Variation in morphological traits of a selection of Indonesian winged bean accessions (Psophocarpus tetragonolobus) and its analysis to assess genetic diversity among accessions. Biodiversitas 21: 2991-3000. In Indonesia, winged bean (Psophocarpus tetragonolobus (L.) DC.) is a traditional vegetable crop grown mainly for its edible green pods. Plant breeding programs aim to produce cultivars with high production and good nutritional qualities. The objective of this present study was to assess genetic diversity among 21 selected Indonesian winged bean lines based on observation of morphological characters. This was the first step in determining an appropriate breeding program for the development of improved vegetable cultivars. Twelve qualitative characters and eight quantitative variables were assessed for each of the 21 lines. Categorical differences among lines were observed in characters such as leaflet, pod and seed shape, pod surface texture, anthocyanin pigmentation of stem, flowers and pods. Several of these characters are useful as genetic markers, and cluster analysis of the 21 lines on the basis of qualitative characters enabled two distinct groupings to be identified. Quantitative variation across line means was also high for several of the quantitative variables (a coefficient of variation > 25% for pod length, number of pods per plant, and total pod weight per plant). Principal component analysis applied to the eight variables accounted for 86% of the total variation in just three components with eigenvalues > 1. On Component 1, the characters number of days to first open flower, pod length and pod weight were closely aligned with total weight of pods per plant. Number of pods per plant was not closely aligned with weight of pods per plant. This study has enabled broad differences between groups of lines to be categorized and has identified particular lines with characteristics that recommend them for inclusion as parents in inheritance studies designed to elucidate the contribution that individual characters make to overall productivity, attractiveness, and nutrition of this useful, high protein, vegetable species.

Bifidobacteria spp. are a group of Lactic Acid Bacteria commonly found in the gastrointestinal tract of adults and infants. LAB are known as probiotics and have many health benefits. This research aimed to isolate Bifidobacteria from infant stool, identify, explore their diversity, and screen their potential as probiotics. Stool samples were collected from 3 healthy infants in Banyumas Regency. The potential screening included lysozyme resistance, antimicrobial activity, and exopolysaccharide production. A total of 7 Bifidobacterium species were isolated from infant stool: B. catenulatum, B. minimum, B. indicum, B. dentium, B. asteroides, B. galicum, and B. coerinum. B. indicum isolates (Bb3F and Bb1B) had the greatest inhibition activity against Escherichia coli and Candida albicans with 10.80 and 9.70 mm, respectively. Bifidobacteria isolates were resistant to lysozyme from egg whites up to 200 µg.mL-1. B. catenulatum Bb1A isolate had the highest yield of exopolysaccharide production with 74 mg.L-1. Among them, three Bifidobacterium strains (Bb1B, Bb2A, and Bb2E) were considered potentially as probiotics.

Benthic foraminifera as ecological indicators in the Tunda Island Waters Serang District, Banten Province, Indonesia. Biodiversitas 21: 3142-3148. Coral reef communities all across Southeast Asia are experiencing intense pressures from extensive tourism and massive coastal development. A prediction regarding coral reefs’ capacity to recover is a necessity since it is constantly exposed to damaging events such as mass bleaching and increased erosion. Tunda Island (which is highly rich in biodiversity and include coral reefs, mangrove forests, seagrass, and algae meadows) was sampled at 10 different sites in August 2019. Coral reefs are highly influenced by ecological factors such as temperature variability, salinity, DO, and pH. The assessments were conducted using the FORAM Index (FI) to accurately predict the capacity of coral reefs to recover based on relative abundances of symbiont-bearing larger benthic foraminifera in reef sediments. The FI values ranged from 3.7 to 7.9 with a median of 4.7 and the average of 5 indicates that the water quality should support the recovery of reefs by reef-building corals and symbiont-bearing larger benthic foraminifers. The lowest FI value sampled was recorded from a site with extensive human activities (FI=3.4), while the highest one was recorded from a site with minimal to none human activities (FI=7.9), which indicates that the deterioration of water quality is caused by wastes from tourism activities near the coastal area.

Morphometric and DNA Barcoding of endemic Halmaheran walking shark (Hemiscyllium halmahera, Allen, 2013) in North Maluku, Indonesia. Biodiversitas 21: 3331-3343. The walking shark (Hemiscyllium halmahera) commonly referred to as bamboo shark, is a shark species that belongs to the Hemiscyllidae family which is endemic in North Maluku, and Indonesian waters. These Hemiscyllium species are generally confined to the Australian-New Guinea region, save a specie which originates from Halmahera, an Indonesian island. Ever since its discovery, sufficient information on its existence has been largely unavailable. Hence, the purpose of this study is (1) to assess the morphology, using meristic measurements and length-weight relationship, and (2) to determine the molecular characteristics through DNA barcoding, phylogenetic reconstruction, and genetic diversity. The samples used in this research were obtained from Halmahera Archipelago, North Maluku, including Halmahera, Tidore, Ternate, Maitara, and Guraici in several expeditions between March 2017 and January 2018. 36 individuals in total were measured for morphometric parameters including Total Length (TL), Standard Length (SL), Head Length (HL), Body Circumference (BC), and Body Weight (BW). Out of a total of 36 sharks, tissue samples were obtained from 32 individuals for molecular identification. After the required measurements were obtained, all individuals sharks were released back to their habitat safely. Equations of linear regression were used to analyze the relationship between length and weight. Morphometric measurements indicated that the average total length of the walking shark samples obtained, were not more than 75± cm for both male and female specimens. The length-weight analysis revealed a positive allometric growth pattern in H. halmahera species, where weight was gained faster than an increase in length. The results of the DNA fragment length analysis from all samples were 793-bp. All 32 tissue samples identified as H. halmahera consisted of 3 samples from Ternate Island, 14 samples from Maitara Island, 9 samples from Guraici Island (South Halmahera), 10 samples from Central Halmahera, and 2 samples from Tidore Island. These results confirmed that DNA subsequence analysis indicates that H. halmahera is different from those sequences extracted from Genbank in the Australia-New Guinea region. This study successfully identified H. halmahera based on their morphological and molecular characters, and has therefore contributed to international genetic database (Barcoding Of Life Database-BOLD) for further studies.

Mangrove forests are crucial to gastropods providing breeding and nursery grounds, food sources, and shelter from predators. They are useful bioindicators of environmental pollution. Consequently, it is crucial to understand the species richness of gastropods mangrove environments. This study aims to provide information about the species composition of mangrove snails in the South Coast of Pamekasan (Madura Island), East Java, Indonesia. Samplings were performed at low tide from 4 stations in December 2019 using a transect (10m x 10m). All samples were identified using reference books and published journal. We found a total of 10 family consist of Assimineidae, Cerithiidae, Ellobiidae, Littorinidae, Muricidae, Neritidae, Olividae, Planaxidae, Potamididae, and Trochidae with consist of 15 Genus, i.e., Assiminea, Clypeomorus, Cerithium, Rhinoclavis, Laemodonta, Littorarina, Tenguella, Semiricinula, Nerita, Clithon, Oliva, Planaxis, Pirenella, Telescopium, and Monodonta. The most common substrate where mangrove snails were found was rocks, sandy, and around the tree and roots of mangrove. All gastropods in this study found alive during our surveys and predominantly mangrove associated. We assume that there are still many snails that have not been surveyed at the location. We suggest to the future researcher to survey through different sampling efforts and sampling methods such as quantitative or semi-quantitative methods.

Abstract. Umiyati R, Millati R, Ariyanto T, Hidayah C. 2020. Calophyllum inophyllum extracts as a natural enhancer for improving physical properties of bioplastics and antimicrobial activity. Biodiversitas 21: 3294-3302. The utilization of Calophyllum inophyllum (or nyamplung) cake (NC) is still limited, only as fertilizer and animal feed. This research aimed to evaluate C. inophyllum (or nyamplung) cake extract (NCE) and its utilization as an antimicrobial agent in packaging material. Bioplastics were prepared by the thermo-compression method. The results showed that the NC and NCE contained tannins and amide groups. They had antimicrobial activities toward Escherichia coli and Staphylococcus aureus. Only NCE had antifungal toward Aspergillus nigger. Therefore, HPS bioplastic containing NCE exhibited antibacterial. Besides, NCE and NC contained carbonyl and amide groups. As a result, NCE addition was able to significantly increase tensile strength (?) of bioplastic, in which it was about 1.5 times higher compared to control. But NCE did not have a significant effect on elongation at break (?) and modulus of elasticity (Y). Water solubility (WS) and water vapor permeability (WVP) were reduced with an increase in NCE concentration. HPS-NCE bioplastics show a finer and more homogeneous texture compared to control bioplastic. The NCE shows the ability as a natural enhancer for improving bioplastics properties as well as function as an antibacterial agent against Escherichia coli and Staphylococcus aureus bacteria in bioplastics.

Abstract. Adam M, Ahmed AA, Yagi A, Yagi S. 2020. Ethnobotanical investigation on medicinal plants used against human ailments in Erkowit and Sinkat areas, Eastern Sudan. Biodiversitas 21: 3255-3262. The present study provides ethnobotanical data for the traditionally used medicinal plants of Erkowit and Sinkat, Eastern Sudan. The survey data were collected from 53 traditional healers through a survey carried out in February-December 2018. Fifty-eight plant species represent 34 flowering families and one Parmeliaceae lichen that are used for therapeutic purposes in the selected area. The families of highest prevalence were Euphorbiaceae (8 species) followed by Leguminosae (7 species). To validate information on the use of plants,: Use Value (UV), Fidelity Level (FL), and Informants’ Consensus Factor (ICF) were applied. The most commonly used species was Adansonia digitata with a UV of 0.89. The majority of the informants agreed on the use of Oxalis anthelmintica to treat anemia and Psidium guajava in the treatment of tuberculosis and cough, with a very high FL (92% and 91% respectively). All informants agreed on the effectiveness of Withania somnifera in treating breast inflammation and cancer, with top ICF of 1. These medicinal plants play an important role in the available healthcare provision in the study area. This preliminary study should be further continued into scientific assessment of their therapeutic use and safety through different biological activity tests and toxicological properties.

Abstract. Risna YK, Harimurti S, Wihandoyo, Widodo. 2020. Screening for Probiotic of lactic acid bacteria isolated from the digestive tract of a native Aceh duck (Anas platyrhynchos). Biodiversitas 21: 3001-3007. The lactic acid bacteria (LAB) isolated from the digestive tract of a native Aceh duck has emerged as a potential probiotic supplement for duck feed. In this study, we isolated the LAB from the digestive tract of an Aceh duck (Anas platyrhynchos) and identified the species and bacterial characteristics. Additionally, we isolated the LAB used in this study from the crop, proventriculus, ventriculus, duodenum, jejunum, ileum, and cecum of the native Aceh duck. Identification included the examination of morphology and physiology, followed by molecular identification by using 16S rRNA and sequence similarity tests for the bile salt hydrolase (bsh) gene. Bacterial isolation from the digestive tract of this duck resulted in 19 isolates with gram-positive, negative catalase, and non-motile characteristics. Of the 19 isolates, 11 isolates exhibited the ability to grow at 15°C, 37°C, and 45°C, with optimum growth at 37°C. From 11 isolates, only 6 were identified by using 16S rRNA primers and 5 were identified by using BSH primers. The identified LAB included Lactobacillus plantarum, Lactobacillus fermentum, and Pediococcus acidilactici. Hence, we conclude that Lactobacillus sp and Pediococcus sp. are potential probiotics that can be isolated and administered for duck feed.

Abstract. Tynsong H, Dkhar M, Tiwari BK. 2020. Review: Traditional ecological knowledge of tribal communities of North East India. Biodiversitas 21: 3209-3224. Traditional ecological knowledge (TEK) refers to the knowledge, innovations and practices of indigenous and tribal people relating to the understanding of structure and functioning of neighboring natural ecosystems and their use for human welfare. In this paper, we have reviewed the literature relating to TEK held by the tribal and other traditional societies of North East India. The region is very rich in TEK pertaining to species, ecosystems, and their interactions including their sustainable management and utilization in forestry, fisheries, agriculture, food, crafts, dye, and health care. Some of these TEK based knowledge systems are at par or even superior to the resource management practices evolved under the ambit of modern knowledge. Though a substantial chunk of TEK of tribal communities of North East India has been documented, huge treasures of such knowledge remain unreported and hence are on the verge of being lost.

Abstract

Abstract. Triwidodo H, Listihani. 2020. High impact of PGPR on biostatistic of Aphis craccivora (Hemiptera: Aphididae) on yardlong bean. Biodiversitas 21: 4016-4021. Plant growth-promoting rhizobacteria (PGPR) are root colonizing bacteria that live and develop near plant roots, which boost plant growth and act as antagonist agents. This research is aimed to asses the effect of PGPR on biostatistics of Aphis craccivora Koch on yardlong bean (Vigna unguiculata subsp. sesquipedalis). The research was conducted at WiSH Laboratory in Bogor, Indonesia, by tested PGPR  mixture of Rhizobium, Bacillus polymyxa, and Pseudomonas fluorescens. The research discovered that the application with seed treatment and watered of PGPR affects the biology of Aphis craccivora in terms of lengthening its stadia of second instar nymph, lengthening its life cycle, and declining its fecundity. The application of PGPR also affects the biostatistics of Aphis craccivora. PGPR applied plants have a gross reproduction rate (GRR), net reproduction rate (Ro) and intrinsic increase rate (r) of Aphis craccivora that are lower than control. PGPR could slower the doubled time (DT) of Aphis craccivora to become lengthier than control. PGPR could improve plant growth: it could increase the number and length of roots, number of root nodules, plant height, number of leaves, as well as length and density of trichome.
