Abstract

Three types of doping drugs, morphine, heroin, and codeine were analyzed qualitatively in urine using GCMS: HP5MS column containing a nonpolar substance, phenylmethyl siloxane, with a temperature range of 90-250ºC and a rate of increase of 10ºC/minute. Diethyl ether was used to extract morphine, codeine, and heroin. The extractions were performed at pH 8, the organic phase was evaporated, followed by derivatization. Morphine and its derivatives were treated with a solution of N-methyl-N-trimethylsilyl-trifluoroacetamide and N-methyl-bis-trifluoroacetamide. The results of the gas chromatographic analysis showed that the retention times of morphine, heroin, and codeine the retention times were 8.01, 7.32, 6.21 minutes, while those of their derivatives were 8.10, 8.10, 7.95 minutes respectively. The identification of each drug using mass spectrometry revealed that their mass spectra were identical to the standard. The mass spectra of the derivatives: morphine-TMS and codeine-TMS showed a resemblance index of 93% respectively compound to the standard, with a detection limit of 2 ug/ml.

Abstract

Human insulin is a hormone that regulate blood glucose in human. Due to the vital role of insulin and the trend of diabetic case globally, insulin demand has been rise in the world. Indonesia as a country that had diabetic patient more than 10 million people in 2017 is still imported raw material of human insulin. The development of production process of human insulin is needed in order to support government objective to be independence in medicine aspect. Human insulin precursor (HIP) expressed from a Pichia pastoris X33/pD902-IP had been developed and optimised in a small scale. However, the scaling up in fermenter 10L has not been studied. Using a 10L-fermenter the fermentation system of P.pastoris X33/pD902-IP was developed. Fermentation done in 120 hours using a basal salt medium (half concentration) for the vegetative and induction media. To induce HIP expression, methanol is fed by pulse strategy with a gradient concentration 1-3% for 48 hours. The dry cell weight (DCW) and HIP titer were 72 g/L and 192 mg/L, respectively. This development is the first fermentation process of HIP in fermenter 10L in Indonesia.

Abstract

Reading is an important activity that must be present in the science learning process, especially reading with meaningful activities, one of which is critical argumentation reading activity. Reading is very closely related to student motivation while reading. This motivation plays an important role in supporting the success of reading goals. So far, research related to students' reading motivation has rarely been carried out, especially for critical argumentation reading activities. Most of the research was conducted at the elementary and secondary school levels. Therefore, this research will be conducted with the aim of analyzing students' motivation while reading using critical argumentation activities. This study involved 83 science teacher candidate students. In practice, students were divided into three classes, namely group experimental class (N = 29), individual experimental class (N = 29), and control class (N = 25). The research was conducted using the think aloud protocol method, in which students were asked questions while reading the text, and recorded all the answers orally. The results showed that students' motivation during reading as part of a self-regulation strategy showed better results when students read using the critical argumentation reading model. In addition, students are able to carry out self-regulation strategies from a better motivational aspect if reading activities are carried out in groups.
