Abstract

The research aimed to assess genetic variations, estimated heritability and genetic gain in resin yield of P. merkusii. The study was conducted on a half-sib progeny test of 4-year-old P. merkusii resin yield plantation at compartment 37C RPH Lumbir BPKH Samudra KPH Banyumas Barat, using incompletely-block-row-column design by sub-line system (sub-line SSO Jember 1, SSO Sumedang 1, Sulawesi 1, Sulawesi 2 and Jatim 2). Variables measured were average of resin yield from two drillings on the right and left trunk using 10 mm drill bit at 50 cm height from the ground. Genetic variation for resin yield between family was significant different except for sub-line SSO Jember 1. Estimated component variances of family for the sub-line SSO Sumedang 1, Sulawesi 1, Sulawesi 2 dan Jatim 2 were 8.92%; 9.87%; 7.53% and 6.74%, respectively. Individual heritability values were 0.39; 0.42; 0.36; 0.29 and family heritability were 0.59; 0.49; 0.50; 0.48 respectively for the sub-line SSO Sumedang 1, Sulawesi 1, Sulawesi 2 and Jatim 2. Expected genetic gains in resin yield were 14.5%, 10.5% and 6.6% using intensity of selection of 10%, 25% and 50%, respectively. Sub-line Sulawesi 1 had the highest estimated genetic gain and sub-line Jatim 2 was the lowest

Abstract

Research for developing Acacia hybrid (Acacia mangium x Acacia auriculiformis) breeding strategy through establishing hybrid seed orchard (HSO) has been started by BBPPBPTH. Two hectares of HSO was established in Wonogiri, Central Java to produce the Acacia hybrid seed. The orchard was laid out using an alternating rows design among the two pure parent species with spacing of 1 m x 3 m: 1 m within the species and 3 m between the species. The purpose of this study is to evaluate flowering synchronization and seed production in first year flowering session of four years age mother trees in the HSO. The unit area of observation was set up in 0.2 ha within the HSO. During the first flowering session, 100 mother trees (31%) were observed showing synchronized flowering time among the two parents species: 50 mother trees for Acacia mangium and 50 mother trees for A. auriculiformis. Seed production was still low in which only 38 out of 50 trees of A. mangium mother trees produced seed, whereas selected A. auriculiformis mother trees did not produce seed. The average viability of the collected seed was moderately low at 59.2%. Subsequent verification through DNA marker using SCAR and SSR revealed less than 1% of the collected seed were identified as true Acacia hybrid.

Abstract

In-Vitro contamination of plant tissue cultures by bacteria is one of the most serious problem in plant micropropagation. Ulin (Eusideroxylon zwageri) bacterial contamination in vitro may survive surface sterization of the explant because they are in interior tissue and some Bacterial spores can also survive the sterilization procedure even if they are un the tissue surface. Bacteria  secrete some sort of glycocalyx, an auter viscous covering of fibers extending from the Bacterium. This Study was conducted to examine the colony thickness and to observe the bacteria glycocalyx contaminant of ulin in vitro folowing three surface explant sterization treatments with fungisida, detergent, alkhohol,HgCl2  and Na2CLO3 as antimicrobial agents. The longer the sterilization time the lesser the bacterial colony thickness farmed. Transmission binocular microscopy showed that distribution of glycocalyx material surrounding coccus ulin bacterial contaminant cells were capsulated like Steptococcus lactis or Enterobacter aerogenes.

Abstract

Plants that have mycorrhizal symbiosis tend to be more resistant to the drought because external hyphae of mycorrhizal will expand the surface of water uptake and it can infiltrate into capillary pores so that water uptake for the host plant need will increase. This study aimed to know the response and adaptation mechanisms of mangium in facing drought stress, to investigate mycorrhizal roles in overcoming drought stress on mangium, and to select the origin of mangium seed source that is tolerant to drought stress. The study used mangium seedlings from four different seed sources and was arranged in split plot pattern of randomized block design, with three factors. The result showed that mangium root infected by mycorrhizal decreased along with increasing level of watering intervals. Mycorrhiza increased height-diameter growth and total biomass of mangium for the entire seed source. Total biomass growth of seven month mangium was significantly influenced by the origin of seed source. The highest total biomass was derived from F-1 Wonogiri (7.14 grams) and followed by Group B (6.82 grams), Group C (6.21 g) and Group A (5.84 g). Adaptation mechanisms of mangium seedlings during drought stress were keeping the water status of the plant, improving plant roots system, and thickening the leaf blades.

Abstract

Diospyros rumphii Bakh. is one of tropical commerciall wood species grouped in ebony. Ebony is the name of some species of commercial timber from genus of Diospyros (Ebenaceae). Seven species of them are commercial wood, which in the trade is known as ebony namely; D.celebica, D.rumphii, D.pilosanthera, D.lolin, D.ebenum, D.ferrea and D.macrophylla. Two of seven species ebony have the highest economic values namely; D.celebica and D.rumphii. Nowadays, the population D. rumphii in the nature are decreases due to over harvesting in the past. This study aims to determine genetic diversity of D. rumphii within populations, between populations and its relationship. The samples of D. rumphii in this study from four populations in North Sulawesi i.e Bitung, Talise, Talaud and Bolaang Mongondow. Genetic diversity was detected by isoenzym markers with seven enzyme i.e Glutamate oxaloacetate transmirase (GOT), Shikimate dehydrogenase (SHD), Esterase (EST), Acid phoshatase (ACP), Diaphorase (DIA), 6-Phosphogluconate dehydrogenase (6PG) and Glutamate dehydrogenase (GDH). The results showed that there are three enzym was detected polymorphisme i.e EST, GOT and GDH. Genetic diversity generated by three enzyme systems who are controlled by 5 loci and 15 alleles. The average percentage of polymorphic loci was100%. The number of total genetic diversity is 0.64,which are distributed within population 0.59 (91.24%) and between population 0.05(8.76%). Cluster analysis using Unweighted Pair Group Method with Aritmatic Means (UPGMA) based on Nei standard genetic distance showed three clusters:Bitung and Bolaang Mongondow, Talise, and Talaud.

Abstract

The breeding program of Melaleuca cajuputi subsp. cajuputi in Indonesia  was started  in 1994 by the Institute for Biotechnology and Forest Tree Improvement in cooperation  with CSIRO-Australia. An assessment  of progeny test of M. cajuputi at Paliyan at  23 months and at Sukun-Ponorogo at 36 months old were conducted to ascertain the  patterns of genetic variation in growth and foliar oil   and to  estimate genetic parameters in growth and foliar oil .  Based on the evaluation, genetic variation on growth and oil yield were high. Genetic gain estimation for growth and oil yield were 11.83 to 20.38 % in the progeny test seedling seed orchard of M. Cajuputi at Sukun, Ponorogo.

Abstract

Flowering stimulation were investigated to accelerate deployment of E. occidentalis. It was found that extended daylength (16-h) in juvenile plants stimulate flowers of very limited individuals/family with less number of buds than normal inflorescences. Extended daylength (16-h) promote flowering of most families from 2 year-old E. occidentalis plants. In combination with paclobutrazol (1 mg. a.i/mm), number of buds were doubled. Under 16-h daylength, juvenil plants were taller up to 25 weeks than under natural light but shorter for family 96-LM that produced more flowers than others. There were no height differences between 16-h daylength and in combination with paclobutrazol in 2-year old plants.

Abstract

Pinus radiata is the major softwood species planted in New Zealand and Australia. Endophyte fungi that have been extensively studied in conifers needles mainly based on fungal isolation and also rarely conducted in Pinus radiata. This study aimed to observe the optimum condition of P. radiata needles sample for direct extraction of fungal DNA and to identify the fungi associated with P. radiata needles in Plenty plantations by a direct PCR approach. Room temperature dried needles produced clear and consistent DNA amplification compare to fresh needles, and needles dried on drying room or drying machine. Capnodiales was the most diverse group and no fungal pathogen detected in this study. DNA sequence data from direct DNA extraction of P. radiata needles provided sufficient discrimination and identification of fungal species.

Abstract

Gyrinops versteegii (Gilg.) Domke is including one of superior agarwood-producing plants and naturally growing in Eastern Indonesia as Nusa Tenggara and Papua. Indonesia has been trading agarwood products both domestically and overseas which one of them is agarwood produced by G.versteegii. This study purpose was to develop an in vitro culture method for mass propagation of G. versteegii. Shoot induction conducted on MS medium supplemented with Benzyl Amino Purine (BAP) 0.7; 1.0; 1.5; and 2.0 mg/l. The shoots multiplication conducted on MS medium supplemented with the best concentration of BAP from shoot induction phase. The rooting of shoots conducted on half strength MS medium supplemented with interaction of Napthalene Acetic Acid (NAA) 0.01 mg/l with concentration of Indole-3-Butyric Acid (IBA) 1.0; 2.0; 3.0; and 4.0 mg/l. Epicotyl explant with a given concentration level of BAP 0.7 mg/l produce the highest rates number of shoots and shoot length compared to other explant respectively 4.8 shoots and 0.41 cm within 6 weeks. The best explant developments in the best medium able to promote the growth of the length and number of shoots are 0.28 shoots and 0.3 cm within 4 weeks. Explants easiest, quickest and most high sprouting ability as a factor of success in terms of multiplication is epicotyl. The combination treatment of material explant with concentration of BAP only affect to growth of shoots length. The combination treatment of NAA with concentration of IBA has no effect against root formation and growth rootlength.

Abstract

Micropropagation technique is applied to produce large number of plants. Juvenile explants and culture medium are two crucial factors that influence the success.The aims of this study were to determine culture medium that was effective to induce shoot and to produce plantlests from embryo culture of Pinus merkusii. Screening of five cytokines and five basal medium were carried aut. Then, explants wee cultured on the best cytokine and basal medium for plant regeneration. The results showed that BAP was the best cytokine type; LP and GD medium were the best medium compositions for shoots induction. GD medium supplemented by high consentration BAP showed the best medium for shoots induction. Solid GD medium containing 0,25uM BAO was effective for multiple shoots. Liquid GD medium without plant hormone was the best composition for shoots elongation. Half strength of GD medium without plan hormone gave the best result for rooting.
