abstract

This reseach aimed to determine the proper cassava drying technique to produce suitable flour properties for food product applications. The method used was proximate analysis and calculation of Gross Energy (GE) value. Furthermore, the results of the study were analyzed using the Two Way ANOVA statistical test followed by the real difference test using the Tukey analysis. Results of the analysis showed that there was no significant difference between the two drying techniques (α0.05). To get good quality flour in the application of food products made from Nuabosi cassava flour is to use oven drying and sun drying techniques. The novelty of this study is that there has been no previous research on the quality of Nuabosi cassava flour using oven drying and sun drying techniques. The benefit of this research is to support the government in the food diversification program on foods made from Nuabosi cassava flour, where Nuabosi cassava is one of the leading types of food in Ende district.

Abstract

Endophytic bacteria are beneficial microorganisms that interact with host plants without causing any interference or damage to plants. This research aimed to obtain endophytic bacteria isolated from the root tissue of rice plants (Oryza sativa L.) which have potential to produce IAA hormones,, identify the endophytic bacteria in morphologically and physiologically, and analysis of the selected isolate 16S rRNA genes. Based on the results of this research, there  were six endophytic bacteria isolates obtained. They have high morphological diversity and differen ability producing IAA hormones. The highest concentration of IAA (425 ppm) was obtained  isolates from  EAP3. Isolate EAP3 also produce inhibit the growth of Xanthomonas oryzae with a 5.2 mm inhibition zone. Based on the biochemical test, EAP3 had 60% similarity with Enterobacter asburiae. Analysis of the 16S rRNA gene showed that EAP3 had the highest similarity with Enterobacter asburiae strain U4 by 99%. This research data is considered as new information about the potential of endophytic bacteria from the roots of rice plants (Oryza sativa L.) which is capable of producing IAA hormones and is able to inhibit pathogenic bacteria. This research provides information that can be used as a basis for developing endophytic bacteria as biological fertilizers.

Abstract

Ganoderma sp. Banyumas 1 isolate that reffered as Ganoderma sp. is a new discovered isolate from Banyumas, Central Java, Indonesia expected to have a potential properties of antioxidant of medicinal mushroom. This study aimed to determine the antioxidant potential and the appropriate solvent for it’s extracting from Ganoderma sp. This research result showed that ethyl acetate was able to extract as many as 15.57%, while etanol was only able to extract 3.87% active compounds from dried 28 days old Ganoderma sp. mycelium cultivated in the Mushroom Complete Medium (MCM). Extract of ethyl acetate (non-polar) extraction of mycelium of Ganoderma sp. had a potential character as an antioxidant source and performed a better result than from ethanolic (polar) extraction as shown in the IC50 value. Extract from ethyl acetate extraction had an average IC50 value smaller than  from ethanolic extract (581.80 1285.67). Extract from ethyl acetate extraction resulted in a higher amount of phenol than that ethanolic extract 29.23 57.67. Inhibition percentage of both extracts at 65% was known to occur at concentration of 1000 ppm for ethyl acetate extract and 2000 ppm for ethanolic extract. An important finding was that ethyl acetate can be used as appropriate solvent for extracting antioxidant compound better than ethanolic. In conclusion, the mycelium extract of Ganoderma sp. extracted with ethyl acetate and ethanol as solvent is potential to be used as a source of natural antioxidants. This research result has benefit in developing potency of local resources as herbal resources.

Abstract

Roadside trees along the national highway in Barangay Perrelos in Carcar City are trees that considered a heritage, and yet are affected by urban developments in the province. The study determined the profile of the roadside trees as well as the knowledge, attitudes, and conservation efforts of the locals in the area. Questionnaires and interviews were given to 50 respondents, and data gathered were analyzed using ANOVA, Pearson r and thematic analysis. Ecological indices such as abundance percentage and species richness were also used by the study. Results revealed that there are 108 identified roadside trees, where the species richness is 0.962 and acacia trees are the most abundant (46.30%). Knowledge and attitudes of the respondents were high and low-to-moderate, respectively. Conservation practices towards the roadside trees included pruning, no burning near the trees and making of diversion roads, all adhering with existing environmental laws of the country. Tree abundance and richness, knowledge acquisition, attitude inculcation, and conservation efforts are manifestations of the educational values of roadside trees to the people. Research on these educational values of trees is novel as it provides importance on cultural heritage, environmental protection, and sustainable planning in the area. Implications to environmental education are formulated for the Philippine elementary and secondary education Science teaching.

Abstract

Meniran (Phyllanthus niruri), patikan kebo (Euphorbia hirta), and benalu (Loranthus sp.) have often been used by people as medicinal plants. This research aimed to measure the levels of flavonoids, phenolics, and ascorbic acid compounds, as well as the Free Radical Scavenging (FRS) activity of ethyl acetate extract from the mixture of P. niruri, E. hirta, and Loranthus sp. The FRS activity was measured with the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The level of compounds was measured by using the spectrophotometry method with specific reagents. The result of the FRS activity in ethyl acetate extract from the mixture of P. niruri, E. hirta, and Loranthus sp. varied depending on its dose. The measure of FRS in the P. niruri: E. hirta: Loranthus sp 0:0:1 (K-OOL) composition showed a strong result with a value of IC50 97.2 ± 2.1 ppm, while in the 0:0.5:0.5 (K-OEL) composition it was moderate with a value of IC50 147.6 ± 6.5 ppm. The other compositions showed weak and inactive results. The K-OOL composition had the highest flavonoid and phenolic content that were 298.8±0.00 mg QE/g extract and 141.5±2.85 mg GAE/g extract respectively. The composition with the highest ascorbic acid content (298.8±0.00 mg/g extract) was K-OEO composition. So far, research on medicinal plants is still limited to one type of plant. The combination of several types of plants in several formulations allows obtaining a composition that can produce maximum antioxidant capacity. Therefore, this research is expected to produce a combination formulation of various types of medicinal plants that have the K-OOL composition very strong antioxidant activity and can be used as herbal medicines.

Abstract

Artemisia cina is one of a member of genus Artemisia that has potential as a medicinal plant. However, the levels of Artemisia medicinal bioactive compounds are very low. Polyploidization is an alternative method that can enhance of growth and secondary metabolite productions of plants. The aims of this research were to determine the effect of polyploid induction using colchicine and plant growth regulator toward plant growth, the chlorophyll, kaemferol and quercetin contents of A. cina. Four different A. cina used in this research consisted of two diploid genotypes (TWN and KJT) and two polyploid genotypes (J and M). Induction of mutant polyploid was conducted using colchicine and combination of plant growth regulator benziladenyl (BA) and 2,4-dichlorophenoxyacetic acid (2,4-D). The measured plant growth parameters were dry weight, leaf area, and plant height. The chlorophyll content of leaves was determined spectrophotometrically, and flavonoid content determined using HPLC. The result showed that the polyploid genotype (M) significantly decreased leaf and root growth compared to the KJT and TWN. In A. cina plants, polyploidization using colchicine is more effective in increasing the biomass than using combination plant growth regulator BA and 2,4-D. The flavonoid content of KJT was the lowest, and significantly different compared to the other plants. This study provides new information about the effect of polyploid on growth and flavonoid content in A. cina. This can be  useful information to develop A. cina to become a medicinal plant.

Abstract

Endophytic bacteria isolated from some various kind of plants are able to yield some active compounds which have a role as an antibacterial compound. This work aimed to isolate and to screen the Endophytic bacteria from Faloak seed in its charge in inhibiting two kinds of pathogenic bacteria, Staphylococcus aureus and Escherichia coli. There were six isolates of Endophytic bacteria isolated in this work. According to the screening result, one isolate which had the most potential antibacterial activity (marked by the formation of inhibition zone) against S. aureus and E. coli. That most potential isolate was then tested and identified for both biochemical properties and molecular 16S rRNA gene. The result of this study showed that the endophytic bacteria isolate of Faloak seed with the code of S1 had the similarity with Enterobacter xiangfangensis strain 10-17 by 93 %. The research about endophytic bacteria of Faloak plants was never conducted before. Thus this research was expected to give information about the potential of antimicrobial ability Faloak plants which can be utilized in the discovery of new antibiotic compounds which in the future are expected to overcome the problem of microorganism resistance to antibiotics. The use of endophytic bacteria is expected to prevents the extinction of Faloak plants due to excessive use.

Abstract

Tujuan penelitian adalah menghasilkan model meksint korefsi dengan pendekatan Jalajah Alam Sekitar pada pembelajaran Struktur Tubuh Hewan sehingga dapat dimanfaatkan sebagai salah satu alternatif kegiatan perkuliahan Struktur Tubuh Hewan. Metode yang digunakan dalam penelitian ini adalah Research Development dengan tahapan sebagai berikut: planning, design, dan development dengan 2 tahap evaluasi yaitu: uji alpha, uji betha, dan uji lapangan. Hasil uji alpha pada model pembelajaran meksint korefsi oleh ahli pembelajaran biologi memperoleh skor sebesar 3.13 dan hasil uji betha oleh mahasiswa sebagai pengguna memperoleh skor sebesar 3.64. Hasil uji t test antara dua rombel mahasiswa responden uji lapangan diperoleh thitung= 2.94 ≥ ttabel= 2.39 (α = 0.01; n = 60). Dengan demikian model meksint korefsi dengan pendekatan Jalajah Alam Sekitar layak digunakan sebagai salah satu alternatif pilihan model pembelajaran dalam proses perkuliahan Struktur Tubuh Hewan, sehingga disarankan perlu adanya proses diseminasi model tersebut The purpose of the study was to develop the meksint korefsi model with Jelajah Alam Sekitar* approach in the Animal Anatomy, for the alternative model in Animal Anatomy lecture. This is a Research and Development, with the following stages: planning, design and development with three stages of evaluation are: alpha testing, beta testing, and field testing. Alpha test results on the learning model meksint korefsi by expert of instructional biologists with scores was 3.13 and beta test results by the students as users gain score of  3.64. Field test results were analyzed by t-test statistical tvalue = 2.94 ≥ ttable = 2.39 (α = 0:01, n = 60). The result showed that the meksint korefsi model with Jelajah Alam Sekitar* approach was a feasible model in the learning of Animal Anatomy, so it was suggested to disseminate this model.
