Abstract. Amini F, Bayat L, Hosseinkhani S. 2016. Influence of preharvest nano calcium applications on postharvest of sweet pepper (Capsicum annum). Nusantara Bioscience 8: 215-220. The effect of preharvest nano calcium treatment on the quality, physicochemical parameters and antioxidative enzyme activities of sweet pepper fruits were evaluated. For this purpose, sweet pepper plants treated with three concentrations (0, 4 and 8 g/L) of nano calcium in five steps (50, 60, 70, 80 and 90 days after pepper planting). 100 days after pepper planting, fruits were harvested and packaged in plastic bags and stored at 20±3 0C, then total soluble solids, pH, weight loss, carotenoids, electrolyte leakage, lipid peroxidation, titrable acidity, ascorbic acid, chlorophyll, calcium, protein and the activities of total antioxidant enzymes, catalase, and guaiacol peroxidase were evaluated at 0, 7, 14 and 21 days of storage period. Nano calcium treated fruits had lower levels of total soluble solids, pH, weight loss, carotenoids, electrolyte leakage, lipid peroxidation and higher levels of titrable acidity, ascorbic acid, chlorophyll, calcium and protein compared to the control plants. The increase percentage of titrable acidity, ascorbic acid, chlorophyll a, chlorophyll b, total chlorophylls, calcium and protein contents in plants treated with 4 g/L nano calcium and 21 days storage period were calculated 68.86%, 41.89%, 81.17%, 78.57%, 80.81%, 21.46% and 83.30% respectively. The maximum activities of total antioxidant enzymes, catalase and guailcol peroxidase were observed in 21 days control plants that application of nano calcium decreased these antioxidant activities. In most parameters 4 g/L concentration was more effective. Therefore, commercial application of preharvest nano calcium (4 g/L) can be considered for the maintenance of quality of sweet peppers during storage and marketing.

In silico and in vivo wound healing studies of ursolic acid isolated from Clematis gouriana against GSK-3 beta. Nusantara Bioscience 8: 232-244. Clematis gouriana Roxb. (Ranunculaceae) is an endemic medicinal plant of Western Ghats. Ursolic acid (UA) was isolated from the methanolic extract (ME), which was characterized by spectral studies, and was subjected to in silico studies on Glycogen synthase kinase 3-? (GSK3-?) protein, to inhibit protein function. UA showed least binding energy (-13.457 ?G) towards the binding site and molecular dynamics studies showed minimum potential energy and greater stability with target protein. Thus, UA may be considered as a potential inhibitor of the GSK3-? protein. The wound-healing activity was assessed by excision, incision, and dead space wound models on Wistar strain rats. The results of excision, incision, and dead space wound models showed significant activity. There was a significant increase in skin-breaking strength in rats treated with UA (562.36 ± 7.60 g). The effects of ME and UA on the dead space wound model were pronounced, with a significant increase in the weight of granulation tissue (26.33 ± 0.25 mg), tensile strength (647.00 ± 0.71 g), and hydroxyproline content (1793.83 ± 0.64 µg/100 g). Thus, UA-treated animals showed excellent wound-healing activity. Animals treated with nitrofurazone, ME and UA showed increased collagenation and decreased accumulation of macrophages at the site of injury.

Abstract. Cahyaningsih R. 2016. Characterization of phenotypic diversity of a medicinal plant Lunasia amara. Nusantara Bioscience 8: 245-251. Characterization of phenotypic diversity is an important first step in the conservation of plant species and in plant breeding for human purposes. Lunasia amara Blanco (‘kemaitan’, ‘sanrego’) is a medicinal plant native to Indonesia, used traditionally as an aphrodisiac. However, there is little knowledge about genetic diversity in this species, and even its taxonomic status remains under review. The study reported here describes and classifies phenotypic diversity among Lunasia amara and its relatives, focusing on vegetative characters assessed in the extensive collection of the species held in Indonesian Botanic Gardens. Based on results of a similarity analysis using SIMQUAL (Similarity for Qualitative Data) employing SM (Simple Matching Coefficients), the accessions exhibit low phenotypic diversity as indicated by high similarity coefficients. Nevertheless, the accessions could be divided into two not very distinctive groups. Based on PCA (Principal Component Analysis), none of the observed characters were of outstanding significance in partitioning the diversity among the accessions. Due to their high similarity value, and despite the wide geographic area represented in the accessions, the study lends support to the belief that they belong to a single taxon, Lunasia amara var. amara. For conservation of Lunasia spp, this study suggests further collection activity is not a priority. Studies on the quantitative content of medicinally active constituents as well as phylogenetic studies of molecular diversity would test this conclusion and support a plant breeding program to develop the usefulness of this plant for human purposes.

Nusantara Bioscience 8: 252-257. This research was aimed to investigate the antimicrobial potential of the leaves extract from Carica papaya, Ipomoea aquatica, Piper betle and Alpinia galanga stem against Aeromonas hydrophila, Pseudomonas sp., Escherichia coli, and Saprolegnia spp.; the pathogenic microbes in aquatics. These plants were dried and extracted with water and ethanol, separately. The extract was then tested for its inhibition effect to the microbial by in vitro agar disc diffusion (ADD) at the concentration of 100, 200, 400, 600, 800, and 1,000 ppm and minimal inhibitory concentration (MIC) at the concentration of 600, 800, and 1,000 ppm. The result showed that water or ethanol extract f of plants showed potent inhibitor against the aquatic microbial. The ethanol extract of P. betle showed the highest inhibition at 800 ppm, with inhibition zones to A. hydrophila, and Pseudomonas sp. 12.3 mm, E. coli, and Saprolegnia spp. 12.7 mm. One thousand ppm ethanol extract of P. betle could inhibit the best microbes growth using MIC method to A. hydrophila 189.7 cfu/mL, Pseudomonas sp. 253.0 cfu/mL, Saprolegnia spp.169.3 cfu/mL, and 800 ppm to E. coli 172.7 cfu/mL.

Abstract. Rahayu EMD. 2016. Handling and propagation of Dendrobium ‘Iriana Jokowi’ in Bogor Botanic Gardens, Indonesia. Nusantara Bioscience 8: 258-263. Dendrobium ‘Iriana Jokowi’ is a new hybrid in Indonesia which has not been mass propagated yet. Mass propagation is necessary to ensure seedling availability. The orchid has been grown in the Orchids Greenhouse of Bogor Botanic Gardens since July 30, 2015. Handling and care for Dendrobium ‘Iriana Jokowi’, includes watering, provision of fertilizer, fungicides, and insecticides. Propagation has already been done is using seed culture. The seeds of Dendrobium ‘Iriana Jokowi’ were sown in four types of culture media, which are modified Vacin & Went (VW), modified Hyponex (HS), modified Knudson’s C (KC), and Knudson’s C (KCA). The seeds were able to germinate in all four tested media. The highest germination rate in the first 3 months after sowing (3 MAS) were found in KCA media, which are 93.21%. Germinated seed was then subcultured to four types of media in order to induce the growth of leaves and roots. The media used were T1A, T1V, HS, and KCA. After 5 months of planting (5 MAP), result showed that all four media were able to provide significant effect on the number of leaves and roots. The highest number of average leaves grown was 4.8 and found in T1A media. On the other hand, the highest number of average of roots grown was found in T1V media (4,2 roots).

Short Communication: Preservation effect of Javanese turmeric and red ginger essential oils on coated frozen patin fillets. Nusantara Bioscience 8: 264-267. Due to antimicrobial and antioxidant properties of essential oil, Javanese turmeric and red ginger potentially used as preservation agents on food. Cassava starch-based edible coating enriched with Javanese turmeric and red ginger essential oils were applied to maintaining of patin fillets quality during frozen storage. Fillets quality were analyzed based on microbiological (total plate count) and chemical properties (total volatile bases; thiobarbituric acid value). Different concentration of Javanese turmeric and red ginger essential oil enrichment were studied (0, 0.1, and 1% (v/v of distilled water)). Javanese turmeric and red ginger essential oils enrichment on coating solution significantly inhibited microbial proliferation, retarded protein deterioration and delayed lipid oxidation in fillet during frozen storage for four months. Increasing essential oil concentration showed on the increase in maintaining fish quality. The results indicated that Javanese turmeric and red ginger essential oil addition on cassava starch-based edible coating could maintain quality and extend the shelf life of frozen fish fillets and can be used as an alternative fish fillets preservation.

Abstract. Utami R, Widowati E, Christy A. 2016. Screening and characterization of amylase enzyme in sweet orange (Citrus sinensis) juice clarification. Nusantara Bioscience 8: 268-272. The amylum compound of sweet orange cloudify the produced juice. Clarification by amylase could remove the cloudiness. The aimed of this study were screening the amylolytic bacteria from spoiled potato which are potential in clarification of sweet orange juice, and characterization of the resulted amylase (optimum pH and temperature, pH and thermal stability, KM and Vmax). The results showed that from thirteen bacterial isolates, three isolates (K-7, K-8, and K-11) produced enzymes that showed the best clarification activity on the sweet orange juice. The optimum pH of the isolate K-7, K-8, and K-11 enzymes were found to be 7, 7 and 6, respectively. The optimum temperature of the isolate K-7, K-8, and K-11 enzymes were found to be 35, 40 and 35°C, respectively. Enzymes of isolate K-7, K-8, and K-11 were stable in the pH range 3-6, 5-6, and 7-8, respectively. The enzyme of isolate K-7 and K-11 were stable at temperature 40-60°C while enzyme of isolate K-8 stable at temperature 40-70° C. Km values of isolate K-7, K-8, and K-11 enzymes were 0.2756 mg/mL, 0.0888 mg/mL, and 0.0956 mg/mL, respectively. Vmax values of isolate K-7, K-8, and K-11 enzyme were 0.0325 U/mL, 0.0164 U/mL, and 0.0134 U/mL.

Abstract. Suryantini. 2016. Effect of phosphorus, organic and biological fertilizer on yield of mungbean (Vigna radiata) under two cropping patterns. Nusantara Bioscience 8: 273-277. The research investigated the effect of phosphorus rates, organic and biological fertilizer on productivity of mungbean grown after rice (cropping pattern: rice-rice-mungbean) and after soybean (cropping pattern: rice-soybean-mungbean). Treatment was arranged in a split plot design with three replications. The main plots were (i) without organic and biological fertilizer, (ii) organic soil treatment (OST) and (iii) phosphate solubilizing biofertilizer (PSB). The subplots were phosphorus (P) fertilizer rates: 0, 100, 150 and 200 kg SP36/ha. The results revealed that the grain yield of mungbean grown after soybean was not affected by the application of OST or PSB, but it was increased by fertilizer P at 200 kg SP36/ha with an increase of 68%, from 0.60 t/ha (control) to 1.01 t/ha. Conversely, the grain yield of mungbean grown after rice was significantly affected by the interaction between fertilizer P and OST or PSB. The highest yield was obtained from the combination treatments of 150 kg SP36/ha + PSB and 150 kg SP36/ha + OST with an increase of 78% and 71% over control respectively. Increased grain yield was supported by the increase in number of pods, 100 seeds weight, and plant P uptake.

Nusantara Bioscience 8: 278-282.  Good packaging is necessary to prevent food spoilage before and after processing. The edible film maintains food quality and eco-friendly packaging. Cassava starch-based edible film is capable of being developed because of the approaching appearance of plastic packaging. The addition of essential oils can be used as an alternative to improve the antimicrobial properties of the edible film. Lemongrass oil consists of active compounds that shown antimicrobial activity. Therefore, the characteristics of cassava starch-based edible film incorporated with lemongrass oil were investigated. Concentration of lemongrass oils varied at 0%, 0.25%, 0.5%, 0.75% and 1%. The analyzed edible film characteristics include thickness, water vapor transmission rate, tensile strength, elongation, and antimicrobial activity. The results showed that the addition of lemongrass oil increased the thickness, tensile strength, and antimicrobial activity but decreased the elongation of edible films. The water vapor transmission rate of edible films was not affected by the addition of lemongrass oil.

Abstract

Abstract. Rasuluntari IN, Muhammad DRA, Praseptiangga D. 2016. Panelist acceptance level on milk chocolate bar with cinnamon (Cinnamomum burmannii) powder addition. Nusantara Bioscience 8: 297-300. Among confectionery products, milk chocolate bar is the most popular product. The development of Indonesian cocoa agroindustry and chocolate signature with special characteristics are required since Indonesia is the third largest cocoa producing country in the world. The effects of cinnamon powder addition in milk chocolate and its impact on the consumer acceptance of milk chocolate were evaluated in this study. Cinnamon-enriched milk chocolates were formulated by completely randomized design (CRD) with one factor which was concentration of cinnamon powder (5%; 10%; and 15%). Sensory evaluation (color, aroma, taste, appearance, overall) were determined by hedonic test with scoring method. The results showed that the level of panelist acceptance decreased with increasing concentrations of cinnamon powder added. Milk chocolate bar with the addition of 5% of cinnamon powder had the highest level of acceptance and preferences for some of the sensory attributes evaluated.
