Abstract

Geobacillus thermoleovorans IT-08 is a Gram positive, thermophilic bacterium that can utilize xylan as a sole source of carbon. This strain was isolated from Gunung Pancar hot spring, Bogor, West Java, Indonesia. A plasmid genomic library in Escherichia coli DH5α was constructed and screened for xylanase activity. One positive clone, namely DH5α (pTP510) has been isolated, sequenced and showed putative exo-xylanase (exo-xyl), β-xylosidase (xyl), and α-L-arabinofuranosidase (abfa) genes (Genebank Accession No.DQ387047, DQ345777 and DQ387046 respectively). Each gene encoded 604, 511 and 502 amino acids, respectively. The BLAST search for protein database revealed that Abfa was high similar with GH51 family Abfa of Geobacillus stearothermophilus T6, but Xyl and Exo-Xyl were slight similar with GH43 family (25-34%) respectively. The deduced protein had a molecular weight of about 70 kDa (Exo-Xyl), and 60 kDa (Xyl and Abfa). These showed good accordance with the calculated molecular weight of each protein (68.64 kDa for Exo-xyl, 57.99 kDa for Xyl and 57.03 kDa for Abfa) from deduced amino acid sequence.

Abstract

Cross-linked chitosan with 78,12% deacetylation degree, synthesed from chitosan bead with 135,982% swelling degree were conducted. Chitosan bead has prepared from chitosan that was dissolved with watery acetic acid and coagulated with sodium hydroxide. Chitosan with 79,6% deacetylation degree was prepared from chitin that was submitted to a deacetylation reaction. Chitin were applied, isolated from giant tiger shrimp that was submitted to a demineral and deproteination reaction. Functional group of the cross-linked chitosan with glutaraldehyde were investigated by the Infra Red spectroscopy and deacetylation degree were investigated by the base line methods. Chitin, chitosan and chitosan bead have the same structure. The difference between chitin and chitosan was the deacetylation degree and chitosan bead in the physical shape. The deacetylation degree and swelling degree of chitosan bead was decreased by cross-linking process.

Abstract

The aim of the study was to describe the changes of sacharide monomer and functional groups of Fe-dietary fiber complex at boiling with acidic medium. The research was designed as the Pre test-Post test Control Group, the treatment of variation pH (pH 3 and 7) and boiling time (raw and 35 minutes). The dependent variables were sacharide monomer of dietary fiber and functional groups of Fe-dietary fiber complex. FeSO4. 7 H2O was added as source of mineral Fe. The results of the study showed: 1) At pH 3-boiling time 35’ treatment, no changes to kinds of dietary fiber sacharide monomer, however changes of monomer content. 2) At pH 3-35’ treatment, the Fe binding both to lignin and cellulose at –OH groups, but it was not able to bind hemicellulose. At raw-pH 7, the Fe binding to lignin at –OH, -COOH groups; to cellulose at –OH, -CH2-O-CH2-, -COO groups; while to hemicellulose at –OH groups.

Abstract

Sucrose as the major transported form of fixed carbon must be translocated from source tissue to the sites of consumption and storage or sink tissues. The translocation of sucrose is facilitated by some distinct sucrose transporters proteins (SUT). To study sucrose transporters in sugarcane, we had conducted isolation and characterization of gene encoding sucrose transporter protein. The isolation was performed with RT-PCR method using total RNA isolated from sugarcane leaf and primer designed from conservative region of SUTcDNAs of SoSUT2A (accession number AY65599), OsSUT-1(accession number AAP54842, OsSUT-1 mRNA (accession number XM 46477). Based on the conservative amino acids sequences of QILQQFA and MGKTEPV, the corresponding sequences of the primers for RT-PCR were: P1, (forward) 5’CAGATCCTTCAACAGTTCGC-3’ and P2 (reverse) 5’-TGCCCTTTGTCTCCGGAACC-3’, respectively. Agarose gel electrophoresis shown a clear single 0.5 kb cDNA band of the PCR product. Thus, the DNA was cloned into pGEMT vector (Promega) for further analysis. Sequence determination of the PCR product revealed a nucleotides sequence of 543 bp in length and has a high homology around 89%, 87.3 % and 84.8 % with maize ZmSUT-1, sugarcane SoSUT 2A and rice OsSUT-1 mRNA, respectively. We designated the cDNA as SoSUT2 and the nucleotide sequence have been submitted to GenBank data base under accession number bankit 734628. By using PSORT analysis the fragment of cDNA-SoSUT2 encoded protein may be located in the endoplasmic reticulum. To have a better understanding, the expression of SoSUT2 gene in sugarcane was determinate by RT-PCR method using total RNA isolated from leaf, petioles, stem and root and visualized the PCR product in agarose gel electrophoresis. Based on the cDNA bands intensity, it can be illustrated that the expression of SoSUT2 gene were found highest in sugarcane leafs then petioles and stem, but the expression was not found in root. Although the SoSUT2-cDNA has not been isolated in full size, the results suggest the presence of gene family of SUT in sugarcane.

Abstract

Shorea accuminatissima, which is known locally as Meranti kuning, is one species of Dipterocarpaceae family. This plant contains compounds that are expected to have antioxidant activity. This research was carried out to examine the radical scavenger activity of polar fraction of methanolic extract from the tree bark of S. accuminatissima by using DPPH (2,2-difenil- 1-pikrilhidrazil) method with vitamin C as comparator. Powder of tree bark of S. accuminatissima was macerated in methanol. The methanolic extract was then evaporated to obtained dry extract. Extract with concentration of 200, 100, 50, 25 and 10 µg/mL were used to test its radical scavenger activity. The result showed that polar fraction of methanolic extract and vitamin has IC value of 319.83 and 3.72 µg/mL, respectively.

Abstract

Recent   observations   suggest   that   lectin-like   toxins,   such   as   endotoxins   from  Bacillus   thuringiensis  are sequestered inside the gut of immune-induced insects providing tolerance to Bt-formulations. To investigate the induction   and   sequestration   process,   we   used   toxin   and   lectins   that   bind   to   galactosamine   (Gal)   and   N-acetylagalactosamine (GalNAc) to identify binding sites in the midgut of induced and non-induced insects using confocal   microscopy.   Our   observations   suggest   that   immune   and   metabolic   hemolymph   components   are transported across the gut epithelium and accumulate inside the gut lumen in Bt-tolerant insects. Co-location of immune components and toxin indicates that some of the toxin is inactivated by coagulation reactions inside the gut lumen before it can reach the brush border membrane

Abstract

In this research Crude Palm Oil (CPO) and methanol were reacted by applying 5 % basic catalyst (KOH) and adding diethyl ether as co-solvent to produce methyl ester (biodiesel). Co-solvent was added in order to form one-phase reaction mixture, and then higher reaction rate was expected compare with two-phase system. Reaction was carried out batch wise in 1 litre glass reactor stirred continuously at 300 rpm. The objective of this research is to obtain the characteristic of biodiesel product. The advantage using this methode was showed by several characteristics of the biodiesel product. The density and viscosity had achieved the commercial biodiesel standard, this biodiesel product has higher flash point of the product compared with solar, which means lower risk factor during storage. The very low sulfur content and the pour point of product that was layed below the maximum allowable limit, are environmentally favorable.

Abstract

There are two different outliers, i.e outlier in observations and outlier in models. The existing outlier detection method in models is using common Likelihood method. The limitation of this method is the optimal value produced might be not the real optimal values. This research yields a method for outlier detection in multivariate linear models with Likelihood Displacement Statistic-Lagrange method (LDL method). This method uses multiplier Lagrange with constraint the confidence interval of parameter’s vector. This parameter’s vector is obtained from the data set which is outlier free. This parameter estimation process uses numerical method with Karush-Kuhn Tucker condition in nonlinear programming. This method compares between LDL value and the table F value that follows the distribution of F value to indentify the outlier in models.

Abstract

Population genetics is a branch of biology which studies about the gene composition from population and the change of the gene composition is effect from some factors. One of them is lethal gene factor. The change of gene composition will influence the genotype probabilities in the population. In this paper discussed about determining the genotype for the probability of the n-th offspring genotypes in dihybrid mating by observing linkage between the two loci. The mating occurred randomly and without concern ethics in mating. This research was done by making mathematics model to determine allele pair, using difference equation, then from this model will be determined genotypes probability. The result show that the mating happened normally had the same genotype probability of each generation, meanwhile in abnormal mating, the genotype probability whose had lethal gene would decrease and the genotype probability whose did not have lethal gene would increase in each generation.Keywords : Difference equation, dihybrid mating, lethal gene, population genetics, probability

Abstract

This study purposed to determine the effect of MSG on the character of cellulose acetate membranes. This membrane preparation using phase inversion technique. The results showed that the higher the concentration of MSG, which is the character of membrane permeability coefficients and higher water flux, whereas for the character of the membrane density test and 100-200 kDa dextran rejection further down. To test the FTIR proved that there are no additives or solvents are trapped in a membrane. Keywords : Cellulose Acetate, phase inversion, MSG, FTIR
