Biomass accumulation and carbon sequestration potential in varying tree species, ages and densities in Gunung Bromo Education Forest, Central Java, Indonesia. Biodiversitas 23: 5093-5100. Forest biomass plays an important role in carbon storage to mitigate climate change. While many studies have investigated the carbon stock of various forests, adding knowledge in the context of education forest might enrich the importance of this forest as a carbon pool besides its role for education purposes. Gunung Bromo Education Forest in Karanganyar Regency, Central Java, Indonesia consists of several tree cover types with each type having a different ability to absorb carbon dioxide in the atmosphere. This research aimed to determine the accumulation of biomass in Gunung Bromo Education Forest and to investigate the potential for carbon sequestration across different tree species, age classes and densities. Three species of tree (i.e. pine, Indonesian rosewood and mahogany) with varying ages were measured and calculated the biomass (i.e. tree, litter and understorey) and total carbon sequestration potentials (i.e. tree, litter and understorey, and soil organic carbon). This study used purposive sampling method across 9 combinations of tree cover type and age classes, each with 3 replication, resulting in a total of 27 sampling points. The results showed pine stands planted in 1973 had the highest tree biomass of 461.08 t ha-1 and while the pine agroforest planted in 2016 and Indonesian rosewood agroforest planted in 2018 had the lowest tree biomass with 1.02 t ha-1 and 0.39 t ha-1, respectively. Similarly, the pine stands planted in 1973 had the highest total carbon sequestration of 372.68 t ha-1 and the lowest in the pine agroforest planted in 2016 and Indonesian rosewood agroforest planted in 2018 with 187.11 t ha-1 and 193.58 t ha-1 respectively. The results of this study strengthen the common agreement in previous carbon studies that tree age strongly affects biomass accumulation and carbon sequestration, in which the older the plant, the higher the carbon sequestration potential than that of younger plants.

Antibiotic sensitivity profile of multidrug-resistant (MDR) Escherichia coli isolated from dairy cow's milk in Probolinggo, Indonesia. Biodiversitas 23: 4971-4976. The presence of resistant bacteria in animal products such as milk can be a new threat because it is directly related to the human food chain. Resistant Escherichia coli has been widely studied and detected in farms in developing countries. The aim of present study was to determine the antibiotic resistance profile of E. coli bacteria from dairy cows taken during the milking process from several dairy farms in Probolinggo district, Indonesia. A total of 150 milk samples were obtained from farms and E. coli was isolated and identified on Eosin methylene blue (EMB) media and biochemical test, such as Triple Sugar Iron Agar (TSIA) and indole test, methyl red test, Voges-Proskauer test, and citrate test (IMViC) were also performed. The antibiotic sensitivity profile was screened using the Kirby-Bauer test and results were interpreted according to the CLSI standard. The results showed that 124/150 (82.67%) E. coli bacteria exhibited highest percentage of antibiotic resistance to tetracycline (13.71%), streptomycin (9.68%), trimethoprim (8.87%), chloramphenicol (0.87%), and aztreonam (1.61%). A total of 9/124 (7.26%) E. coli isolates were detected as multidrug-resistant (MDR) and 1/9 (0.81%) E. coli isolate was suspected as extended spectrum ?-lactamase (ESBL) bacteria which was resistant to aztreonam antibiotic. Thus, the threat of multidrug-resistant (MDR) E. coli can come from milk which can affect public health.

Physio-biochemical, molecular characterization, and phage susceptibility of Ralstonia pseudosolanacearum associated with tomato (Solanum lycopersicum) and eggplant (Solanum melongena). Biodiversitas 23: 5149-5158. Ralstonia solanacearum is a plant pathogen that causes bacterial wilt diseases in various plant species. The high diversity of these bacteria strains is the basis for their grouping into the R. solanacearum species complex (RSSC). RSSC associated with tomato and eggplant in Indonesia are commonly known as R. solanacearum. It is necessary to characterize R. solanacearum which infects tomatoes and eggplant according to the latest classification. The high diversity of strains also affects their susceptibility to bacteriophages as host-specific biocontrol agents. This research was conducted by characterizing R. solanacearum isolates from different locations based on their physio-biochemical properties, biovar, virulence, phylotype, sequevar, and susceptibility to 12 bacteriophage isolates. The phylotype identification was carried out using multiplex polymerase chain reaction with several specific primers for R. solanacearum. Amplification and sequencing based on the egl gene region were carried out to determine the sequevar of R. solanacearum isolates. As a result, isolates RS18, RS19, RS23, and RS24 had morphological, physio-biochemical, and biovar characteristics according to Ralstonia pseudosolanacearum phylotype I, biovar 3. The two most virulent isolates, RS19 and RS24 were identified as sequevar 14. RS19 and RS24 isolates were susceptible to 7 of the 12 bacteriophage isolates used in this study. Variations of bacteriophage isolates resulted in different plaque morphology which could be attributed to R. pseudosolanacearum susceptibility and bacteriophage virulence.

Short Communication: Antimicrobial of lemongrass (Cymbopogon citratus L.) volatile oil and cytotoxic effects against L20B and MCF-7cell lines. Biodiversitas 23: 5298-5301. Lemongrass (Cymbopogon citratus L.) has been used in different countries in folk remedies for coughs, malaria, pneumonia, and others for many years. The development of bacterial resistance to available antibiotics has obligated finding for new agents to serve as potent antibacterial drugs. The present investigation deals with the effect of volatile lemongrass oil cultivated in Iraq, on different bacterial species and evaluates the cytotoxic activity of the extracted oil on L20B and MCF-7cell cancer cell lines. The plant samples were collected from the college of science /Baghdad University /scientific garden and classified as C. citratus L. by the plant herbarium at the same college. The volatile plant oil was extracted from fresh leaves at the laboratory of the plant biotechnology department of Biotechnology Research Center at Al-Nahrain university/Iraq. With the aid of the Clevenger apparatus, a hydro-distillation method was employed to quantify the lemon grass’s volatile oil. The extracted essential oil and the plant crud maceration were screened for their antibacterial activity against two Gram-negative bacteria (Escherichia coli, and Vibrio cholera) and two Gram-positive bacteria (Bacillus cereus and Staphylococcus aureus) using the well-diffusion method and disc diffusion method. The biological survey also included the cytotoxic effect of oil subjected to the anticancer activity in vitro on two cancer cell lines; L20B mouse cell line that expresses the genes for human cellular receptors for polioviruses, and the second line was the MCF-7 breast cancer cell line. As a result of this screening study, it was shown that the plant seemed to be rich in essential oil content. The Iraqi cultivated plant produced 1.5%v/w essential oil. The volatile oil affected both Gram-negative and Gram-positive strains in comparison to the crud plant extract among the selected bacterial cultures, the highest antibacterial activity was recorded against the Gram-positive strain S. aureus by well diffusion method. Besides, the plant oil showed an inhibitory effect on L20B cell line with a percent inhibitory growth rate reaching 47.1% at 1.125 ?l/mL of the oil concentration. While for the other cell line, MCF-7 cell line, the inhibitory growth rate percentage appeared for almost all concentrations in comparison with control after 24 hours, and even at a concentration of 0.3125?l/mL, the inhibitory growth rate percentage reached up to 86%. This study was conducted to high lightened the benefits of this plant as little study had been done for an Iraqi cultivated plant and the results showed the potent biological effects of the plant especially the volatile oil as an antimicrobial and as a potent cancer inhibitory agent.

Abstract. Pradipta S, Siswoyo TA, Ubaidillah M. 2023. Nutraceuticals and bioactive properties of local Java pigmented rice. Biodiversitas 24: 571-582. Pigmented rice is used in nutraceuticals and has health benefits, on the other side, the characterization of local pigmented rice has not been widely observed. This research aimed to determine the nutraceutical and bioactive properties of ten pigmented Indonesian rice varieties (Segreg, Merah SP, Gogo Niti II, Mansur. Merah Wangi, MS Pendek, Super Manggis, Lamongan I, Aek Sibundong, and Seksek). The proximate composition of pigmented rice was performed based on standard methods. The antioxidant activity was carried out using DPPH radical scavenging activity. Morphological characteristics of these pigmented rice were the weight of 1000 grains ranges 17.47-23.10 g, with slender, medium, and bold shape categories. The proximate analysis revealed a significant difference (p<0.05) among cultivars, excluding the lipid. Amylose content varied between 17.43-25.71%. The lowest amylose cultivar had a high peak, trough, and final viscosities but a low pasting temperature. The gelatinization enthalpy was 0.83-1.93 J/g, and the thermal properties of the onset were between 78.6-83.60ºC. The peak was 81.90-86.20ºC, and the conclusion temperature was between 86.00-89.80ºC. The functional properties of the water absorption index were 5.1-7.42 g/g. The water solubility index was 2.32-7.32%, the oil absorption index was 0.92-1.67 g/g, and the bulk density was 0.56-0.70 g/cm3. The total phenolic range was 2.69-9.51 mg GAE/g, and flavonoids were between 0.61-2.31 mg QE/g. Antioxidant activity has been positively correlated with the percentage of flavonoids in phenolics.

Abstract. Anastasia L, Kodrat EA, Victor H, Sanjaya A, Pinontoan R. 2022. Isolation and characterization of indigenous amylolytic enzyme-producing Aspergillus sp. from sweet-flavored tapai. Biodiversitas 23: 5580-5586. Aspergillus sp. are able to produce abundant amylolytic enzymes that may have a crucial role in producing the sweet flavor of tapai. This study was aimed to evaluate the amylolytic activity of Aspergillus sp. isolated from a widely known sweet tapai from Jember, East Java, Indonesia. The fungi isolated from sweet tapai were identified under notable morphological parameters, and its amylolytic activities were evaluated by utilizing various starch-based media, such as potato peel, potato dextrose broth, cassava, and maize. Optimum pH and temperature of partially purified amylolytic enzymes were evaluated using DNS assay. The result of this study suggested that TM3 fungal isolate isolated from sweet tapai belonged to Aspergillus sp. section Nigri. Aspergillus sp. TM3 grown in the potato peel medium displayed the highest amylolytic activity, followed by those grown in maize, potato dextrose broth, and cassava media. Further characterization of partially purified amylolytic enzymes from the isolate showed that optimal pH for the amylolytic activity was at pH 4.59 while the optimal temperature was around 54°C. Furthermore, analysis by thin-layer chromatography revealed that the amylolytic enzymes starch hydrolysis yielded dextrose as the main product and oligosaccharides as the side product, which indicated that Aspergillus sp. TM3 may have both alpha-amylase and glucoamylase involved in its amylolytic activity. This study explored the role of Aspergillus sp. TM3 in producing the sweet flavor of tapai.

Composition and abundance level of pest mites in jasmine gambier (Jasminum officinale) plantation of Purbalingga, Central Java, Indonesia affected by some abiotic factors. Biodiversitas 23: 5227-5232. The decline in productivity of gambier jasmine flowers is not only caused by the reduction of gambier jasmine land area but also insect, caterpillar and pest mite attacks. Types of pest mites and their population dynamics are closely related to abiotic factor conditions such as temperature, humidity, rainfall, season, an abundance of predatory mites and types of plant cultivars. The study aimed to determine the types of pest mites and some abiotic factors that affect their abundance in gambier jasmine plants. The research method was a survey with a purposive sampling technique. Sampling locations were in Cipawon village, Bukateja sub-district, Purbalingga district, and Central Java Province, Indonesia. The obtained data were analyzed by analysis of variance at an error rate of 0.05. The types of pest mites that were identified consisted of 8 species, namely Brevipalpus phoenicis, B. californicus, B. papayensis, B. obovatus, Tetranychus urticae, T. kanzawai, T. cinnabarinus and Tyrophagus putrescentiae. The type of pest mite always found at each sampling point was B. phoenicis, with an abundance range of 0.027 to 0.067. The results of the analysis of the variance of pest mite abundance in gambier jasmine plantations showed that abundance of individuals between pest mite species did not significantly differ at all sampling points. Based on multivariate analysis, it was noted that trichome density had the most influence on pest mite abundance with a population model of Y = 0.399 + 0.043a + 0.012b.

Waterbirds can serve as bio-indicators for their surrounding environments, indicating issues such as habitat degradation and climate change. One factor that contributes to successful reproduction for many bird species is parental behavior during the brood care stage. The painted stork (Mycteria leucocephala) is a waterbird species that provide biparental care throughout the breeding season. Although many studies have focused on populations of this species, information about their behavior, particularly during nestling rearing, remains lacking. Hence, this study was conducted to examine the parental behavior of painted storks between the early and late stages of brood care in two different colonies located in Peninsular Malaysia: Tasik Shah Alam (TSA) in Selangor and Upper Bisa Putrajaya Wetland (UBPW) in Putrajaya from May to September 2016. Three pairs of painted storks were monitored in every site, and seven behaviors were identified: roosting, preening, nest repair, wing opening, flying, feeding and brooding. Of all the behaviors recorded, flying, roosting and wing-opening behavior were the most frequently observed behaviors in both study areas during the early stage of brood care. However, painted storks were found to spend most of their time flying when the chicks had grown up by the late stage. This may be due to the food demand for the parents and their chicks. As the nestlings grow bigger, more food is required before the nestling can fledge independently. This is supported by increased feeding frequency observed from the early to late stages. Males were found to have a higher frequency of all behaviors than females. This data facilitates a greater understanding of painted stork behavior, which can help in their conservation and management processes.

EG6.4: Molecular identification, larvicidal toxicity, and mechanism of actions Biodiversitas 23: 5431-5438. This research was carried out for molecular identification, as well as the determination and mechanism of action of larvicidal toxicity of Bacillus sp. EG6.4 isolated from breeding sites of Aedes aegypti from Gresik, East Java, Indonesia. Bacillus sp. EG6.4 was a Gram-positive endospore-forming bacteria. Molecular species identification using 16S rRNA gene sequencing showed that isolate had 97.89% similarity with Bacillus mojavensis. The isolate showed larvicidal toxicity against A. aegypti larvae. The lethal concentration 50% (LC50) values ??at 24 and 48hours exposure were 8.99±1.01 ×107 cells/mL and 8.43±1.01 ×107 cells/mL, respectively, while lethal time 50% (LT50) value was 11.9±1.1 hours. Production of chitinolytic enzymes or biosurfactants, chitinolytic and hemolytic assays were conducted to determine the larvicidal mechanism. As a result, Bacillus sp. EG6.4 showed hemolytic, but not chitinolytic activity, indicating its potency to produce biosurfactants. Transmission Electron Microscopy (TEM) result showed that isolate had oval-shaped endospores located subterminal with massive-shape parasporal inclusions. The detection of srfA-D gene showed that isolate produced surfactin biosynthesis thioesterase. Thus, Bacillus sp. EG6.4 produced biosurfactant that potentially to be developed as a biocontrol agent for disease vectors and plant pathogens.

Abstract. Priskila C, Vidian V, Sanjaya A, Sugata M, Pinontoan R. 2022. Thrombolytic potential in bacteria isolated from fermented durian tempoyak. Biodiversitas 23: 5731-5737. Cardiovascular disease is one of the leading causes of death worldwide and is primarily caused by dysregulation in the blood clotting system leading to the formation of unwanted blood clots (thrombi). Thrombolytic therapy, which aims to break down unwanted blood clots, employs various agents possessing thrombolytic activity. However, the harmful side effects of conventional thrombolytic agents necessitate the exploration of new and safer agents with high thrombolytic activities. Thrombolytic agents can be obtained from various sources, including fermented foods. Tempoyak is a condiment made from fermented durian, which is found in some regions of Indonesia, and is reported to contain beneficial microorganisms. The aim of this study was to isolate and identify thrombolytic agents from tempoyak. After several pure microbial colonies were successfully isolated from tempoyak, they were screened for proteolytic activity on skim milk agar medium. The isolated colonies with proteolytic activity were then subjected to several tests, including the whole blood clot lysis test, euglobulin clot lysis test, fibrin plate method, and zymography. Subsequently, the isolates with thrombolytic and fibrinolytic activity potential were identified in terms of their morphology and biochemistry. In this study, three bacteria with thrombolytic activity potential were successfully isolated. Fibrin zymography indicated that the molecular weights of the proteins that play roles in the fibrinolytic activity ranged from approximately 30 to 153 kDa, which were suspected to correspond to serine protease, cysteine protease, and/or metalloprotease. All three isolates were identified as belonging to the genus Corynebacterium. This is the first report discussing the thrombolytic and fibrinolytic activities of Corynebacterium sp. derived from fermented food.
