Abstract

Type I hypersensitivity reactions, also known as allergic reactions or anaphylactic reactions. The mechanism of anaphylaxis involves the release of mediators from mediator cells. The degranulation of mast cells and basophils causes an exaggerated immune response and causes the signs and symptoms of anaphylaxis. Red dragon fruit peel contains secondary metabolites such as triterpenoid flavonoids and steroids. This study was an in vitro study of the ethanol extract of red dragon fruit peel (Hylocereus polyrhizuz) on the mast cells of male white mice induced by antigen. Five variations of the concentration used are 25; 50; 100; 200; 400 µg/mL. The percentage of mast cell degranulation by red dragon fruit peel extract at concentrations of 25; 50; 100; 200 dan 400 µg/mL is 75.27; 69.05; 64.04, 56.61; 45.17; dan 27.88%. Abstract

Cocrystal is one form of modification in increasing the solubility of meloxicam which is included in Biopharmaceutics Classifications System (BCS) II. Cocrystal is a multicomponent system with a stoichiometric ratio between the active ingredient and the coformer which are bound in the crystals lattice to form hydrogen bonds. Design of new drugs can be done through an in silico program to optimize the parent compound before the synthesis of derivative compounds. This study aims to predict which coformers are most stable in the formation of crystals. Cocrystal formation is done by drawing a two-dimensional structure from meloxicam and coformer using ChemBioDraw Professional 16.0 software from CambridgeSoft®. The prediction will result in the amount of bond energy formed between meloxicam with coformer. The smaller the bond energy, the more stable the meaning of the bond. The smaller the bond energy formed, the more stable the bond is. Stable bonds have a high probability of forming cocrystals meloxicam. Hydrogen bonds occur between hydrogen atoms and other atoms that have high electronegativities such as O and N atoms which have lone pairs of electrons. This electronegativity difference makes H atoms tightly bound to O and N atoms so that the hydrogen bonds in the meloxicam cocrystal are tightly bound and stable. From the results of the study showed that the most stable coformer can form cocrystals with a small bond energy that can produce bonds with meloxicam, namely urea.

Abstract

S.aureus infection was found to be around 5%-35% and cases of S.aureus resistance increased by 22.8% due to people being less compliant in treatment. One of the preventions of resistance is to return to the use of natural materials, especially plants that have been scientifically and empirically proven, one of which is Mahkota Dewa Fruit (Phaleria macrocarpa) and Green Meniran Herb (Phyllanthus niruri L.). The study was conducted using the True Experimental method using S.aureus bacteria. 3 treatment groups were used, namely positive control (Amoxicillin), negative control (DMSO), as well as Mahkota Dewa extract (Phaleria macrocarpa) and Green Meniran Extract (Phyllanthus niruri L.). Using the disc diffusion method and the dilution method to determine the magnitude of the inhibition zone, the value of MIC, and MBC. The extract of the Crown of God Fruit, The Green Meniran Herb, as well as the combination of the two extracts have successive bacterial inhibitory power with an average of 18.02 mm; 15.95 mm; and 15.09 mm against S.aureus. Testing by the dilution method obtained a KHM value of 60% (0.06g/mL) in each extract. KBM is not obtained in each extract.

Abstract

The safety of using herbal plant extracts is essential for COVID-19 patients who consume herbal plants daily to increase immunity and overcome the symptoms of the disease. One of the herbal plants used by the community in COVID-19 therapy is Qusthul Hindi (Saussurea costus). The public needs to know the safety of these herbal remedies when consuming them. Therefore it is essential to conduct a toxicity test of Hindi Qusthul extract. Secondary metabolite compounds present in plant extracts can be tested for toxicity in vitro using the Brine shrimp lethality test (BSLT) method using Artemia salina Leach larvae to determine LC50 values. The BSLT method is used to determine biological activities in extract plants, such as cytotoxic activity, phototoxic, enzyme inhibition, and ion regulation, using Artemia salina Leach shrimp larvae as bioindicators. The BSLT method is carried out by observing the mortality rate in Artemia salina Leach shrimp larvae which are given extracts from plants and then incubated for 1x24 hours. The results obtained from this study as an LC50 (Lethal concentration) value of 532.78 ppm. LC50 (Lethal Concentration) value of 532.78 ppm indicates that the concentration of the compound can cause death in Artemia salina Leach by as much as 50%.
