Abstract

Abstract. Tapilatu RF, Wona H, Siburian RHS, Saleda ST. 2020. Heavy metals contaminants in the eggs and temperatures of nesting beaches of sea turtles in Kaimana, West Papua, Indonesia. Biodiversitas 21: 4582-4590. Etna Bay and Venu Island in Kaimana, West Papua, Indonesia are two of many areas that sea turtles use during the nesting season. Here, we report data on heavy metals contaminants from a sample of eggs of green (Chelonia mydas) and hawksbill (Eretmochelys imbricata) turtles collected from a subset of two nests during the 2016 nesting season at Venu Island, Kaimana, West Papua, Indonesia. Three heavy metals contaminants (i.e. mercury, cadmium, arsenic) found in eggs exceeded the established safety limits for human consumption. Other contaminants such as lead, zinc, manganese, iron, and copper were found below the established safety standards. There is an implication of the containment of heavy metals in sea turtle eggs to human health in Kaimana when it is consumed. It is highly recommended that the harvesting of turtle eggs and adults of all of sea turtles at Kaimana be totally prohibited. In addition, overall, beach temperatures in Lakahia fluctuated at a larger range between 26.5o and 33.8oC than at Venu island that fluctuated between 25.9o and 30.2oC. Mean daily temperatures of in-situ nests during the middle third of the incubation period were above the pivotal temperature of 29.0oC for temperature-dependent sex determination suggesting a female bias may already exist. The chemical contamination found in the eggs of C. mydas and E. imbricata at Venu Island is thought to assist sea turtle conservation initiatives in the area. The conservation program at the prime nesting habitat for green and hawksbill turtles on the small islands west of Kaimana such as Venu Island should be intensified to reverse the decline and increase population size of nesting sea turtle species. It is recommended that education and outreach be implemented in surrounding communities in Kaimana to raise awareness about marine turtle conservation.

Abstract. Faramayuda F, Mariani TS, Elfahmi, Sukrasno. 2020. Short Communication: Callus induction in purple and white-purple varieties of Orthosiphon aristatus (Blume) Miq. Biodiversitas 21: 4967-4972. Orthosiphon aristatus (Blume) Miq. are known to have many benefits, including stimulating urine expenditure (diuretics) and dissolving kidney stones. O. aristatus widely planted in Indonesia are purple and white-purple. The main secondary metabolite components of O. aristatus are sinensetin, rosmarinic acid, and eupatorin. One of the initial steps to increase secondary metabolites in O. aristatus is by induction of callus using plant tissue, which later can be developed into a culture suspension for secondary metabolites. The materials used are the leaf of two varieties of O. aristatus that have been sterilized and grown on Murashige and Skoog media with growth regulatory 2,4-dichlorophenoxyacetic acid at a concentration of 0.4;1.0; 2.0 mg/L. The identification of secondary metabolites of callus was carried out by thin-layer chromatography. The best growth regulating agent for callus induction on the leaves of purple and white-purple varieties of O. aristatus is 2,4-D 0.4 mg/L on Murashige and Skoog media. These media can grow callus at a faster time, friable, and slightly white-yellow color. The identification of secondary metabolites in callus acetone extract showed the presence of sinensetin and rosmarinic acid.

Abstract

Sicydiinae gobies have been widely collected and identified throughout tropical indo-pacific, except in Karama River, Mamuju, West Sulawesi, Indonesia. Penja was the Gobi fish’s local name, where the pelvic disc is adherent to the belly. This study aims to describe the habitat and growth patterns of genus Sicyopterus in Karama river, which passes through three sub-districts from upstream to downstream. The research was conducted in Karama River from four sites: Kalumpang, Bonehau, Arassi, and Kalonding. Fish growth was described by Deroberts and William model, and continued with habitat characteristics. The total number of fish from four sites about 71 adults Gobiidae fish, identified as family Gobiidae: Sicyopterus longifilis and Sicyopterus pugnans. The average growth pattern in all sites is isometric negative (b<3), except in the Kalumpang sites, growth pattern for Sicyopterus longifilis is isometric (b=3) whereas in Sicyopterus pugnans in the Kalumpang sites growth pattern is positive allometric (b>3).

Abstract. Ruiz MA, Acosta-López C, Soto-Vivas A. 2021. Short Communication: Morphometric variations in two populations of Limnophora marginata Stein, 1904 (Diptera, Muscidae) in Ecuador. Biodiversitas 22: 2654-2657. Limnophora includes 230 species, eleven of which are present in Ecuador. There are few studies on the bioecology of these species in the country. However, recent studies indicate that Limnophora marginata Stein, 1904 as the most abundant species in the Evergreen High Montane Forest (EHMF) located at 3400 masl in Tocachi parish, in contrast to a low abundance in the Dry Forest (DF) at 2500 masl in Malchingui parish, Pedro Moncayo Canton. This study was described the wing morphogeometric variation of L. marginata associated with these two habitats. Thirty-two wings were photographed: 16 specimens in EHMF and 16 in DF. The coordinates configurations (x,y) were registered and aligned through Generalized Procrustes Analysis. The centroid size between habitats was significantly different (Kruskal-Wallis; p?0.005); EHMF specimens were larger than DF. Reclassification a posteriori was perfect in DF specimens (100%), and almost perfect in EHMF (93.75%). The landmarks that most contributed to the variation between habitats were: Interception between M and wing border, interception dm-cu, and interception between Cu and dm-cu. The wing morphometric variations observed in L. marginata could be associated with phenotypic plasticity.

Abstract

Perception, attitude, and motive of local community towards forest conversion to plantation in Dharmasraya District, West Sumatra, Indonesia. Biodiversitas 21: 4903-4910. Forest conversion in Dharmasraya District massively occurred from 2000 to 2014. In 2000, forest area reached 86% of 33,550 ha. In 2014, forest cover reduced to only 16% with an increase in plantation area (rubber and oil palm), covering 59% of the total area. This study was aimed to examine the perception, attitude, and motive of the local community regarding forest conversion to the plantation. This study was located in PFMU (Production Forest Management Unit) Dharmasraya West Sumatra, which included a production forest area. This study was conducted from February to August 2018 with a case study approach. A total of 40 households, was selected as respondents. Snowball sampling was applied to interview the key informants. Data were analyzed using the interactive model, which included data reduction, data presentation, also conclusion drawing and verification. The study result indicated that forest is owned by the local community based upon the customary law, and the state does not have the right to manage and claim forest ownership. In terms of the economic aspect, the community benefited greatly from wood availability in the forest as the source of income. According to the local community, the conversion of forests into plantation did not have a significant effect on the environment. In fact, the local community agreed that land-use change from forest to plantation will provide greater benefit than preserving the forest. The expansion of plantation was found to be the motive for land clearance by cutting trees to obtain ownership over the forest.

Abstract. Melia S, Juliyarsi I, Kurnia YF, Pratama YE, Pratama DR. 2020. The quality of fermented goat milk produced by Pediococcus acidilactici BK01 on refrigerator temperature. Biodiversitas 21: 4591-4596. This research aimed to determine the quality of chemical and microbiological milk of goats fermented with Pediococcus acidilactici BK01 with the storage time on refrigerator temperature. The method used is the experimental method of Completely Randomized Design with five times the treatment of goat milk fermentation at the refrigerator temperature is over 0, 7, 14, 21, and 28 days with four repetitions. The results showed long-lasting storage of real effect (P < 0.05) on water content, pH, titratable acid, total lactic acid bacteria, and total plate count, but no noticeable effect (P > 0.05) on the protein levels and milk fat fermentation during the resulting storage. The value of moisture content during storage ranges between 85.88-84.92%, pH 4.48-4.28 , 3.69-3.49% fat content, 3.53-3.58% protein content, as well as the count of titratable acid ranging from 1.52-1.73%. The whole colonies of lactic acid bacteria reached between 9.106 log CFU/mL to 10.376 log CFU/mL, and the total plate count (TPC) value during storage experienced an increase from 2.653 log CFU/mL to 4.012 log CFU/mL. Based on the results of the study can be concluded that the milk of goat fermentation with P. acidilactici BK01 can maintain quality until the retention period of 28 days with the viability of lactic acid bacteria that meet the category as probiotics and still liked by the consumer.

Biochemical identification and molecular characterization of Klebsiella pneumoniae isolated from street foods and drinks in Yogyakarta, Indonesia using 16S rRNA gene. Biodiversitas 22: 5452-5458. Yogyakarta is best known as a student city in Indonesia. However, the fact that students spend almost full day in the school gave rise to the sale of a great variety of foods and drinks in its surrounding streets. Unfortunately, most street vendors do not pay attention to food hygiene. This study aims to identify the biochemical and molecular isolates of Klebsiella pneumoniae isolated from street foods and drinks in Yogyakarta using 16S rRNA gene. Data were collected from 120 samples of street foods and drinks sold at schools and several public places. The samples were enumerated using CCA medium to isolate suspected Klebsiella spp. colonies, then physiologically screened and identified using the API-20E kit. Positive isolates were isolated and amplified using the 16S rRNA gene marker for sequencing, which was then compared with the GenBank database to establish molecular identity. The results of biochemical identification obtained 11 isolates confirmed as K. pneumoniae and 1 isolate as K. oxytoca. The phylogenetic analysis showed that only 10 isolates were determined as K. pneumoniae with the reference sequences of K. pneumoniae from infected patients' respiratory and urinary tract. Furthermore, 2 isolates identified as Citrobacter sp. and Micrococcus sp., which are contaminated with Klebsiella spp. were highly found in street foods and drinks. Therefore, hygiene and sanitation need to be implemented when processing and serving these foods.

Abstract. Kusbiyanto, Bhagawati D, Nuryanto A. 2020. DNA barcoding of crustacean larvae in Segara Anakan, Cilacap, Central Java, Indonesia using cytochrome c oxidase gene. Biodiversitas 21: 4878-4887. Species-level identification of crustacean larvae is challenging due to morphological constraints. DNA barcoding offers a precise method to solve the problems. That method has never been applied to crustacean larvae from the eastern of Segara Anakan, Cilacap, Central Java, Indonesia. This study aims to identify crustacean larvae in the eastern of Segara Anakan using the cytochrome c oxidase subunit I (COI) gene as a barcode marker. Larvae morphotypes were identified under a binocular microscope. The COI gene was sequenced from one individual of each morphotype. Microscopic observation placed the samples into 15 morphotypes. DNA barcoding placed twelve morphotypes as Crustacea with sequence homologies from 72.21% to 99.21%. Intra-species genetic divergences between samples and reference species ranged between 0.9% and 31.9%, while genetic distance ranged from 0.0% to 17.80%. Intra-species genetic divergences ranged between 0.00% and 3.9%, while genetic distance ranged from 0.00% to 3.8%. The phylogenetic tree proved the monophyly between samples and reference species and showed clear separation among species. All parameters proved that nine morphotypes were identified into species level and were counted for five species. Three morphotypes were identified into the genus level and were counted for three genera. Eight species of crustacean larvae were successfully identified using the cytochrome c oxidase subunit 1 gene.

Salinity-induced changes in the morphology, physiology, and anatomy of seeds and seedlings of smooth narra (Pterocarpus indicus Willd. f. indicus). Biodiversitas 21: 5146-5154. Pterocarpus indicus Willd. forma indicus is a commonly planted species for reforestation in the Philippines regardless of the area’s condition. Since the species could survive in areas even with harsh conditions, it was hypothesized that the species may also thrive in areas having substantial amount of salt, especially during its early growth. This study assessed the morphological, physiological, and anatomical responses of seeds and seedlings of P. indicus Willd. f. indicus (Fabaceae), a native species in the Philippines to varying salt (NaCl) concentrations (0-300 mM). Based on the results, P. indicus f. indicus grew significantly in terms of height, number of leaves, leaf area, root collar diameter (RCD), root nodules, and root-shoot ratio in the control and at 100 mM NaCl. On the other hand, the growth of the species was suppressed at 300 mM NaCl. The physiological characteristics (germination, survival, and photosynthetic rates) were also high in the control compared to the NaCl treatments. As for the seed anatomical characteristics, the radicle cells remained undamaged, with high cell number and longer cells in the control, but showed damaged and reduced cell number and length at 300 mM NaCl. This indicates that P. indicus f. indicus can only tolerate moderate saline soil conditions. Hence, this result needs to be considered in reforestation efforts of the country using this species.

Abstract. Ikhsan M, Priyambodo S, Nurmansyah A, Hendarjanti H, Sahari B. 2020. Species diversity, abundance and damaged caused by rats in oil palm plantation in West and Central Sulawesi, Indonesia. Biodiversitas 21: 5632-5639. Oil palm is one of the plantation commodities which has an important role in Indonesia’s economy. Indonesia is the largest producer and exporter of oil palm in the world with its export value capable of supplying 55.78% of global consumption. Rats are wild animals that are important pests for human life, both in agriculture plantations and urban areas. Rats can damage crops at various stages of plant growth. This study was aimed to determine the species abundance of rats in oil palm plantations, calculate the severity of crop damage due to rats infestation. Three blocks of mature plants were used in OC and OH plantation blocks were used to calculated, crop damage caused by rats, Identify the species of rats, and estimated rats abundance. The results of these studies showed there were four species of rats found in the oil palm plantation, namely Rattus tiomanicus, R. tanezumi, Rattus sp, and Maxomys hellwaldii. However, only R. tiomanicus and R. tanezumi were dominant species. There were significant differences in the number of, sex, and body weight between R. tiomanicus and R. tanezumi. The OC 12 block had the lowest infestation intensity than the other blocks since it was well maintained. The OH 02 block had the highest infestation intensity, infestation area, and abundance due to a lack of maintenance.
