Abstract

Justicia gendarussa Burm.f., a medicinal plant, is Acanthaceae that has many functions. Furthermore, the compounds in gendarussa must be produced in high quantity and quality by applying callus culture method. Accordingly, it is important to study the effects of plant growth regulators (2,4-D, IBA, and BAP) on callus induction of gendarussa leaves. This research design utilized a factorial design with two factors (2,4-D and IBA: 0.5, 1, 1.5 mg/L and BAP: 0.5, 1, 1.5, 2 mg/L). The experiment consisted of 24 treatments, each of which was repeated 3 times. Observation was carried out in 6 weeks. Data on the time of callus formation, percentage of explants formed callus, and callus morphology were analyzed descriptively,while data on fresh and dry weight were analyzed by Two-Way ANOVA (? = 0.5). Interestingly, the results showed that various concentration of plant growth regulators (2,4-D, IBA, and BAP) affected callus induction from leaf explants of gendarussa. We concluded that the most optimal treatment combination of concentration of plant growth regulators in inducing callus from leaf explants of gendarussa is 1.5 mg/L 2,4-D and 2 mg/L BAP with a relatively long period of callus formation at the earliest, i.e. on day 5, 2.247 g of fresh weight, 0.108 gof dry weight, white callus translucent, and friable. Moreover, the optimum treatment will be used to produce secondary metabolite and seed synthetic by cell suspension culture.

Abstract

Gandarusa (Justicia gendarussa Burm.f.) is an Indonesian medicinal plant that has many benefits as drug and male contracetive. For industrial needs, Gandarusa must be available in large quantity. Hairy root culture is one of methode to produce phytochemistry compound. The objective of the study was to examine the effect of various density of Agrobacterium rhizogenes strain LB510 on hairy roots induction of gandarusa (Justicia gendarussa Burm.f.) leaf plant. Leaf explants were inoculated in MS liquid medium with various density of OD600 = 0.1; 0.2; 0.3; 0.4; and 0.5. Explants were co-cultivated for 2 days on MS solid medium without any hormone then sub-cultured on MS solid medium containing antibiotic cefotaxim 300 ppm, in dark condition. The data were analyzed descriptively and statistically. The results showed that various density of Agrobacterium rhizogenes strain LB510 was affected the lenght of hairy roots induction of J. gendarussa Burm.f., but these was not effected toward lenght formation time and number of hairy root. The treatment of OD600 0.2 was the best treatment for hairy root induction on Justicia gendarussa Burm. f. This data could be used for optimized the quality of methode of hairy root induction.

Abstract

Abstrak Tujuan dari penelitian ini adalah menentukan pengaruh kombinasi NAA dan 2,4-D dengan BAP terhadap perkembangan kultur daun dan konsentrasi yang sesuai untuk induksi kalus tiga kultivar Aglaonema sp. Kultivar-kultivar yang digunakan adalah Dynamic Ruby, Snow White, dan Siam Aurora. Eksplan daun dikulturkan pada medium MS padat dengan perlakuan kombinasi NAA dan 2,4-D dengan BAP. Hasil pengamatan minggu kedelapan menunjukkan ada pengaruh perlakuan zat pengatur tumbuh terhadap perubahan bentuk eksplan daun. Eksplan daun melengkung, bergelombang, membengkak, warna eksplan memucat dan membentuk kalus, tetapi hanya Aglaonema sp. cv. Dynamic Ruby yang mampu membentuk kalus. Kombinasi zat pengatur tumbuh 0,1 ppm 2,4-D and 1 ppm BAP adalah kombinasi yang sesuai untuk induksi kalus untuk Aglaonema sp. cv. Dynamic Ruby. Kata Kunci: Aglaonema, NAA, 2,4-D, BAP, kalus   Abstract   The objective of this study were to determine the effect of combination of NAA and 2,4-D with BAP toward leaf culture development and to know the best concentration of growth regulator substance to induce callus on leaf explant of three Aglaonema cultivars. The cultivars were Dynamic Ruby, Snow White, and Siam Aurora. Leaf explants were cultured on solid MS medium with addition of various concentration combination of NAA and 2,4-D with BAP. Result of eight week observation gave significantly effect to changes shape leaf. Leaf blade was curved or rolled up, swelled, leaf color became pale strands, and formed callus, but the only cultivar which formed callus was Aglaonema sp. cv. Dynamic Ruby. The conclusion of this study was combination of growth regulator substance 0,1 ppm 2,4-D and 1 ppm BAP are the appropriate combination on callus inducing for Aglaonema sp. cv. Dynamic Ruby. Abstract

Abstrak Penelitian ini bertujuan untuk menginduksi akar rambut eksplan daun Gandarusa (Justicia gendarussa Burm.f.) dengan perlakuan perbedaan lama waktu infeksi Agrobacterium rhizogenes strain YM072001 dan A4T dan mengevaluasi lama waktu infeksi terbaik untuk induksi akar rambut. Eksplan daun diinokulasi di media MS (Murashige dan Skoog) cair yang berisi bakteri Agrobacterium rhizogenes dengan OD600 = 0,1 selama 10, 20, 30, 40, 50, dan 60 menit. Kokultivasi di MS padat selama 2 hari, lalu disubkultur ke media MS padat. Data dianalisis secara deskriptif. Akar rambut mulai tumbuh pada minggu ketiga. Induksi akar rambut berhasil pada eksplan dengan perlakuan strain YMB072001 dengan lama waktu infeksi 20, 40, dan 50 menit. Strain A4T juga berhasil menginduksi eksplan membentuk akar rambut dengan perlakuan 10 menit. Lama waktu infeksi terbaik untuk strain YMB072001 adalah 20 menit dan untuk strain A4T adalah 10 menit. Kata kunci: Agrobacterium rhizogenes, akar rambut, Justicia gendarussa Burm. f., waktu infeksi   Abstract The objective of this study were to induce hairy root of Gandarusa leaf explants by differences of Agrobacterium rhizogenes infection time treatment and to evaluate the best infection time for induction. Leaf explants were inoculated on MS (Murashige dan Skoog) liquid medium with bacterial concentrations of OD600 = 0,1 for 10, 20, 30, 40, 50, and 60 minutes and 2 days co-cultivated on MS0 solid medium then sub cultured on MS0 solid medium. Data were analyzed descriptively. Hairy roots were growing on the third week. Explants were successfully induced by strain YMB 072001 at 20, 40, and 50 minutes treatment. A4T strain was successfully induced by 10 minute of treatment. The best infection time for hairy root induction in gandarusa leaf explants for YMB 072001 strains was 20 minutes and for A4T strain was 10 minutes. Abstract

The aim of this research was to know the morphological variety of flower in Chrysanthemum morifolium Ramat through visual observation which use digital camera and then was analyzed descriptively. Varieties of Chrysanthemum morifolium Ramat in this research were C. morifolium var reagen pink, C. morifolium var puma purple, C. morifolium var evergreen, C. morifolium var boris becker, C. morifolium var stroika, C. morifolium var tiger, C. morifolium var remix purple, C. morifolium var jaguar red, C. morifolium var rhino white, C. morifolium var pasopati, C. morifolium var towntalk. From those 11 varieties showed the morphological differences of the flowers, which are the color of ray floret, the quantity of ray floret, the quantity of ray floret layers, the shape of ray floret, the tip of ray floret, the quantity of disk floret, the position of disk floret, the diameter of flower, the length of ray floret, the width of ray floret, the diameter of involucrum and the disk shape. However, this research also look the similarities between the varieties, which are the edge of ray floret (flat), the color of involucrum is green, the adhering of inter-involucrum (star like), the tip of involucrum (dull), transparent coating involucrum, the quantity of involucrum layers (2 layers), the color of the corolla tubes(yellow), the branched and yellow pistil, and yellow anther. From the results it can be concluded that Chrysanthemum morifolium Ramat has multiple the morphological differences and similarities of the flower morphology between the varieties.

Abstract

The aim of this research was to know the effect of 3-indole-acetic acid (IAA) and 6-bensilaminopurine (BAP) and optimum concentration of IAA+BAP toward growth and development of the leaf culture of Aglaonema sp. var. Siam Pearl, Aglaonema sp. var. Lady Valentine, and Aglaonema sp. var. Lipstik. The explants were taken from the leaf. The explants were cultured in solid MS medium with addition various concentration of IAA 0,5 mg/l, 2 mg/l, 3 mg/l, 5 mg/l, and 8 mg/l and BAP 5 mg/l. Observation had been conducted for 16 weeks to know the response the development of explants leaves such as the periphery of explants coloured of brown, leaves explants of arch shaped, the colour of explants to change for pale, and to form callus. The data were include of the number of explants which able to form callus, time of callus formation, the colour and texture callus, and stage of development explants were analyzed descriptively. The result of 16 weeks observation showed that the addition of IAA and BAP on the leaves explants to give the different effects. Aglaonema sp. var. Lipstik can form callus by treatment of 5 mg/l IAA:5mg/l BAP, and 8 mg/l IAA:5 mg BAP. The colour of callus are clear, the shape of globular, with friable textures.

Abstract

Pandemi Covid-19 menuntut masyarakat di dunia untuk hidup sehat, hingga saat ini belum ada obat atau vaksin yang efektif untuk menghentikan laju penyebaran Covid-19. Indonesia memiliki kearifan lokal untuk memanfaatkan tanaman sebagai alternatif pengobatan. Penggunaan tanaman sebagai pengobatan tradisional menjadi salah satu solusi hidup sehat di masa pandemi Covid-19. Tanaman memiliki banyak khasiat yang bisa dimanfaatkan untuk meningkatkan sistem imun tubuh dan mencegah penyebaran Covid-19. Namun informasi tersebut masih banyak yang belum terdokumentasi dan menyebar kepada seluruh masyarakat dalam berbagai macam kalangan usia. Penelitian studi etnomedisin telah dilakukan untuk membantu mendokumentasikan dan memberikan informasi terkait manfaat tanaman yang ada di sekitar kita khususnya untuk pencegahan Covid-19. Metode yang digunakan adalah snowball sampling, hasil yang didapatkan adalah terdapat >100 jenis tanaman yang dimanfaatkan oleh masyarakat untuk pengobatan tradisional. Tanaman seperti jahe, kunyit, dan tempuyung memiliki potensi untuk mencegah penyebaran coronavirus. Hasil penelitian yang diperoleh disosialisasikan kepada masyarakat luas secara nasional melalui sarana zoom meeting.

Abstract

Direct extraction and conversion of microalgal lipid to biodiesel in one step process is developed to initiate an efficiently method in the biofuel production. The method that called as in-situ process had been applied in the biodiesel production from microalgae Chlorella vulgaris lipid by involving of sonication assisting. Various parameters that affect the in-situ transesterification were investigated to find the optimum conditions including power of sonication, co-solvent use, and biomass amount. The experiment was performed by acidic catalyst The use of sonication in 25kHz/270W yielded the highest biodiesel product. The apply of co-solvent n-hexane significantly increased the yield biodiesel rather than no co-solvent. The biodiesel yield of 10.39 % obtained when the process was accompanied by the n-hexane.as co-solvent with increasing level as 47% rather than without co-solvent. The in situ process in the ratio biomass - methanol 3:50 that assisted by sonication for 60 mins produced the highest biodiesel, which was 20,31 % w/w. The yield was higher than assisting with reflux or combination of reflux-sonication. The component of biodiesel yielded in the process consisting of 7,10,13-hexadecatrienoic methyl ester; pentadecanoic acid, 14-methyl-methyl ester; 9,12,15-octadecatrienoic acid-methyl ester; and eicosanoic acid-methyl ester. Joining of reflux-sonication exhibited a low biodiesel product. Disadvantage method from the combination may inhibit on lipid extraction or FAME transformation, the exact cause needs to be looked for in the future.
