Abstract

Based on some findings that some plants are tolerant to contaminated media, this research was conducted to study more thoroughly about characters and potencies of some of them as hyperaccumulators. Three of the most tolerant plants were studied in this research i.e Centrocema pubescence, Calopogonium mucunoides,and Micania cordata. The plants were grown in different waste media, i.e. tailing from PT. Aneka Tambang (ANTAM) and people mine waste. Both waste have different characters, physically and chemically. Waste of PT ANTAM major contaminant was cianide (Cn) whereas people mine waste major contaminant was mercury (Hg). This different characters resulted in different plant responses. The plants grown under PT ANTAM waste media gave better performance than that grown under people mine waste media. The most tolerant species was C. pubescence followed by M. cordata and C. mucunoides. Ability in metal accumulation of C. mucunoides was the highest, followed by M. cordata and C. pubescence.The results raised some-prospects for phytoremediation technology for rehabilitating contaminated mined lands. © 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: Centrocema pubescence, Calopogonium mucunoides, Micania cordata, hyperaccumulators, phytoremediation

Abstract

The potential of phosphate solubilizing bacteria/PSB (Bacillus megaterium, B. pantothenticus, Chromobacterium lividum and Klebsiella aerogenes) were used as biofertilizer to increase the fresh leaf production of green mustard (Brasica caventis Oed.). An experiment was conducted at green house condition. The experiment were used 18 treatments such as single isolate of potential PSB (A,B,C,D), inoculants contain two isolates of potential PSB (E,F,G,H,I,J), inoculants contain three isolates of potential PSB (K, L, M, N), inoculants contain four isolate of potential PSB (O), chemistry fertilizer (P = control 1), organic fertilizer (Q = control 2), and without fertilizer (R = control 3). The treatments were arranged in Completely Randomized Design (CRD) with 5 replications. The result showed that the inoculants of potential PSB increased the fresh plant production of green mustard. The mix of four isolates of potential PSB (inoculants O) was the best to increase the fresh plant production of green mustard until 32.87% than other PSB inoculants, 207.84% than control 1,217.23% than control 2, and 930.60% than control 3. © 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: BPF/PSB, Wamena Biological Garden, Brasica caventis Oed.

Abstract

Nugraha Y, Utami DW, Rosdianti I, Ardie SW, Ghulammahdi M, Suwarno, Aswidinnoor H. 2016. Markers-traits association for iron toxicity tolerance in selected Indonesian rice varieties. Biodiversitas 17: 753-763. Ferrous iron toxicity is a mineral disorder frequently occurring under flooded soils condition where rice is cultivated. Here we study identification the Single Nucleotide Polymorphism (SNPs) markers associated with iron toxicity tolerance characters. The phenotypical data was collected from exploiting of twenty-four rice genotypes that were grown under Yoshida + 0.2% agar solution with treatment of 400 mg. L-1 Fe2+ and control conditions. The same genotypes were grown in iron toxicity acute and control sites at Taman Bogo, Lampung Province, Indonesia. The Principle Component Analysis (PCA) of the phenotypic data showed that 18 rice genotypes were selected representing grouping of related characters to iron toxicity condition. The genotyping of selected genotypes was carried out using multiplexes of 384 SNPs Golden Gate Illumina© assay. We identified, TBGI380435 which located on 14.45 Mbp of chromosome 9 was associated to leaf bronzing and relative shoot weight characters in the greenhouse experiment. The marker was associated with heavy metal transport detoxification (HTDT). The results are expected to assist in locating the potential candidate genes or Fe toxicity tolerance and to allow for precise marker-assisted selection. This research will serve for rice improvement through marker-assisted breeding and genomic selection in Indonesia. Abstract

Research on the red rice fermented by Monascus purpureus had been done. The rice consisted of 2 cultivars such as Bah Butong and BP 1804 IF 9. The aim of the research was to know the content of the pigment and lovastatin of the fermentation result/ angkak. Angkak was powdered by using blender. To measure the content of pigment, the powder was extracted by methanol. By using spectrophotometer the content of the pigment could be measured with 390 nm wave lengths for yellow pigment and 500 nm for the red pigment. For lovastatin the powder was extracted by acetonitrile and H2SO4. By using HPLC the content of lovastatin could be measured. The results showed that the highest pigment content for yellow pigment was on the PB 1804 IF 9 cultivar, and red pigment was on the Bah Butong cultivar. The highest lovastatin content was on the BP 1804 IF 9.Key words: Monascus purpureus, red rice, pigment, lovastatin.

Abstract

Several isolates of Lactobacillus had been isolated from Indonesian fermented foods. Four isolates where isolated from tongcai (TT2), saguer drink (Sg.Mnd.N2), pindang ikan selar (PSl1) and sawi asin (S5) had been used in this research. The aim of the research was to get Lactobacillus isolates as probiotic candidate and to know its resistance to low pH. The treatment were several concentration of low pH of MRS broth medium, i.e. 2; 2.5; 3; and 6.5 (optimal pH). The optical density (OD) was measured after 24, 48, 72, and 96 hours respectively, with three replication by using spectrophotometer (λ = 600 nm). The results showed that all Lactobacillus isolates (i.e. TT2, Sg.Mnd.N2, PSl1, and S5) could been used as probiotic candidate because they were resistant to all of low pH used. © 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: low pH, Lactobacillus, probiotic

Abstract

The aims of the research were to inventory the flora of Sulawesi, especially orchids in the Mount Tinombala Natural Reserve and to collect the plant materials for planting as a collection plants in Bali Botanical Garden. The method used in this research was explorative method at the place with altitude more than 700 m above sea level. The result of the research was 50 collection number of orchids which was consist of 72 specimens, 19 families and 24 species found in the Mount Tinombala Natural Reserve, Tolitoli Regency, Central Sulawesi. Two numbers of them that unidentified yet called as a genus dubious. Abstract

The research on the using of Areca catechu seeds and roots extract contain selenium which fermented by Acetobacter–Saccharomyces consortium as a gargle had been conducted. The aim of the research was to know the influence of Se content of the seed and the root of Areca catechu by fermenting of Acetobacter–Saccharomyces consortium on the growth of Streptococcus mutans bacteria. The research consisted of four stages i.e. fermented the extract, isolated S. mutans, tested the activity of the antimicrobial and analyzed selenium concentration in the extract. The result showed that the concentration of selenium has fluctuation. The highest content of selenium of the seeds extract at 2.2 g/L concentration was 3.05 ppb on the fifteenth day while on the root extract at 8.8 g/L was 14.03 ppb on the twenty first day fermentation. Both of the extract has potential used as antiseptic gargle. Abstract

Phytopharmaceuticals and phytochemicals evaluation on two species of Calophyllum (C. inophyllum and C. soulatri) had been done. Phytopharmaceuticals had been carried out by macro- and microscopic observation and also phytochemical screening by Ciuley Method (1984). Macromorphology of two species of Calophyllum was quite different. Leaf surface of C. inophyllum were obovate lamina, entire margin, obtuse apex, symmetrically base, petiole 1.5-2.0 cm, upper surface were green, shine, glabrous, occurrence of oil gland, and lower surface were long (hirsute), midrib, pinnate venation. C. soulatri were ovate lamina, entire margin, acute apex, symmetrically base, petiole 1.5-2.0 cm, upper surface were green but not shine, glabrous, no occurrence of oil glands and lower surface were long (hirsute), midrib, pinnate venation, green, and glabrous. Leaf dimension of C. inophyllum (± 20x10 cm2) was wider than C. soulatri (± 10x5 cm2). Leaf micromorphology of two species was also different in stomata type, C. inophyllum was paracytic type while C. soulatri was anisocytic type. Oil glands in C. inophyllum were higher than C. soulatri. There were no hair glands that found in these species. Micromorphology bark identified of two species showed that the number of oil glands in C. inophyllum is more than C. Soulatri, Ca oxalate of C. inophyllum is in simple and twin forms which C. soulatri is loose aggregates of numerous prisms which have grown together to form a roughly spherical mass which has projecting points and angles all over surface. Phytochemical screening identified that these species contents are have similar chemical groups (volatile oil, fatty acids, steroids/ triterpenes, tannin, flavonoids and reducing sugar). © 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: Calophyllum inophyllum, C. soulatri, Clusiaceae, phytopharmaceuticals and phytochemicals evaluation.

Abstract

Kedawung (Parkia roxburgii G. Don.) is one of plants that originally could be found in almost all parts of Java Island. Due to lack of attention, at present situation this plant could be categorized as an endangered species. This plant species distributed widely in Africa. In several African countries, this plant has an important position either as food resource or alternative medicine. In Indonesia, traditionally kedawung tree often use as medicine, especially as part of “jamu gendong” formulation. This work was focused on the study of phytosterol content and it distribution. The research result show that almost all parts of kedawung tree have a significant content of phytosterol, which is dominated by beta-sitosterol. Abstract

Plant response to drought stress depends on the physiological and biochemical response of the individual plant. Plant resistant toward the drought stress is controlled by so many genes that identification toward the drought stress resistant plant could use one or several characters related to drought. In order to find out the relationship between stomata density and the drought stress resistant plant, observation on the stomata density at the lower leave epidermis of various rice (Oryza sativa L.) somaclones originated from radiated callus using gamma ray. The observed varieties are Gajahmungkur, Towuti, and IR 64. The plant observed is the ones selected drought resistant by using PEG, root penetration test to paraffin layer, and proline content test. Stomata density observation on several somaclone shows that the radiation treatment to the three varieties may change the stomata width, length and density. Stomata with the lower density than the control shows the higher resistant to the drought viewed from the variable of filled grain, at the stress treatment given in the experiment. Gajahmungkur variety, which is basically higher land rice, has the lower density of stomata than that of Towuti and IR 64. In conclusion, the stomata density can be applied as one indicator on the stress resistant level of rice. © 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: stomata, drought resistant, rice (Oryza sativa L.).
