Abstract

Strawberries were a lot of holticulture commodities consumed by the people of Indonesia. Strawberry consumption continues to increase over time in Indonesia but not matched by production. This it because the strawberry is native to subtropical climate so that production in Indonesia limited in the plateau regional still affected by the tropical climate. Therefore, it is necessary to increase the production of both quality and quantity of strawberries, one with plant breeding through the induction of colchicine. The objective of this research were to determine the differences in phenotypic characters of strawberry plants with the control and induction results to determine the length of time and the induction colchicine 0,05% and 0,01% on the Festival cultivar of strawberry most effective. Research were done by inducing with 0,05% and 0,01% colchicine in leaves, roots 24 hours and 36 hour, the root and leaves of 24 and 36 hour. The observed for 6 months long leaf growth, leaf width,number of leaves, stem circumference, plant height, bloom vast flower and volumes of the fruit. The results indicate that the induction of the leaf growth at the optimal parameter leaf lenghth, leaf width and number of leaves. While 24-hour induction of root growth at optimum height and volume of fruitplants, then root induction at 36 hour to flowers bloom. Induction in root and leaves 36 hour optimum growth on trunck circumference. From this study it could be concluded that there are differences in the character of the strawberry crop cultivar Festival phenotype induction results colchicine 0,05% and 0,01% with control. Induction for 36 hours and induction on leaves was most effective for induction of strawberry cultivar Festival.Keywords: character phenotype, colchicine, festival

Abstract

A random amplified polymorphic DNA (RAPD) marker linked to powdery mildew resistance gene (Pm-I) in melon PI 371795 was reported. However, the RAPD marker has problem in scoring. To detect powdery mildew resistance gene (Pm-I) in melon accurately, the RAPD marker was cloned and sequenced to design sequence characterized amplified region (SCAR) markers. SCAPMAR5 marker derived from pUBC411 primer yielded a single DNA band at 1061 bp. Segregation of SCAPMAR5 marker in bulk of F2 plants demonstrated that the marker was co-segregated with RAPD marker from which the SCAR marker was originated. Moreover, results of SCAR analysis in diverse melons showed SCAPMAR5 primers obtained a single 1061 bp linked to Pm-I in resistant melon PI 371795 and PMAR5. On the other hand, SCAPMAR5 failed to detect Pm-I in susceptible melons. Results of this study revealed that SCAR analysis not only confirmed melons that had been clearly scored for resistance to Pm-I evaluated by RAPD markers, but also clarified the ambiguous resistance results obtained by the RAPD markers.   Key words: Cucumis melo L., Pm-I, RAPD, SCAPMAR5

Abstract

Strawberry cultivation in Indonesia is centred at Citrus and Subtropical Fruit Research Institute, which collects different cultivars of Fragaria spp. Information on classification based on phenotypic characters of Fragaria spp. has not been studied as a whole. The purpose of the present study was to identify and determine phylogenic relationships of nine cultivars of strawberries based on the anatomical and morphological characters. The present study identified morphologically nine strawberry cultivars by referring to the IPGRI (1986) and the UPOV (2012) and Hofer et al. (2012). Anatomical preparations of roots, stems and leaves were made by the embedding method. Phylogenetic relationships were determined using MVSP software with UPGMA algorithm through the Gower General Similarity Coefficient method. Subsequently, the principal components were analyzed using the Euclidian Biplots algorithm depicted in a Scatter plot. Results showed that there were 37 anatomical and morphological characters that play a role in the characterization of nine cultivars (25 synthetic characters and 12 diagnostics characters) where the shape of the fruit and type of attachment of the flowers were the main characters distinguishing among the nine cultivars studied. The phylogenetic relationship produced two clusters with 79% similarity index. The closely related cultivars were Festival and Rosa Linda with a similarity index of 94%, and Earlibrite and Aerut with a similarity index of 86%

Abstract

Lahan kritis berkapur, karst, memberikan cekaman abiotik pada tanaman karena hambatan hidrasi dan nutrisi. Asam absisat (ABA) adalah hormon yang diekspresikan tumbuhan pada kondisi cekaman abiotik. CmBG1 merupakan salah satu gen peregulasi hormon ABA pada melon yang akan terakumulasi saat tumbuhan mengalami cekaman. Tujuan penelitian ini adalah mendeskripsikan pengaruh lahan kritis karst terhadap ekspresi gen CmBG1 secara kualitatif dan kuantitatif melon hasil turunan kultivar TACAPA, yaitu kultivar PT dan AT yang ditanam pada medium tanah karst dari wilayah Agroekosistem II dan III Yogyakarta (Gunungsewu, Dlingo, maupun Sentolo). cDNA library diperoleh dari reverse transcription isolat RNA. cDNA diamplifi kasi dengan primer spesifi k, kemudian dispektrofotometri pada ?260 nm untuk mengetahui konsentrasi gen CmBG1. Ekspresi gen dianalisis dengan real time PCR dengan gen referensi Cm-actin. Uji kualitatif dilakukan dengan elektroforesis gel agarosa 1,5%. Hasil penelitian menunjukkan gen CmBG1 terdeteksi dengan ukuran ±1258 bp pada kultivar PT dan AT. Konsentrasi gen CmBG1 melalui spektrofotometri menunjukkan semua kultivar yang ditanam pada media kontrol memiliki konsentrasi yang lebih rendah bila dibandingkan media tanam dengan perlakuan lahan kritis baik Gunungsewu, Dlingo, maupun Sentolo. Hasil ini sama dengan uji ekspresi gen CmBG1 menggunakan analisis kuantitatif real time PCR. Karsts critical land gives abiotic stresses in plants because of hydration and nutrition disturbances. Abscisic acid (ABA) is a hormone that expressed in the plant in abiotic stress conditions. CmBG1 is one of the regulatory genes encoding hormone ABA in melon plants which is accumulated in stress condition. The purpose of this study was to describe the infl uence of karsts critical land on the genes expression of CmBG1 melon cultivars PT and AT qualitatively and quantitatively. The plants were grown in medium karst land of Agroecosystems II and III of Yogyakarta (Gunungsewu, Dlingo, and Sentolo). Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplifi ed using specifi c primer. Spectrophotometry was conducted in ?260 nm and electrophoresis run in 1.5% agarose gel. Control band and reference gene chosen in Real Time PCR was Cm-Actin. CmBGI band (± 1258 bp) was showed both on PT and AT. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar PT and AT melons when they are grown under stress condition. This result show similarly when using real time PCR.

Abstract

Strawberry (Fragaria spp.) has a high economic value and various benefits, but the production of strawberry plants in Indonesia is still low in terms of both quantity and quality. Strawberry plant breeding can be done in various techniques, one of which is polyploidization. Polyploidization by an induction of colchicine at various concentrations in strawberry crops cv. California and Festival resulted in superior phenotype characteristics. To prove the existence of a change in ploidy in strawberry, then research at the molecular level needs to be done. The purposes of this study were to find out changes in ploidy of strawberry plants using CAPS molecular markers and to detect the polymorphism in strawberry plants quickly. The samples used were young leaves. Main procedure was the cutting of the amplified DNA using restriction enzymes of TaqI and HaeIII. The results showed that CAPS molecular markers were capable of detecting polymorphism quickly and efficiently in strawberry plants. Specific bands among strawberry plants having undergone polyploidization and those not having undergone polyploidization can be seen on the differences in monomorphic or polymorphic bands between the control plants and treated plants.

Abstract

CmBGI is the enzymatic genes encoding β-glucosidase that involved in Abscisic Acid (ABA) metabolism of Cucumis melo L. β-glucosidase promotes the accumulation of glucose, fructose, and sucrose, and it might act as a regulator that mediates melon fruit ripening both climacteric and nonclimacteric. ABA mediates adaptive responses to abiotic and biotic stresses. Agricultural Balitbang in 1997 showed that there were approximately 158.600 ha of degraded land scattered in three zones of agroecosystems in Yogyakarta (DIY). One of them is Dlingo Bantul area which has a karst type critical land area. Karst provides stress to the certain plant growth. One way to conserve critical land is making this area for agriculture. Cultivar TACAPA and TA were superior melons that have been developed by Genetic Laboratory of Biology Faculty UGM. This preliminary research was conducted to examine molecular characterization of CmBGI gene expression in cultivar TACAPA and TA which are planted in normal condition medium and in critical land medium treatment. Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplified using specific primer. Spectrophotometry was conducted in λ260 nm and electrophoresis run in 1.5% agarose gel. Control of band chosen was Cm-Actin. CmBGI gene concentration of TACAPA and TA in normal condition medium are in succession 578.5 and 579.4 μg/ml then for critical land medium treatment 743.4 and 773.5 μg/ml. CmBGI band was showed both of TACAPA and TA as ± 1258 bp. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar TACAPA and TA melons when they are grown under stress condition.

Abstract

Indonesia is known as a country rich in various agricultural and plantation products, including vegetables. One effort to improve quality of the production by identifying and characterizing chromosomes. The purpose study to characterize number, shape and size of the chromosomes in Solanaceae family. The study using modified Squash method. Chromosome preparation including fixation, maceration, staining, and analyzed using Image Raster 3 application, then Corel Draw X6. The results showed study to characterize the number, shape and size in the Capsicum annum L. Tm 999 (2n=20m+4sm and 2,41 s/d 9,99 µm) then Capsicum frutescens L. Kencana (2n=16m+8sm and 3,12 s/d 10,49 µm). In Solanum lycopersicum Or Diana and Marta (2n=16m+8sm and 3,12 s/d 10,49 µm) then (2n=22m+2sm and 2,65 s/d 7,28 μm). In Lycopersicon esculentum Mill. Juliet and Tropical Ruby (2n=18m+6sm and 0,77 s/d 1,81 μm) then (2n=20m+4sm and 0,39 s/d 0,94 μm). Solanum melongena L. Or Valerie and Mustang (2n=24m and 0.29 s/d 0.87 μm) then (2n=22m+2sm and 0,5 s/d 1.28 μm). In Solanum melongena L. Kania and Pulus (2n=20m+4sm and 0.64 s/d 3.43 μm) then (2n=16m+8sm and 0.84 s/d 2.51 μm). In Solanum melongena L. Jeno and Planet Hijau (2n=18m+6sm and 0.65 s/d 2.423 μm) then (2n=14m+10sm and 0.46 s/d 1.73 μm). In Capsicum annum var. Grossum L. Red Star and Purple Star (2n=22m+2sm and 3.19 s/d 8.49 μm) then (2n=24m and 5.5 s/d 11.92 μm).
