Abstract. Elfiati D, Susilowati A, Modes C, Rachmat HH. 2019. Morphological and molecular identification of cellulolytic fungi associated with local raru species. Biodiversitas 20: 2348-2354. This study is aimed to isolate and identify cellulolytic fungi from different local Raru species in Central Tapanuli, North Sumatra, Indonesia. Cellulolytic fungi are fungi that can degrade natural polymer materials such as cellulose. The fungal species were isolated from soil under three local raru species. The samples inoculating (1 mL) from serial dilutions (10-2 to 10-5) on Carboxy Methyl Cellulose (CMC) plates. The plates were incubated at 28 ?C for one week, then the fungal colonies were observed and pure cultures were maintained. Fungi with cellulolytic activity were characterized by the formation of clear zones around the colonies on CMC media plus congo red (0.1%). The identification of fungi was carried out by molecular identification of the isolated fungi at the species level. The extracted fungal DNA was amplified by PCR using specific internal transcribed spacer primer (ITS1/ITS4). The PCR products were sequenced and compared with the other related sequences in Gen Bank (NCBI). 20 isolates were successfully isolated and nine isolates had cellulolytic activity. Six fungal species from 20 isolates were identified as: Aspergillus sp., Penicillium sp., Aspergillus terreus, Penicillium mariae-crucis, Aspergillus tanneri and Penicillium citrinum. The results showed that Aspergillus terreus was the most abundant fungus obtained from all the raru stand.

Abstract. Cahyanto T, Efendi M, Shofara RM. 2019. Short Communication: Floristic survey of vascular plant in the submontane forest of Mt. Burangrang Nature Reserve, West Java, Indonesia. Biodiversitas 20: 2197-2205. A floristic survey was conducted in submontane forest of Block Pulus ??Mount Burangrang West Java. The objectives of the study were to inventory vascular plant and do quantitative measurements of floristic composition as well as their structure vegetation in the submontane forest of Nature Reserves Mt. Burangrang, Purwakarta West Java. Samples were recorded using exploration methods, in the hiking traill of Mt. Burangrang, from 946 to 1110 m asl. Vegetation analysis was done using sampling plots methods, with plot size of 500 m2 in four locations. Result was that 208 species of vascular plant consisting of basal family of angiosperm (1 species), magnoliids (21 species), monocots (33 species), eudicots (1 species), superrosids (1 species), rosids (74 species), superasterids (5 species), and asterids (47), added with 25 species of pterydophytes were found in the area. The three families of plants are Lauraceae (10 species), Urticaceae (9 species), and Rubiaceae (8 species) dominating those areas. Fourteen species belong to IUCN red list: Least concern/LC (12 species), Vulnerable/VU (1 species), and endangered/EN (1 species). Furthermore, Castanopsis argentea A.DC, Pinanga javana Blume and Amorphophallus decus-silvae Backer & Aldrew belonging to protected plants are also found in the area. Based on the assessment of analysis vegetation, the forest has experienced disturbance, the density of trees is commonly  low and has a lot of gaps. Many vacant lots are found. On the other side, there is the presence of invasive plant species that may slow down a succession into climax growth of local plant.

Abstract. Mau YS, Ndiwa ASS, Markus JER, Arsa IGBA. 2019. Agronomic performance and drought tolerance level of sweet potato hybrids grown in Kupang, East Nusa Tenggara, Indonesia. Biodiversitas 20: 2187-2196. This study was conducted to evaluate agronomic performance and to assess drought tolerance level of purple and orange-fleshed F1 sweet potato hybrids in dryland during dry season in Kupang, East Nusa Tenggara. The study was carried out in Integrated Archipelagic Dryland Field Laboratory of Universitas Nusa Cendana, Kupang, from May to October 2018. Two sets of experiment were conducted, the first was for evaluation of agronomic performance, and the second was for evaluation of drought tolerance of the sweet potato genotypes. The first experiment assessed the agronomic performance of 20 sweet potato genotypes in a Randomized Block Design. The second experiment employed a Split Plot Design consisted of irrigation level as the main plot and sweet potato genotypes as subplot treatments. The main plot consisted of no stress (normal) and water-stressed conditions while the sub-plot comprised of 20 genotypes of the sweet potato. The observed agronomic performance variables included growth and yield parameters. Tuber yields in no stress and water-stressed conditions were observed and used for assessment of drought tolerance. The results showed that, in the agronomic performance experiment, the sweet potato genotypes differed significantly in all the observed agronomic performance variables including growth, yield contributing, and yield variables. UNC2016.Cil/JPV.01 and UNC2016.Cil/JPV.05 were short maturing genotypes with only about 90 days to harvesting while the rest of the genotypes were harvested in 120 to 150 days. The genotypes were also significantly varied in tuber yield; the highest was observed in UNC2016.JPV/KDL.11 (43.38 t ha-1). In the drought tolerance evaluation experiment, tuber yields in no stress and stressed condition were, respectively, 23.42 t ha-1 and 7.08 t ha-1. Six F1 hybrids were classified and selected as drought-tolerant genotypes with high yielding performance in both no stress and water-stressed conditions.

Biodiversitas 20: 2603-2611. Fruit as a source of food has been mostly underrated, while in fact play a role in providing nutritional security as well as contributing to the integrity of local ecosystems. The declining significance of local indigenous fruits, often referred to as underutilized fruits, in rural populations may have an unprecedented consequence to the availability of high-quality resources for the wider society. This article, therefore, sees this importance by documenting the existence of underutilized fruit-trees across indigenous communities, often acknowledged to be the chaperones of local biodiversity. Using a combined method of interviews, exploratory observation and vegetation analysis in seven indigenous communities in West Java Province, this study investigated the role and position of fruit-tree species in their ecological landscapes and communities' knowledge systems. The article documents 75 fruit tree species, 38 out of which are categorized as underutilized. The proportion of this group of species to the total fruit tree species varied between 0 to 45%. Further analysis found that the underutilized fruit trees were far smaller in abundance compared to commercial timber, fruit, and other trees. We conclude that efforts to uplift the conservation value of these fruit-trees can begin via promotion and market development by multiple stakeholders.

Genome group classification and diversity analysis of talas and rutai banana, two local cultivars from East Kalimantan, based on morphological characters. Biodiversitas 20: 2355-2367. Talas and rutai banana are two local cultivars grown in East Kalimantan and long-time used as dessert or cooking bananas for traditional food and cakes, but unfortunately, their taxonomic and genetic status are still little known. This study was conducted to explore their taxonomic and genetic status through morphological observation, genomic group classification and diversity analysis of talas and rutai banana compared to other recognized and identified bananas in Indonesia, i.e., liar/monyet, Ambon, tanduk and klutuk banana. In total 108 morphological characters were observed on site of growing area using the “Descriptors for Banana (Musa spp.)” morphological indicators. The genome group classification was carried out using Simmonds and Shepherd scoring system toward fifteen key banana characters. The diversity analysis was performed using Hierarchy Cluster Analysis at MINITAB 17.1 software based on the selected characters of the highest Principal Component Analysis (PCA) values. The morphological observation showed that the characters of talas and rutai banana are much closely related to those of belonging to M. acuminata although some characters showed balbisiana-type. In addition, liar/monyet and klutuk banana morphological characters were exactly matched with the M. acuminata and M. balbisiana accessions. Furthermore, most of Ambon banana morphological characters are similar to M. acuminata, but tanduk banana had a combination characters between M. acuminate and M. balbisiana. The genome group classification showed that talas and rutai banana are the AAB genotype, while liar/monyet, Ambon, tanduk, and klutuk were grouped into AA, AAA, AB, and BB genotype, respectively. The diversity analysis showed that talas and rutai banana have a very high similarity about 75% and closed related to AA/AAA genotype i.e., liar/monyet and Ambon with similarities level of 45%.

Short Communication: Digenea parasites in giant mudskipper (Periopthalmodon schlosseri) in Kuala Lupak Village, South Kalimantan, Indonesia. Biodiversitas 20: 3782-3787. Parasites are organisms that impart detrimental effects on their host. Monogenea which are generally ectoparasites and digenea which are endoparasites are the most common parasites which invade fish. This research was aimed to find out the existence of monogenean and digenean parasites as well as their species in giant mudskipper or timpakul fish (Periopthalmodon schlosseri) in Kuala Lupak Village, South Kalimantan, Indonesia. Employing 25 giant mudskipper which was randomly collected from Kuala Lupak Village, South Kalimantan, this study explored the presence of ectoparasites and endoparasites on the mucous, fins, gills, intestines, stomach, and meat of the fish. The results showed that only digenean parasites were found in the giant mudskipper (Periopthalmodon schlosseri). The digenean parasites found to come from Opecoeloides genus and Buchepalidae family, metacercariae cysts, and parasites of the nematode phylum. Monogenea parasites were not found due to low fish-density, fish behavior that is rarely in the water, and adequate nutrition which is the opposite of growth-supporting factors of monogenean. Parasitic infection could occur because the fish eat some intermediate hosts of digenea, such as snails and other fish. Besides, the condition of waters that have been contaminated with digenean eggs allows the life cycle of the digenean to continue in fish.

Biodiversitas 20: 2004-2010. Insulin-like growth factor 1 (IGF-1) is defined as an essential gene for growth. Therefore, this research aims to identify the polymorphism of the IGF-1 gene from ayam ketawa with the dangdut and slow type vocal characteristics. Twenty male chickens were selected as an animal model. The sample consisted of 10 chickens with the dangdut type and 10 chickens with the slow type. The chickens were all obtained from five areas of the Pinrang district. The RFLP and HRM methods were selected to be used in this research. Results showed that the IGF-1 gene was 622 bp in length, while polymorphism analysis showed that the dangdut and slow type are homozygotic. The unrestricted DNA fragment indicated it after reaction of the restriction enzyme Pst1. Moreover, it confirmed that ayam ketawa with the dangdut or slow type had a similar sequence.

Short Communication: Use of Geographic Information System for mapping of Aquilaria malaccensis land suitability in North Sumatra, Indonesia. Biodiversitas 20: 2561-2568.  The Geographic Information System (GIS) can be applicated in the field of forestry and agriculture, such as for mapping as part of land evaluation. The land suitability of Aquilaria malaccensis in the agroforestry land of North Sumatra Province is important.  Commonly, A. malaccensis were planted by the community in Langkat and Deli Serdang District. This study aimed to map the land suitability for A. malaccensis in Telaga Village, Sei Bingai Sub District, Langkat District, Peria-ria Village and Sari Laba Jahe Village, Biru-biru Sub District, Deli Serdang District. A survey method was used to collect soil samples in the field. Land suitability classification (LSC) for A. malaccensis was evaluated based on the matching method.  The results showed that the actual land suitability classes for A. malaccensis in Telaga Village were moderately suitable (S2) and marginally suitable (S3). The actual land suitability classes for A. malaccensis in Peria-ria Village and Sari Laba Jahe Village were moderately suitable (S2), marginally suitable (S3) and not suitable (N). There were several limiting factors in land suitability evaluation in the area, namely: nutrient retention (nr), erosion hazard (eh), and root zone medium (rc).

Abstract. Sofiyanti N, Iriani D, Fitmawati. 2019. The first record of grass-like fern, Schizaea dichotoma (L.) J. Sm. (Schizaeaceae) from Lingga Island, Indonesia: Its morphological, anatomical and palynological study. Biodiversitas 20: 2651-2660. Schizaea dichotoma (L.) J.Sm. is one of fern species from Family Schizaeaceae. This species is grass-like, dimorphic with fan-shaped fronds, and well-known with local name Rumput Merak. The first record of this species is reported from Lingga Island, Riau Islands Province, Indonesia. The aim of this study was to examine the morphology, anatomy and spore characteristics of this fern. This study had been carried from June to December 2017. Samples were collected using exploration method, documented, and prepared for herbarium specimen. In this study, we examined the morphological characters, anatomical characters of rhizome, stipes, and pinnae of both fertile and sterile fronds, as well as spore feature. The anatomical preparations were carried out using paraffin method, while spore specimens using acetolysis method. The anatomy of rhizome, stipes, and pinnae, as well as spore characters, were then observed and documented using digital microscope Celeron. S. dichotoma is dimorphic fern with grass-like habit, fan-shaped laminae that repeatedly forked end in sorospore in fertile frond. The rhizome has vascular bundle surrounded by star-shaped sclerenchymatous layer. The cross-section of stipes of both fertile and sterile fronds are "M" shaped with a vascular bundle located in the dorsal side, while the leaves have a vascular bundle in the middle of midrib. The type of vascular bundles and stele of those organs are similar, i.a. collateral closed and protostele. We observed The Casparian strip in the endodermis layer of rhizome, stipes, and leaves, as well as at cortex and among the xylem. The spore of S. dichotoma is monolete and large spore class (ca. 59.5 ± 2.4 x 35.86 ± 2.01 µm).

Leaf chlorophyll content in North Sulawesi (Indonesia) local rice cultivars subjected to polyethylene glycol (PEG) 8000-induced water deficit at the vegetative phase. Biodiversitas 20: 2462-2467. Climate change can result in drought stress in the environment and this condition reduces rice production. Polyethylene glycol (PEG) is used to induce water deficit, because it is able to decrease water potential in the solution. The present study aimed to evaluate the physiological response of North Sulawesi local rice (cvs. Superwin, Ombong, Burungan, and Temo) to PEG 8000-induced water deficit based on the leaf chlorophylls (a, b and total chl.) content at the vegetative phase. The results of this study showed that PEG 8000-induced water deficit with media water potential (WP) -0.25 and -0.5 MPa decreased the content of leaf total chlorophyll and leaf chlorophyll a. The longer treatment period could reduce the contents of leaf total chlorophyll, chlorophyll a and chlorophyll b. The content of leaf chlorophyll total and chlorophyll a were potential physiological indicators for North Sulawesi local rice response to PEG 8000-induced water deficit.
