Abstract

The exploration of several active compounds of medicinal plants Aegle marmelos Correa (Rutaceae) have found marmin that strongly inhibit the histamine release from mast cells. The present study investigated the antagonism phenomena of marmin on 5-hydroxy triptamine and histamine receptors in guinea-pig ileum smooth muscle. The study was performed using isolated-organ technique in Tyrode buffer medium. The agonists used to sensitize the receptors were histamine and 5-hydroxy triptamine. The type of antagonism was identified by considering: 1). the decrease of pD2 or Emax values of agonists due to pretreatment of marmin, and 2) Schild regression method. In the study, marmin (10 and 100 mM) decreased the maximum contraction effect of 5-HT by 40% and 60%, respectively. However, marmin did not influence the pD2 value of 5-HT . On the other side, marmin did not influence the maximum contraction effect of histamine, however, decreased the pD2 value of histamine. Based on Schild regression method, the pA2 value of marmin was 0.8016. In conclusion, marmin non-competitively antagonized 5-HT 4 receptor, and competitively antagonized histamine receptor in ileum smooth muscle.

Abstract

The aims of this research are to determine the effects of tartaric acid, sodium bicarbonate and their interaction on the physical properties of effervescent granule made by dry granulation and also to determine the optimum composition area of tartaric acid and sodium bicarbonate that produce qualified effervescent granule. The experimental study employed factorial design with two factors, i.e. tartaric acid and sodium bicarbonate, and two levels, i.e. high level and low level. The formulas were optimized on their physical properties, i.e. the granules flow rate, moisture content, disintegration time and pH of the solution. The data were statistically analyzed using Yate’s treatment with 95% level of confidence. The results show that tartaric acid was dominant in determining the disintegration time of the effervescent granules, while sodium bicarbonate was dominant in determining the moisture content of the effervescent granules. Interaction between tartaric acid and sodium bicarbonate significantly determined the flow rate of the effervescent granules and the pH of the solution. The optimum composition area of tartaric acid and sodium bicarbonate was not produced because there was one physical property of the effervescent granule that was not fulfilled, i.e. the granules moisture content.

Abstract

Many lactic acid bacteria (LAB) are known to produce antimicrobial peptides, termed bacteriocins. Bacteriocins are ribosomally synthesized antimicrobial peptides which kill closely related bacteria. Bacteria producing bacteriocin are also known to produce bacteriocin immunity protein (bip). This study aimed to identify bacteriocins activity in an attempt to isolate the bacteriocin and the bip. In this study, genus Streptococcus and Weissella from our LAB collection were screened on well diffusion agar for antagonism assay. Growth inhibition zone observation has been carried out by antagonism assay using Leu. mesenteroides TISTR 120, Leu. mesenteroides JCM 6124, Staphylococcus aureus FNCC 0047, Bacillus subtilis FNCC 0061, Escherichia coli FNCC 0183, Salmonella typhi FNCC 0165, Pseudomonas aeruginosa FNCC 0063, and Listeria monocytogenes FNCC 0156 as indicator strains. Protease K, catalase, and trypsin were also used following the first screening assay for confirmation test. Results showed that both Strep. macedonicus MBF10-2 and W. confusa MBF8-1 posses bacteriocin activity although both against Leu. mesenteroides only, the TISTR 120 and JCM 6124 indicator strains

Abstract

Green betle leaf (Piper betle L.) and red betle leaf (Piper cf. fragile Benth.) were empirically used for treatment such as anti-microbial, anti-diabetic, antiseptic and resolve nosebleeds and have antioxidant activity. Phytochemical screening and antioxidant activity test have been carried out on (water and ethanol 70%) extracts of both green and red betle leaf. Phytochemical screening showed that both water and ethanol 70% extracts of both green and red betle leaf contain secondary metabolite compounds, flavonoid, alkaloid, tannin, steroid and triterpenoid, the presence of which may produce the antioxidant activity. The DPPH free radical inhibition method showed that antioxidant activity of water extract of both green betle and red betle (IC50) were 36.02 μg/mL and 60.35 μg/mL, whereas the antioxidant activity of 70% ethanol extract (IC50) were 10.59 μg/mL and 28.05 μg/mL, respectively. The highest antioxidant activity was in 70% ethanol extract of green betle (IC50 = 10.59 μg/mL), but was lower than the vitamin C (IC50 = 7.39 μg/mL) or quercetin (IC50 = 2.89 μg/mL) as positive controls. The statistical tests using twowaysanalysis of variance followed by Tukey test showed that there are significant differences between the green and red betle’s, and between water and ethanol 70% extratcs as indicated by the P value < α (α = 0.05).

Abstract

This research investigated the antioxidant activity of prenylated flavonoid compound from the ichloromethane extract of Artocarpus communis leaves on 1,1-diphenyl-2-picryl hydrazyl free radicals scavenging assay. The dichoromethane extract of Artocarpus communis leaves was fractionated by column chromatography techniques on silica gel, and each fraction was assayed for its activity. The active compound, 1-(2,4-dihydroxyphenyl)-3-[8-hydroxy-2-methyl-2-(4-methyl-3-pentenyl)-2H-1-benzopyran-5-yl] 1-propanone, showed moderate antioxidant activity with IC50 54.6 μg mL-1.

Abstract

This research had been conducted to verify the activity of ethanolic extract of Pegagan or Centella asiatica Urban (CA) from Indonesia as immune enhancing with the activity and capacity of peritoneal macrophages as parameters tested. Sprague Dawley male rats were divided into 4 groups of 5 rats each. Dose 1, dose 2 and dose 3 groups were treated with extract at the dose of 10, 20 and 40 mg/200 g b.w. p.o. for 15 consecutive days, respectively, and the control group was supplemented with carrier. One day after the last treatment, all rats were sacrificed after induced by Staphylococcus epidermidis (109 CFU per rat) i.p. and then the activity and capacity of peritoneal macrophages were examined with giemsa staining microscopically. The results showed that the immune-enhancing effect of the extract was a dose dependent manner. Based on statistical analysis (ANOVA, p < 0.05), the administration of CA extract at the dose of 40 mg/200 g bw demonstrated the highest result and differed from control group significantly (96% of macrophage activity and 61 bacteria of macrophage capacity). From these experiments could be concluded that the ethanolic extract of CA had potential to be developed as immune-enhancing agent. However, it was still needed to conduct further study for elaborating the mechanism of immune enhancer completely.

Abstract

The barks of Pentaspadon motleyi Hook. f. (Anacardiaceae) were collected from the Poso district in Central Sulawesi, Indonesia, in 1992 which it has been used traditionally as a remedy for malaria and tuberculosis in the local area. It was found that a mixture of (+)-sesamin (1) and (-)-sesamin (2) in a ratio of 100:86 were present together with lupeol in the bark of Pentaspadon motleyi. This was the first evidence of sesamin being isolated in almost equivalent amounts of (+)- and (-)-forms from a single plant species, and indicates that the biosynthetic pathway of sesamin in the plant is not enantioselective or fuzzy.

Abstract

Preliminary studies have been carried out phytochemical test acetone leaf extract Garcinialateriflora Blume., and toxicity test against shrimp larvae (Artemia salina Leach) and murine cell lineP388. Isolates which showed the highest mortality are proceeded further for fractionation, isolation andpurification. The structure identification of active campound carriedout by analyzing the spectrum of IR ,GC-MS (Gas chromatography mass spectrometer) and proton nuclear magnetic resonance spectrometer(1H-NMR ) and (13C-NMR ). The results showed that the most active against the larval shrimp Artemiasalina L. with LC50 value of 18.03 ug/mL is b-asaron, IC50 of test results on cell line murine P-388 is 47.0ug/mL. This compound was isolated first time from the plant Garcinia lateriflora BL.

Abstract

Diuretics use as first treatment in hypertension and heart failure. Empirically, community use belimbi as diuretic. Preliminary study to explore the potency of bilimbi ethanolic extract as a natural diuretic was conducted using ninety male mice which divided into 6 groups. Each group consist of three sub-group of 5 mice respectively for three replications. Group 1 was given orally saline solution of tween 80, Group 2 was given orally urea solution 500 mg/kg BW, Group 3 was given orally furosemide 3 mg/kg BW, Group 4 was given orally extract of bilimbi 2.5 g/kg BW, Group 5 was given orally extract of bilimbi 5 g/kg BW, and Group 6 was given orally aquadest. The diuretic activity was determined by measuring cumulative urine volume every 1 hour during 5 hours post administration. The result showed that 2.5 g/kg BW had a strong diuretics activity 2 hours post administration and reached its maximum activity 3 hours post administration. At the peak of diuretic activity, extract of bilimbi 2.5 g/kg BW increased diuretic effect by 219% as compared to the aquadest and 41,5% as compared to the furosemide. It is concluded that extract of bilimbi has a potency as a natural diuretic.

Abstract

The juice of Ananas comosus L. contains bromelain which has been known to defoliate dead skin cells. Investigation on five gel formulations containing 17%, 33%, 50%, 67% and 78% of the juice of Ananas comosus L with 4% of HPMC as gelling agent, have been carried out. The stability of all formula were examined by the accelerated test method at room temperature and 40 oC for 6 weeks. Parameters observed were organoleptic, homogeneity, syneresis, viscosity, flow behavior, spreading ability, and pH. Results indicated that the higher the juice concentration in the gel would reduce the gel stability progressively. Gel formula using 4% of HPMC containing 17% or 33% of Ananas comosus juice can be produced while maintaining good physical stability for 6 weeks. The products were pale yellow to yellow in color, homogeneous, and have viscosity of 13000-45800 cps, pH 4.0-4.9, pseudo plastic flow, and spreading ability of 2732.58 to 4534.16 mm².
