Morphological and molecular characteristics of the species Cosmocerca commutata and C. ornata (Nematoda: Cosmocercidae) in Uzbekistan. Biodiversitas 24: 4609-4616. In studies on the helminth fauna of amphibians conducted by numerous researchers, two species of the genus Cosmocerca, namely Cosmocerca ornata (Dujardin, 1845) Diesing, 1861 and C. commutata (Diesing, 1851), have emerged as the most common and extensively studied. This paper elucidates the morphology and molecular characteristics of two nematode species, Cosmocerca commutata and C. ornata, found in the intestines of Pelophylax sp. and Bufotes pewzowi in the Ferghana Valley of Uzbekistan. The captured amphibians were individually kept in plastic bags with water and vegetation until the examination. It is the first record of B. pewzowi as a host for both species C. commutata and C. ornata. Standard methods were used for the fixation and processing of nematodes. These nematodes ?orphologically differ in body size, relative length of spicules and gubernaculum (C. commutata has twice the length of gubernaculum and spicules compared to C. ornata), shape and number of plectanes (C. commutata has 8 pairs of plectans and 4 pairs of caudal papillae, while C. ornata has 5 pairs of plectanes and 3 pairs of caudal papillae), and tail morphology and length. The morphological evidence supports that C. commutata and C. ornata commonly parasitize anurans in Uzbekistan. This study provides molecular data of both species and their sequences differed by 1.5% (6 bp) in the ITS1+5.8S+ITS2 region. Each species of them formed a well-supported clade in the phylogenetic tree. Our findings contribute to the comprehension of species and genetic diversity of the genus Cosmocerca in Uzbekistan.

Abstract. Praptiwi, Palupi KD, Ilyas M, Marlina L, Fathoni A. 2023. Diversity of endophytic fungi isolated from tree spinach Cnidoscolus aconitifolius (Euphorbiaceae) and their bioactivities. Biodiversitas 24: 6229-6239. Endophytic fungi associated with medicinal plants have been known as a reservoir for therapeutic compounds. Cnidoscolus aconitifolius I.M. Johns (Euphorbiaceae), which possesses several local names, including papaya jepang in Indonesia, has been historically used as traditional medicine. However, research on the bioactivity of endophytic fungi associated with it has yet to be conducted. The purposes of the study were to isolate endophytic fungi from C. aconitifolius and determine their bioactivity, i.e., antibacterial activity against Escherichia coli and S. aureus and free radical scavenging activity representing antioxidant activity. Identification of endophytic fungi was carried out based on their morphological characteristics. Total phenolic and flavonoid contents were determined by the spectrophotometric method. The qualitative antibacterial and antioxidant bioactivity screening was performed using the TLC-bioautography method. The quantitative analysis of antibacterial and antioxidant activity was conducted by microdilution to obtain IC50 and MIC values. In total, 14 endophytic fungal isolates were successfully isolated. Antioxidant analysis showed that one isolate had strong antioxidant activity, and one had moderate growth inhibition activity against E. coli. The total phenolic content strongly correlates with the AAI value as an indicator of antioxidant activity. The findings of this study reveal that Phomopsis PjBg-1, an endophytic fungus from C. aconitifolius, is identified as Phomopsis sp. LH243 possesses strong antioxidant activity and moderate antibacterial activity against E. coli. Endophytic fungi from C. aconitifolius have the prospect of being an alternative to antioxidants and antibacterials. The isolation of compounds responsible for bioactivities needs to be further studied.

Abstract. Wiadnya DGR, Harlyan LI, Rahman MA, Mustikarani SMI, Nadhiroh ENS, Taufani WT. 2023. Stock status and supporting species of anchovy fisheries in the northern of East Java, Indonesia. Biodiversitas 24: 4775-4782. Anchovy, a commercial fish with high demand in local and international markets, was selected as the Unit of Assessment (UoA) of the Fisheries Improvement Project (FIP) based on the priority fisheries consultation process under the Marine Stewardship Council certification in Indonesia. One of the criteria is the availability of stock status and information about supporting species. This research was conducted from January to May 2022 at Lekok Fishing Port, Pasuruan, and Karangagung Fishing Port, Tuban, using primary data collection methods, including a fishery survey to obtain data species composition data and fishing coordinates. The secondary data were the catch-effort data from 2013 to 2021 collected from both fishing ports. Data analysis was carried out by the surplus production models, Schaefer and Fox model, to determine the total allowable catch (TAC) and the exploitation rates of anchovy fisheries. The participatory mapping method was carried out to obtain data on the distribution of fishing grounds and catch composition. The exploitation rate of anchovy (Stolephorus commersonnii) and large anchovy (Encrasicholina punctifer) landed in the Lekok fishing port was 76% and 75% (moderate), respectively. In this situation, fishing efforts can still be increased, as TAC value for anchovy fisheries' sustainability. On the other hand, the exploitation rate of anchovy in the northern waters of Tuban is 105% (over-exploited), which is necessary to reduce fishing efforts for its sustainability. The classification of supporting species resulted in 19 identified species in the Lekok fishing port, covering three target species: two endangered threatened and protected (ETP) species, two main primary species, six minor primary species, one major secondary species, and five minor secondary species. While in Karangagung fishing port, eight species were identified, consisting of 1 major primary species, five minor primary species, and two minor secondary species. The supporting species' information is required to complete FIP Anchovy fisheries.

Pathogenicity of entomopathogenic fungi to eggs, larvae, and adults and their effects on development of Aedes albopictus. Biodiversitas 24: 4766-4774. No information is available on the effect of fungi on the development of Aedes albopictus and the effectiveness of fungi in killing its eggs, larvae, and adults. The aim of this research was to determine the pathogenicity of entomopathogenic fungi on the eggs, larvae, and adults of Ae. albopictus and to investigate the effects of the fungi on the development of Ae. albopictus. The fungal species identified molecularly were Aedes albopictus, Beauveria bassiana, Metarhizium anisopliae, Penicillium citrinum, and Talaromyces diversus. The results showed that entomopathogenic fungi negatively affected the development of Ae. albopictus and could decrease the eggs laid by adult females and shorten adult longevity. M. anisopliae was the most pathogenic fungal species (mortality rate 100%) for eggs, larvae, pupae, and adults of Ae. albopictus. However, B. bassiana, P. citrinum, and T. diversus were also found pathogenic to them. The effect of eggs treated with entomopathogenic fungi negatively affected the larval, pupal, and adult stages of Ae. albopictus. These findings highlighted that M. anisopliae, B. bassiana, P. citrinum, and T. diversus have insecticidal activity against Ae. albopictus eggs, larvae, pupae, and adults. Therefore, further investigation is needed to develop these fungal species into ovicides, larvicides, and adulticides to control Ae. albopictus.

Short-term dynamics and growth parameters of cyanobacteria and microcystins in freshwater from the Sidi-Yacoub dam, North-east of Algeria. Biodiversitas 24: 5598-5609. The study aimed to establish the inventory of the blue-green algae species and monitoring the monthly evolution of their specific and total cell biovolume in Sidi-Yacoub dam, located northwest of Algeria from February 2021 to January 2022. At the same time, a monthly assessment of particulate and dissolved microcystin concentrations, chlorophyll pigment concentrations, and certain physicochemical parameters has been conducted. Eight species were listed, cited in descending order of their average relative abundance are: Microcystis aeruginosa (54.05%); Oscillatoria limosa (23.68%); Merismopedia sp. (8.34%); Anabaena cylindrica (3.53%); Aphanizomenon sp. (2.41%); Microcystis botrys (2.66%); Woronichinia sp. (2.95%) and Chroococcus limneticus (2.34%). The maximum value of the total biovolume, was recorded during December (27.4x106 µm3.L-1). The maximum concentration of chlorophyll-a and phycocyanin were respectively 55.26 and 47.46 µg L-1, recorded in September 2021. The microcystins detected where mainly in the cell-bound form, reaching a maximum concentration of 0.134 µg L-1 in October 2021. The principal component analysis shows a strong positive correlation between the total monthly cell biovolume, particulate microcystin (MCP) and some physicochemical parameters. M. aeruginosa has been identified as the primary producer of microcystins found in the eutrophic waters of the dam, with these microcystins being predominantly maintained in an intracellular form throughout most of the year.

Molecular detection of Babesia infection in cattle in Yogyakarta, Indonesia. Biodiversitas 24: 4192-4198. Babesiosis is a tick-borne disease caused by hemoprotozoa that poses a significant threat to livestock production worldwide, including in Indonesia. This study aimed to assess the prevalence and molecular characterization of Babesia sp., the causative agent of babesiosis, in cattle from multiple regions in Central Java, Indonesia. The disease has had substantial negative economic impacts, highlighting the need for accurate prevalence data and effective disease control measures. A total of 13 blood samples were collected from cattle exhibiting symptoms of hematuria and babesiosis. The samples were obtained from smallholder farmers who reported these cases to local veterinarians in various regions of Yogyakarta, Indonesia. The farmers were selected based on their proximity to veterinary clinics and willingness to participate in the study. Upon sample collection, each blood sample was subjected to microscopic examination using Giemsa-stained blood smears. The examination aimed to identify the presence of Babesia parasites within the red blood cells. Positive samples, indicating the presence of Babesia infection, were further analyzed by molecular assay. Molecular tests were performed using Polymerase Chain Reaction (PCR) to detect the DNA of Babesia sp. Two specific genes were targeted: cytochrome b oxidase (cytb) and 18S small subunit ribosomal RNA (18S rRNA). PCR amplification was carried out following established protocols, and the resulting products were visualized using gel electrophoresis to confirm the presence of Babesia DNA. Statistical analysis was conducted to compare the sensitivity and efficacy of the two PCR methods (cytb and 18S rRNA). The data obtained from this study contributes to our understanding of the occurrence of babesiosis in Central Java's cattle population. The findings underscore the need for comprehensive babesiosis disease surveys to obtain accurate prevalence estimates and facilitate the development of effective disease control strategies. Moreover, the more sensitive amplification targeting the cytb gene holds promise for improved diagnostic and surveillance efforts. These insights are crucial for combating babesiosis and mitigating its economic impact on livestock production.

Abstract. Maghfiroh RN, Suwarno WB, Sobir, Gunawan E. 2023. Assessment of relationship among traits and genotypes for melon (Cucumis melo) breeding. Biodiversitas 24: 4523-4531. Melon (Cucumis melo L.) is widely consumed as fresh fruit or juice, and therefore deeper analyses of fruit quality are needed for future breeding purposes. Breeders have a role in providing improved melon varieties that suit consumer ideotypes or preferences. This study aimed to (i) evaluate the performance of melon genotypes, (ii) estimate the correlation between quantitative traits, and (iii) determine the clustering among genotypes based on plant and fruit traits. This research evaluated 24 genotypes from inodorus, reticulatus, and makuwa groups that were carried out from March to June 2022 at a greenhouse in the Tajur experimental station of the Center for Tropical Horticulture Studies, Bogor, Indonesia. IPB HD 2-111 and IPB HD 2-100 had good quality based on soluble solids content, fruit shape, flesh texture, and rind color, and G30 was relatively superior on flesh thickness, fruit weight, and vitamin C. Based on the multiple regression analysis, it was known that petal width, petiole length, and days to harvest were significantly affected the proportion of fruit cavity width to the fruit diameter. The adjusted coefficient of determination (R2) was 0.569. Cluster analysis grouped 23 genotypes into three main clusters based on the neighbor-joining method. These results may be useful for selection in melon breeding programs.

Abstract. Nuralieva U, Tajiyev K, Sheralieva Z, Toishimanov M, Moldakhmetova G, Temirbayeva K, Tajieva A. 2023. Geometric morphometric characteristics of Apis mellifera honeybee in Kazakhstan. Biodiversitas 24: 4402-4409. This study was conducted in the whole beekeeping region of Kazakhstan. We researched morphological studies using (45 colony bees in 5 main regions in various geographical and climatic zone environmental conditions. The obtained data were compared with the Apis mellifera carnica reference samples, Apis mellifera caucasica, and Apis mellifera mellifera. Morphometric identification was carried out using 19 landmarks by IdentiFly software. Almost all colonies showed belonging to the lineage C (about 85%), by subspecies to A. m. carnica, A. m. caucasica showed 11%, A. m. mellifera belonged to only 4%. Furthermore, using multivariate principal component analysis compared honeybee lineages between reference samples. There were significant differences in wing landmarks between lineages and subspecies (Mahalanobis square distance). By Mahalanobis, distances between the evolutionary lineages M differ from line C by 15.93 and from line O by 19.03 units. In contrast, the distance between lines C and O is insignificant, with satisfactory reliability. The forewing wing size evaluated centroid size, which showed the highest degree of honeybees in the west and south Kazakhstan zones. The percentage of variation in wing centroids was relatively small between analyzed samples (less than 1%) but extremely changed for reference samples A. m. carnica, A. m. caucasica, A. m. mellifera, where log centroid sizes showed 1.33%, 4.18%, and 4.67%, respectively. The results showed significant differences between populations in different climatic zones.

Abanfan matilon, a local wisdom in marine ethnoconservation system on the coast of Liki Island, Sarmi District, Papua, Indonesia. Biodiversitas 24: 4693-4701. Liki Island is the outermost part of Sarmi District, Papua, Indonesia, which is directly facing the Pacific Ocean and bordering Papua New Guinea. This island is unique in its geographical features and is characterized by the local wisdom of the people known as Abanfan matilon. Therefore, this study aimed to document the knowledge, participation of people, and the role of the local wisdom of Abanfan matilon in the marine ethnoconservation system. The study was conducted from June to November 2022 using a survey method based on semi-structural interviews. The results of the study of 50 respondents showed that (i) the community knowledge about local wisdom of Abanfan matilon, possed by the Liki Island village community, had transcended from one generation to another through oral stories from parents, (ii) the role of the community in implementing local wisdom was substantial, starting from the planning, preparation, and implementation up to the harvesting activities through local traditional ceremonies, and (iii) the implementation of local wisdom of Abanfan matilon was carried out by community leaders from various sectors, including traditional, church, or local government. In conclusion, local wisdom traditions in Abanfan matilon exhibited the characteristics of a marine ethnoconservation system.

Biodiversitas 24: 5343-5351. The seaweed industry is strategically important in Indonesia, comprising over 40% of the nation's aquaculture annual production. Despite the industry's promising growth, various challenges and problems remain, including intense epiphyte infestation. Hence, investigating the economic value of these epiphytes could provide new opportunities for potential industrial applications. Epiphytic algae is commonly found growing on commercial seaweeds which causes decreased biomass and increased risk of crop failure. Information regarding these epiphytic algae remains limited. This study investigates the molecular identification of the abundant epiphytic macroalgae found in one of the largest seaweed farms in, Seriwe Bay, Lombok, Indonesia. In addition, the epiphytic bioactive activity is also evaluated for further industrial potential. Molecular identification with the mitochondrial marker COX1 identifies the epiphytic macroalgae as Gracilaria edulis (S.G.Gmel.) P.C.Silva. The Indonesian G. edulis is closely related to the G. edulis specimens from Malaysia (JQ026083.1), Philippines (KY995636.1 and KY995635.1), and Thailand (JQ026088.1). The outgroup used was the G. edulis specimen from India (KP099563.1) because it shows the most distinct relationship to the other specimens. Extracted agar of Seriwe G. edulis shows moderate yield (21%) and low gel strength (134 g/cm2). The phytochemical content analyses show that G. edulis agar has a TPC value of 3.65 ± 0.52 mg GAE/g and promising antioxidant activity (DPPH IC50 = 797.40 ± 1.50 µg/mL; ABTS IC50 = 558.40 ± 1.44 µg/mL). Further phytochemical profiling with GCMS shows various promising major constituents such as tetradecanoic acid, neophyte diene, pentadecanoic acid, and hexadecanoic acid. Therefore, the findings suggest that G. edulis displays potential applications in the functional food and cosmetic industry.
