Mapping spatial-temporal skipjack tuna habitat as a reference for Fish Aggregating Devices (FADs) settings in Makassar Strait, Indonesia. Biodiversitas 22: 3637-3647. Skipjack tuna (Katsuwonus pelamis) has a high economic value in the international market. Catching skipjack tuna using fish aggregating devices (FADs) without knowing its habitat characteristics can damage the ecosystem. This study aimed to determine suitable fishing areas for setting skipjack’s FADs. The data used included that on catch, sea surface temperature (SST), and sea surface chlorophyll-a (SSC) in the Makassar Strait obtained for 2017-2019. The generalized additive model (GAM) and empirical cumulative distribution function (ECDF) analyses were used to investigate the skipjack’s tuna habitat. A pelagic habitat index (PHI), with PHI > 75%, was applied to determine suitable FAD positions. The gravity center of the skipjack tuna habitat for ten months (January-October 2020) was calculated to validate the model’s results. The results showed that the optimum SST range was from 28.78°C to 31.25°C, while the SSC from 0.18 to 0.28 mg m-3. The best skipjack habitats in the southern Makassar Strait are criterion 4 (PHI > 90%) and criterion 3 (PHI = 85-90%), having a relatively high consistency of the average PHI values. These results can help determine the optimal positions for setting FADs to benefit the global management and sustainable development of skipjack tuna fisheries.

Effect of three composts with active ingredients of Pseudomonas fluorescens on the development of white root disease and production of rubber plants. Biodiversitas 22: 3237-3242. White root disease (WRD) caused by Rigidoporus lignosus is a very dangerous disease and a scourge for rubber farmers because it can result in decreased production and kill rubber plants. This research was conducted to observe the impact of compost enriched with the biological agent Pseudomonas fluorescens on the development of white root disease and production in rubber plants. The results showed that the application of compost with active ingredient of P. fluorescens isolates A and B reduced the severity of white root disease in plants with mild, moderate and severe infections, 34.12%, 29.31% and 57.21% respectively. Application of compost with P. fluorescens isolates A and B, either singly or in combination, can increase latex production. The treatment of giving compost enriched with P. fluorescens isolates AR and ABR on rubber plants infected with mild WRD resulted in the highest latex production of 406 gm and 402.74 gm per plant, respectively. These two treatments did not differ from each other but were significantly different from the other treatments and controls.

Abstract

Postharvest losses are a significant concern for tomato breeding associated with their short fruit shelf life. Sletr1-2 is a new ethylene receptor mutant that has a prominent character in a prolonged fruit shelf life. This research aimed to estimate the combining ability of Sletr1-2 mutant and determine the selection method for future breeding associated with the fruit shelf-life and yield. Four lines of tropical tomato, i.e., 'Intan,' 'Mirah,' 'Ratna' and 'Mutiara,' were crossed with the wild type Micro-Tom (WT-MT) and Sletr1-2 mutant tomato using a line x tester mating design. A randomized complete block design with four replications was used to evaluate twelve F1 and their parents. The study revealed significant differences in the GCA of the line and tester but not for SCA. The lines and testers contributed more to total variance than their interaction. 'Intan' and Sletr1-2 mutants had the greatest fruit shelf-life combiners, with additive gene action being the most prevalent. Simple phenotypic selection or pure line selection from selected crosses in advanced generations would be preferable. 'Mutiara' was the best combiner for yield and plant growth, with non-additive gene action was the most common. The breeding strategy that considered dominance, overdominance, and epistasis was preferred.

Short Communication: First record of the cassava lace bug Vatiga illudens (Drake, 1922) (Hemiptera: Heteroptera: Tingidae) from East Java, Indonesia. Biodiversitas 22: 2870-2876. The cassava lace bug, Vatiga illudens (Drake, 1922) (Hemiptera: Heteroptera: Tingidae) is recorded from Indonesia for the first time. This species is found on the abaxial surface of leaves of cassava (Manihot esculenta Crantz, Euphorbiaceae) in several plantations of East Java and heavily infest this plant. Vatiga illudens is native to the Neotropical Region and seems to be the alien species in the Oriental Region. This tingid has already spread over a wide region of East Java, suggesting that the invasion of the lace bug endangers the other regions cultivating cassava.

Species distribution of Enterobacteriaceae and non-Enterobacteriaceae responsible for urinary tract infections at the Zainoel Abidin Hospital in Banda Aceh, Indonesia. Biodiversitas 22: 3313-3318. Urinary tract infection (UTI) is an infection that occurs along the urinary tract caused by pathogenic bacteria. Enterobacteriaceae and non-Enterobacteriaceae are generally normal flora originating from the large intestine and vaginal mucosa. This study aimed to assess the distribution of Enterobacteriaceae and non -Enterobacteriaceae pathogens that cause UTI based on species diversity, patient age, gender, and hospital units, and to evaluate their antibiotic sensitivity to uropathogens in ZAH. In total, there were 284 isolates classified as UTI pathogens with 178 Enterobacteriaceae isolates dominated by Escherichia coli and 107 non- Enterobacteriaceae isolates. Non- Enterobacteriaceae consisted of 77 isolates of Gram-positive dominated by Enterococcus faecalis and Enterococcus facium, and 29 isolates of Gram-negative dominated by Acinetobacter baumannii. The highest UTI ratio was found in the elderly (56-65 years), namely 66.2% Enterobacteriaceae and 33.8% non-Enterobacteriaceae isolates. Based on gender, females were infected with Enterobacteriaceae (66.86%) and non-Enterobacteriaceae (37.41%). Most of UTI patients in the internal medicine unit were infected with Enterobacteriaceae (55.36%) and non-Enterobacteriaceae (44.64%). Amikacin, meropenem, and levofloxacin were antibiotics that had high sensitivity against Enterobacterial and non-Enterobacterial uropathogens. The emergence of Gram-positive uropathogens need to be concerned as these groups start emerging. It is also important to monitor regularly the antibiotic susceptibility of bacterial UTI pathogens to ensure the efficacy of current UTI treatments and minimize the UTI incidence in nosocomial settings.

Abstract. Nasrika E, Retnonongsih A. 2021. Inter Simple Sequence Repeat molecular markers to reveal the genetic diversity of superior durian of Gunungpati, Semarang, Indonesia. Biodiversitas 22: 4054-4059. Kalimantan is the center of many tropical fruits, including durian. One of the edible durians and favorited by many is Durio zibethinus, which has many superior varieties. Almost all regions in Indonesia have superior durian varieties; for example, superior durian Gunungpati from Gunungpati Sub-district, the center for producing durian in Semarang City, Central Java, Indonesia. ISSR analysis was carried out to reveal the genetic diversity of the superior durian Gunungpati. DNA isolation of 16 superior durians used the modified CTAB method. The genomic DNA was amplified using 10 ISSR primers and then electrophoresed using 2% agarose gel. Data were analyzed using NTSYS PC version 2.02. The resulting allele has a relative size of 220 bp-1800 bp, with 87.9% are polymorphic. The similarity coefficient of 16 varieties was ranged between 0.54-0.88. All the superior durian varieties examined were different accessions so that each variety has the potential to be registered as a new variety of superior Indonesian durian. Specific alleles are found in G1, G3, G7, G8, and G13, which can be an identity of these varieties.

Abstract. Maser WH, Sinaga MZE. 2021. Evaluation of proximate composition, microbial load and sensory characteristics of instant holat as traditional spice in North Sumatra. Biodiversitas 22: 3440-3445. Holat is the name of traditional food from North Sumatra, Indonesia which has astringent flavor derived from the bark of Phyllanthus emblica. The aims of this study were to determine the proximate composition and evaluate microbial load, and sensory characteristics of instant holat spice. There were 12 treatments, namely the combination of different amounts of bark (15 g, 25 g, 35 g, 45 g) and drying time (20 h, 22 h, 24 h). The proximate composition (moisture, ash, protein, fat, crude fiber, fat, and total carbohydrate content), microbial analysis (total plate count; yeast and mold count), and sensory characteristics (aroma, color, taste, and overall acceptability) were carried out. The results showed that water, protein, fat, crude fiber, and total carbohydrates contents were all significantly different (p < 0.05), except ash content. There was no yeast, mold, and bacterial growth on total plate count. There were no significant differences in all sensory characteristics (aroma, color, taste, and overall acceptability) of instant holat before and after brewing (p ? 0.05). The treatment of F4-24 was the best formula and has the highest score that might be developed as a product of instant holat spice. Further study should be conducted to determine the effect by increasing the amount of bark and evaluating its bioactivity.

Abstract. Pebriani MA, Barus TA, Ilyas S. 2021. Fish diversity and heavy metal accumulation of Pb, Cu and Zn after Mount Sinabung Eruption in Benuken River, North Sumatra, Indonesia. Biodiversitas 23: 187-194. The Benuken River is a river that has been affected by the volcanic ash eruption of Mount Sinabung, one of the active volcanoes in Indonesia. This study further examines the diversity of fish species and the analysis of their tissues with regard to the heavy metal content of each species in the Benuken River, Berastepu Village, Karo Regency after the eruption of Mount Sinabung using atomic absorption spectroscopy (AAS). The fish and water sampling were carried out in March 2019 with two sampling points using the electrofishing technique. Furthermore, four species were documented, including Spotted barb (Puntius binotatus), River loaches (Homaloptera nebulosa), Mahseer (Tor soro), and Striped snakehead (Channa striata). The diversity index of the fish community from two study sites ranged between 0.012<H' <0.023 or was categorized as very low in diversity. Meanwhile, the heavy metals concentrations of Cu, Pb and Zn in the water of Benuken River were below tolerance level based on the Governmental Regulations No. 82 year 2001. The average heavy metal concentrations in fish tissue were also safe for consumption based on the standard quality by FAO/WHO (2004) with 0.0049 ppm, 0.0056 ppm and 0.3852 ppm of Pb, Cu and Zn, respectively.

Abstract. Samanhudi, Sakya AT, Setyawati A, Muawanah M. 2022. Agarwood (Aquilaria malaccensis) diversity conservation by in vitro culture with IAA and yeast extract. Biodiversitas 23: 2457-2463. Agarwood (Aquilaria malaccensis) is a scare plant with high economic value and many benefits. Tissue culture is an effective way of producing plants in large quantities, in a short time with uniform results. This study aims to get the proper concentration of IAA and yeast extract for agarwood growth in vitro. The research was conducted in December 2019-June 2020 at the Laboratory of Plant Physiology and Plant Biotechnology, Faculty of Agriculture, Universitas Sebelas Maret, Surakarta. The design used is Randomized Complete Block Design (RCBD) in 2 factors: first factor was IAA concentration with 4 level: 0; 0,5; 1; 1.5 mgL-1 and second factor was yeast extract concentration with 4 levels: 0; 200; 400; 800 mgL-1. Obtained 16 combinations of treatments repeated 3 times so that there are 48 units of experiments. Observed variables: the time of appearance of shoots, the number of shoots, the height of shoots, the number of leaves, and the number of roots. Root count data is described and data other than root count is analyzed ANOVA, if real influence is further tested DMRT 5%. The IAA affects the height of shoots and the number of agarwood leaves in vitro rooted plantlets has 5 plantlets.

Abstract. Naive MAK, Demayo CG, Alejandro GJD. 2021. Short Communication: New insights into the morphology and distribution of the Philippine endemic Etlingera pilosa (Zingiberaceae). Biodiversitas 22: 3175-3179. Etlingera pilosa Poulsen & Docot, only known in Negros Oriental and Negros Occidental of Visayas islands, Philippines, is recorded in the island of Mindanao for the first time, thus, extending its distributional range. The species is characterized by having sericeous ligule; leaves subsessile, narrowly obovate, coriaceous, glabrous adaxially, densely pubescent abaxially; inflorescence up to 12 cm long; flowers yellow, 3.0–3.2 cm long; subglobose to obovoid, pilose, with wart like blunt spines fruit; and seeds irregularly subglobose to oblate, black, fully embedded in juicy sour-sweet white aril. This species closely resembles E. pubimarginata but differs in having densely pubescent abaxial leaf, sericeous ligule and subglobose to obovoid, pilose, with wart like blunt spines fruit. Here, we provide a detailed description, comparison with its allied species, colored photographs to aid identification, a distribution map, an IUCN conservation assessment and a taxonomic key to the Mindanao Etlingera species.
