Abstract

Bufonidae is widely distributed amphibian family around the world, including Indonesia (Sumatra) and Asia. Sumatra is an island which has separated because of sea level increase and create gene flows barrier for amphibian in Sumatra and Asia. The purpose of this research is to analyse Bufonidae phylogenetic which exist in several locations in Sumatra and Asia based on gene cytochrome b analysis. Samples were collected from six localities in West Sumatra. DNA extraction and amplification has been done in Genetics and Cell Biology Laboratory, Andalas University, whereas DNA sequencing were analyzed at MacroGen USA DNA Sequencing Laboratory, South Korea. About 14 tissue samples of Bufonidae from West Sumatra has been analyzed. The relationship of Bufonidae phylogenetics in Sumatra and Asia were divided into three main clusters. Duttaphrynus melanostictus in Sumatra is closer to Duttaphrynus melanostictus from India with 0,3-0,5% sequence divergence and it is a group of paraphyletic with Duttaphrynus melanostictus from Vietnam, Taiwan, and China.

Abstract

The true frog species, Odorrana hosii, found in West Sumatera Indonesia, has high morphological differentiation and also estimated has high genetic variation. A total of 35 O. hosii at a seven location have analyzed using DNA microsatellite markers. Genetic variation of five microsatellite loci provided the highest value of expected heterozygosity (He) for the population in Padang (He= 0.618), while the lowest was the population in Merapi (He= 0.427). There are genetic differences in moderate levels among populations of O. hosii in West Sumatra (FST= 0.108) with inbreeding intrapopulation value (FIS= -0.559), and high value of gene flow among the populations (Nm = 2.061). This study becomes the first molecular data for establishing effective population management conservation.

Abstract

Ploidization of Mystacoleucus padangensis Bleeker, 1852 (Pisces: Cyprinid) with heat shock induction has been carried out from January to September 2014 at Genetics and Cytology Laboratory, Biology Department of Faculty of Mathematics and Natural Sciences, Andalas University. This study aimed to find suitable temperature for ploidizatiom on M. padangensis. The study used Completely Randomized Design method with five treatments and five replications. The treatments were various water temperatures (i.e. 27oC as control, 36oC, 38oC, 40oC, and 42oC). Abstract

Species Inventory of the Cyprinid fishes at Batang Nareh River, Padang Pariaman using descriptive method was conducted from January to March 2013. Seven species of Cyprinid that belonging to five genera and 253 individuals were collected from three stasiun at Batang Nareh River. The species were Tor tambra CV, T. tambroides Blkr, Rasbora reticulata M. web & de Bfrt, R. laterastriata Blkr, Lobocheilos falcifer CV, Osteochilus hasseltii Blkr, and Puntius binotatus CV. Tor tambra and T. tambroides were reported as high economic value species for local people.Keywords: Inventory, Cyprinid, Batang Nareh River.

Abstract

The morphological analysis of Puntius binotatus Valenciennes 1842 (Pisces: Cyprinidae) collected from several locations in West Sumatra has been conducted from April to June 2012. Several morphological characters were measured and analyzed using principal component analysis (PCA) and UPGMA. This study found significant morphological variations of the standard length (SL), head length (HL), least depth of caudal peduncle (HCPD), length of caudal peduncle (LCPD), eye diameter (ED), caudal fin length (CFL), caudal fin length up (CFL up), caudal fin length bottom (CFL bottom), length of maxillary barbels (LMB) and length of snout barbels (LSB). Those variations were more likely influenced by elevations and western-eastern side of Barisan Mountain in Sumatra.Keywords: Barisan Mountain, cyprinidae, morphological characters, P. binotatus

Abstract

Chromosomal characteristics of catfish Hemibagrus lacustrinus, known as Baung fish, a new species of bagrid catfish from the upriver stream of Batang Sianok, West Sumatra, that recently described by Ng and Kottelat, 2013 was examined from the gills of 5 healthy specimens using general squash method. The result showed that diploid chromosome number of this species was 2n=56. Thekaryotype consisted of 28 metacentric (m), 10 submetacentric (sm), 8 subtelocentric (st) and 10 acrocentric (a) chromosomes. The chromosome formula can be represented as 2n= 28m+ 10sm+ 8st+ 10a. The largest chromosome in this species is a pair of submetacentric chromosomes and the smallest pair is acrocentric chromosomes. The present study is the first report on the karyology of this particular species and helping to support the karyotype data of the genus Hemibagrus. This study will also open for several future studies to explore this species.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi serangga penyerbuk pada tanaman kopi (Coffea arabica L.). Penelitian ini dilakukan dari bulan April hingga September 2016 di Kabupaten Dairi. Sampling dilakukan pada enam lokasi yang tersebar pada tiga Kecamatan yaitu Sumbul, Parbuluan, dan Siempat Nempu Hulu. Masing-masing lokasi terdiri dari lima plot, setiap plot berukuran 7 x 7 meter. Sampling dilakukan ketika hari cerah pada pukul 07.00 hingga 12.00 WIB. Serangga yang berhasil ditangkap dimasukkan ke dalam killing bottle yang mengandung etil asetat. Selanjutnya dimasukkan ke dalam kertas papilot dan disimpan ke dalam box plastik untuk kemudian diidentifikasi. Identifikasi dilakukan di Laboratorium Biosistematika Jurusan Biologi Universitas Negeri Medan. Berdasarkan hasil identifikasi diperoleh sebanyak lima belas spesies yang termasuk dalam tiga Ordo, yaitu Lepidoptera Diptera, dan Hymenoptera serta tujuh famili. Lebah madu Apis cerana merupakan spesies yang paling banyak ditemukan. Kata kunci:  Serangga penyerbuk, Kopi, Apis cerana, Diptera, Hymenoptera, Lepidotera

Abstract

Research on the genetic variation of Anguilla spp. in the waters of the West Coast of West Sumatra was carried out from February to June 2022. This study used the RAPD (random amplified polymorphic DNA) method. In this study, two Anguilla species were found, namely A. marmorata and A. bicolor bicolor. The highest heterozygosity value of A. marmorata in the Tarusan River was 0,2018, and A. bicolor bicolor in Mentawai was 0,2340. The values of gene flow (Nm) in A. marmorata and A. bicolor bicolor were 0.7592 and 1.6318. The values of genetic differentiation (GST) in A. marmorata and A. bicolor bicolor were 0.3971 and 0.2345, respectively. This shows that the value of genetic variation of A. marmorata and A. bicolor bicolor in several rivers on the west coast of West Sumatra is low.
