Abstract

The aim of this research was to determine the viability of bacterial cells on different cryoprotectant agents. Three kind of cryoprotectant agents and two types of mixed cryprotectant were applied to evaluate the ability of bacteria to growth on Agar medium. Four isolates were used namely Escherichia coli ATCC 35218, Staphylococcus aureus ATCC 25923, Shigella flexneri ATCC 12022 and Bacillus subtilis ATCC 6051. All strains were preserved in 10% skimmed milk, 10% glycerol, 10% DMSO, 10% glycerol:10% skimmed milk and 10% DMSO:10% skimmed milk at -80°C for sixty days. Immediately after preserved, the bacteria were tested their viability at day 30 and 60 during storage. The enumeration of samples taken from cultures in NA media. Better survival of bacterial cells after freezing was shown in all tested cryprotectant agents. Highest microbial viability was shown in 10% skimmed milk, 10% glycerol and mixed 10%DMSO:10% skimmed milk. Hence, those three types of cryoprotectant agents can be recommended to preserve bacterial cell for long term storage.

Abstract

Central Sulawesi is a province produce eels (Anguilla marmorata) in Indonesia. Eels is the one commodity that has a high economic value. In the last years, eels trading traffic through the airport had increased. Fish health status to be one of the important issues in trade of eels. Genus Aeromonas is disease-causing pathogen on eels and as the etiologic agent responsible for a variety of infectious complications pathogenic bacteria. The purpose of this study is identify the various species of the genus Aeromonas on the eels which are traffic through Mutiara Sis Al jufri Airport Palu. This observation have been from April to December 2014. Specimens Anguilla sp. was obtained from supplier in the Palu city. Laboratory examination performed use conventional methods (Biochemistry) and performed in Microbiology Laboratory of Fish Quarantine Palu. The result showed from 396 eels have been identified, 128 of them are of the genus Aeromonas (32.32%). Aeromonas were identified some species among other: Aeromonas sobria, Aeromonas hydrophilla, and Aeromonas caviae. A.sobria is higher percentage in April and August 2014 (57,14%), A. hydrophilla in December 2014 (92,85%), and A. caviae August 2014 (21,43%). The existence of pathogenic bacteria in eels this should be a concern to prevent widespread dissemination through trade traffic.

Abstract

This study aims to acquire the diesel oil degrading microbia in Pare-Pare port. This study was conducted at the Laboratory of Microbiology, Department of Biology, Faculty of Science and Technology, UIN Alauddin Makassar. The method is descriptive exploratory study. The data was analyzed by tables and graphs. The results showed that three isolates of bacteria isolated from the Pare-Pare Bay that can degrade diesel oil as Bacillus sp., Psedomonas aeruginosa, and Alkaligenes feacalis. P. aeruginosa is the best bacteria in degrading diesel oil.

Abstract

Micorrhyza arbuscula fungi is one of endomicorrhyza group with hiphae stucture called arbuscule. Micorrhyza fungi able to associate with various plants include crop plant. This research aim to know the genus of micorrhyza arbuscule fungi on crop plants root and to know micorrhyza structure on it. The result showed that the highest diversity of micorrhyza was found on potato root, then on strawberry root and the last on tomato root. There are 16 genus was found, they are 11 genus Glomus, 4 genus Acaulospora and 1 genus Ambispora. The arbuscule and vesicle structure was also observed.

Abstract

Phalaenopsis amabilis (L.) Blume or Moth Orchid is Indonesia’s native orchid. In its cultivation, the crown rot which is caused by F. oxysporum is one of diseases that usually attack P. amabilis. The aim of this research is to assess the potency of endophytic fungi Sistotrema sp., Ceratorhiza sp. and Moniliopsis sp., that were isolated from healthy P. amabilis roots, in inhibiting F. oxysporum growth in vitro. The experiment of antagonist test was arranged in complete randomized design by three replications. The in vitro antagonist test was done on PDA and the parameter was the percentage inhibitation of F. oxysporum growth by endophytic fungi. The collected data were analyzed by one way anova and DMRT. The result showed that the three endophytic fungi isolated from P. amabilis roots had the potency in inhibiting F. oxysporum. In vitro antagonism test result showed that Moniliopsis sp. had the highest inhibition percentage (71,80%) among Ceratorhiza sp. (67,70%) and Sistotrema sp. (54,87%).

Abstract

P. susannae (L.) Raf. is terrestrial orchid grown on the floor of Wonosadi forest. They belong to endangered species of orchid. Those orchids have grown well on degraded leaf litter and surround the grass. So, these plant were interesting to be elucidated especially for their nutrient availability. The was investigate the possibility of yeast associated with terrestrial orchids and cooperate in the provision of nutrients. The research objective was to obtain cellulolytic yeast on soil of root system P. susannae (L.) Raf. The research began with rhizosphere soil sampling on orchids root system P. susannae (L.) Raf. in two place in Forest Wonosadi. Soil samples were analyzed in the laboratory of microbiology, including the determination of the population of cellulolytic yeast and yeast isolated from soil of root system P. susannae (L.). directly from root system using Carboxy methyl cellulose medium (CMC), incubated for 5 days. Colonies were counted for the determination of population growth. Yeast colonies were grown separately isolated as a single culture. Sorting trough growth experiments using a form of cellulose (cellulose microcrystalline, leaf litter and paper) was different. Isolates were grown on microcrystalline cellulose selected for further testing based on the ability to use CMC. Activity was determined by the use of cellulose determine reducing sugar levels (glucose) was released using a spectrophotometer (OD 540 nm). The results showed that the rhizosphere soil P. susannae (L.) Raf. containing a yeast population (17x102 CFU and 2.3 x 102 CFU) and was dominated by the four isolates (K2, K16, A9 and A10) cellulolytic yeasts. The results of the identification of the four isolated of yeasts like Saccharomyces sp. (K2) and Candida sp. (K16, A9 and A10).

Abstract

This research was aimed to use tofu as organic feed for catfish and thus to create a product of organic fish feed made of tofu. Research was done in four steps, (I) Preparation of materials andequipment, (II) Production of organic fish feed, (III) Analysis of carbohydrate, protein, and lipidcontent in organic fish feed, (IV) Measurement of catfish weight. The parameters observed in thisstudy are carbohydrate, protein, and lipid content of organic catfish feed, and the weight of organiccatfish. Organic fish feed made from 100% tofu dreg was used as control (AT0). Feed used fortreatment were made of tofu dreg-fish head (AT1), tofu dreg shrimp head (AT2), and tofu dreg-bran(AT3). The result showed that the best treatment to produce organic fish feed were AT1 (42.01%)and AT2 (43.05%) which showed high content of carbohydrate, protein, and lipid. Based ongrowth, AT1 and AT2 also showed the best percentage rate of growth and survival. AT2 treatmentshowed greater weight than AT1 but not significantly different.

Abstract

Cauda of Periophthalmus gracilis rarely used for locomotion in land. The objective is to know skeleton structure of caudal fin complex. Skeleton staining used Alizarin red’s-alcian blue modified from Inouye (1976). Periophthalmus gracilis’s caudal fin complex have 8 os preural centrum, 2 os epural, os parhypural, 5 os hypural and branched lepidotrichia. The caudal used to support movement in land. Periophthalmus gracilis used it for balancing and support in land.

Abstract

This research aims to determine the effect of Moringa rod extract toward the conception rate of male ICR mice (Mus musculus). This study is an experimental study using completely randomizeddesign (CRD), which consists of one control (A0) and four treatments (A1, A2, A3 and A4) with adose of 75 mg/kg. Treatment is given for 1 week, 2 weeks, reversibility for one week and twoweeks.Provision of intra peritonial with a volume of 0.5 cc/mice. The parameters observed as anindicator is the number of corpus luteum, implantation percentage, percentage of live fetuses andthe percentage of intra uterine death in female mice as testmating pairs in conception rates of malemice. The results showed that the extract of Moringa rod with 75 mg/kg dose given to male micesignificantly affect the percentage of implantation and the percentage of live fetuses in female mice.But did not significantly affect the amount of the corpus luteum and the percentage of intra-uterinedeath. From the results, this study concluded that the extract of Moringa rodeffect on conceptionrates male ICR mice. The 75 mg/kgdose has not shown resilience fertility in male mice eventhough their use is stopped for up to three weeks. In other words, the moringa rod extract mayinterfere with the reproductive function of male ICR mice (Mus musculus), potentially as an activeingredientofanti-fertility

Abstract

As many as 50 million people worldwide are currently experiencing a deficiency of vitamin A and about 250000-500000 malnourished children in developing countries become blind each year from vitamin A deficiency, with the highest prevalence in Southeast Asia and Africa which will result in visual impairment, and increased child mortality and pregnant women. Melon can be one of the solutions for higher productivity and fruit may contain carotene (pro-vitamin A) with varying levels. In the years 2009-2011 Genetics laboratory, faculty of Biology UGM has produced a new melon cultivar called Melodi Gama 1 (MG 1). MG 1 phenotype has a rounded character with rather thick rind and hard, green and fleshy with a smooth orange flesh color. The purpose of this research was to study and analyze the nutritional content of MG 1 compared to commercial cultivars from multilocation and multiseason cultivation. The methods were used vitamin A quantitative analysis by extraction and spectrophotometer, whereas vitamin C in quantitative analysis using the iodine titration method. The results obtained in the analysis of total carotene in the dry season for the MG 1 has the highest value compared with 4 other commercial cultivars, it was 706.61 μg/100gr for cultivation in Yogyakarta and 292.16 μg/100gr for cultivation in Magetan. In the rainy season total carotene MG 1 was 59.88 μg/100gr for cultivation in Yogyakarta, and 20.76 μg/100gr for cultivation in Magetan. The results of the quantitative analysis of vitamin C MG 1 in the dry season was 7.50 mg/100gr for cultivation in Yogyakarta and 16.50 mg/100gr for cultivation in Magetan, while the rainy season was 8.55 mg/100gr for cultivation in Yogyakarta and 6.75 mg/100gr for cultivation in Magetan. Based on these results it can be concluded that the content of Vitamin A and C from Melodi Gama 1 cultivar is higher in the dry season than in the rainy season.
