Arthrospira production technology in catfish waste media can be an alternative to reduce environmental pollution. However, some environmental factors such as nutrition, light, and water content can influence characterization of Arthrospira at the genetic and physiologic level. Arthrospira platensis is one of the phycocyanin-producing cyanobacteria and can be cultured using catfish culture wastewater. Water quality especially pH and salinity can effect of growth rate and residue of phycocyanin from Arthrospira platensis. This study aimed to identify the species and morphological forms of Arthrospira cultured using technical fertilizer and waste media, as well as to know the phylogenetic trees between species in this study and the GeneBank based on the 16S rRNA gene, and determine the optimum of pH and salinity required in the medium of catfish culture wastewater to phycocyanin maximum production of Arthrospira. The optimation of pH and salinity method used Completely Randomized Design (CRD) factorial with 2 factors consisting of the first factor with 3 treatments and the second factor with 4 treatments and 3 replications. The first factor was pH of culture medium i.e. pH 6.5 ± 0.2, pH 8.5 ± 0.2 and pH 10.5 ± 0.2. The second factor was salinity of culture medium, that was salinity 0 ppt (parts per thousand/‰), 10 ppt, 20 ppt, and 30 ppt. Parameters observed in Arthrospira include density, growth rate, rendement of phycocyanin, and decreased total nitrogen and phosphate content in culture media. The results showed that morphology Arthrospira cultured on technical fertilizer media (AF) had a longer and helix filament compared to Arthrospira cultured on waste media (AW) which showed several linear and shorter filaments. Both samples have a genetic distance of 0.068 (6.8%). Phylogenetic trees indicated that AF had a close relationship with Arthrospira platensis petH from Japan (bootstrap value 95%). While AW formed a separate sub-cluster of AF isolates and Arthrospira platensis petH from Japan (bootstrap value of 85%). The best treatment in this study was P2S3 (pH 8.5 ± 0.2 with salinity 20 ppt), which produced 0.867 grams maximum density, growth rate of 22.026 %.day-1 and 11.347 mg.g1 rendement of phycocyanin.

A pictorial key for the identification of beetle (Order: Coleoptera) and diversity study in selected area within Pelangai Forest Reserve, Negeri Sembilan, Malaysia. Biodiversitas 22: 947-955. Beetles or Coleoptera were largely distributed in Asia, especially in Malaysia. The beetle was important in the ecosystem as a decomposer, pollinator, and bio-indicator of the habitat. However, the beetle could also be a potential pest to crops or plants that cause damage to many plantations. This study aimed to establish a key pictorial for beetle identification based on selected characteristics and to determine the beetle diversity, evenness, and genus similarity in Pelangai Preserved Forest, Negeri Sembilan at three different sites i.e., forest, open area, and the sub forested area. This study was carried out between February and March 2020. The Shannon Evenness Index (E) calculated species evenness and the Simpson’s Index (D) assessed the diversity of beetles in this research. The results obtained were 67 individuals of beetles comprised of 7 families and 13 genera. The most found beetle was genus Apogonia sp. (16.4%), followed by genus Epilachna sp. (14.9%), and genus Adoretus sp. A (14.9%) while the least found beetle was genus Blabephorus sp. (1.5%). Site C or sub forested area had the highest species similarity (E= 0.913), followed by Site B or open area (E= 0.912) and Site A or forest area with the lowest species similarity (E= 0.908). This showed that the species in the sub forested area were evenly distributed compared to the forest and open area. As for diversity index, sub forest had the highest diversity (D= 0.860), followed by open area (D= 0.845) and forest area with the lowest diversity (D= 0.188). In conclusion, the beetle from genus Rhyncophorus sp. was the most distinct among other beetles due to the presence of snout and visible head which make it easier to identify. Beetle from genus Apogonia sp. was most found in sub forested areas due to high vegetation and medium sun exposure. Sample collection can be increased in future study by using dung trap sample collection and longer sampling time.

Abstract. Dhakal L, Aryal B, Joshi GP, Pant DR. 2020. Nutritional potential of selected species of Arisaema Mart. from Nepal. Biodiversitas 21: 5703-5709. Species of Arisaema Mart. are believed to be poisonous, but few species are used as food in different communities. Therefore, an assessment of nutritional properties of six selected taxa of Arisaema including three edible taxa was made in order to find out their nutritional potential. Methanolic or aqueous extracts of samples (rhizomes and aerial parts) were analyzed for various nutritional and anti-nutritional parameters. On nutritional basis, the total starch content and total reducing sugar were found highest (25.71±0.04mg/g powder) and (46.73±0.06 mg/g extract), respectively in aerial parts of A. echinatum. Total protein and phosphorus content was highest in rhizome of A. tortuosum var. curvatum. Similarly, extracts of aerial parts of A. tortuosum var. curvatum also possessed highest amount of total phenolic content (52.67±0.24mg GAE/g), total flavonoid content (14.13 ± 0.03 mg QE/g), and also demonstrated best antioxidant activity (IC50, 108.59±0.54 µg/mL) in terms of DPPH radical scavenging activity among the selected taxa. Inhibition of ?-amylase and ?-glucosidase in vitro was found to be highest (78.82 %) in rhizome extracts of A. echinatum and extracts of aerial parts of A. concinnum (34.82 %), respectively.

Abstract. Martiwi INA, Nugroho LH, Daryono BS, Susandarini R. 2020. Genotypic variability and relationships of Sorghum bicolor accessions from Java Island, Indonesia based on IRAP markers. Biodiversitas 21: 5637-5643. Sorghum (Sorghum bicolor (L.) Moench) is a multifunctional crop with high adaptability to various habitats and thus it is important to assess the genotypic variability of this crop species. This study aimed to reveal the genotypic variability and relationships of sorghum accessions using inter-retrotransposon amplified polymorphism (IRAP) molecular markers. Twenty-three sorghum accessions were collected from smallholder farmers in three provinces of Java Island during the 2019 planting season. The IRAP profiles generated using 10 primer combinations produced 233 scorable bands, with degree of polymorphism 75-96.9%. Cluster analysis using the neighbor-joining (NJ) method showed the formation of three groups corresponds to geographic origin of samples. The NJ dendrogram structure indicated high genotypic variability, and was supported by distribution pattern of accessions on the scatterplot constructed using principal coordinate analysis. The grouping of accessions did not correspond to classification of botanical races. Each group recognized from cluster analysis and principal coordinate analysis consisted of various races namely Bicolor, Guinea, Caudatum, and Kaffir. The fact that each group consisted of various botanical races indicated local adaptation and the maintenance of genotypic identity through farmers’ selection in traditional cultivation practices. It could be concluded, therefore, that geographical proximity and the practice of seed exchange between different regions showed considerable influence on the genotypic relationships of sorghum accessions.

Abstract. Sembiring M, Munawaroh H, Mukhlis, Hidayat B, Sabrina T. 2021. Soil macrofauna diversity in andisol after eight years of Mount Sinabung eruption in Sumatra, Indonesia. Biodiversitas 22: 3024-3030. The eruption of Mount Sinabung resulted in volcanic ash covering the soil of various thicknesses. That will affect the population and diversity of macrofauna in it. This research aimed to determine the Andisol soil macrofauna in Karo District with various thicknesses of volcanic ash covering from Mount Sinabung. This research was conducted in May 2019. Plots were placed in four locations, Location I: processed land (0 cm), Location II: Land covered by thin ash (?2 cm), Location III: Land covered by medium ash (2-5 cm), Location IV: Land covered by thick ash (?5 cm). Sampling was conducted by using the Pitfall trap, Monolith squared, and Hand sorting methods. The research results indicated that the thicker the volcanic ash covering the soil surface, it would reduce soil moisture, soil water content, organic C, and soil pH, but on the other hand, increase the soil temperature. A total of 20 species were able to live on the Andisols affected by the eruption of Mount Sinabung.

Abstract. Wijaya IDS, Defiani MR. 2021. Diversity and distribution of figs (Ficus: Moraceae) in Gianyar District, Bali, Indonesia. Biodiversitas 22: 233-246. Fig (Ficus) is an important species in tropical regions, but there is a lack of updated data on fig diversity in Bali. This study of fig diversity and distribution in Gianyar District, Bali, Indonesia was conducted to record the alpha-diversity of figs. The intensive exploration method was applied at seven locations representing the seven sub-districts of Gianyar. Habitats and coordinates were recorded, and the appropriate samples were preserved as dried or spirit specimens. The results showed that there were 23 identified species of figs from six subgenera: Ficus (1), Pharmacosycea (2), Sycidium (5), Sycomorus (5), Synoecia (2), and Urostigma (9). Based on location, three species had restricted distribution: F. fulva, F. tinctoria ssp. globosa, F. virens, and F. subcordata, whereas six species were adapted in every location: F. montana, F. hispida, F. septica, F. pumila, F. benjamina, and F. superba. The figs were primarily distributed near bodies of water, especially riverbanks.

Abstract. Fathir A, Haikal M, Wahyudi D. 2021. Ethnobotanical study of medicinal plants used for maintaining stamina in Madura ethnic, East Java, Indonesia. Biodiversitas 22: 386-392. The use of herbal medicine that expands rapidly across the world opens a lot of opportunities for drug exploration and discovery through ethnobotanical study. Therefore, countries with high level of biological and cultural diversity like Indonesia have great opportunities to achieve this goal, and even Indonesia has its own term for a group of herbal medicine, called jamu. This research aimed to conduct ethnobotanical study of jamu used by Madura ethnic in Pamekasan District, Madura Island, Indonesia by focusing on the specific use of it for maintaining stamina. Structured interviews with purposively selected respondents of 80 knowledgeable elder women were performed to gain insights about the medicinal herb used by them. Our study found 10 recipes of jamu used by Madura ethnic for maintaining stamina. These recipes contain 19 medicinal plants belonging to 16 genera and 11 families, with the family of Zingiberaceae contributed to the largest share of species used with eight species. Turmeric (Curcuma longa) was the species with the highest use-value by the respondents and the rhizomes were the most favorable plant organ used. In conclusion, the diversity of medicinal plants used by Madura ethnic could contribute to the development of new plant-based drugs, especially those for maintaining stamina.

Abstract. Niode NJ, Adji A, Rimbing J, Tulung M, Tallei TE. 2021. Composition and diversity of bacteria from giant Asian honeybee Apis dorsata gut. Biodiversitas 22: 906-912. As a social insect, honeybee possesses a unique gut bacteria community. Therefore, we need to understand the composition and diversity of bacteria in Apis dorsata gut, the largest honeybee species that live in the forest, especially in Southeast Asia. The present study aimed to investigate the gut bacteria of A. dorsata using a metabarcoding approach. The DNA barcode region used in this approach was the V3-V4 region of 16S rRNA. Honeybees were caught in a forest area located in Tareran, South Minahasa, North Sulawesi, Indonesia. There were 11 phyla identified from the gut of A. dorsata. The most abundant phyla were Firmicutes (95.8%), Proteobacteria (3.7%), and Actinobacteria (0.4%). The class Bacilli was responsible for 94.5% of the total bacterial population, the dominant family was Bacillaceae (87.2%), and the dominant genus was Bacillus (87%). Simpson (1-D) 0.24 and Shannon diversity index 0.98 indicated that the diversity of the genera was low. A nonsignificant number of species belong to the lactic acid bacteria was also detected, which may have certain benefits that need to be investigated further.

Ganoderma diversity from smallholder oil palm plantations in peatlands of Kampar District, Indonesia, based on mycelia morphology and somatic incompatibility. Biodiversitas 22: 16-22. Basal Stem Rot disease is caused by the pathogenic fungus Ganoderma boninense which has caused major economic losses in the palm oil industry. Ganoderma boninense has been reported not only infecting crops in the field, but also attacking at the immature phase of the plant even in the nursery. Studies related to Ganoderma diversity in oil palm plantations in Riau, Indonesia have not been widely reported. Ganoderma genetic diversity is important because its provide information regarding the mechanism of infection and the spatial distribution. The Ganoderma spp. isolates were collected from three blocks of smallholder oil palm plantation in Deli Makmur Village, Kampar District, Riau Province, Indonesia and brought to Plant Disease Laboratory, Faculty of Agriculture, Riau University, Pekanbaru, Indonesia. This experimental research was conducted using six Ganoderma isolates (Gan1, Gan2, Gan3, Gan4, Gan5, and Gan6) on the parameters of colony diameter and growth speed, while for the parameters of the morphological diversity of Ganoderma mycelium and the diversity based on somatic incompatibility assay was carried out descriptively. The results of this study show that oil palm plants in smallholder plantations in Kampar District are infected by two groups of Ganoderma based on morphology characteristics using UPGMA dendrogram, but different among isolates based on genetically using somatic incompatibility assay. For the next confirmation, it is necessary to further identify whether the six isolates are different species using molecular identification.

Abstract. Kalsum U, Susanto S, Junaedi A, Khumaida N, Purnawawati H. 2021. Flowering and fruit quality characteristics in some seeded and seedless pummelo cultivars. Biodiversitas 22: 379-386. Most of the pummelo cultivars with seeds, while other parts of it are seedless. Each cultivar is thought to have his characteristics. This study aimed to obtain more detailed information related to the difference in flowering characteristics and fruit quality of seeded and seedless pummelo cultivars. This experiment used a randomized block design with four treatments, i.e., seeded pummelo (Adas Duku and Bali Merah 1) and seedless pummelo (Bali Merah 2 and Jawa 1). The results showed an irregular pattern in the flowering and harvesting period in both the seeded and seedless groups. Adas Duku blooms twice whereas Bali Merah 1 and the two cultivars of the seedless group bloom only once a year. The highest total flower number from the four cultivars was Adas Duku and followed by Jawa 1>Bali Merah 1>Bali Merah 2. Seeded pummelo was not significant difference with seedless pummelo on several variables, except the total titrable acidity (TTA) content (seeded group > seedless group). Adas Duku became the best cultivar compared to the other three cultivars in several characteristics, such as the lowest peel thickness, the highest in the edible portion, the total soluble solids (TSS) and TTA of flesh fruit.
