Abstract

Soybean pod damaged by pod sucking bug (Riptortus linearis) is one of the constraint within soybean yield improvement in Indonesia. The research aimed was to identify the resistance of soybean genotypes to pod sucking pest. The experiment was conducted in Iletri’s screen house from March to June 2015. The experiment was arranged in Randomized Completely Block Design using 10 soybean genotypes with three replicates. The soybean resistance was evaluated using no-choice test. Data collected on number of pod/plant, number of seed/plant, seed weight per plant, number of attacked pod/plant, and number of attacked seed/plant. The result showed that the lowest percentage both of pod and seed damage was G511H/Anjasmoro//Anjasmoro-2-8, that was 25.83 % and 19.12 %, respectively. Based on the value of the percentage of seed damage, there were five susceptible genotypes, three moderately resistant, and two resistant genotypes. Based on the value of the percentage of pod damage, showed four susceptible genotypes, five moderately resistant, and a resistant genotype. G511H/Anjasmoro//Anjasmoro-2-8 was the only resistant genotype, and it could be used as a genetic source in the improvement of soybean resistance to pod sucking bug.How to CiteKrisnawati, A., Bayu, M. Abstract

Fungi have a lot of important function in the bionetwork resembling to decomposition and even nutrient cycling with respect to its symbiotic relationships with trees and other plants. This study aimed to assess the diversity and distribution of bracket fungi in Mt. Kilakiron, Portulin, Pangantucan, Bukidnon. Thus, floristic survey was conducted in one of the mountain of Mindanao to provide information as to what and how many species of bracket fungi are present in Mt. Kilakioron, Bukidnon, Philippines.  40 total species belonging to 17 genera and 5 family was collected and assessed. 12 individuals of Trametes versicolor were observe which favors dwellings with low temperature and high altitude are. Additionally, study revealed that most types of bracket fungi initiate in the site are decomposers and saprophytic in nature which contributes a lot in the process of putrefaction. The species of bracket fungi in Mt. Kilakiron clearly shows abundant and provides a large contribution to the ecosystem having high Shannon diversity index. The significance of each environmental factor is discussed in relationship to the ecology of the species. Bracket fungi are reliant on the climatic and conservational settings which makes them as excellent natural pointers of ecosystem in the forests of Mindanao Island, Philippines.

Abstract

Acanthuridae is a marine fish family with some members are known as ornamental species. Previous studies has been reported the diversity of Acanthuridae. However, there was no study on the phylogenetic relationship of the Acanthuridae from Ujunggenteng and Taman Manalusu southern coast of West Java. The purpose of this study was to provide information about the phylogenetic relationship among ornamental fishes under the Acanthuridae family of Ujunggenteng Beach Sukabumi and Taman Manalusu Beach Garut, West Java. The phylogenetic tree was reconstructed based on morphological characters using maximum parsimony algorithm in software PAUP with 1000 pseudoreplicates and with Halichoeres marginatus selected as an outgroup species. The phylogenetic tree had a consistency index of 0.6429. The value means that the tree was highly reliable due to its low homoplasy. Acanthuridae formed a monophyletic clade compared to the outgroup species by having seven synapomorphic characters. Acanthurid clade was divided into two subclades at generic levels by nine derived characters; three apomorphic characters in Acanthurus and six apomorphic characters in Naso. The tree indicates that Naso brevirostris and Naso lituratus represents basal group, while all species within genus Acanthurus are the most advanced species.  This study is the first to provide the data about the evolutionary relationship of ornamental acanthurid fish from the southern coast of West Java. Data on closely related species is one of the essential scientific basis on making policies regarding the sustainable use of those closely related species.

Abstract

Tawangmangu tangerine (Citrus reticulate Blanco subsp. tawangmangu) is one of prime local fruit from Tawangmangu region, Karanganyar, Central Java. This tangerine have good appearance, easy to peeled, sweet flavored fruit, and high production rate. However at 1984 Tawangmangu tangerine run into depreciation of population caused by CVPD (Citrus Vein Phloem Degeneration). As a result, this variety listed as endangeredThe effort of consevation by culture tissue technique can order to obtain prime seeds that free from virus or disease. This study was aimed to understand the effect of IBA and BAP in culture medium on growth of apical and lateral shoot of Tawangmangu tangerine. The treatment provided were IBA (0 ppm, 0.5 ppm, and 1 ppm) and BAP (0 ppm, 1 ppm and 2 ppm). The treatment in this research was using complete randomized factorial design in 9 different treatment. This research resulted that added combination of IBA and BAP affecting growth of Tawangmangu tangerine but the interaction of IBA and BAP did not happen. The highest shoot length occurs on treatment IBA 1 ppm+BAP 2 ppm while when the fastest shoot appears occurs on treatment IBA 0 ppm+ BAP 2 ppm and the highest shoot formed on treatment IBA 0.5 ppm+BAP 2 ppm. Benefit of the research Efforts to conserve Tawangmangu orange plants need to be done to maintain the diversity of germplasm and support the cultivation in a manner. One technique that can support the conservation of plants is by tissue culture techniques.

Abstract

Penelitian bertujuan untuk mendeskripsikan perlunya pengembangan penyedap rasa alternatif berbahan hidrolisat tempe dan proses pembuatannya, menguji jenis asam amino, kadar protein dan uji organoleptik. Metode eksperimen digunakan pada penelitian ini. Pengujian jenis asam amino melalui teknik kromatografi, kadar protein terlarut melalui metode Biuret dan uji organoleptik produk dibandingkan penyedap rasa sintetis. Hasil yang didapatkan adalah pengembangan penyedap rasa hidrolisat tempe perlu dilakukan karena alasan dampak mengkonsumsi penyedap rasa sintetis, pengembangan teknologi, sumber penghasilan dan potensi penelitian. Proses pembuatannya melalui penambahan sari nanas pada tempe yang telah dikukus dan diblender dengan perbandingan tempe:air:sari nanas 1: 0,5:0,5. Pengovenan selama 2 jam pada suhu 55ºC, penambahan dektrin + NaCl (masing-masing sebanyak 0,5 gr/100 gr tempe), pengovenan kembali selama 2 jam pada suhu 55ºC. Perlakuan B2 (pengovenan pada suhu 55ºC selama 2 jam) menghasilkan asam glutamat. Produk ini berpotensi membentuk monosodium glutamat dengan penambahan NaCl. Jumlah protein terlarut dipengaruhi oleh lama pengovenan dan suhu. Produk hidrolisat tempe sebagai penyedap rasa menghasilkan rerata kesukaan warna (3,3 = menarik), aroma (3,9 = sangat suka) dan menghasilkan cita rasa sama jika ditambahakan sebanyak 2 kali dibandingkan penyedap rasa sintetis.This research was aimed to describe the need for the development of alternative flavor made from hydrolyzated tempe and its manufacturing process, to test the types of amino acid, to measure the protein levels and to test the favor organoleptically. This was an experimental study. The types of amino acid was tested using chromatographic technique, the level of soluble protein was tested using Biuret method and the products were compared organoleptically. The study showed that the development of hydrolyzated tempe flavoring needs to be performed by the reason of the impact of consuming synthetic flavors, the technology development and the research potential. The manufacturing process was done by adding pineapple juice in steamed and blended tempe (the ratio of tempe:water:pineapple juice was 1:0.5:0.5), and then baked for 2 hours at the temperature of 55ºC, added with NaCl + dextrine (each as much as 0.5 g/100 g tempe), baked again for another 2 hours at 55ºC. The treatment B2 (baked at temperature of 55ºC for 2 hours) produced glutamic acid. This product has potentially forms monosodium glutamate with the addition of NaCl. The total soluble protein was affected by the length of the baking time and the temperature. The products i.e. the hydrolyzated tempe as the flavor enhancer yielded an average color preference of 3.3 (i.e. interesting), aroma of 3.9 (i.e. like much), and produced the same taste when using as much as twice volume of the product compared to the synthetic flavors.

Abstract

Justicia gendarussa Burm.f., a medicinal plant, is Acanthaceae that has many functions. Furthermore, the compounds in gendarussa must be produced in high quantity and quality by applying callus culture method. Accordingly, it is important to study the effects of plant growth regulators (2,4-D, IBA, and BAP) on callus induction of gendarussa leaves. This research design utilized a factorial design with two factors (2,4-D and IBA: 0.5, 1, 1.5 mg/L and BAP: 0.5, 1, 1.5, 2 mg/L). The experiment consisted of 24 treatments, each of which was repeated 3 times. Observation was carried out in 6 weeks. Data on the time of callus formation, percentage of explants formed callus, and callus morphology were analyzed descriptively,while data on fresh and dry weight were analyzed by Two-Way ANOVA (α = 0.5). Interestingly, the results showed that various concentration of plant growth regulators (2,4-D, IBA, and BAP) affected callus induction from leaf explants of gendarussa. We concluded that the most optimal treatment combination of concentration of plant growth regulators in inducing callus from leaf explants of gendarussa is 1.5 mg/L 2,4-D and 2 mg/L BAP with a relatively long period of callus formation at the earliest, i.e. on day 5, 2.247 g of fresh weight, 0.108 gof dry weight, white callus translucent, and friable. Moreover, the optimum treatment will be used to produce secondary metabolite and seed synthetic by cell suspension culture.

Abstract

Indonesia had a high number of malaria cases where the treatments are only perceived by 10% of patients in health facilities. The resistance of Plasmodium sp. to drugs causes the difficulty of controlling malaria. The use of irradiation to produce malaria vaccine material is being explored until nowadays. In previously study that dose rate of gamma irradiation delivered to parasite was at the low dose rate (380 Gy/hour) which produced several doses that not be able to attenuate the parasite. This research aimed to increase dose rate (740 Gy/hour) and to determine the dose irradiation effect of 150-250 Gy to the protein profile, erythrocyte and leukocyte numbers in mouse blood infected by P. berghei erythrocytic stage. Results showed that there was an influence of dose rate of 740 Gy/hour for doses of  150-250 Gy in treated mice and no difference in erythrocyte, leucocyte and body weight with negative control. Dose of 150 Gy was the optimal dose to attenuate parasites, whereas doses more than 150 Gy effectively killed the parasites. The changes of protein profile marked by the appearance of higher number of bands at molecular weight of 30-37 kDa that predicted as merozoite surface protein of  P. berghei and/or antibodies elicited in mouse.How to CiteRosadi, I., Syaifudin, M., Elfidasari, D. Abstract

Antioxidants play a crucial role in human health owing to their ability to neutralize oxidative reactions. Kitolod (Isotomalongiflora) is one of the medicinal plants that can be used as a source of antioxidant. The aim of study wasto compare the content of antioxidant compounds (flavonoids, phenolics, chlorophylls, carotenoids, and ascorbic acid) in various organs of the kitolod plant. The plant organs studied includingroots, stems, leaves, flowers, and fruits. The extraction of each plant organ wasconducted by maceration technique using ethanol as a solvent at the room temperature for 48 hours. The analysis of total flavonoids, phenolics, and ascorbic acid was performed by colorimetry method using AlCl3, Folin, and sulfosalicyclic acid reagent respectively, while chlorophylls and carotenoids was determined by direct colorimetry method with methanol as a solvent. The antioxidant activity of various plant organs of kitolod was analyzed using the DPPH method. The highest total flavonoid, phenolic, chlorophyll, and carotenoid compounds werefound in leaves, respectively by 10.48, 1.46, 7.25, and 56.98 ppm. The highest ascorbic acid content and antioxidant activity were obtained fromfruits.The research findings provide new and important information aboutthe contents and antioxidant activity of the secondary metabolites (flavonoid, phenolic, chlorophyll, carotenoid, and ascorbic acid) in each organ of kitolod plant. The information from the results of this study can be used to increase the medicinal value ofkitolod plants.

Abstract

Dayak Tomun is a traditional society living in the Lamandau Regency of Central Borneo. The Dayak society Tomun has local wisdom in the treatment and care of pregnant women and the brithing process utilizies medicinal plants. The traditional medicines were passed down from the ancestors of the Dayak. The purpose of the research was to discover, assess, and inventory medicinal plant used in the treatment and care of pregnant and postpartum women by the Dayak Tomun. The method used was a semi-structured interview and direct participatory observation of six key informants professional activities in the treatment of Dayak Tomun in the Lopus Villages. As many as 17 species of medicinal plants with 14 families. Found of these 23.53% were of member of the family Zingiberaceae. Traditional herbal treatment of pregnant and postpartum by Dayak Tomun societies many are formulated by using part of the leaves (52%), boiled (37.50%) and consumed with a drink (28%). Treatment and care of pregnant and postpartum women in Dayak Tomun should be applied and preserved so that local knowledge of Dayak Tomun society in the Lopus Villages is not lost to the next generation. The research provided information to the society about the utilization of plants used in treatment for women.

Abstract

Preservation of ovarian tissue from severely injured or dead valuable animals has the potential to preserve female germ cells of animals. The ability to mature and fertilize of oocytes from preserved ovary of endangered species will allow us to sustain genetic and global biodiversities. The aims of this study were to investigate the viability of oocytes collected from the preserved ovary and its potential utilization for the production of cat embryos followed by in vitro maturation and fertilization. Ovary was preserved immediately in phosphate buffer saline (PBS) at 4 °C for 24 or 48 hours. The quality and viability of oocytes after the maturation process were identified microscopically using aceto-orcein staining. Biological function of the oocytes was evaluated by using in vitro culture technique for the maturation and fertilization rate in CR1aa medium culture. The results showed that the percentage of oocytes collected from preserved ovary for 24 and 48 hours that remained at the stage of metaphase-II were 29.4% and 21.9% respectively. Fertilization rates produced in the IVF using oocytes collected from ovary preserved for 24 or 48 hours were significantly lower (30%) than that of unpreserved control (36.7%). In conclusion, female germ cells of cat ovary preserved at 4 °C in PBS for 2 days were still viable for in vitro fertilization and thus can be utilized for in vitro production of cat embryos. Information obtained can be used as a basis of knowledge of using a combination of physiological reagent and cold-based preservation technique in modern reproductive technology for animals.
