Biodiversitas 23: 1635-1647. Human disturbance in the form of sand mining and grass harvesting activities in the area of Mount Merapi is an important component of the ecological process alongside the eruption. The group of terrestrial mammals (TM) is one of the essential elements that are affected by this disturbance. Therefore, the objectives of this study were to determine the response of the TM in various types of habitats in areas affected by eruption disturbances >20 years (ERL), eruption < 20 years (ERE), mining (MIN), grazing (GRA), and non-disturbance areas (NDA) in Gunung Merapi National Park (GMNP). Camera traps were used to identify the abundance of each TM. Vegetation data were also used to represent habitat conditions and were taken on a plot of 0.04 Ha. The analysis results showed that the highest abundance of TM is at the NDA site (14.4), while the lowest is at the MIN site (2.08). The site with the highest level of TM diversity is ERL (1.97) and the lowest is NDA (1.52). Furthermore, there is a significant difference in TM abundance between the MIN and ERL sites, which indicates that each disturbance reacts differently to the site. Based on the results, mining disturbances negatively influenced the abundance of TM in GMNP, therefore, managers need to regulate mining activities and make efforts to restore the affected areas.

Biodiversitas 23: 2405-2410. Shallot (Allium ascalonicum L.) is one of the leading horticultural and spice commodities in Indonesia. Assembling shallot varieties to get a good seed quality requires a high genetic diversity which can be analyzed using molecular markers. This study aims to identify the genetic diversity of shallots from several locations in North Sumatra, Indonesia based on Random Amplified Polymorphic DNA (RAPD) markers. This research was conducted at the Biotechnology Laboratory, Faculty of Agriculture, University of Sumatera Utara, Medan from September to October 2021. A total of 11 shallot varieties from several locations in North Sumatra were genetically analyzed based on RAPD markers with OPA-13, OPB-07, OPD-20, OPM-01 primers. The results showed that 11 shallot varieties originating from several locations in North Sumatra had a high genetic diversity with the presence of two main groups at a dissimilarity coefficient of 76%. These beneficial results can be used as a complement to morphological markers in the genetic study of shallots for breeders to decide what genotypes will be crossed to make new genetic combinations in the development of shallots. This study reports the success of the shallot varieties’ fingerprints using RAPD markers.

Short Communication: Metabolites of Albizia inhibit in vitro growth of phosphate solubilizing microbial consortia isolated from tea garden soil of Darjeeling hills, India. Biodiversitas 23: 2865-2870. Phosphate solubilizing microorganisms (PSM) play a crucial role in maintaining the nutritional status and fertility of the soil. PSMs help in solubilizing the metallic phosphate salts of soil into available phosphate anions for easy uptake by plants. However, this beneficial group of microorganisms also face different challenges for survival from its immediate surroundings. This study was carried out to assess the effect of shade tree litters on the PSM consortia isolated from different tea garden soils of Darjeeling hills, since shade trees are an integral part of tea plantations. Albizia odoratissima, Albizia chinensis and Albizia procera, widely used in tea gardens as shade trees were selected. GC-MS analysis was carried out to detect the metabolites produced by the litters. Twenty-three compounds exhibiting antimicrobial activities were detected. Major peak was found in A. odoratissima, followed by A. chinensisand least in A. procera. Compounds like 1-heptanol, 2-propyl-, neophytadiene, phytol and squalene were common in all three extracts and are considered to provided antimicrobial activity to Albizia. A proportional relation has been observed between magnitude of inhibition zones and peak area percentage in Albizia spp. PSM isolates from shade tree gardens were observed to be more tolerant toward the leaf extracts.

Determination of free radical scavenging activity, phenolic and flavonoid content of seven corn cultivars from the Southwest Maluku District, Indonesia. Biodiversitas 23: 5974-5981. The variation of corn (Zea mays L.) kernel might be considered as a surrogate indicator of the phytochemical and bioactive content. This study aimed to determine the phenolics, flavonoids, and free radical scavenging activity of seven local corn cultivars from Kisar Island, Southwest Maluku District in Indonesia. Sample extraction was done by the maceration method. Flavonoid content was determined using the Dowd method, and total phenolic content was performed using the Folin-Ciocalteu method. Free radical scavenging activity was determined using the DPPH radical scavenging method. One-way ANOVA was performed with Duncan multiple range tests to assess the differences between corn seed cultivars, and p < 0.05 were regarded as statistically significant. The results show that the total flavonoid content varied from 8.463 mg QE/mg (Putih cultivar) to 40.075 mg QE/mg (Kuning Dalam cultivar). Total polyphenol content also varied between 105.63 mg GAE/mg (Merah Delima Tongkol Putih cultivar) and 136.41 mg GAE/mg (Pulut cultivar). The antioxidant activity by DPPH assay determined that the seven local corn cultivars had IC50 values ??that were similar to quercetin (used as control), some were even lower. The IC50 value of the seven local corn cultivars ranged from 5.56 to 16.96 µg/mL, while quercetin had an IC50 value of 11.97 µg/mL. It was observed that the Merah Delima Tongkol Cokelat, Merah Delima Tongkol Putih, and Putih cultivars had comparable antioxidant activity to the control quercetin. The Merah Darah, Kuning Genjah, and Kuning Dalam cultivars exhibited a statistically significant higher antioxidant capacity than quercetin. These results can contribute to the selection of corn cultivars to harvest for commercial purposes and potential nutraceutical applications.

Short Communication: Genetic variation of oceanic manta ray (Mobula birostris) based on mtDNA data in the Savu Sea, Indonesia. Biodiversitas 23: 1700-1706. The Savu Sea, one of Indonesia's top conservation priorities, is home to various marine charismatic species, including the oceanic manta ray (Mobula birostris), whose conservation status is currently endangered and is protected by the Indonesian government. However, due to domestic and global demand for its fishery products, as well as shortcomings in fisheries management, this species is still poached and bycaught in the Savu Sea. Understanding their population structure is important to achieve effective conservation and fisheries management strategies that will have a positive impact on preserving their population in this area. This study aims to reveal the genetic variation of oceanic manta rays in the Savu Sea. Thirty samples from three locations in the Savu Sea were successfully preserved from East Flores (24), West Manggarai (4), and Rote Ndao (2) and then analyzed using ND5 locus from Mithocondiral DNA (mtDNA). The result indicated a close genetic relationship between three locations (East Flores, West Manggarai, and Rote Ndao) based on the phylogenetic tree and Analysis of Molecular Variance (AMOVA) result with value of 0.05158 (P-value = 0.62268) indicated as a single population. In conclusion, the findings of this study provide some insight into the possibility of manta ray populations in the Savu Sea having strong connectivity between areas, which is critical information for regulators and managers to integrate conservation and management strategies within the Savu Sea.

Expression of heterosis, heterobeltiosis, and gene action in quantitative characters on soybean (Glycine max). Biodiversitas 23: 1745-1751. Most soybean (Glycine max (L.) Merrill) varieties have been created by crossing two or more parental genotypes. The study aimed to quantify the value of heterosis, heterobeltiosis, the degree of dominance as well as the gene action of the agronomic characters of F1 plants derived from the four soybean cross combinations. All the parents and crosses were evaluated in the screen house of the Indonesian Legume and Tuber Crops Research Institute (Malang, East Java, Indonesia) during 2021. The estimation for mid-parent heterosis (hMP), high-parent heterosis or heterobeltiosis (hHP), degree of dominance, and gene action was done for sevenquantitative characters (plant height, number of branches, number of nodes, number of filled pods, number of empty pods, weight of 10 seeds, and seed yield). The performance of the quantitative characters differs among cross combinations. The estimated value of heterosis mid-parent (hMP) for F1 crosses from four parental combinations ranged from -61.20 to 27.57, heterobeltiosis (hHP) ranged from -71.17 to 9.84, and the degree of dominance ranged from -6.45 to 19.40. The crosses of Derap 1 × Gepak Kuning and Dering 1 × Gepak Kuning showed a positive hMP and hHP for seed yield character. Thus, these hybrids could be used to develop high-yielding varieties with desirable traits in soybean. Seed yield character in the cross of Derap 1 × Gepak Kuning exhibited the overdominance type of gene action, meanwhile, the Dering 1 × Gepak Kuning was incomplete dominance.

Abstract. Utaminingrum W, Nofrianti, Hartanti D. 2022. Diversity and use of medicinal plants for traditional women's health care in Northern Banyumas. Biodiversitas 23: 1970-1976. This study aimed to record the diversity and utilization of medicinal plants for maintaining women's health in Northern Banyumas (Central Java, Indonesia) using qualitative and quantitative approaches. Semi-structured interviews with 97 informants were conducted to collect data on local plant names, plant parts, herbal preparation, and application of plants used for traditional women's health care. The value and importance of the plants to the people in the surveyed areas were quantitatively determined by the species use-value (SUV), the relative frequency of citation (RFC), and fidelity level (FL). The local community cited 21 plant species belonging to 10 families. The most frequently used plant part, preparation, and application were fruits and rhizomes (32.6% each), infusion (83.7%), and oral (88.4%), respectively. There were four use-category of the plants, i.e., post-partum, pre-menstrual syndrome, lactation stimulant, and leukorrhoea. The most valuable plants were Curcuma longa L., Piper betle L., and Kaempferia galanga L. with SUV of 0.41, 0.34, and 0.21, respectively. The C. longa and P. betle were also recorded as the most important plants with RFC values of 0.63 and 0.40. These findings confirm the status of Zingiberaceae as the main component of jamu (traditional medicine) to maintain women's health.

Abstract. Watuwaya BK, Syamsu JA, Budiman, Useng D. 2022. Forage productivity in native grasslands of Haharu Sub-district, East Sumba District, Indonesia. Biodiversitas 23: 362-1368. The identification and management of native grasslands are important to ensure the availability of beef cattle feed in smallholder farms. This study aims to identify the existing native grasslands in Haharu Sub-district, East Sumba District, East Nusa Tenggara Province, Indonesia and analyze the botanical composition, biomass and the carrying capacity of the native grasslands. We used the combination of remote sensing approach and field survey to identify and measure the productivity of native grasslands. Sentinel 2A imagery was used to identify the area of existing native grasslands using Supervised Classification - Maximum Likelihood method. Meanwhile, the productivity of native grasslands was measured using the Dry Weight Rank method. The results of remote sensing and spatial analysis showed that the existing area of native grasslands in Haharu Sub-district was 324,10 km2 with an Overall Accuracy of 98.63% and Kappa Accuracy of 0.98. The vegetation analysis showed that the botanical composition of native grasslands consisted of grasses 94.42%, legumes 3.55% and weeds 2.07%. The carrying capacity of the studied grassland was 0.68 AU/ha/month in the wet season. In conclusion, the productivity of existing native grasslands in the wet season is still low and special efforts are needed to improve the quality.

Sumatran tiger identification and phylogenetic analysis based on the CO1 gene: molecular forensic application. Biodiversitas 23: 1788-1794. Wild animal hunting, especially in the Sumatran tiger (Panthera tigris sumatrae), has been caused the population decline. Regulation and law enforcement have been implemented even though it does not affect optimal because of the trickery of poachers and illegal traders. Sometimes, the evidence of P. t. sumatrae derivative products, for example, bones, nails, skins, hair, and other body parts, cannot be properly identified to raise the cases. However, genetic markers, such as the CO1 gene, have successfully identified illegal trafficking samples. This study used 20 samples, consisting of seven samples (four preserved hairs, two claws, one bone) that were suspected of P. t. sumatrae collected from illegal wildlife trade cases in West Sumatra, Indonesia. Other thirteen samples were twelve blood and one hair of P. t. sumatrae samples were collected from the Dharmasraya Sumatran Tiger Rehabilitation Center (PR-HSD). All samples were isolated, Polymerase Chain Reaction (PCR), sequenced, and 999 base pairs (bp) of the CO1 gene sequences were analyzed. In addition, National Center for Biotechnology Information (NCBI) data sequences including two P. t. sumatrae sequences, four P. t. altaica sequences, one P. t. amoyensis sequence, one P. t. corbetti sequence, three P. pardus sequences, and one Felis catus sequence were collected for comparison and supporting data. The result confirmed that all samples in this study were P. t. sumatrae. We determined those depending on similarity value which was 99.60%-99.70% with P. tigris reference sequence (NC_010642.1) and 99.90%-100% with P. t. sumatrae (JF357969.1). Phylogenetic analysis supported species identification with average intraspecies sequence divergence was 0 to 0.4% and presented the monophyletic group. This study was the first and most recent report to use seized samples to identify P. t. sumatrae based on the CO1 gene in West Sumatra, Indonesia.

Abstract. Suwardi AB, Navia ZI, Harmawan T, Seprianto, Syamsuardi, Mukhtar E. 2022. Diversity of wild edible fruit plant species and their threatened status in the Aceh Province, Indonesia. Biodiversitas 23: 1311-1319. Aceh Province, Indonesia is home to a diverse range of tropical fruit plants. However, increased anthropogenic activities such as population growth and natural disasters resulted in various biological resources such as wild edible fruit plants being under threat. The study aimed to assess the diversity and threatened status of wild edible fruit plant species in the Aceh Province of Indonesia. The study was conducted in six regencies, namely Aceh Tamiang, Aceh Timur, Aceh Selatan, Aceh Utara, Aceh Barat, and Aceh Tengah. At each study area, a line transect of 1,000 meters was laid from the forest's edge into the forest. Wild edible fruit plant species discovered along the transect were collected and given local names. A total of 129 wild edible fruit plant species at six studied areas in the Aceh Province belonged to 38 families. The highest number of fruit plant species was recorded in the district of Aceh Tamiang, i.e., 63 species, followed by Aceh Selatan (56 species), Aceh Barat (46 species), Aceh Timur (37 species), Aceh Utara (31 species), and Aceh Tengah (23 species). Mangifera foetida Lour., Mangifera odorata Griff., Artocarpus integer (Thunb.) Merr., Ficus altissima Blume, Ficus fistulosa Reinw. ex Blume, Syzygium cumini (L.) Skeels, Passiflora foetida L., and Pometia pinnata J.R. Forst. & G. Forst. were the most frequently recorded wild fruit plant species in all regencies. From 129 wild edible fruit species recorded in the study areas, 35% are currently classified as Least Concern, 6% as Vulnerable, 3% as Near Threatened, 2% as Low Risk, 2% as Data Deficient, and 52% of species in the IUCN red list had No Available Data. Community-based conservation through domestication of wild edible fruit plant species can be one of the management strategies for the conservation of wild edible fruit plants in Aceh Province.
