Biodiversitas 23: 4570-4577. Mangifera rufocostata Kosterm is known as a medicinal plant for antidiabetic. Therefore, intensive use of the bark of this plant results in a significant decline in the plant population which can eventually cause the plant to become extinct. This study aimed to determine the potentially suitable cultivation region of M. rufocostata using Maximum Entropy (MaxEnt) modelling. Twenty-three environmental variables including bioclimatic, soil type, slope, altitude, and solar radiation have been used for the development of distribution modeling. The results showed that precipitation seasonality, precipitation, mean temperature, and solar radiation are important variables in the development of the habitat suitability model. Suitable habitat locations for M. rufocostata include the Hulu Sungai Tengah, Hulu Sungai Selatan and Hulu Sungai Utara Regency, South Kalimantan Province, Indonesia. The MaxEnt model provided an area under curve (AUC) value of 0.959, indicating that MaxEnt is accurate and informative for the prediction of habitat suitability of M. rufocostata. The results indicate that this suitability prediction model may be applied used for future management, monitoring, cultivation and conservation of M. rufocostata.

Identification of Tambelo (Bactronophorus thoracites) in Wamesa Waters, Manokwari, West Papua, Indonesia. Biodiversitas 23: 6021-6030. Tambelo, otherwise known as shipworm, is a marine bivalve mollusk comprising 16 genera in the Teridinidae family, of which Teredo is the most frequently mentioned. Furthermore, the only species in the genus Bactronophorus is the wood borer, Bactronophorus thoracites (Teredinidae), which has a wide distribution in the Indo-West Pacific biogeographical region. Identifying shipworm species is difficult due to their high diversity and the limited information about their taxonomic characteristics, which often leads to identification errors by collectors. Papuan people consume tambelo as a substitute for side dishes because they are believed to be able to cure various diseases. Therefore, this study aims to establish by morphology and molecular analysis that tambelo belongs to the B. thoracites of the Teredinidae family. Tambelo was identified using the morphological method of observing the physical characterization of the palette, which is unique between genera. It was collected from a mangrove forest near Wamesa Beach, South Manokwari, Manokwari Regency, Indonesia. Meanwhile, this study adopted PCR, an identification method that uses DNA from a small number of samples and can provide a DNA sample sequence. Morphological analysis based on pallet characterization showed that the characteristics of the tambelo pallet were similar to those of the B. thoracites based on the identification book. In addition, the molecular phylogenetic analysis using two mitochondrial protein-coding genes, cytochrome c oxidase subunit I (COI), showed tambelo was 91% similar to B. thoracites from mangroves environment on the west coast of Peninsular Malaysia, Malaysia.

Prey composition and correlation between morphometry and prey biomass weight of the endemic Nepenthes bicalcarata in Kalimantan, Indonesia. Biodiversitas 23: 5453-5460. Nepenthes bicalcarata Hook.f. is an endemic plant of the north-western part of Borneo, which is threatened by the conversion of its habitat. The understanding of the ecology of N. bicalcarata in regard to its prey is limited. This research aims to analyze the prey composition of N. bicalcarata and to investigate the correlation between pitcher morphometry and prey biomass weight. This research was done from November 2021 to March 2022 in a peat swamp primary forest in Sintang District, West Kalimantan Province, Indonesia. We used a quantitative descriptive approach and exploration method to gather data from N. bicalcarata in-situ habitat. The morphometry data of N. bicalcarata pitchers were collected in the field while the biomass trapped in N. bicalcarata pitcher liquid was sampled with a plastic bag and preserved with 70% of alcohol. Seven upper pitchers and 39 lower pitchers were recorded in this research. The composition of N. bicalcarata preys was dominated by Formicidae family including C. schmitzi, Polyrhachis sp. and Crematogaster sp.. Besides that, there were also mosquito larvae, ladybugs larvae, Ceratopogonidae larvae and some unidentified flying arthropods as the prey of N. bicalcarata. We found the plant’s parts in the lower pitchers of N. bicalcarata in the peat swamp forest, suggesting a possible mechanism of this plant to extract plant’s parts as alternative nutrient sources, yet this needs further research and intensive field observation. The correlation between pitcher volume and prey biomass weight was significant with P-value =0.00 (<0.01) and regression is formulated as Y (prey weight) = -0.1727 + 0.0039X (pitcher volume). This means that any increase in N. bicalcarata’s pitcher volume will increase the weight of the biomass linearly.

Diversity of the Tabuhan Island coral reef fish revealed by DNA barcoding and implication on conservation strategy in Banyuwangi, Indonesia. Biodiversitas 23: 4844-4851. Tabuhan Island, Banyuwangi, Indonesia, is one of the mainstays of coastal tourism with the charm of coral reefs and has a fairly high potential for reef fish diversity. Coral reef ecosystems provide suitable habitats for reef fish to spawning ground, feeding ground, and nursery ground which provide suitable nurturing areas. Here, studies have been carried out on the diversity of reef fish by molecular approaches. The molecular identification approach provides accuracy in identification to the species level. In this study, samples of reef fish species from Tabuhan Island waters were identified molecularly in the mitochondrial DNA region of the cytochrome c oxidase subunit I (COI). The identification results showed that 53 specimens had been identified, and some of the themes were registered in the GenBank database to strengthen the genetic information of reef fish in the tropical region of Indonesia. A total of 53 specimens were identified, spread over 49 species, 3 orders, and 17 families dominated by reef fish groups from Labridae (20 species). The phylogenetic tree reconstruction shows that the family collects several species, but some species are classified as paraphyletic. The results of this molecular identification have also succeeded in registering 35 COI sequences in the Genbank database. The mtDNA sequence data is very important and becomes the basis for the genetic conservation resources in coral reef ecosystems.

Urine biochemical profile analysis and transabdominal ultrasonography for pregnancy detection in the endangered lowland anoa (Bubalus depressicornis). Biodiversitas 23: 6056-6061. The population of lowland anoas (Bubalus depressicornis) has decreased significantly due to illegal hunting and habitat loss. As such, the urgency in developing successful ex-situ conservation programs cannot be overstated. Ex-situ breeding programs of anoas, however, are hindered by difficulties in early pregnancy detection. The failure to detect pregnancy at an early stage often results in miscarriage due to repeated breeding attempts of unconfirmed pregnant females. Moreover, frequent false heat during 5 to 6 months of pregnancy corroborated the repeated breeding of pregnant females. Therefore, early detection techniques are needed to improve breeding success. The present study evaluated the use of urine biochemical profiles as a minimally invasive approach for early pregnancy detection and transabdominal ultrasonography as a confirmation tool of false heat in four lowland anoas. Statistical analysis showed that glucose and protein level were significantly elevated during pregnancy since the 0 days of post-mating (p<0.05) and consistently higher compared to the non-pregnant period. Meanwhile, transabdominal ultrasonography successfully identified organ structures and development by the 155th, 176th, 180th, and 191st days of pregnancy. The present study suggests that urine biochemical data provide a promising, minimally invasive approach to early pregnancy detection in anoas. Incorporating urine data and ultrasonography can significantly help to maintain pregnancy by validating false heat.

Abstract. Suriya C, Satian C, Visut B, Supadha K. 2022. Diversity of freshwater fish at Sago Palm Wetlands, Nakhon Si Thammarat Province, Thailand. Biodiversitas 23: 6335-6344. Studies on the diversity of freshwater fish at Sago Palm Wetlands, Nakhon Si Thammarat Province, Thailand, were conducted in the period from March 2020 to November 2020. Fish samples were collected, encompassing the hot-dry season (March to June) and the rainy season (September to November) at 4 sampling zones, with 5 stations for each zone. Fish were caught using a seine net of 1 cm mesh size, 1.5m in length, and 1m in width. All data were analyzed to find out the fish diversity indices. The multivariate method was used to determine cluster analysis. A total of 3634 fish specimens, representing 62 species belonging to 24 families, were recorded during the sampling period. Cyprinidae was the major family, with Trichopsis vittatus containing the highest index of relative importance (IRI). The species diversity index of fish was in the range of 0.66 to 2.67; the evenness index was in the range of 0.64 to 0.95, and the species richness index ranged from 0.36 to 6.86, indicating an intermediate-low uniformity, with a medium species number in the area. The cluster analysis of the Bray-Curtis similarity index shows A division of the fish community into 2 clusters in the hot-dry season and 3 clusters in the rainy season, having a similarity percentage ranging between 42% and 95%, respectively. Also documented were one introduced species and one endangered fish species during sampling. The findings suggest that Sago Palm Wetlands is still inhabited by numerous fish species and should be conserved to ensure its aquatic animal richness for the future. This data provides information about the fish habitat in the Sago Palm Wetlands and can be used to update the checklists of fish species in the Tapi River, the Pak Phanang River, and the Trang River Basins. It also can be useful for the planning of fishing activities and the provision of guidelines for future research and conservation purposes.

Biodiversitas 23: 5217-5226. The grouper stocks in Kwandang Bay, Sulawesi Sea, Indonesia, are dominated by orange-spotted grouper (Epinephelus coioides) and coral trout (Plectropomus leopardus). However, the Kwandang Bay grouper fishery is currently experiencing serious challenges due to population decline resulting in a decline in both the average size of fish caught and the production volume. This study was conducted in Kwandang Bay, Sulawesi Sea, from January to December 2021. The population dynamics parameters estimated were growth, recruitment, mortality, exploitation rate and yield per recruit. The results showed that the total length (TL) of E. coioides and P. leopardus ranged from 16.50-101 cm and 17.90-60 cm, respectively, with Von Bertalanffy K-coefficients of 0.38 and 0.69, respectively. Recruitment occurred during most of the year, with respective recruitment value ranges of 0.99-17.11% and 0.08-19.22%. Natural and fishing mortality was 0.69/year-1 and 0.87/year-1 for E. coioides, 1.16/year-1 and 1.47/year-1 for P. leopardus. Beverton and Holt yield per recruit analysis indicated recruitment overfishing of both E. coioides and P. leopardus. Fishing regulations are needed to maintain a sustainable grouper fishery, including a 25% reduction in fishing vessels and gear, to enable the groupers of Kwandang Bay to spawn at least once during their life cycle.

Abstract. Vasandani SR, Tan WA. 2022. Diversity of bacterial phenol hydroxylase-encoding genes from gasoline-contaminated silt soil. Biodiversitas 23: 5502-5506. Phenol is an aromatic compound often used as a raw material or intermediate in various industries. Improper handling and disposal may lead to the accumulation of this hazardous compound. Bioremediation is the most viable method to remove phenol from contaminated environment. In this study, the diversity of genes that encode for phenol hydroxylase, a key enzyme in phenol degradation, was assessed in gasoline-contaminated soil from a commercial gas station in Central Jakarta, Indonesia. Partial phenol hydroxylase-encoding gene library was constructed using a pair of universal primer in the pGEM®-Teasy vector. A total of 30 recombinant clones were obtained and sequenced to analyze the genetic diversity of this gene. Obtained clones were 86-99% identical to phenol hydroxylase-related proteins. Phylogenetic tree analysis on amino acid sequences derived from our library revealed that 56.7% of the cloned fragments were closely related to Proteobacteriodota and 20.0% of them were clustered with Actinomycetota. The rest 23.3% of the clones formed a cluster separate from any of the reference sequences, possibly indicating the presence of novel phenol hydroxylase genes.

Biodiversitas 23: 5267-5277. The worsening environmental problems have become a serious concern prompting humanity to search for solutions aiming for environmental management through understanding the relationship between human-environmental attitude and behavior with the belief that humans are the prime contributor to both - problem and the solution. By acknowledging this existing situation, this study was designed to assess the senior high school student’s level of understanding of and attitude toward the environment and the effects of both variables on their ecological behavior. A sequential strategy under mixed-method research was used in the study. A survey questionnaire containing both qualitative and quantitative questions was distributed to and answered by the respondents, followed by a focus group discussion that solidified and substantiated the quantitative result. Analysis of the responses demonstrated that students’ environmental attitudes were uncertain and connoted that they have intermediate attitudes and were all observed to be connected to their intermediate ecological behavior. Further, it revealed anthropocentric attitudes were dominant among students. This environmental attitude, together with human dominance over nature, denoted correlation along ecological behavior toward conservation policies, environmental threats and support for population growth policies, while eco-centric attitudes do not correlate to ecological behavior. In conclusion, the limited and ambiguous knowledge of senior high school students on the rule of human and nature need reinforcement for them to develop a good quality viewpoint about nature and willingly advocate environmental protection through their actions.

Abstract. Pikulthong V, Bhirompan S, Dechkla M, Mokkapan K, Boonman N, Wanna C, Machana K, Phakpaknam S. 2022. Local herbs for pain relief in the area of Tumbon Khao Hin Son, Chachoengsao, Thailand. Biodiversitas 23: 5012-5019. Local herbs have been used in Thailand for years. However, data collection on the use of local herbs in particular areas has not been thorough. This study aimed to survey local herbs treating pain relief in the area of Tumbon Khao Hin Son, Amphoe Phanom Sarakham, Chachoengsao, Thailand. Semi-structured interviews were used. Purposive sampling was used to determine the sample group. In the survey and study on botanical characteristics, from November 2019 - March 2020, 68 plants were identified and classified into 29 families. The local herbs which were mostly used for pain relief included 7 species from the Acanthaceae family, 6 types from Fabaceae, Lamiaceae, and 5 types from Zingiberaceae. The leaves of herbaceous plants had the highest frequency of use. A remedy, Ko-Klan, was found and prepared using a boiling method that could relieve pain. The surveyed herbs were used to treat 4 main symptoms which were body pain, stomachache, headache, and toothache. It was found that 36 species of herbs, or a majority of them, were used to directly relieve the pain. The interviews showed that 27% of the respondents had some home-grown herbs which were used for cooking and basic illness treatment. The age of the respondents was relevant to the use of herbs. The findings in this study will be the foundation for local herb studies which support and are aware of Thai local wisdom and knowledge as guidance to alter herbs for pharmaceutical purposes in the future.
