Abstract

Penelitian ini bertujuan menginduksi dan meregenerasi embrio somatik dari embrio zigotik buah pepaya muda kultivar Eksotika II. Kegiatan dilaksanakan di laboratorium Kultur Jaringan Jabatan Sains Tanaman, Universiti Putra Malaysia pada bulan Januari sampai Agustus 2001. Penelitian ini menggunakan rancangan acak lengkap dengan perlakuan kombinasi beberapa konsentrasi 2,4-D (1,0; 2,5 dan 5,0 mg/l) dan BAP (0; 0,001; 0,005; dan 0,01 mg/l). Hasil penelitian menunjukkan bahwa media MS yang mengandung 5,0 mg/l 2,4-D membentuk embrio somatik tertinggi (74,55%) dan kombinasi dari 2,4-D 1,0 mg/l dan BAP 0,01 mg/l menghasilkan kalus tertinggi (52,58%) pada 8 minggu setelah kultur. Jumlah embrio somatik per eksplan terbanyak (66,61) diperoleh pada media MS dengan 5,0 mg/l 2,4-D dan 0,01 mg/l BAP. Pemasakan embrio diperoleh dengan memindahkan embrio globular ke media padat MS tanpa zat pengatur tumbuh. Empat minggu setelah kultur, 42% embrio somatik berkecambah setelah embrio masak. Plantlet siap diaklimatisasi pada 8 minggu setelah kultur ke media perkecambahan. Sistem perbanyakan dan regenerasi embrio somatik pepaya ini dapat menunjang keberhasilan pemuliaan tanaman pepaya modern melalui transformasi genetikThe experiments with the objectives to induce and regenerate somatic embryogenesis were carried out at the Tissue Culture Laboratory of Crop Science Department, Universiti Putra Malaysia from January to August 2001. The experiments involved the induction of somatic embryogenesis from immature zygotic embryos of papaya cv. Eksotika II and regeneration of plantlets from the somatic embryos. A nonfactorial completely randomized design was used as experimental design with treatments of the 2.4-D combinations concentrations (1.0; 2.5 and 5.0 mg/l) and BAP concentrations (0; 0.001; 0.005 and 0.01 mg/l). The results showed that MS medium supplemented with 2,4-D 5.0 mg/l promoted the highest percentage of somatic embryogenesis (74.55%), while the combination of 1.0 mg/l 2,4-D and 0.01 mg/l BAP produced the highest percentage of callus formation (52.58%) after 8 weeks of culture. The highest number of somatic embryos per explant (66.61) was obtained when 5.0 mg/l 2,4-D and 0.01 mg/l BAP were added into MS medium. Maturation of somatic embryos was achieved on transfering the globular somatic embryos derived from zigotic embryo to solid MS medium without growth regulator. After 4 weeks of culture, 42% germinated somatic embryo was occurred following maturation of somatic embryos. Plantlets were ready for acclimatization to germination medium 8 weeks after culture. Somatic embryogenesis system could enhance the successful of the modern papaya breeding program through genetic transformation.

Abstract

Kabupaten Solok dicanangkan sebagai salah satu sentra produksi bawang merah untuk mengimbangi sentra produksi utama di Pulau Jawa yang produksinya selalu berfluktuasi. Beberapa kelebihan dan kekurangan dimiliki oleh Kabupaten Solok dalam mengembangkan potensinya sebagai sentra bawang merah nasional. Penelitian bertujuan untuk merumuskan strategi pengembangan Kabupaten Solok sebagai sentra bawang merah nasional. Penelitian dilakukan dari bulan Agustus hingga Desember 2017 bertempat di Kabupaten Solok. Data primer berupa atribut-atribut faktor internal kekuatan dan kelemahan serta faktor-faktor eksternal peluang dan ancaman diidentifikasi melalui Focussed Group Discussion (FGD) yang dilakukan dengan peserta para pemangku kepentingan di Kabupaten Solok sekaligus untuk melakukan kuantifikasi dari atribut-atribut yang telah teridentifikasi. Analisis dilakukan dengan pendekatan SWOT, yaitu menggunakan perhitungan IFE, EFE, matriks IE, matriks SPACE, dan matriks SWOT. Terdapat enam atribut kekuatan, delapan atribut kelemahan, enam atribut peluang, dan lima atribut ancaman. Perhitungan IFE dan EFE memperlihatkan bahwa faktor internal merupakan faktor yang paling dominan, dengan faktor kekuatan merupakan faktor yang paling dominan dibandingkan kelemahan. Posisi Kabupaten Solok dalam matriks IE dan matriks SPACE masing-masing berada dalam posisi jaga dan pertahankan serta mendukung strategi agresif. Strategi yang perlu diterapkan adalah strategi memaksimalkan kekuatan untuk menangkap peluang yang tersedia (S-O) di antaranya pengembangan teknologi PTT bawang merah spesifik lokasi, pengembangan teknologi pascapanen bawang merah, membuka pasar baru selain yang sudah ada, dan membina penangkar bawang merah. Beberapa dukungan teknologi dan inovasi Badan Penelitian dan Pengembangan Pertanian sebagai peluang yang tersedia juga perlu diterapkan untuk memaksimalkan kekuatan yang dimiliki Kabupaten Solok untuk berkembang sebagai sentra produksi bawang merah nasional.KeywordsKabupaten Solok; Bawang merah; Strategi; SWOTAbstractSolok Regency is planned as one of the production centers of shallot to complement the main production centers on Java Island which production always fluctuating. Some advantages and disadvantages are owned by Solok in developing its potential. This study aims to formulating strategy to develop Solok Regency as a national shallot production center. The research was conducted from August till December 2017 at Solok District. Primary data such as internal factor attributes like strengths and weaknesses as well as external factors like opportunity and threat are identified through Focus Group Discussion (FGD) which conducted with participants of stakeholders in Solok to quantify the attributes that have been identified. The analysis then conducted with the SWOT approaches using IFE, EFE, IE matrix, SPACE matrix, and SWOT matrix. There are six attributes of strength, eight attributes of weakness, six attributes of opportunity, and five attributes of threat. The calculation of IFE and EFE shows that internal factor is the most dominant factor, with strength is the most dominant internal factor. The position of Solok Regency in IE is in a position to guard and defend while in the SPACE matrix the position of Solok Regency is to support an aggressive strategy. The strategies that needs to be implemented is strategy to maximize the power to capture the available opportunities (S-O) such as the development of location-specific shallot integrated crop management technology, the development of shallot post-harvest technology, create new markets other than existing ones, and develop shalot breeder. Some IAARD’s technology and innovation support as available opportunities also need to be applied to maximize the power of Solok to grow as a national production center for shallot.

Abstract

ABSTRAK. Penelitian tentang pola sebaran vertikal Thrips parvispinus Karny pada tanaman paprika dilaksanakan dari bulan Maret - Desember 2003 di Balai Penelitian Tanaman Sayuran di Lembang (1.250 m dpl). Tujuannya adalah untuk mengetahui bagian tanaman paprika yang paling disukai oleh T. parvispinus. Bagian tanaman yang diuji meliputi bunga, daun pucuk, daun atas, daun tengah, dan daun bawah. Hasil penelitian menunjukkan bahwa nimfa T. parvispinus paling banyak dijumpai pada daun atas, sedangkan imago pada daun pucuk. Dengan demikian, daun atas dan daun pucuk dapat dipilih sebagai unit contoh dalam kegiatan pengamatan trips.ABSTRACT. Prabaningrum, L. and T.K. Moekasan. 2008. A Study on vertical distribution of T. parvispinus on sweet pepper was conducted from March to December 2003 at the Indonesian Vegetables Research Institute, Lembang (1,250 m asl.). The aim of the study was to determine which part of the plant that most prefered by T. parvispinus. The plant parts tested were flower, shoot, upper leaf, middle leaf, and lower leaf. The results indicated that the highest population of nymph was found on upper leaf and the highest population of imago was found on shoot. Therefore, upper leaf and shoot can be chosen as sampling units in monitoring of thrips.

Abstract

ABSTRAK. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Balai Penelitian Tanaman Sayuran pada bulan Maret-Juni 2005, dengan tujuan untuk mengetahui pengaruh penambahan zat pengatur tumbuh BAP dan pikloram pada media dasar MS dan B5 terhadap pertumbuhan tunas bawang merah kultivar Sumenep. Bahan tanaman yang digunakan adalah jaringan meristematik sedangkan media tumbuh yang diuji adalah media dasar MS dan B5 dengan konsentrasi BAP (0, 1, dan 2 mg/l), pikloram (0, 0,1, dan 0,2 mg/l). Hasil penelitian menunjukkan bahwa ada perbedaan pertumbuhan plantlet akibat penambahan zat pengatur tumbuh BAP dan pikloram. Persentase daun normal terbesar didapat dari perlakuan B5 tanpa pikloram dan BAP, media MS tanpa pikloram. Pertumbuhan akar plantlet lebih baik yang dikulturkan pada media MS tanpa hormon atau dengan penambahan hormon dengan konsentrasi terendah.ABSTRACT. Karjadi, A.K. and Buchory, A. 2008. The`experiment was conducted in Tissue Culture Laboratory of Indonesian Vegetable Research Institute in Lembang on March-June 2005. Objective of the experiment was to find out the effect of additional BAP and picloram to basal medium MS and B5 on shoot induction of shallot cv. Sumenep. The treatments were culturing meristem tissue (shoot tip) on medium MS and B5 with concentration of BAP (0, 1, and 2 mg/l), and picloram (0, 0,1, and 0,2 mg/l). The results showed that there were differences on plantlet growth with addition of BAP and picloram. Meristematic tissue of shallot cv. Sumenep could be proliferated on medium MS or B5. The highest percentage of normal leaf growth was obtained by B5 medium without picloram or BAP, and MS medium without picloram. The best roots growth was found in culture of MS medium without or with low concentration of hormone.

Abstract

Tujuan dari penelitian ini untuk mengetahui hasil to tal tomat, kubis, serangan OPT, dan nilai kesetaraanlahan (NKL) dalam sistem tumpangsari tomat dan kubis. Tumpangsari yang dicoba adalah tomat monokultur, kubismonokultur, tumpangsari tomat + kubis, tumpangsari tomat + kubis + kubis + tomat, dan tumpangsari tomat + tomat +kubis + tomat + tomat. Rancangan percobaan yang digunakan adalah acak kelompok dan ulangan lima kali. Hasilpenelitian menunjukkan bahwa tumpangsari antara tomat + tomat + kubis + tomat + tomat merupakan kombinasiterbaik dan dapat menekan populasi hama Plutella xylostella sebesar 97% dan Crocilodomia binotalis sebesar 76,2%.Secara kuantitatif produksi tomat maupun kubis yang ditanam sistem ganda (intercropping) lebih tinggi daripadaditanam secara tunggal. Sistem penanaman tomat dan kubis secara tumpangsari memberikan keuntungan karena nilaidari NKL > 1, keuntungan tertinggi diperoleh dari sistem tumpangsari tomat + kubis + kubis + tomat sebesar Rp44.420.000,-/ha.Ef fect of intercropping be tween to mato and cab -bage to pests de vel op ment and yield. Abstract

ABSTRAK. Penelitian bertujuan untuk mengetahui media, suhu, dan lama blansing yang optimum sebelum pengeringan terhadap mutu lobak kering. Penelitian dilakukan pada bulan Oktober sampai November 2004. Metode penelitian yang digunakan adalah metode eksperimen di laboratorium menggunakan rancangan kelompok pola petak terpisah dengan 3 ulangan. Petak utama yaitu media blansing yang terdiri dari (1) air dan (2) uap. Anak petak yaitu kombinasi suhu dan lama blansing yang terdiri dari (1) suhu 65°C selama 15 menit, (2) 65°C selama 30 menit, (3) 75°C selama 10 menit, (4) 75°C selama 20 menit, (5) 85°C selama 5 menit, dan (6) 85°C selama 10 menit. Hasil penelitian menunjukkan bahwa terjadi interaksi antara media dan kombinasi suhu dan lama blansing terhadap rendemen, kadar air, dan vitamin C. Dari uji organoleptik perlakuan yang terbaik adalah media uap pada suhu 75°C selama 10 menit dengan hasil lobak kering yang disukai (nilai 3,73). Lobak kering hasil perlakuan tersebut mempunyai kadar air 8,33%, rendemen 3,75%, rasio rehidrasi 281,67%, dan vitamin C 567,25 mg/100 g.ABSTRACT. Asgar, A. and D. Musaddad. 2008. The Effect of Medium, Temperature, and Blanching Time on the Dried-Radish Quality. The objective of this research was to find out the effect of medium, temperature, and blanching time on the characteristic of dried-radish. Experiment was conducted at Indonesian Vegetables Research Institute from October to November 2004. This research was arranged in a split plot design with 3 replications and followed by Duncant’s test. The main plot was blanching medium, consisted of (1) water and (2) steam. While the subplot was temperature and time of blanching, consisted of (1) 65°C and 15 minutes, (2) 65°C and 30 minutes, (3) 75°C and 10 minutes, (4) 75°C and 20 minutes, (5) 85°C and 5 minutes, and (6) 85°C and 10 minutes. The results indicated that there was interaction between medium and the combination of temperature and duration of blanching. Organoleptics test showed that steam blanching at 75°C for 10 minutes gave the most prefered dried radish (3.73), with moisture content of 8.33%, dry matter of 3.75%, rehydration ratio of 281.67%, and vitamin C of 567.25 mg/100 g.

Abstract

ABSTRAK. Pengendalian kimiawi terhadap hama D. citri, vektor penyakit CVPD sudah banyak dilakukan, namun ada beberapa kelemahan dan efek langsung yang terjadi, antara lain biaya besar, resistensi serangga, dan pencemaran lingkungan. Penggunaan agens hayati berupa patogen untuk mengendalikan D. citri merupakan salah satu pendekatan ekologi dalam mengatasi masalah ini. Entomopatogen H. citriformis merupakan salah satu pilihan, di samping entomopatogen M. anisopliae dan B. bassiana yang sudah dikenal lebih dulu. Beberapa pengamat hama di lapang menduga infeksi alami H. citriformis lebih cepat mematikan D. citri bila ada jamur entomopatogen lain yang sudah menginfeksi lebih dahulu. Tujuan penelitian adalah mengetahui patogenisitas H. citriformis terhadap imago D. citri dengan cara eka dan dwiinfeksi. Percobaan dilakukan di Laboratoium Mikologi Balai Penelitian Tanaman Jeruk dan Buah Subtropika Tlekung dan kebun jeruk milik petani di Jombang. Perlakuan yang di uji adalah H. citriformis, B. bassiana, M. anisopliae, dan kombinasinya. Rancangan yang digunakan adalah acak lengkap dengan 3 ulangan. Hasil penelitian menunjukkan bahwa pada dwiinfeksi B. bassiana dan H. citriformis menunjukkan hubungan sinergisme dan menyebabkan persentase kematian D. citri lebih tinggi disusul dengan ekainfeksi H. citriformis.ABSTRACT. The chemical control of D. citri, CVPD disease vector’s was one of major method applied in the field, but it has several side effects, such as of insect resistance or environmental pollution. Control measures of D. citri by using biological agents have the potency to reduce insecticide application, especially the use of H. citriformis entomopathogen, besides M. anisopliae and B. bassiana, that were popular before. Several field pest observers indicated that natural infection of H. citriformis could accelerate the mortality of D. citri if combined with other entomopathogens. The objectives of this study was to measure entomopathogenicity of H. citriformis in controlling D. citri in combination with other entomopathogens. The research was conducted at the Micology Laboratory, Indonesian Citrus and Subtropic Fruit Research Institute and Jombang citrus farmer field. The treatments tested were H. citriformis, B. bassiana, M. anisopliae and their combination. The randomized block design with 3 replications was used in this experiment. The results showed that double infection of B. bassiana and H. citriformis was most sinergism than others treatments, which caused highest mortality of D. citri, followed by single infection of H. citriformis.

Abstract

ABSTRAK. Alpinia purpurata merupakan salah satu komoditas tanaman hias tropis yang termasuk baru dari famili Zingiberaceae. Tujuan penelitian ini adalah mengetahui pengaruh pupuk P dan K dalam memacu pertumbuhan vegetatif dan generatif A. purpurata. Penelitian dilaksanakan di rumah sere KP. Segunung, Balai Penelitian Tanaman Hias mulai bulan September 2004 sampai dengan Agustus 2005. Rancangan percobaan menggunakan acak kelompok pola faktorial dengan 2 faktor dan 3 ulangan. Pemupukan P (0, 36, 72) kg P2O5/ha) sebagai faktor pertama dan pemupukan K (0, 60, 120, 180) kg K2O/ha sebagai faktor kedua. Hasil penelitian menunjukkan bahwa tidak ada interaksi antara pemberian pupuk P dan K terhadap pertumbuhan tanaman A. purpurata. Namun pemberian pupuk K (60 kg K2O/ha) meningkatkan produksi bunga/plot tertinggi (27,33 tangkai) pada bulan Februari 2005 demikian pula K 120 kg K2O/ha) nyata meningkatkan diameter bunga mekar tertinggi (3,97 cm). Sedangkan pemberian K nyata meningkatkan panjang daun (25,30 cm) dan diameter bunga mekar (3,97 cm).ABSTRACT. Utami, P.K., R. Tedjasarwana, and D. Herlina. 2006. Growth and flower quality improvement of A. purpurata through fertilization application of phosphate and potassium. Alpinia purpurata is one of the new tropical ornamental plants from Zingiberaceae family. The objective of the experiment was to determine the effect phosphate and potassium fertilizer application on promoting vegetative and generative growth of Alpinia. The experiment was conducted in the screenhouse at Segunung field station (1,100 m asl) at Indonesian Ornamental Crops Research Institute, Cianjur, from September 2004 to August 2005. The plots were arranged in a factorial completely randomized design with 3 replications and 2 factors. The first factor comprised of 3 level phosphate dosage i.e. 0, 36, 72 kg P2O5/ha. The second factor consisted of 4 level potassium dosage i.e. 0, 60, 120, and 180 kg K2O/ha. The results showed that there was no significant interaction between phosphate and potassium fertilizer application, but the use 60 kg K2O/ha increased of flower production on February 2005. K120 kg K2O/ha significantly increased of opened flower diameter (3.97 cm), and flower bud diameter (1.17 cm). Mean while, K application at 120 kg K2O and 60 kg K2O significantly increased the leaf length and mature flower (3.97 cm).

ABSTRAK. Sifat ketahanan terhadap penyakit merupakan salah satu kriteria dalam seleksi progeni hasil persilangan untuk pelepasan varietas baru krisan. Untuk mendapatkan klon-klon unggul krisan tahan karat, sejumlah progeni telah dievaluasi ketahanannya. Penelitian dilaksanakan di Balai Penelitian Tanaman Hias (1.100 m dpl) dari bulan Oktober 2002 hingga September 2003 untuk mengevaluasi 13 aksesi yang terdiri dari 12 klon krisan, yaitu nomor 151.13, 159.79, 162.7, 164.28, 164.37, 164.64, 164.82, 164.88, 164.97, 164.102, 164.156, 165.12, dan 1 varietas kontrol (cv. White Reagent) terhadap penyakit karat. Hasil penelitian menunjukkan bahwa awal terjadinya gejala dan insidensi penyakit bervariasi pada klon-klon yang dicoba. Berdasarkan kriteria tingkat ketahanan, terdapat 2 klon, yaitu No. 151.13 dan 164.64, dikategorikan sangat tahan terhadap penyakit karat. Selain itu terdapat 2 klon yang termasuk kriteria tahan dan 7 klon yang dikategorikan agak tahan. Sedangkan cv. White Reagent dan klon No. 164.37 termasuk dalam kategori agak rentan terhadap penyakit karat.ABSTRACT. Budiarto, K., I. B. Rahardjo, and Suhardi. 2008. Resistance to major disease is one of important criteria in the selection of chrysanthemum for new released varieties. Japanese white rust is considered one of constrained-diseases and become major problems in commercial chrysanthemum growers in Indonesia up to this moment. Thus, selection of progenies against this disease became important. The research was conducted from October 2002 to September 2003 to evaluate 13 accessions of chrysanthemum, comprised of 12 promising clones of no. 151.13, 159.79, 162.7, 164.28, 164.37, 164.64, 164.82, 164.88, 164.97, 164.102, 164.156, 165.12, and 1 commercial variety (cv. White Reagent) as control to white rust disease. The results of the experiments showed that the initial symptoms and white rust incidence were varied among clones tested. Among the 13 accessions, 2 clones (no. 151.13 and 164.64) were considered strongly resistant. The other 2 and 7 clones were included to the criteria of resistant and moderate resistant, respectively. While cv. White Reagent (control) and clone no. 164.37 were categorized as moderate susceptible.

Abstract

ABSTRAK. Tanaman jeruk mengandung metabolit sekunder flavonoid, karotenoid, dan limonoid yang banyakterdapat dalam daun, kulit buah, biji, dan pulp. Penelitian bertujuan mengetahui kandungan flavonoid dan limonoid padaberbagai fase pertumbuhan tanaman jeruk Kalamondin dan Purut serta mendapatkan informasi kandungan limonoidpada fase embrio dan planlet hasil perbanyakan in vitro melalui embriogenesis somatik. Penelitian dilakukan di BalaiPenelitian Tanaman Jeruk dan Buah Subtropika (Balitjestro) sejak bulan Mei sampai dengan Desember 2009. Ruanglingkup penelitian terdiri atas (1) identifikasi metabolit sekunder yaitu flavonoid dan limonoid pada berbagai fasepertumbuhan tanaman jeruk Kalamondin dan Purut dan (2) identifikasi limonoid pada fase embrio dan planlet tanamanjeruk Kalamondin yang diperbanyak dengan metode embriogenesis somatik secara in vitro. Analisis kandunganmetabolit sekunder dilakukan di Unit Layanan Pengujian, Fakultas Farmasi, Universitas Airlangga. Hasil penelitianmenunjukkan bahwa kandungan flavonoid dan limonoid dapat diproduksi dari berbagai bagian tanaman, seperti padapulp, biji, kulit buah, dan daun pada berbagai fase pertumbuhan jeruk Purut dan Kalamondin. Kandungan flavonoidpada jeruk Purut dan Kalamondin tertinggi terdapat pada buah tua, masing-masing 18,8 ppm. Kandungan limonoidpada jeruk Purut hanya terdeteksi pada daun pendukung buah tua (1 ppm) dan biji (61 ppm), sedangkan pada jenisKalamondin hanya terdeteksi pada biji yaitu sebesar 74 ppm.ABSTRACT. The aimsof this research were to determine flavonoid and limonoid contents in every growth phase of Kalamondin and Purutand the limonoid contents in embryo and plantlet phases derived from in vitro somatic embryogenesis. The researchwas conducted in Indonesian Citrus and Subtropical Fruits Research Institute (ICISFRI) from May to December 2009.The research consisted of two activities as follows: (1) analyses of flavonoid and limonoid contents in every growthphase of Kalamondin and Purut and (2) analyses of the limonoid contents in embryos and plantlet proliferated fromsomatic embryogenesis culture. Flavonoid and limonoid contents were analyzed at the Assessment Service Unit,Faculty of Pharmacy, Airlangga University. The results showed that flavonoid and limonoid compounds could beproduced in all parts of plant i.e, such as pulp, seeds, peel of fruit, and leaves from every growth phase of Kalamondinand Purut. In Purut and Kalamondin, the highest flavonoid content was obtained from ripen fruit, with concentration18.8 ppm. Liminoid content in Purut was detected only in leaf supporting ripen fruit (1 ppm) and seeds (61 ppm),and in Kalamondin was only in seeds with concentration 74 ppm.
