Abstract

Six resveratrol oligomers, namely (-)- ε-viniferin (1), (-)-laevifonol (2), (-)-vaticanol B (3), (-)-hemsleyanol D (4), (+)-isohopeaphenol (5), and (-)-hopeaphenol (6) have been isolated from the acetone extract of the stem bark of Shorea brunnescens. The structure determination of these compounds were based on spectroscopic data (UV, IR, 1H NMR), and compared with literature data. The cytotoxicity of isolated compounds against murine leukemia P-388 cells showed the IC50 values as 18.1, 23.2, 41.9, 12.3, 39.7, and 5.5 µM, respectively.

Pengelolaan Pembelajaran Lesson Study Mata Pelajaran Ipa Di SMA Negeri 1 Nogosari Kabupaten Boyolali Haryoto1), Marlupi Dwi Lestari2), Nugroho Adi Suseno3) 1)Staf Pengajar Progdi Magister Manajemen Pendidikan Program Pascasarjana Universitas Muhammadiyah Surakarta2)Guru Sekolah Menengah Atas Negeri 1 Nogosari, Kabupaten Boyolali3)Guru Sekolah Menengah Pertama Negeri 1, Kaliwungu, Kabupaten Semarang Abstract Quality of education in Nogosari High School still lower because there is use to function educational production approach which too centripetal at education input, less is paying attention of learning process, to increase quality of education conducted by real stages there are learning process of pass with lesson study approach. Learning process of lesson study is conducted represent learning process collaboration by teachers through three activities there are planning, implementation, and reflection. Planning is started identified the problem, analysis of problem, determination of problem priority to be solved, election the lesson model, making lesson plan, cozening method, physic appliance and assessment conducted together by collaboration. The implementation of lesson is conducted together, someone become teacher model and other is observation of learning process; observer may not bother of learning process, observer duty note important problem in lesson. The reflection is conducted after ending lesson, its analyzing learning process, teachers’ model submit what experienced by learning moment, while observer submit result of observation, then analyzed and looked for the way out of the problem of faced at the moment of lesson  implementation. Learning Process of lesson study in Nogosari High School implemented by collaboration between ‘science teacher’ and ‘study teacher’ which start planning until implementation, as for reflection is implemented every saturday of meeting learn or Team Working of Teacher. Result of learning for lesson study in Nogosari High School can improve the quality of education.  Keywords: Lesson Study

Abstract

Kulit kacang tanah mengandung luteolin yang berpotensi sebagai antiinflamasi secara in vitro dan in vivo. Penelitian ini bertujuan untuk mengetahui efek antiinflamasi ekstrak etanol kulit kacang tanah (Arachis hypogaea L.) pada tikus jantan galur Wistar yang diinduksi karagenin Penelitian menggunakan rancangan acak lengkap pola searah. Dua puluh lima ekor tikus putih jantan galur Wistar dibagi menjadi 5 kelompok. Kontrol negatif menggunakan CMC-Na 0,5% 2,5 mL/200 gBB, kontrol positif menggunakan Na-diklofenak 6,75 mg/kgBB. Ekstrak etanol kulit kacang tanah yang diujikan yaitu dosis 50, 100 dan 200 mg/kgBB. Larutan uji diberikan secara peroral dengan volume pemberian 2,5 mL/200 gBB 1 jam sebelum kaki hewan uji diradangkan dengan karagenin 1% secara subplantar. Pengukuran volume kaki tikus dilakukan tiap 0,5 jam selama 6 jam. Data volume udem dihitung nilai AUC (Area Under the Curve) rata-rata volume udem terhadap waktu dan persen daya antiinflamasi. Data dianalisis dengan anava satu jalan dan dilanjutkan uji LSD (Least Significant Difference) dengan taraf kepercayaan 95%.  Hasil penelitian menunjukkan bahwa ekstrak etanol etanol kulit kacang tanah (Arachis hypogae L.) dengan dosis 50, 100 dan 200 mg/kgBB mempunyai persen daya antiinflamasi berturut-turut sebesar 16,33%; 26,39% dan 31,70%, sedangkan Na-diklofenak 6,75 mg/kgBB sebesar 32,24%.Kata kunci: kulit kacang tanah (Arachis hypogaea L.), ekstrak etanol, antiinflamasi

Abstract

Artocarpus had been used as traditional medicine for cancer, cirrhosis, malaria, hypertension, and diabetes. This research aimed to evaluate the cytotoxic activity of methanol leaf extract of Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi against MCF-7 breast cancer cell. The leaf extracts of Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi were obtained by the method of maceration with methanol. Phytochemical screening wasperformed by Thin Layer Chromatography with the use of silica GF-254 for stationary phase and ethyl acetate: n-hexane (2:8) for mobile phase. The cytotoxic test was conducted by MTT with diverse concentrations of extract, namely 500, 250, 125, 62.5, and 31.25 μg/mL. The results of cytotoxic test were measured by ELISA reader. Based on phytochemical screening, tannin, and alkaloids were found in three extracts. Cytotoxic test demonstrated that those extracts had low cytotoxic activity against MCF-7 breast cancer cells, with IC50 of 120.85; 339.46 and 530.88 μg/mL, respectively, for Artocarpus altilis, Artocarpus heterophyllus, and Artocarpus camansi.

Abstract

Abstract

Previous research has shown that some compounds in leaves and seeds of sugar apple have a cytotoxic activity. The aim of this research was to determine the cytotoxicity of semipolar fraction of ethanolic extract from sugar apple stem bark (Annona squamosa L.) on T47D cancer cells. The semipolar fraction of ethanolic extract from sugar apple stem bark was collected by fractionation using Vacuum Liquid Chromatography (VLC) with hexane:ethyl acetic (9:1, 8:2, 7:3, and  6:4)  as  mobile  phase.  Cytotoxicity from the fractions of five different concentration namely; 25, 50, 100, 150, and 250, µg/mL was measured by MTT assay. The potency of the cytotoxicity was defined by the ability of the fraction to inhibit the growth of T47D cells indicated by the value of IC50. Qualitative analysis of contained compounds in the fraction was done by Thin Layer Chromatography (TLC) method using silica gel F 254 as a stationary phase and hexane:ethyl acetic (7:3) as a mobile phase. UV 254 and 366 nm lamp also Dragendorff, citroboric, and FeCl3 spray reagents were used to visualize the spots of the secondary metabolites. The result proved that the semipolar fraction of ethanolic extract from sugar apple stem bark showed potential cytotoxicity on T47D cancer cells with IC50value of 70,77 µg/mL. Qualitative analysis showed that the fraction contained flavonoids and alkaloids which is presumably responsible for its cytotoxic activity.

Abstract

The program of community service began with the socialization of applications on the village government and partners. Furthermore, it introduced machinery/technology to create technology on partners. The purpose of the community service activity was to provide the skills in the production of cumi-cumi products from sweet potato to members of Aisyiyah, PKK, and Karang Taruna in Karangbangun Village - Karanganyar District. The community service program began with a socialization program by the village government and partners. Furthermore, machines and technology were introduced to partners. The training of cumi-cumi product making and packaging was given to the partners, as well as production assistance, to ensure that partners were skilled at processing the products independently. The results showed that the partners were skilled at producing cumi-cumi products and using the machines. Two new UMKMs were formed in the community service program. Partners have been packing the products with labeled, marketed the products locally, as well as getting income from these sales.

Abstract

Antibacterial activity in vivo study of ethanol extract nonpolar fraction of inggu stem (Ruta angustifolia [L.] Pers) was conducted on Staphylococcus aureus and Streptococcus mutans. It had a purpose to determine the effect of non polar fraction of ethanol extract of inggu stem on animals that had been infected by bacteria Staphylococcus aureus and Streptococcus mutans. Inggu stem powder was macerated with ethanol 96% and fractionated by vacuum liquid chromatography using hexane : chloroform 6:4, 5:5, 4:6, 3:7. Inggu stem was tested with in vivo antibacterial activity on mice with various doses of 0.3, 1.2 and 2.14 g/kg. Intraperitoneal fluid of mice had been treated was cultured on agar media and colonies of formed bacteria were counted. Antibacterial activity on Staphylococcus aureus bacteria has an average percentage of inhibition for colony are respectively 73, 81 and 97 %., whereas that of the Streptococcus mutans bacteria are 41,81 and 97% respectively. Identification of thin layer chromatography fraction in nonpolar compounds of ethanol extract of inggu stem is indicated by flavonoids, terpenoids, and alkaloids.

Abstract

Mangrove plants as traditional medicine have long been used by the society for the therapy of anticancer diseases. So far, the potency of mangrove plants as anticancer has not been studied intensively. The methanol extract of R. mucronata stem bark has cytotoxic activity on myeloma cells. This study aims to determine the isolates of the ethyl acetate fraction of the methanol extract of the stem bark of R. mucronata and R. apiculata, and to examine the potency of cytotoxic activity against MCF-7 and T47D cells. The powder of the stem bark of R. mucronata and R. apiculata were extracted with methanol by maceration, and then the methanol extract was fractionated consecutively using n-hexane, ethyl acetate, and methanol. Ethyl acetate fraction was then purified by column chromatography with a diameter of 2 cm and a height of 30 cm with a static phase in the form of silica gel 60 (0.063-0.2 mm) Merck 7734 and eluent n-hexane:ethyl acetate (6: 4, 5: 5, 6: 4, 7: 3, 8: 2, 9: 1) and n-hexane: methanol (9,5: 0,5) 10 ml each. The eluates that produce the same number and pattern of stains were combined into one isolate. Then the eluate was evaporated at room temperature. Then the purification results of the extract R. mucronata were isolated using preparative TLC with 0.25 mm GF254 silica gel as static phase and n-hexane mobile phase: ethyl acetate (6: 4), while the apiculata extract with n-hexane mobile phase: ethyl acetate (5: 5). The results of the stain separation of the compound were scraped off and separated from the static phase using a solvent. The purity of isolates compound was examined using TLC in the static phase of silica gel GF254 0.25 mm. The qualitative test results of the ethyl acetate fraction of R. apiculata stem bark with an NMR spectrophotometer showed a composition of cis-3- (3,5-dihydroxyphenyl) acrylic acid, while the ethyl acetate fraction of R. mucronata stem bark contained n-hexan-3-ol compounds. The cytotoxicity test of ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol extract was carried out on MCF-7 and T47D cells using the MTT method. The results proved that the ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol fraction from the ethanol extract of R. mucronata and R. apiculata stem bark, based on cytotoxic activity, were inactive against MCF-7 and T47D cells.
