Abstract. Yusup DS, Mahardika IG, Suarna IW, Giri INA. 2020. Feeding preference and growth response of early adults abalone, Haliotis squamata on some macroalgae. Biodiversitas 21: 4369-4375. Study on early adult Haliotis squamata fresh feed has evaluated some macroalgae, however, a study on Enteromorpha sp., Halymenia sp., and Hypnea sp. have not been carried out. This study was proposed to evaluate the feeding preference (feed response and feed intake) and the growth performance of early adult H. squamata on some macroalgae. Seven species of macroalgae were evaluated as unpair feeding choice and the experiment design employed was completely randomized design. The results showed that H. squamata responded variously to the seven macroalgae species and the food deprivation (fasting) data showed mounting response to all macroalgae. Ulva lactuca and Enteromorpha sp. were the most responded which up to four times higher compared to the least responded i.e. Halymenia sp. and Sargassum sp. The highest to the lowest palatable feed intake (FI) were Gracillaria sp, Eucheuma spinosum, Ulva lactuca, Halymenia sp., Enteromorpha sp., Sargassum sp. and Hypnea sp. respectively. The first four palatable macroalgae resulted in various growth responses of H. squamata. Except for Halymenia sp., the feed intake (FI) rate showed an opposite linear correlation to growth response and FCR. The daily growth rate (g/day) was Ulva lactuca (0.104), Enteropmorpha sp. (0.085), Gracillaria sp. (0.084), and Halymenia sp. (0.016).

Abstract. Gandomkar H, Shekarabi SPH, Abdolhay HA, Nazari S, Mehrjan MS. 2020. Genetic structure of the Capoeta aculeata populations inferred from microsatellite DNA loci. Biodiversitas 21: 4565-4570. The present study aimed to investigate the genetic variation of Capoeta aculeata on the basis of DNA microsatellite loci from three rivers (Beshar, Khersan, Maroun) in Kohgiluyeh and Boyer-Ahmad Province in Iran. DNA from fin clips of 120 specimens extracted and was examined with eight microsatellite markers. Genetic differences between the populations were discerned by pairwise comparison based on allelic distribution. The average number of alleles per locus ranged from 4 to 14, while the average observed heterozygosity (Ho) at various loci varied between 0.212 to 0.579, implying that a moderate level of genetic variation. Among three populations, Maroun River population displayed the highest level of variability in terms of heterozygosity. Tests of Hardy-Weinberg showed that the microsatellite loci deviated significantly in the populations. The results indicate that some of the populations were significantly differentiated from one another based on pairwise FST estimates. Genetic distance-based measures supported the clustering of Maroun, Beshar, and Khersan rivers. The neighbor-joining dendrogram topology constructed on the basis of genetic distances among populations supported observed division between the populations. The non-significant differentiation between C. aculeata samples from Beshar and Khersan can be explained by a relative disconnection of these two populations and/or small amounts of gene flow.

Abstract. Pearson MW. 2021. A pilot study into ecological burning in forests as part of a species conservation plan. Biodiversitas 22: 1296-1303. Fire management and the process of managing communities has become an increasingly topical subject due to recent events. The role of fire for fuel reduction with areas set aside for conservation may not be compatible as a seed recruitment tool for Allocasuarina robusta. Presently, fire as a recruitment tool on A. robusta is not well understood. The pilot study aims to examine the latent seedbank ability to contribute to the conservation of A. robusta when ecological burning is part of a fire management plan. The investigation examined the soil seed bank beneath A. robusta before and after an ecological burn as a greenhouse experiment. The experiment results demonstrated that a diverse range of species responds to a fire that may out-compete the seed recruitment from A. robusta. The results indicate that A. robusta may not be recruiting through seed. Competition appears to be a factor that may limit the ability of A. robusta to recruit from seed.

Short Communication: Investigating the environmental impacts of long-term monoculture of sugarcane farming in Indonesia through DPSIR framework. Biodiversitas 21: 4945-4958. An increasing trend of sugar demand in Indonesia due to the rising population has forced the government to boost its national sugarcane production through intensification program. Long-term monoculture system has long been practiced by sugarcane growers in Indonesia, particularly by large sugar companies for more than 30 years. This farming method bolsters the government’s program in scaling-up national sugar production. Through a literature study, the present study analyzed the impacts of long-term sugarcane monoculture in Indonesia on agroecosystem functions by using the Driver-Pressure-State-Impact-Response (DPSIR) framework. Results showed that long-term sugarcane monoculture leads to decreased soil quality, lowered hydrological functions, reduced agrobiodiversity, and increased greenhouse gas emissions. Those conditions corresponded to reduced sugarcane yield and productivity, increased pests and diseases, decreased income gained by growers, higher dependencies on chemicals, and higher cultivation costs. In the end, we proposed several sustainable crop management to mitigate the detrimental effects of sugarcane monoculture practice in Indonesia. These include performing crop break or rotation with legume or the other cash crop, intercropping, green harvesting and trash blanket, precision agriculture methods, and soil amendment with organic matters. However, some constraints in implementing those sustainable crop management, such as inadequate knowledge and capital, should be considered. The information given in this study can be used by sugarcane growers or companies, policymakers, and sugarcane-related stakeholders as considerations to improve sugarcane productivity while at the same time minimizing its impact on the environment.

Biodiversitas 21: 4234-4239. Rice production in Indonesia faces a potential threat of pests and diseases. Biointensive Integrated Pest Management (Bio IPM) is a new approach based on natural and biological control, which effectively controls pests and diseases. However, the impact of this technology on soil microbes and endophytic fungi is almost unknown. The objective of the study was to investigate the effect of Bio IPM on abundance of soil microbes, endophytic fungi, and blast disease under field conditions. Three pest management types were tested i.e. conventional, reduced pesticides, and Bio IPM. Field experiment was set in four replications in randomized complete block design. Experiments were carried out in two seasons i.e. dry and wet season. Variables assessed during experiment were amount of soil bacteria, soil fungi, phosphate solubilizing bacteria, colonization of total leaf endophytes, colonization of Nigrospora sp., and blast disease severity. The results showed that the abundance of soil bacteria and phosphate solubilizing bacteria were significantly increased with Bio IPM whereas it was not found to be significant in soil fungi. Total endophytic fungi and Nigrospora sp. were found higher in Bio IPM especially in early growth stages. Bio IPM controlled neck blast more effectively than conventional and reduced pesticides, but it did not show any significant effect on leaf blast disease.

The effectiveness of 60Co gamma ray exposure to the reproductive systems of rat (Rattus argentiventer) as sterile male technique. Biodiversitas 21: 3805-3810. Various strategies have been implemented to control pests, however, these do not able to reduce rat attacks on rice plants. Therefore, the sterile male technique (SMT) using 60Co gamma radiation was introduced. A group of rats (Rattus argentiventer) of 2 months age were irradiated with 1-6 Gy doses (dose rate of 99.5631 cGy/min) and the quantity, morphology, and viability of spermatozoa of rats were assessed with standard procedures. The results showed that the concentration of spermatozoa was decreased with increasing radiation doses where for lower than 4 Gy were categorized as normal (>20 million/mL). The morphology of spermatozoa also decreased with increasing radiation dose where under 3 Gy were in normal (> 50%). Spermatozoa viability was also decreased where for doses 1 and 2 Gy were categorized as normal. Statistical analysis showed a significant difference (p<0.05) between the exposed group to control in spermatozoa quantity, morphology, and viability. This finding is quite similar with other experiments on the reproductive appearance of rat based on spermatozoa after irradiation. It is concluded that dose of 3 Gy that caused changes in spermatozoa with its viability still above 50% is the most appropriate dose for SMT of rice field rats.

Local knowledge of traditional medicinal plant use and education system on their young Ammatoa Kajang tribe in South Sulawesi, Indonesia. Biodiversitas 21: 3989-4002. Kajang tribal community in Indonesia has been using plants as traditional medicine for long periods of time; however, this traditional knowledge has not been appropriately documented. Therefore, to prevent the knowledge from being degraded or vanished, it needs to be documented. This study aimed to (i) document the local knowledge of Ammatoa Kajang tribe on the use of traditional medicinal plants, (ii) describe the education system of their young generation, and (iii) conduct Informant Consensus Factor (ICF) and Use Value (UV) analyses. The research data were obtained from 114 informants through semi-structured interviews. This research uses descriptive analysis, quantitative ICF, and UV. A total of 104 plant species from 50 families were identified. Parts of plants that were frequently used by the tribal community were leaves (48.56%). Before using the medicinal plants, the people customarily boiled (33.09%) the plants. The youth received education through informal education that became the highest level of education delivered orally by the family (51.75%). The highest ICF was found in the Digestive System Diseases category, and the highest UV was observed in Piper betle.

Abstract. Maulidi A, Purnaningsih T, Maulina A, Gunawan YE, Rizki M. 2020. Short Communication: Herpetofauna diversity at University of Palangka Raya, Indonesia. Biodiversitas 21: 4509-4514. Research on herpetofauna at the University of Palangka Raya (UPR) is vital as there has been no official data so far and remains to be anecdotal evidence from various members of the university. The present study aimed to analyze and report the herpetofauna diversity on the Tunjung Nyaho Campus. It was conducted between October 2019 and November 2019. Herpetofauna data were collected using the Visual Encounter Survey (VES) method. Diurnal and nocturnal surveys were carried out between 07:00-10:00 and 19:00-22:00, respectively. The results showed that as many as 12 species of herpetofauna were present, including six species of reptiles from six families and six species of amphibians from four families. Comparative diversity analyses of these species, unburned, post-burnt, and around building, indicated that the unburned area had a relatively high diversity (2.06) compared to the other two (0.86 and 0.88, respectively). However, the evenness index (E) for the herpetofauna community in all three locations was > 0.70. The resulting species list from this study consisted of two species with threatened statuses under the IUCN Red List, i.e., Ophiophagus hannah (VU), and Cuora amboinensis (EN).

Abstract. Ho VT, Nguyen MP. 2020. An in silico approach for evaluation of rbcL and matK loci for DNA barcoding of Cucurbitaceae family. Biodiversitas 21: 3879-3885. DNA barcodes have been used intensively to discriminate different species in Cucurbitaceae family. The main of this study is to evaluate the effectiveness of rbcL and matK loci for 16 species of Cucurbitaceae family by using in silico approach. For analysis, sequences were firstly retrieved from NCBI and then calculated for sequence parameters. Sequences were then aligned and constructed phylogenetic try and examined for species resolution ability. The obtained data show the variability of resolving capacity among species. rbcL region is suitable for distinguishing five species namely S. edule, M. cochinchinensis, L. aegyptiaca, C. melo, and C. pepo, whereas matK locus is more proper for different five species consisting of M. balsamina, M. cochinchinensis, M. charantia, S. edule, and C. sativus. The resolving power is improved sharply by analyzing the rbcL + matK combination with up to nine species consisting of C. lanatus, B. hispida, C. melo, C. sativus, C. pepo, C. agryrosperma, L. aegyptiaca, S. edule, and M. cochinchinensis. Therefore, the integration of rbcL and matK loci may improve the competence of assessing genetic relatedness at species level of members in Cucurbitaceae family. The obtained information could be important for choosing proper DNA barcode loci for phylogenetic study of this crop family.

Abstract. Miftahudin, Putri RE, Chikmawati T. 2020. Vegetative morphophysiological responses of four rice cultivars to drought stress. Biodiversitas 21: 3727-3734. Each rice genotype develops certain morphophysiological responses to drought stress. The study aimed to analyze the morphophysiological responses of vegetative aspect of four rice cultivars to drought stress. A 10% Polyethylene glycol-6000 was added to a Yoshida nutrient solution medium as a drought stress stimulant for four rice cultivars, i.e., IR64, Hawara Bunar, Situbagendit, and Inpago 10. Fourteen-days-old rice seedlings were grown on the media with and without drought stress treatment for 9 days, and morphophysiological characters of vegetative aspects were observed. Drought stress inhibited the shoot growth of cv. Hawara Bunar, but increased shoot growth of cv. Inpago 10. The physiological responses in the form of leaf relative water content, proline, malondialdehyde (MDA), and total chlorophyll contents in cv. Hawara Bunar was inversely proportional to those of cv. IR64 showed an inferior response to drought stress. The rice cv. Hawara Bunar might develop better response mechanisms to drought than that of cv. IR64. The physiological responses of cvs. Situbagendit and Inpago 10 were in between the other two cultivars. We conclude that the variation of morphophysiological responses to drought stress among rice cultivars is an indicator of tolerance capability to drought that could be used as early-growth selection criteria in rice breeding programs for drought tolerance.
