Short Communication: Analysis of detected metabolites compounds from the crude extract of Rimau Gerga Lebong orangesfruit (Citrus reticulata ‘RGL’) using LC-QTOF-MS/MS. Biodiversitas 23: 3778-3783. Rimau Gerga Lebong (RGL) oranges are the leading commodity in Bengkulu Province. However, the content of bioactive compounds in RGL oranges has not been widely reported. This study aims to identify the flavonoid, alkaloids, and tannins from the ethanol extract of RGL oranges. The results of further identification will be used as a reference in further research on molecular docking in various biological activities. Chemical compounds were identified using qualitative tandem liquid chromatography quadrupole time of flight mass spectrometry (LC-QTOF-MS/MS). The measurement of vitamin C levels was carried out using High-Performance Liquid Chromatography (HPLC). The results showed 25 identified compounds, consisting of 6 compounds belonging to the alkaloid group, 17 from the flavonoid group, and 2 from the tannin group. The alkaloid group compounds were 2?, 3?, 6exo-trihydroxy nortopane, 2?, 3?-dihydroxy nortopane, isosalsolin,3?-dihydrocadambine, chelidimerine, and oleracein D. The compounds from identified flavonoid group were chebuloside, neokurarinol, rutinoside, flavonone, glucoside, licochalcone, viscumneoside, and pinnatifinoside. The identified tannins are corilagin and moracin. These compounds that have been identified have biological activity potential as antioxidants, anti-asthma, anti-dementia, and anticancer. The vitamin C content of the ethanol extract of RGL oranges was 0.02±0.0001%. This vitamin C supports the antioxidant activity of RGL orange.

Genetic diversity and structure of kola tree (Cola nitida) clones germplasm in Côte d’Ivoire using Single Nucleotide Polymorphism markers. Biodiversitas 23: 4677-4685. The kola nut comes from the cola tree (Cola nitida [Vent.] Schott & Endl.), a product of economic interest to Côte d’Ivoire. The country is the leading producer and exporter. However, much of its collection has been destroyed due to urbanization and deforestation. This has resulted in the loss of much of its genetic diversity. This study aimed to determine genetic diversity and structure of C. nitida in Côte d’Ivoire using Single Nucleotide Polymorphism (SNPs) markers. Thirty clones from Côte d’Ivoire (14 clones) and Nigeria (16 clones) cultivated in CNRA (Centre National de Recherche Agronomique) were analyzed using 145 SNPs. GBS (Genotyping by Sequencing) technique was used for the genotyping. The results confirmed the existence of both origins (Côte d’Ivoire and Nigeria) by analyzing clones’ genetic structure and the principal coordinate analysis. The genetic diversity obtained was higher within origins than among origins. The two origins showed moderate differentiation (FST = 0.11) and the genetic distance was 0.46. The 30 clones were subdivided into six subgroups partially overlapping the geographical origins. Genetic differentiation among subgroups was moderate to very strong (FST ranging from 0.10 to 0.26). Four clones from Nigeria were close to those from Côte d’Ivoire. This genetic structuring highlights the possibilities for genetic improvement of kola trees in Côte d’Ivoire.

Species determination and heavy metal content of sailfin catfish (Pterygoplichthys pardalis) from Tempe Lake, South Sulawesi, Indonesia. Biodiversitas 23: 4409-4417. Sailfin catfish is invasive alien species that has become dominant in Indonesian freshwater, including in Tempe Lake, South Sulawesi, Indonesia. Despite their abundance, these sailfin catfish are not utilized because of suspicions that they are contaminated by heavy metals that can endanger human health. The purposes of this study were to determine the sailfin catfish species present in Tempe Lake and measure the content of heavy metals Hg, Pb, and Cd in various body parts. Fish species were identified based on abdominal patterns, while heavy metal content was meaured by atomic absorption. The heaviest two body parts were the head (34.04%) and flesh (26.64%), heavy metal content detected in the liver (0.0402-0.0928 mg/kg), gut (0.0322-0.0671 mg/kg), gills (0.319-0.0865 mg/kg), dark meat (0.0268-0.0589 mg/kg), white meat (0.0221-0.0470 mg/kg), bones (0.0180-0.0519 mg/kg), and skin/scales (0.0257-0.0675 mg/kg). All of them were below the FAO/WHO 1989, US-EPA 1999, and the Indonesian National Standards. The government health and safety guidelines for fish to be safe for human consumption were Hg <0.005 mg/kg, Pb<0.10 mg/kg and Cd<0.02 mg/kg. Therefore the abundant sailfin catfish Pterygoplichthys pardalis in Tempe Lake could be consumed directly and as ingredients in other food products.

Ethnomedicinal study, phytochemical characterization, and pharmacological confirmation of selected medicinal plant on the northern slope of Mount Wilis, East Java, Indonesia. Biodiversitas 23: 4303-4313. The traditional usage of plants in ethnomedicine is a valuable resource for human primary healthcare. The effectiveness of herbal medicines and the creation of new drugs are proven by pharmacological and phytochemical verification. This study documented the traditional ethnomedicine knowledge (TEK) on the northern slope of Mount Wilis community in utilizing medicinal plants and revealed the ethnopharmacology relevance of selected medicinal plants. Data on TEK and its practices were collected through in-depth and semi-structured interviews with informants who were selected using purposive sampling and snowball sampling techniques. The phytochemical compounds of the selected medicinal plant were identified using the UPLC-Qtof-MS/MS instrument, while their pharmacological activity was measured in antibacterial tests. The results showed that the community continued to apply TEK that had been passed down by the previous generation through a method called Gethok Tular. The medicinal plant species in the study area were diverse, 82 species from 39 families with various modes of preparation were used to treat seven disease categories. Tithonia diversivolia had the highest use value of 0.65 and it contained 23 phytochemical compounds. In addition, this plant has high pharmacological potential for antibacterial agents.

Detailed description of scanning electromagnetic microscope (SEM) of the Holothuria scabra’s ossicles (Holothuroidea: Echinodermata) collected from Pesawaran waters, Lampung, Indonesia. Biodiversitas 23: 3697-3704. Ossicles are the microscopic calcified endoskeleton that are unique across sea cucumber species. Furthermore, their integrity is maintained after physical and chemical processes that make them a desired attribute for identification purposes.  Understanding ossicle characteristic especially for the sandfish H. scabra juvenilles is important during mariculture and prevent fraud due to their high economic value. This study aims to identify wild H. scabra ossicles from Pesawaran waters, Lampung Province, Indonesia using scanning electron microscopes (SEM) to get more detailed morphology of their ossicles. Seven specimens of H. scabra were collected in Pesawaran, Sari Ringgung and Mutun, around Pesawaran waters. Morphology identification were carried out by observing the body appearance and tentacles of the specimens as well as measuring their weights and lengths. The observation of the ossicles was performed using a light microscope (LEICA ICC50 HD) at 400x magnification and a SEM (Jeol-JCM6000). The length of the tested specimens were around 140-300 mm and their weight were approximately 99-579 g. These two parameters were found to exhibit positive correlation (R2 = 0.93; R= 0.96; p = 0.000). All observed specimens displayed peltate tentacles (12-22 tentacles) surrounding their mouths. The correlation between length and tentacle numbers was also positive (R2= 0.53; R= 0.73; p = 0.101). The shape of the ossicles consist of six to fourteen-hole buttons, tables, spiny perforated plates from the cloaca, tube foot rods, tentacle rods, dorsal rods, dorsal perforated spiny rods, I shaped and branched rods.

Southeast Sulawesi Province has natural/artificial inland waters for various usages. For example, freshwater has supported various activities such as agriculture, plantations, inland fisheries, and ecotourism. This study reports the presence of invasive mollusk in freshwater (Melanoides tuberculata) and terrestrial (Achatina fulica) habitats. This study is the first report regarding the spatial distribution of the three invasive mollusk species in the inland waters of Southeast Sulawesi, Indonesia. All administrative areas (17 districts/cities) in Southeast Sulawesi province were invaded by A. fulica and 16 districts/cities by M. tuberculata. These invasive mollusks species are a severe threat to the preservation of native species and human health because they are an intermediate host for several types of parasites, such as the trematode group. The spreading and rapid development of these invasive mollusks is caused by their reproductive ability and resistance to poor environmental quality and airborne dispersal factors by birds, water flow, and commercial activities. This invasive species can directly cause huge economic losses as they become a pest in agricultural activities, plantations, and freshwater fisheries (aquaculture). They also can transmit pathogens and displacement of native species. Therefore, it is recommended to have a preventive effort to tackle the expansion of IAS in Southeast Sulawesi.

Papeja, herbal potions during breastfeeding in Madura (Indonesia): The use, perceived effectiveness, and documentation of the plants used. Biodiversitas 23: 5139-5148. The World Health Organization recommends breastfeeding as the normal infant feeding method. However, many women often face challenges in breastfeeding their infants. One of the challenges may be a real or perceived insufficient milk supply. Poor breast milk production is the common cause of breastfeeding failure in preterm babies. In Indonesia, Madurese women use Papeja (herbal medicines) as a herbal potion to enhance breast milk production. Hence, this study aims to identify the pattern of use, perceived effectiveness, and galactagogues plant component of Papeja taken by mothers during breastfeeding in the Sub-districts of Guluk-Guluk and Pasongsongan, Sumenep District. The data were collected through interviews using a semi-structured questionnaire. Sixty women who had agreed to be interviewed, 18 years or older, breastfeeding or breastfeeding in the past 24 months, and two traditional midwifery in the study area were interviewed. We used simple descriptive statistics to summarize the data on demographic, respondent attitudes, and galactagogues plants. Most participants (n = 48) perceived that the herbal galactagogues effectively promoted breastfeeding adequacy. We identified 46 plants from 27 families to have been used in Papeja. Some of the plants used by the mothers have been recorded as having benefits for enhancing milk supply, some have not had that information, but most have not yet been chemically tested. Thus, it is important to validate the plants scientifically for possible galactagogues properties, which can be harnessed for future use. Since we recorded those plants as ethnomedicinal and therapeutic plants, phytochemical and pharmacological investigations are recommended because this could lead to developing new medicine to address poor milk production among mothers.

Abstract. Pratiwi A, Lestari DA, Romdhonah Y. 2022. Short communication: Germination monitoring of selected Annonaceae seeds: Seed bank collections of Purwodadi Botanic Garden, East Java, Indonesia. Biodiversitas 23: 3567-3572. Monitoring activities are an essential aspect to determine the viability and germination of seeds after storage for a certain period. The seed bank of Purwodadi Botanic Garden collected 20 seeds of 11 different species of Annonaceae. Therefore, this study aimed to examine the seed viability of 11 selected species of Annonaceae after being stored for a certain time. Twenty seeds with three replications, i.e., 60 seeds in total were sown. Meanwhile, for genera of Uvaria consisted of U. purpurea, U. rufa, and Uvaria sp. were sown at 30 seeds in total, which means 10 seeds with three replications. The observed parameters were percentage of germination, seed viability, initial germination, and germination type. The results showed only 4 out of 11 selected species of Annonaceae were germinated, namely A. montana, F. latifolium, U. micranthum, and U. purpurea. These four species have epigeal germination types, and monitoring activities included stock rejuvenation, and evaluation of storage procedures for Alphonsea javanica, Annona glabra, Orophea enneandra, Polyalthia littoralis, U. rufa, and Uvaria sp. Furthermore, P. longifolia requires collection regeneration due to its recalcitrant character, and the number of empty seeds from the cut test was >75%. We suggested that P. longifolia seeds might be better to be stored in the refrigerator.

Isolation and identification of Cr-resistant bacteria from the rhizosphere of Tagetes sp. and their ability to reduce Cr(VI). Biodiversitas 23: 4118-4124. Bioremediation involving the interaction of plants with microorganisms is a technique for improving the environment polluted with chromium hexavalent [Cr(VI)]. Marigolds (Tagetes sp.) are known to be a hyperaccumulator plant for Cr(VI) and there are microbes in its rhizosphere that enable the plant tolerant to heavy metals. The aims of this study were to isolate and identify indigenous rhizobacteria resistant to Cr(VI) from Tagetes sp. and to test the ability of these bacterial isolates to reduce Cr(VI). Three isolates of Cr(VI) resistant bacteria were obtained in this study, namely Micrococcus luteus RT-9, Stenotrophomonas maltophilia RT-12, and Brevundimonas sp. RT-23. M. luteus RT-9 and S. maltophilia RT-12 which were resistant to Cr(VI) 100 mg L-1 and their mixture were tested for their ability to reduce Cr(VI) in a liquid medium. Microbacterium sp. SpR3 obtained by the previous study was used as a reference. The results showed that the isolate of S. maltophilia RT-12 had a higher ability (2.2 mg L-1 h-1) than M. luteus RT-9 (0.9 mg L-1 h-1). S. maltophilia RT-12 had the ability to reduce Cr(VI) similar to Microbacterium sp. SpR3. The mixed inoculation of M. luteus RT-9, S. maltophilia RT-12, and Microbacterium sp. SpR3 did not show a difference in the reduction ability of Cr (VI) compared to pure cultures.

Short Communication: A pilot-study on the occurrence and probable factors influencing the population decline of House Sparrow (Passer domesticus) along an urbanization gradient in Coimbatore district, India. Biodiversitas 23: 3884-3889. The House sparrow is a widely distributed, human-commensal bird species across the globe. Over the past few decades, the population of this species has been reported to be declining at an alarming rate. The primary factor postulated to cause this decline is urbanization which leads to the unavailability of suitable nesting sites, a dearth of invertebrate food, the advent of phone towers, and electromagnetic radiation. The current study aims to document the occurrence, abundance and scrutinize the reasons for the decline of House Sparrows along a gradient of urban to rural in Coimbatore district, India. To estimate the abundance of House Sparrows, we counted them in16-points within a three 3km-radius along an urban-rural gradient. Information on gender, cluster size, detection distance, an angle from the observer, etc. were collected. The questionnaire survey was conducted to understand people's perceptions about the importance of the species and its current status. Along the gradient, we observed an average of 69.8 (±24.9) individuals. The species were abundant in rural and semi-urban areas, which is attributed to the availability of nesting sites, such as roof cavities, eaves, mud-walls, adequate feeding sources, presence of open scrub patches and garbage dumping sites adjacent to the residential spaces. However, sparrows were not present in the urban areas, even in their preferred habitats. According to the questionnaire survey, the likely causes for their absence are an increase in vehicle traffic, modern buildings, high-rise structures, and electromagnetic radiation from cell phone towers.
