Biodiversitas 23: 5989-5994. Schistosomiasis is a zoonotic parasitic disease with Oncomelania hupensis lindoensis, intermediate snail hosts of Schistosoma japonicum. The spread of O. hupensis lindoensis snail habitat was found in the three areas of the Napu, Bada, and Lindu Highlands with an infection rate above 1%. Oncomelania hupensis lindoensis  is primarily cryptic species that are morphologically difficult to identify and distinguish from other species. Consequently, it can be confused with the naming of species. One of the molecular approaches that can be used to identify the Oncomelania spp. quickly and accurately is DNA barcoding using the COI mitochondrial gene. However, the research on identifying Oncomelania spp. in Indonesia is still very limited. Therefore, this study aimed to identify six Oncomelania spp. from Napu and Lindu, Central Sulawesi using COI mitochondrial gene as a molecular marker for DNA barcoding. The method used in this study was a Polymerase Chain Reaction (PCR) method with universal primers, LCO-F and HCO-R. The data obtained were then analyzed using GeneStudio, DNASTAR, BLAST, Identification Engine, Mesquite, MEGAX, and BEAST. The analysis was conducted to obtain similarity, genetic distance and reconstruct a phylogenetic tree. The result revealed that all six samples of Oncomelania spp. collected from Napu and Lindu were identified in one species, namely Oncomelania hupensis lindoensis. This research is very important to be carried out regularly periodically so that it can be used as a basis for Schistosomiasis control program data and related sectors to eradicate snails effectively, efficiently, and on target.

Abstract. Sa-An AC, Quinitio GF, Anasco NC, Traifalgar RFM, Nillos MGG. 2022. Identification and quantification of vitellogenin gene in eyebrow goby (Oxyurichthys ophthalmonema). Biodiversitas 23: 5606-5613. Vitellogenin (VTG) gene has been used as a biomarker for exposure to estrogenic compounds in marine and freshwater fish species. There was limited information about the detection of VTG mRNA in an estuarine environment using a bioindicator fish species. In this study, a quantitative real-time polymerase chain reaction (qPCR) assay was developed for quantification of VTG mRNA normalized to ?-actin in eyebrow goby, Oxyurichthys ophthalmonema. VTG mRNA in the liver of reproductively mature female samples was isolated, amplified, and sequenced. There were 430 bp fragments of VTG, corresponding to a 143 amino acid sequence obtained from eyebrow goby. The partial protein sequence exhibited similar identities with the phosvitin motif of VTG in other gobies. The new qPCR assay was developed through the evaluation of hepatic mRNA levels in nonreproductive male eyebrow goby at 0, 6, 12, and 24 h following intraperitoneal injection of 10 mg kg-1 17?-estradiol. Hepatic mRNA levels were detected 6 h after 17?-estradiol injection and the highest VTG expression was noted 24 h after injection. Results of the present study suggest that male eyebrow goby can be used as indicator species for estrogenic contamination. Likewise, the established qPCR assay can be used to assess recent exposure to estrogenic contaminants in the aquatic environment. The characterization of the amplified VTG gene from eyebrow goby and the application of the qPCR assay developed for eyebrow goby in monitoring EDCs in the field may be considered for future investigation.

Abstract. Syukur A, Zulkifli L, Mahrus H. 2021. Mangrove ecosystem provisioning services for the sustainability and diversity of bird species in the coastal region of Lombok Island, Indonesia. Biodiversitas 24: 1589-1599. Ecological services offered by mangroves are imperative for various faunal species. The present study is focused on assessing the role of mangroves in the sustainability and diversity of bird species in the coastal region of Lombok Island, Indonesia. The aims of the study are (i) to describe the conservation status of bird species and (ii) to describe the value of the ecological index of birds as an indicator of mangrove conservation. The species diversity of birds is assessed by observation, whereas the quadratic transect method is used to assess mangrove vegetation structure. Data analysis has been carried out using univariate statistical analysis, namely the Shanon-Wiener index (H'), Simpson Index (d), Evenness Index (E), Richness Index (R), and linear regression analysis. The genus Rhizophora and Avicennia species have an even distribution in the study area. Rhizophora species in the study area result from revegetation, whereas Avicennia species grow naturally. Altogether, a total of 19 bird species have been observed in the present study. Of these, eight species have a limited distribution, four bird species are Near Threatened (NT), and others are Least Concern (LC) concerning their conservation status. The estimated Diversity Index (H') is ranged between 3.09-3.67, whereas Evenness (E) and Richness (R) index are ranged between 0.72-0.85 and 7.09-10.33, respectively. The ecological index value shows that the research locations have high species diversity and richness, and species are evenly distributed across the sites. Therefore, the value of the index of bird ecology and the distribution of bird species are indicators of mangrove supply services for the preservation of birds at the study site. The study concluded that the diversity of ecological indices and the conservation status of birds are important information for the success of mangrove conservation through revegetation at the study site. However, a comprehensive study is needed on the functional relationship of mangrove supply services with the diversity of bird species to obtain validity and rehabilitation as conservation indicators.

Macroepiphyte biodiversity on Kappaphycus alvarezii surface and its interaction with environment in cultivation centers on Lombok Island, Indonesia. Biodiversitas 23: 6284-6292. The aims of this paper are to investigate the influence of environmental factors on the diversity of macroepiphytes in Kappaphycus alvarezii. Macroepiphyte sampling was carried out at four cultivation centers: Seriwe Bay, Ekas Bay, Gerupuk Bay, and Siwak of Lombok Island, West Nusa Tenggara, Indonesia. Several stations were placed at each location, and at each station were used one to three cultivation units with five replications. Environmental data measurements were carried out at the time of sampling. Some parameters were measured: pH, temperature, salinity, current velocity, brightness, DO, BOD, nitrate, phosphate, and TSS. Data analysis was performed using several diversity indices. In addition, a biplot analysis was performed using PCA to analyze the correlation between environmental factors and environmental factors to macroepiphytes. The results showed that the most dominant species was Polysiphonia spaerocharpa, and the second was Ceramium sp1. The highest richness and abundance were found in Ekas Bay. The highest species dominance was in Siwak. The highest diversity index was in Ekas Bay. The correlation between environmental factors showed that hemeroby harmed depth, phosphate, and DO and positively affected BOD and the thallus surface structure. The principal correlation analysis showed that the abundance of macroepiphyte was influenced by BOD, the roughness of host thallus, and the hemeroby index.

The Green Junglefowl is Indonesian germplasm that is at risk of population decline. Therefore, efforts are needed to protect its existence and survival. Cryopreservation of spermatozoa in the form of frozen semen is required to preserve this germplasm. This research aims to determine the effect of lactose-astaxanthin in preventing DNA fragmentation and intact plasma membrane of Green Junglefowl spermatozoa during the freezing process. A completely randomized design with a 2x4 factorial pattern, including two treatment groups and four dimethylsulfoxide (DMSO) concentration factors, was used. The treatment groups are T1 and T2, consisting of egg yolk phosphate diluent and egg yolk phosphate diluent added with a combination of 0.6% lactose-0.004% astaxanthin, respectively. The four DMSO concentration factors were 4%, 6%, 8%, and 10% respectively. Subsequently, each treatment was repeated four times. The DNA fragmentation and intact plasma membrane of spermatozoa were observed, and the data obtained afterward were analyzed for variance, followed by the Ducan test. The results showed that the addition of 0.6% lactose-0.004% astaxanthin with 6% DMSO had a significantly better effect (P<0.05) on the quality of post-thawing spermatozoa with 19.75±0.96% DNA fragmentation and 53.75±3.86% intact plasma membranes. Therefore, it is recommended that a combination of 0.6% lactose-0.004% astaxanthin at 6% DMSO concentration should be added to Green Junglefowl semen during the freezing process to prevent DNA fragmentation and intact plasma membranes.

Abstract. Saidy AR, Priatmadi BJ, Septiana M. 2022. Diversity in characteristics of tropical peatlands varying in land uses leads to differences in methane and carbon dioxide emissions. Biodiversitas 23: 6293-6301. The use of tropical peatlands for commercial agriculture causes a change in their original function as carbon storage to become sources of methane (CH4) and carbon dioxide (CO2) emissions. Therefore, this research aims to quantify the emission of CH4 and CO2, and peat characteristics in five tropical peatlands with different land uses, namely shrubs-, burned-, Albizia-, spring onion-, and lettuce-peatlands, to determine factors controlling carbon emissions. The results showed that CH4 emission ranged from 0.21 to 0.58 mg C m-2 h-1, with the lowest and the highest obtained in burned- and cultivated-peatlands (spring onion- and lettuce peatlands), respectively. The emission of CO2 ranged from the lowest in burned peatland (34.10-47.06 mg C m-2 h-1) to the highest in shrubs-peatland (136.79-180.87 mg C m-2 h-1). This showed that the diversity in CH4 emissions with different land uses is attributed to variations in the water table, water-filled pore space, ammonium, and nitrate contents. The rates of CO2 emission were controlled by carbohydrate-, fiber-, organic C-, and lignin-contents. This indicated that land and water managements need to be applied to reduce the emissions of CH4 and CO2 in tropical peatlands with different land uses.

Short Communication: Polymorphism of collagen type X (COLX) gene and their association with egg production traits and egg weight in Alabio and Mojosari ducks. Biodiversitas 24: 1518-1523. Type X collagen (COLX) is non-fibrillar collagen expressed in the white isthmus of the avian oviduct and plays a role in egg formation. This study aimed to identify COLX gene polymorphism and its association with egg production and egg weight in Alabio (Anas platyrhynchos Borneo) and Mojosari ducks (Anas platyrhynchos Javanicus). Blood samples of 100 female ducks were collected, and their deoxyribonucleic acid (DNA) was extracted. In addition, a 220 bp fragment of COLX gene was amplified using the polymerase chain reaction (PCR) method and sequenced for single nucleotide polymorphism (SNP) identification and genotyping. Data analysis was performed to calculate genotype and allele frequencies, and an independent sample t-test was used to examine the association between genotype and egg production and egg weight of each individual. Three genotypes (CC, TT, and CT) of SNP g.74C/T were detected in both breeds. Allele C (0.77) was dominant in Alabio population; otherwise, allele T (0.81) dominated Mojosari population. The frequency of CC (0.59) was highest in Alabio ducks, followed by CT (0.37) and TT (0.04), while TT (0.65) was highest in Mojosari ducks, followed by CT (0.33) and CC (0.02). Meanwhile, association analysis showed that ducks with CC genotype had significantly higher egg production at 28 and 34 weeks in Alabio than those with CT genotype (p<0.05). These results indicate that COLX is an effective genetic marker for a breeding program to increase egg production in Alabio ducks.

Abstract. Ivayani, Widiastuti A, Suryanti, Suharjo R, Priyatmojo A. 2022. Short Communication: First report of Xanthomonas sacchari causing rice sheath rot disease in Lampung, Indonesia. Biodiversitas 23: 6463-6470. Rice sheath rot is recently reported as an emerging important rice disease in Indonesia, which can cause yield losses of up to 85%. Due to its potential loss, serious attention should be given on the disease. However, there is still limited information on the causal agent of the disease in Indonesia, especially from plant pathogenic bacteria. One bacterial isolate (LSE 33) was obtained from the sheath rot symptom of rice. Pathogenicity test revealed that bacteria produced symptoms that were similar to those obtained from the field. Based on the sequence analysis of 16S rRNA gene region, isolate LSE 33 was placed within group of the type strain and reference strains of Xanthomonas sacchari. Strain LSE 33 had 99.93% sequence similarity with Xanthomonas sacchari strain SAM 144 and strain AF 10 isolated from rice, and 99.35% similarity with strain LMG 471T isolated from sugarcane. To our knowledge, this is the first report of X. sacchari causing sheath rot on rice in Indonesia.

The F3 is the early selection generation that has a good heritability and heredity pattern in a segregated population. The aims of this study are to identify the effectiveness of forming a selection index based on narrow-sense heritability and factor analysis and to select potential tomato F3 lines to be continued in the F4 generation. The research was arranged by an augmented design combined with a randomized complete block design as the environmental design. The research factors consisted of non-repeated genotypes, i.e. the lines with a limited number of seeds consisting of 75 lines and repeated genotypes as comparison varieties consisting of three varieties (Mawar, Tymoty, and Karina). The lines that were not repeated were divided into 5 blocks, while the check varieties were planted repeatedly in each block. The effective selection criteria in this study were the yield, fruit weight,  number of branches, and total fruit number. The priority character besides yield in this study is fruit weight with high and normal narrow-sense heritability. The index selection results from this study showed that there were a good 32 tomato lines to be continued in the F4 generation. t was considered effective in the selection of tomato lines of the F3 generation.

Abstract. Heryanto FSS, Wirnas D, Ritonga AW. 2022. Diversity of twenty-three sweet corn (Zea mays L. saccharata) varieties in Indonesia. Biodiversitas 23: 6075-6081. One of the most well-known vegetables in Asian countries, including Indonesia. Improving superior sweet corn varieties is important in the face of increasing biotic or abiotic stresses and consumer preferences for agricultural products. Plant breeding programs rely on genetic variability and heritability information. Therefore, this study aimed to quantify the genetic diversity and heritability of 23 commercial sweet corn varieties in Indonesia. The study was conducted at the Leuwikoppo experimental field of the Department of Agronomy, Faculty of Agriculture, IPB University, Bogor, West Java, Indonesia, from August to November 2021. Twenty-three commercial sweet corn varieties (hybrid and open-pollinated) from Indonesia were planted in the field employing a randomized complete block design with 3 replications. Ear shelf life and green ear yield had a high genotypic coefficient of variation (GCV), whereas plant height, ear height, stalk length, 100-grain weight, total soluble solids, green ear shelf life, green ear weight, and ear weight had a moderate GCV. Stem diameter, leaf number, leaf length, leaf width, 50% days to tasseling and silking, anthesis, silking interval, ear length, ear diameter, ear length with the kernel, ear length percent with the kernel, kernel length, and kernel width had a low GCV. High genetic diversity was present among the trialed varieties, indicating their high potential for improvement.
