Review: Fruit collapse and heart rot disease: Pathogen characterization, ultrastructure infections of plant and cell mechanism resistance. Biodiversitas 22: 2477-2488. Fruit collapse and bacterial heart rot are diseases in pineapple caused by Erwinia chrysanthemi (later classified as Dickeya zeae) which are increasingly prevalent in the last decade, causing devastating production loss in pineapple cultivation. Yet, comprehensive knowledge to tackle such diseases is limited, understandably due to the relatively new emerge of the diseases. Here, we review the causes of bacterial heart rot and fruit collapse, stages of infection, typical symptoms and the occurrence of resistance mechanisms in plants. In pineapple, the fruit collapse is noticeable by the release of juice and gas bubbles, also the shell of the fruit that turns into olive-green. Meanwhile, bacterial heart rot is characterized by water-soaked zones on the leaves, the formation of brown streaks on the lamina and in the mesophyll, and light-brown exudate in the blisters. The most common means of penetration into the host plant used by this type of pathogen is through plant natural openings, injuries and wounds, and entire surfaces. Concurrently, plants and fruits develop disease-resistant mechanisms to inhibit infection growth under this pathogenic attack. These mechanisms can be divided into hypersensitive reactions, locally acquired resistance, and systematic acquired resistance. In addition, pathological infections produce an interaction of the cell wall with pectolytic enzymes. Understanding the membrane breakdown process carried out by these enzymes has become critical to a pineapple protection plan. This review suggests that future research to tackle fruit collapse and bacterial heart rot can be focused on disease-resistant mechanisms, and their effects on the cell wall status with an enzymatic characterization.

Assessment of biological and agronomic diversity of seven durum wheat varieties cultivated in the Northeastern region of Algeria. Biodiversitas 22: 1025-1036. The present work was carried out during the 2017/2018 agricultural season, at the experimental station of the ‘Institut Technique des Grandes Cultures’ (ITGC) – El Khroub, Constantine, Algeria. The main objective of this study was the evaluation and the characterization of seven varieties of durum wheat (Triticum durum Desf.) in terms of their behavior and yield. For these reasons, several phenological, morpho-physiological, and biochemical traits, as well as the yield with its components were studied. The obtained results showed a significant variety effect, in particular, for the variables corresponding to the production and to the tolerance. The analysis of the correlation coefficients indicated that the number of spikes per m2, the number of grains spike-1, and the rate of chlorophyll pigments in the leaves were the most related characteristics to grain yield. Indirect selection for breeding productivity through these traits could prove to be effective. Besides, it revealed that tall varieties displayed better resistance to possible hazards; had high relative water content (RWC), accumulated more soluble sugars and proline, as well as, they showed a low level of Malondialdehyde (MDA) compared to other varieties. In addition, this study allowed us to determine many relationships between the studied parameters, that can make a contribution in both the productivity and the adaptation improvement programs of the durum wheat.

As a vector of tungro virus, Nephotettix virescens is one primary pest in rice cultivars. Associated Rice Tungro Spherical Virus provides a helper component for Rice Tungro Bacilliform Virus acquisition produced the severe symptoms in rice. The vector proteins encoded by the virus become a poorly understood transmission of plant viruses, particularly in semi-persistent mechanisms. It is essential to develop new molecules to interrupt the transmission mechanism of plant viruses by insect vectors. This study aimed to detect the helper proteins in vectors with proteomics. Protein separation was performed using SDS-PAGE, and then was identified by the Liquid Chromatography-Mass Spectrometry (LC-MS/MS) to find the candidate proteins. The result showed that actin was identified in N. virescens as responsible protein that related to transmitting tungro viruses into the plant.

Abstract. Withaningsih S, Parikesit, Nasrudin A. 2021. Correlation between landscape structure and distribution of Javan Pangolin (Manis javanica) in an extreme landscape. Biodiversitas 22: 920-932. The Javan Pangolin (Manis javanica) is a unique mammal with hard scales and can roll over when threatened. However, the study of Javan pangolin ecology, particularly using an ecological landscape approach, is limited. Here, a spatial analysis of the presence and distribution of Javan pangolins living in an extreme landscape in Rongga Sub-district, West Bandung District was conducted and was correlated to the landscape structure using a landscape metric approach. A descriptive method was used in conjunction with quantitative statistical analyses using simple linear regressions based on the Pearson correlation coefficient. The variables were features of the extreme landscape structure and the number of Javan pangolin animal signs at the sampling sites. The seven sample sites had variations in land cover classes, and the landscape structure affected the distribution of the Javan pangolins. The pangolin distribution showed a strong, negative correlation with the number of patch types (R2 = 0.628) and a weak, negative correlation with both the landscape heterogeneity (R2 = 0.012) and the percentage of forest cover (R2 = 0.136). Together, the landscape heterogeneity, the number of patch types and the percentage of forest cover negatively affected the distribution of Javan pangolins, showing a strong correlation (R2 = 0.799).

Abstract. Novita I, Oedjijono, Asnani A. 2021. The characteristics of fermented purple sweet potato (Ipomoea batatas) and black rice (Oryza sativa) using UV-irradiated Monascus purpureus. Biodiversitas 22: 684-690. This research aimed to produce Monascus fermented product (MFP) with purple sweet potato (Ipomoea batatas (L.) Lam.) and black rice (Oryza sativa L.) using UV-irradiated Monascus purpureus Went and evaluated the characteristic of its antibacterial activity against Salmonella typhi. M. purpureus was irradiated with UV at l254 nm for 0, 2, 3, and 4 min. The solid-state fermentation process was carried out for 7, 14, and 21 days. The pigments were measured at l390 nm for yellow and l500 nm for red. The ethanol extracts of MFP were analyzed for their antibacterial activity against S. typhi using the Kirby–Bauer method. The results showed that the highest yield of MFP was obtained from MFP–black rice (51.88%) that used UV-irradiated M. purpureus for 2 min and fermentation for 21 days. The highest absorbance value of MFP–purple sweet potato was obtained from UV-irradiated M. purpureus for 3 min, whereas the highest absorbance value of MFP–black rice was obtained with UV-irradiated M. purpureus for 2 min. Ethanol extracts of both MFP3–purple sweet potato and MFP2–black rice showed antibacterial activities against S. typhi with minimum inhibitory concentration values of 0.2 and 0.15 g/mL, respectively. Thin-layer chromatography analysis of the ethanol extract from MFP2–black rice revealed the presence of bioactive saponin and flavonoid. These findings suggest that UV-irradiated M. purpureus was able to use both purple sweet potato and black rice substrates to produce MFP with antibacterial activity against S. typhi.

Abstract. Kakati R, Das N, Bhuyan A, Borah D. 2021. Status of biodiversity in wetlands of Biswanath District of Assam, India. Biodiversitas 22: 453-472. Reports on biodiversity of a region are the firsthand data to understand the assemblage, importance, and to follow conservation inputs. The present study was aimed to document and analyze the wetland biodiversity of Biswanath District of Assam, India. Data was collected from 27 wetland habitats from 2015-2019. A total of 235 taxa were recorded, of which 79 taxa (77 species, one variety, and one subspecies) were vascular plants, 83 water birds, 47 fishes, 7 amphibians, 18 reptiles, and 2 mammals. None of the plants and amphibians fall under any of the threatened categories as per IUCN Red List 2020. However, among the bird species, 2 species are assessed as endangered, 4 species as vulnerable, and 10 species as near threatened. Among the fishes, 2 are vulnerable and 3 near threatened. Among the reptiles one is extinct in the wild (EW), 3 endangered and 3 are vulnerable. Along with the checklist of the present biodiversity, past and present population trends, as well as earlier records from Assam of the threatened species, is provided. The results of this study can be used by stakeholders for species identification and as the baseline taxonomic account for future studies on the conservation of these wetland species in a global context.

Architectural and physical properties of fungus comb from subterranean termite Macrotermes gilvus (Isoptera: Termitidae) mound. Biodiversitas 22: 1627-1634. Subterranean termite Macrotermes gilvus Hagen (Isoptera; Termitidae) is the most widely distributed termite species in Indonesia. This termite species has a unique habit of making fungus comb inside their nest. The fungus comb is a growth substrate for Termitomyces fungi, which provides a nutrient source for the termite. However, there is a lack of scientific information regarding the architecture and physical properties of fungus combs of M. gilvus. A study was conducted to determine the architecture and physical properties of fungus combs of M. gilvus found in Yanlappa Experimental Forest, Bogor, West Java Province, Indonesia. The fungus combs were collected from six of twenty-two nests of M. gilvus found in the rectangular sample plot (150 x 250 m) in the area. The results showed that the fungus comb of M. gilvus was brain-shaped with 44.17 ± 7.36 cm3 in volume and had burrows that interconnected from the surface (6.20 ± 1.06 mm in diameter) to the base of the fungus comb structure (4.32 ± 0.91 mm in diameter). The burrows were interconnected with each other to support cross-ventilation in the fungus comb. White nodules of Termitomyces fungi were found in the fungus comb. Architecturally, the fungus comb consisted of two structural parts, namely fresh comb on the upper part and old comb on the lower part of the fungus comb. The fresh comb possessed a larger volume (48.33 ± 2.89 cm3) and smoother texture (177.88-977.50 nm) than the old comb (40.00 ± 8.66 cm3, 407.49-6762.62 nm). The fresh comb had a larger volume (48.33 ± 2.89 cm3) than the old comb (40.00 ± 8.66 cm3). It was also found a smoother texture in the fresh comb (177.88-977.50 nm) than in the old comb (407.49-6762.62 nm). In terms of color, the fresh comb was darker (reddish) than the old comb (yellowish white). In addition, the density of the old comb was higher (0.87 ± 0.11 g/cm3) than the fresh comb (0.77 ± 0.13 g/cm3) so that the old comb was able to function as a strong foundation for fungus comb.

Climate change impacts agricultural production, including the increase of the yield loss in soybean due to pod shattering. This research aimed to identify the pod shattering resistance and agronomic performance of several soybean genotypes with various pod and seed colors. A total of 100 soybean genotypes were planted in Malang (East Java, Indonesia) during the dry season 2019. The field experiment was laid out in a randomized block design with three replications. The pod shattering resistance was assessed using the oven-dry method. The pod shattering resistance of 97 genotypes were classified into 20 very resistant genotypes, 17 resistant genotypes, 15 moderate genotypes, nine susceptible, and 36 very susceptible genotypes. The pod colors did not affect the pod shattering resistance in soybean. The new findings from this study, i.e., two very resistant genotypes (G42 and G15) which produce high yield and have medium maturity, were suggested to be developed as new cultivars, meanwhile, the other very resistant genotypes can be used in the soybean breeding program to develop shattering resistant cultivar with desirable traits.

Genetic variation of longtail tuna Thunnus tonggol landed in four fish markets in Indonesia based on mitochondrial DNA. Biodiversitas 22: 1644-1651. Longtail tuna (Thunnus tonggol, Family: Scombridae) is an economically valuable neritic species found in tropical and subtropical waters in the Indo-Pacific region. High catch numbers, which have been decreasing, could negatively impact this tuna’s population level. Little research has been conducted on the longtail tuna population in Indonesia. This study aims to determine the genetic diversity and potential population structure of longtail tuna landed in four fish markets in Indonesia (representing three sampling locations because two markets are relatively close to each other) based on sequences of a region of mitochondrial control region (d-loop). A total of 101 samples, out of 110, were identified and confirmed as T. tonggol species by amplifying and sequencing a fragment of d-loop (amplicons ranging from 482 - 523 bp). Neighbor-joining analysis resulted in a topology with all samples grouped into one clade with an average genetic distance of 0.020. Meanwhile, haplotype diversity (Hd) and nucleotide diversity (?) values of the longtail tuna samples were 0.9939 and 0.0192, respectively. The fixation index (Fst) value was -0.00507, with p> 0.05, which indicates that there is no significant population structure among the longtail tuna collected from four fish markets representing three sampling locations. The results of this analysis can be used as basic data in planning sustainable fisheries management efforts.

Abstract. Nugroho LH, Dewatisari WF, Retnaningrum E, Purwestri YA. 2021. The potency of Sansevieria trifasciata and S. cylindrica leaves extracts as an antibacterial against Pseudomonas aeruginosa. Biodiversitas 22: 408-415. Sansevieria trifasciata and Sansevieria cylindrica are the major herbs in Indonesia, which contain several bioactive compounds as potential sources of antibacterial agents. This study aims to evaluate the antibacterial activity of S. trifasciata and S. cylindrica leaves extract and fraction against Pseudomonas aeruginosa and to identify its bioactive compounds. Crude ethanolic extract of S. trifasciata and S. cylindrica leaves were tested for their antimicrobial activity by disk diffusion method against P. aeruginosa. S. trifasciata showed strong antibacterial activity with an inhibition zone of 18.3 mm compared to S. cylindrica. Different concentrations of extract i.e. 4 mg/mL, 8 mg/mL, 16 mg/mL, 32 mg/mL, 64 mg/mL, 128 mg/mL and 256 mg/mL were tested for their minimum inhibitory concentrations (MIC). The MIC results showed that the S. trifasciata extract was able to inhibit bacterial growth at a concentration of 32 mg/mL. Results of vacuum liquid chromatography (VLC) and thin-layer chromatography (TLC) revealed that only fraction 3 showed the highest antibacterial activity at 16 mg/mL. In TLC bioautography analysis, fraction 3 showed a clear zone at Rf 0.93. The phytochemical analysis showed that terpenoid, phenolic, triterpenoid, and flavonoid compounds were found in S. trifasciata extract that were associated with antibacterial activity.
