Abstract

Rabies is a zoonotic disease. Rabies protection level detection was performed using antibody titration. Blood sampling activities in the field require special handling to avoid blood lysis, the sample delivery requires a cold chain with a stable temperature. Alternative method forwhole blood sample delivery using filter paper were carried out on 48 samples from susceptible animals (dogs and cats) transpored through the Soekarno Hatta Agricultural Quarantine Center. This  study was designed to investigate the feasibility of filter paper sampling of blood at temperature of 26 °C to detect of rabies antibodies using indirect method of ELISA. The results of statistical analysis of  Randomized Block Design and Tukey's test showed that the antibody titres of whole blood extracted from filter paper diluted in the PBS T 100 μl were equivalent to antibody titres of serum. Assesment of filter paper capability has sensitivity and specificity as much as 96% and 76%, positive predictive value of 67%, negative predictive value of  94% and reliability level of filter paper were 0.5 is moderate category. This indicates that the filter paper can be used as an alternative method of blood transport medium for rabies ELISA test.

Abstract

Increased broiler production in Bogor Regency leads to the continuous development of traditional chicken collecting facilities and slaughterhouse (CCFS), especially in Bogor City. Pondok Rumput area, located in Kebon Pedes Village, as one of CCFS centers. Pondok Rumput CCFS was first established in 1971. There are currently 25 CCFS in Pondok Rumput. The CCFS in this area are located in densely populated residential area. The CCFS, are located in the middle of the residential area leads to various problems, one of which is environmental pollution.The Study aimed to identify the factors determining the sustainability status of CCFS in Pondok Rumput area, in Tanah Sareal Subdistric, Bogor City. The types of data collected included primary and secondary data. The data analysis used by Multi Dimensional Scalling (MDS) analysis (by using Rapfish software) and prospective analysis. Partial analysis of each dimension concerning the sustainability status of chicken collecting facilities and slaughterhouse in Pondok Rumput showed that ecological dimension were not unsustainable with a value of 24.66. The key attributes were having business license, conformity to slaughtering regulations, entrepreneurs’ attitudes towards relocation, and business feasibility.

Abstract

Food products of animal origin which are free from biological and chemical contamination are an absolute requirement of food safety. Antibiotics residues in the food of animal origin is one of the chemical contaminants in food are harmful/hazardous to human health. Tetracycline is an antibiotic that is often used in the fishing industry. The study aims to detect the occurrence of tetracycline residues in tilapia sold in traditional markets in Kota Yogyakarta. The research was conducted with detection test (detect disease). The study used 61 samples of tilapia fish from 16 traditional markets in Kota Yogyakarta. Tilapia meat samples are prepared according to the Association of Official Analytical Chemistry (AOAC). The existence of tetracycline residues in meat can be detected by high performance liquid chromatography (HPLC). Residue analysis in fish meat was conducted at the Department of Pharmacology, Faculty of Veterinary Medicine of Gadjah Mada University using HPLC Shimadzu version 6.1. The results showed that 24 samples of tilapia meat marketed in Kota Yogyakarta was positively contained tetracycline antibiotic residues. A total of 19 samples contained residues above maximum residue limits (MRL). The conclusion of the study is 31% of tilapia marketed in Kota Yogyakarta contains tetracycline residues above MRL. Surveillance of residues in food of animal origin and further research in terms of reducing antibiotic residue in tilapia meat is highly recommended to improve the quality and safety of food of animal origin.

Abstract

Antibiotics are substances that capable of inhibiting the growth of or killing microorganisms. The presence of residues in the tissues are associated with continuous use of antibiotics for long periods of time. The antibiotic residue that is often detected in freshwater fishery products is tetracycline. One method of analysis of tetracycline residues in fish meat is widely developed using high performance liquid chromatography (HPLC). This research was aimed to validate the method of analysis of tetracycline content in tilapia meat by using HPLC Shimadzu 6.1. The mobile phase consisting of methanol: acetonitrile: oxalic acid (5:15:80) with 1 ml / min flow rate, detector UV Vis with wavelength 355 nm, and C18Shim-pack column size 150 L x 4,6 mm at temperature of 30oC. The result of this research showed values corresponding to validation criteria based on parameters of specificity, precision, accuracy, linearity, limit of detection, and limit of quantification. Tetracycline analysis using HPLC tool has good and acurate validity as the first step in detecting the tetracycline level in tilapia meat.

Abstract

This study was aimed to determine the effect of insulin supplementation in maturation and/or culture medium on nuclear maturation rate and the early bovine embryo development in vitro. Oocytes were collected and matured in maturation media without (IVM I-) or with (IVM I+) 10 ug/µL insulin at incubator 5% CO2 and the temperature of 39 °C, for early embryonic development evaluation, oocytes were divided into 4 groups, without the supplementation of insulin to the maturation medium and culture (IVM I-/IVC I-), insulin supplementation only the maturation medium (IVM I+/IVC I-), insulin supplementation only in the culture medium (IVM I-/IVC I+), and the combination of insulin supplementation to the maturation medium and culture (IVM I+/IVC I+). The result showed that supplementation of insulin to the maturation medium increased number nuclear maturation was higher 87.7% (P<0.05) compared to treatment without supplementation of insulin (70.1%). Cleavage rate in treatment IVM was higher IVM I-/IVC I- (55.8%), IVM I+/IVC I- (64.1%), IVM I-/IVC I+ (59.9%) (P<0.05). Result of the other were showed that early bovine embryo on day-4 cultured (IVC) reached 16 cell on treatment IVM I-/IVC I+(31,9%) and IVM I+/IVC I+ (27,1%) were higher compared to treatment IVM I-/IVC I-(2.9%) and IVM I+/IVC I- (2.5%) (P<0.05). In conclusion, supplementation of insulin to maturation medium and culture medium can increase nuclear maturation rate and improved early embryo cleavage rate.

Abstract

Leptospirosis is a zoonotic disease of global concern, and is caused by pathogenic serovar Leptospira interrogans. Canine Leptospirososis is widespread worldwide, dogs can act as incidental hosts or maintenance hosts for various serovars. The purpose of this research was to identify leptospire serovars that infect healthy and suspected leptospirosis dogs in Yogyakarta. A total of 56 dogs (36 healthy dogs and 20 suspect leptospirosis dogs) sera were taken from cephalica vein as much as 3 ml. Sera were examined for leptospirosis with Microscopic Aglutination Test (MAT) which conducted at the Research Center for Veterinary Science, Bogor. Microscopic Aglutination Test carried out on various Leptospire serovar, namely: Ichterohaemorrhagiae, Javanica, Celledoni, Ballum, Pyogenes, Cynopeteri, Rachmati, Australis, Pomona, Canicola, Grippotyphosa, Bataviae, Hardjo, and Tarrasovi. The results showed that Celledoni serovars infected 25% of healthy dogs and 5% of suspect leptospirosis dogs, Javanica serovar infected 19% of healthy dogs, Bataviae serovars infected 15% of suspect leptospirosis dogs, Grippotyphosa serovar infected 11% of healthy dogs, Tarrasovi serovar infected 10% of suspect leptospirosis dogs, serovars Cynopteri infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, serovar Pyrogenes infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, and serovar Rachmati infects 5% of suspect leptospirosis dogs. Seven healthy dogs (19%) and 2 suspect leptospirosis dogs (10%) were infected with more than 2 leptospire serovars. From the results of this study it can be concluded that Celledoni serovar of Leptospira interrogans infection causes subclinical leptospirosis, while Bataviae serovar infection causes clinical leptospirosis in dogs in Yogyakarta.

Abstract

The phytochemical components of avocado seeds (Persea americana Mill) consist of alkaloids, flavonoids, tannins and saponins, so that they have the potential as antibacterials. The aim of this study was to determine the ability of Persea americana Mill extract to inhibit the growth of Staphylococcus aureus through inhibition zone observation and the ability to accelerate wound healing infected by S. aureus through observation of wound closure and cytopathology.  Persea americana Mill were extracted using maceration method with 70% ethanol. The activities of this extract were tested by diffusion disc agar method. Biomedical tests were carried out on anaestetic Wistar rats whose were S. aureus infected wounds  and treated with Persea Americana Mill extract. Based on the results of the antimicrobial test, avocado seed extract has a inhibitory effect on the growth of S. aureus at a concentration of 6.25% and best at a concentration of 100% compared to the control. Tests in experimental rats showed the effect of wound healing on the 2rd day after giving the extract Persea americana Mill ointment, faster than the control, which was need 4 days of wound closure. Cytopathology results showed that leukocyte activity was more prominent in rats treated with avocado seed extract compared to control rats. Avocado seeds as byproducts have the potential to be new herbal medicines that can be used as antimicrobials of S. aureus which have been confirmed to be resistant to various antibiotics.

Abstract

Egg Drop Syndrome can cause detrimental impacts on breeders due to reducing production and quality of theaffected eggs.This study aim was to determine the seroprevalece ofantibody against Egg Drop Syndrome (EDS) virus in ducks. Antibody titers examination was done from 75blood samplesof ducks thathave not been vaccinatedagainstEDS virus. The duck samples were collected from Tumbak Bayuh Village, Mengwi, Badung. Serological examination was held at the Virology Laboratory,Faculty of Veterinary Medicine, Udayana University by usingHaemagglutination Inhibition (HI) test. HI test results showed that 24 samples were positive contained antibody of the EDS,while 51 samples were negative. Range of EDS antibody titer observed was from24 to 27 HI units.  This results indicates that the ducks have protective antibody titer against EDS virus. The positive serum samples were also tested using HI test against Newcastle Disease (ND) virus with negative result. It can be concluded that the ducks in Tumbak Bayuh Village, Mengwi, Badung have 32% of antibody titer against EDS virus whichmight result from being exposed by EDS virus naturally.

Abstract

Tanah Putih Village has the potential for the development of Bali cattle because it has extensive grazing land and the majority of farmers raise Bali cattle. The aim of the study was to obtain data on the diversity and intensity of gastrointestinal endoparasitic infection in Balinese cattle with an extensive breeding system in Tanah Putih Village, Kupang Timur District, Kupang Regency, East Nusa Tenggara Province. Gastrointestinal endoparasitic diversity was identify using sedimentation and flotation methods while the intensity of endoparasitic infection was carried out using McMaster method, while data on the diversity and rates of gastrointestinal endoparasitic infection were analyzed descriptively. The results of the study on the diversity of gastrointestinal endoparasites found there are five types of nematode worm eggs namely Haemonchus contortus, Oesophagostomum radiatum, Strongyloides papillosus, Bunostomum phlebotomum, and Trichostrongylus axei. Factors that support the diversity of endoparasites are feed sources, livestock populations, and grazing rotation. The average number of worm eggs found based on the total eggs per gram of faeces for Haemonchus contortus as many as 100 eggs and the other four types of worm eggs amounted to 50 eggs. The average number of eggs indicates that endoparasitic infections in these animals fall into the mild intensity category, is influenced by season, feed source, and age of livestock. The five types of gastrointestinal endoparasites found namely Haemonchus contortus, Oesophagostomum radiatum, Strongyloides papillosus, Bunostomum phlebotomum, and Trichostrongylus axei.

Abstract

The study evaluates in vitro effects of Areca catechu crude aqueous extract  as anthelmintics on Ascaridia galli adult mortality rate and morphometry. Naturally infected chickens were collected from slaughtered chickens at local slaughterhouses in Yogyakarta. Chicken’s intestines were carefully examined and transported to the Parasitological Laboratory of Gadjah Mada University, Yogyakarta. Ascaridia galli were collected from intestines (including duodenum, jejunum, and ileum) and put into a petri dish containing 0.62% saline water. There were 60 A. galli in each concentration of Areca catechu crude aqueous extract. Investigation on mortality rate of adult worms at various concentration and observation as well as differences in A. galli morphometry and mortality rate were analyzed using analysis Anova. The figures of the parasites or the parts of the parasites were captured using camera Lucida and then they were measured using both micrometer and curvimeter. The morphology of the parasites was identified to find out their morphometric characteristics.The results of morphometric observation of the A. galli showed that there was a significant difference in the body width and the vulva length of the females and in the body width, the esophageal length and the esophageal width of the males.This research disclosed that Areca catechu crude aqueous extract significantly affected the mortality rate of adult A.galli at various concentration. This research observed that the best concentration to kill the adult worms was 25% Areca catechu crude aqueous extract.
