Abstract

Salmonella typhi is a negative basil Gram bacterium that causes typhoid fever. Flagel of S. typhi contain proteins, as locomotor and it can help bacteria to attach on host cells. Haemagglutinin protein is protein that can agglutinate erythrocytes. The goal of this research is to analize S. typhi flagellin proteins haemagglutination activity Based on ABO blood group. Flagellin proteins isolation method of modified Alexan method (2009) and haemagglutination test method of Finkeltein and Hanne method (1982). Haemagglutination result of ABO blood type show that SLT-1 S. typhi flagellin proteins can’t agglutinate human erythrocytes A, B, and O, but it can agglutinate human erythrocytes until 8 times dilution of AB blood type from 50 μg/μl concentration in 50 μl. BA 07.4 S. typhi flagellin proteins can agglutinate human erythrocytes of A blood type until 16 times dilution, 8 times dilution of AB blood type, 4 times dilution of O blood type, and it can’t agglutinate human erythrocytes B blood type.Keywords: Salmonella typhi, Flagellin Proteins, Haemagglutination, Erythrocytes of ABO System.

Abstract

Entomological study with laboratory analysis done on Anopheles sundaicus mosquitoes as a malaria vector. The aim is to determine the existence of genetic variation in the Anopheles sundaicus species. PCR was used for the molecular identification of An. Sundaicus using COI and ITS2 genes. COI and ITS2 sequences in the study group compared with An. Epiroticus from GenBank. The comparation were done using a multiple sequence alignment program of Bioedit 7.1.9. The phylogenetic tree were constructed using neighbor-joining and bootstrap- tested by MEGA6. ITS2 sequence alignment results showed a single nucleotide substitution that can be used as markers for species determination. Phylogenetic tree analysis based on COI gene showed that An. Sundaicus in this study with isolates from GenBank, separate into two different groups or are polyphyletic. This study proved that An. Sundaicus from field collection was An. Epiroticus species by molecular identification. This study can be used to help public health policy related to vector control.Keywords: PCR, COI, ITS2, An. Sundaicus, An. Epiroticus

Abstract

Background: The prevalence of abortion in Iran in 2014 reached 45.7% of the 2,470 pregnant women. Abortion among farmers exposed to pesticides as much as 9%. Pesticide exposure in pregnant women can cause thyroid dysfunction that causes hypothyroidism causing abortion. Methods: This case-control study involving 30 cases and 30 control in Puskesmas Sidamulya. The variables studied were history of pesticide exposure (duration of exposure, long exposure every day, duration of exposure every week, engaging in agricultural activity). And the incidence of spontaneous abortion, among the variables that cholinesterase levels. Analysis using Chi-square test. Results: There was a relationship with the exposure period the incidence of spontaneous abortion (p=0.001,OR=14.00 CI 95%), There is a relationship between duration of exposure every day with spontaneous abortion (p=0.001,OR=12.250 CI 95%), there is a relationship long exposure every week with Spontaneous abortion (p=0.000,OR=25.375 CI 95%), there is a relationship of involvement in farming activities with the incidence of spontaneous abortion (p=0.004,OR=7.875 95% CI) Conclusion: There is a relationship between the length of exposure with spontaneous abortion (p=0.001, OR=14.00 CI 95%), There is a relationship between duration of exposure every day with spontaneous abortion (p=0.001,OR=12.250 CI 95%), There is a relationship Exposure every week with spontaneous abortion (p=0.000,OR=25.375 CI 95%), There is a relationship of involvement in agricultural activities with spontaneous abortion (p=0.004, OR=7.875 95% CI)Keywords: incidence of spontaneous abortion, history of exposure to pesticides

Abstract

French beans are foodstuff from agricultural products that are useful, for nutrient sources that needs continue to increase to rise as cunsumed by the whole society, so it needs to be improved productivity, for example by the giving of PGPRM. This reseacrh aims to determine the most appropiate time when giving the PGPRM which give growth and yield the best french beans. This reseacrh was conducted at the experiment station Mercu Buana University Faculty of Agroindustri Yogyakarta, located in Gunung Bulu, Sedayu, Bantul, Yogyakarta in February-April 2015. This reseacrh uses a single factor experimental design with 4 treatments were arrenged in the field using a completely randomized design (CRD) with 3 replications, namely Giving PGPRM at the time of preparation of the seed; Giving PGPRM at the planting time; Giving PGPRM one week after planting; and Giving PGPRM during vegetative growth, given once a week. The results from the reseacrh showed that while giving PGPRM (Plant Growth Promoting Rhizospheric Microorganism) effect on growth and yield french bean, that giving PGPRM once a week at the vegetative stage of growth showed a better french beans and giving PGPRM in preparing french bean seeds provide results better.Keywords : French beans, PGPRM (Plant Growth Promoting Rhizospheric Microorganism), The exact timing of PGPR

Abstract

Tifoid fever is endemic disease who caused by Salmonella typhi bacteria and still to be seriously health problem in Indonesia. The treatment still use antibiotic who can cause resistant if uncontrol using. Tamarind (Tamarindus indica) is plant who can use as medicine. The leaves contain flavonoid, tannin, and saponin who can function as antibacteria. The purpose of this research to know inhibition of tamarind leaves to Salmonella typhi who caused tifoid fever in 25% consentration. This research using old tamrind leaves from Rembang. The leaves was drying and powdering, then soxhletation with eter, extraction with maceration and infused method using distiled water with 25% concentration, then freeze dying and make 1, 2, 3, 4, and 5 mg/25 μL concentration per disc in a row. antibacteri tested with Kirby bauer method. The result of this experiment showing all of variance concentrations at disc can inhibit Salmonella typhi BA07.4 in maceration method as well as in infused method in 25% concentration of tamarind leaves. 5 mg/25 μL concentration has large antibacterial essence showed with existence large zone of inhibition 11,5 mm in maceration method and 14 mm in infused method. infused method of 25% tamarind leaves is more antibacterial essence than maceration method.Keyword : Tamarind leaves, Salmonella typhi, Tifoid fever

Abstract

Candida albicans is a normal flora in the oral mucosa, tongue, and palate, but it could become the pathogenic one. If the amounts were excess so that it could cause thrush. Kaffir lime peel infusion contains saponins, tannins, flavonoids, coumarin which are active antifungal compound. The purpose of this study was to examine the effectiveness of kaffir lime peel infusion to the growth of Candida albicans which causes thrush with a consentration of 10%, 15%, 20% and contact time of 5 minutes, 10 minutes, 15 minutes. Method of this study used an experimental research laboratory. Suspension Candida albicans 100 mL with dilution 10-4 of McFarland 0,5 and contacted to the kaffir lime peel infusion 10%, 15%, 20% with three repetitions. After 5 minutes, 10 minutes, and 15 minutes put 100 μL and then inoculated into SGA antibiotics then incubated at 37˚C for 48 hours. Based on the results of this study concluded that kaffir lime peel infusion can inhibit the growth of Candida albicans. The best results was the 20% consentration and contact time of 15 minutes with an average colonies of 3×104 CFU/100 μL. The higher consentration of kaffir lime peel infusion the more able to inhibit the growth of Candida albicans and the longer the contact time kaffir lime peel infusion, the more can inhibit the growth of Candida albicans.Keywords: Infusion, Citrus hystrix DC., Candida albicans.

Abstract

France Stringbean is one of good variaty vegetables which is export commodity. It has high consumption rate so that it needs to be risen of its growth and crops. One of the ways is by using various planting media and giving PGPRM. The purpose of this research is to learn the effects of planting media to the growth and the crops of France String Bean. This research conducted in the experiment garden of Agro Industry Faculty of Yogyakarta Mercu Buana University which is in the 90 meters above the sea level. This research uses Experimental Single Factor with eight different treatments which is arranged into RAL (Rancangan Acak Lengkap) by three times repetitions. The experiment factors are Sand+Soil+Compost (1:1:2) without PGPRM, Sand+Soil+Compost (1:1:1) without PGPRM, Sand+Soil+Compost (2:1:1) without PGPRM, Sand+Soil+Compost (0:1:1) without PGPRM, Sand+Soil+Compost (1:1:2) with PGPRM, Sand+Soil+Compost (1:1:1) with PGPRM, Sand+Soil+Compost (2:1:1) with PGPRM, Sand+Soil+Compost (0:1:1) with PGPRM. The result of this research showed that planting media and PGPRM do not have any differentiations to all variables of the growth and the crops of France String Bean.Keywords : France String Bean, Planting Media, PGPRM, growth, crops
