Abstract

The study aimed to identify variations in morphological and molecular character of sugarcane mutants from the mutation of Ethyl Methane Sulphonate (EMS). It used 21 sugarcane mutants and two non-mutant PS865 plants as controls. The treatments with inundation were carried out on 1-17 mutants and non-mutants, while the treatments without inundation were carried out on 18-21 mutant plants and non-mutants. The tolerance characteristics base on the agronomic characters. The RAPD molecular character was observed to detect changes in genotypes and kinship relationships of the plants tested. The results showed that the characteristics of tolerance to the best inundation were found in mutants 1, mutants 3 and mutants 6 which were characterized by the root volume (cm3), fresh root weight (g), sucrose content and brix value (%). Mutant plants treated with inundation showed higher levels of sucrose and brix values. Whereas the control plants in fats, showed lower levels of sucrose and brix. Changes in sugarcane mutant genotypes from non-mutants based on RAPD markers ranged from 14.7 - 56.7 % which resulted in an average polymorphic band of 35.1 % from 37 DNA bands and produced four main groups based on dendrogram analysis.

Abstract

An evaluation of formulated Bio-PF with the active ingredient of Pseudomonas fluorescens to reduce synthetic fungicide usage in controlling soil borne disease in leather leaf fern under different shelters was studied. The research was conducted at the experimental field of the Indonesian Ornamental Crops Research Institute (IOCRI) from January to December 2015. Different concentrations of Bio-PF were singly and/nor alternately applied with two different synthetic fungicides were applied on leather leaf plants under UV plastic + 70% black-net and 70% black-net shelters. The results showed that the average disease intensity was higher in 70% black-net shelter. Weekly alternate-application of Bio-PF 5 + Carbendazim & Mancozeb and single Carbendazim & Mancozeb gave lower disease intensity than other treatments with the highest percentage of suppression. These two treatments also improved yield in terms of number and proportion of the preferred grade of harvested leaves. Combined Bio-PF 5 + Carbendazim & Mancozeb gave longer vase life and the application reduced synthetic fungicide usage in controlling the respected disease. In term of synthetic fungicides, Carbendazim & Mancozeb gave more consistent effects on disease suppression, plant growth and foliage production than Asilbensolar & Mancozeb.

Abstract

Arthropods biodiversity on a rice ecosystem plays an important role related to the pest management. This research aimed to investigate the diversity and community structure of arthropods in the rice ecosystem. It was conducted in Lembah Seulawah, Aceh Besar Regency, Indonesia from February to August 2016. Sweep net was used to collect insects on four rice plots at 35, 45 and 55 days after transplanting (DAT) as vegetative, transition and generative stages respectively. Result showed that the twenty-five morpho-species found at vegetative stage and it was the highest. They were categorized as phytophagous insects, spiders, predatory insects, parasitoid, and neutral insects. Each arthropod’s category was in the higher abundance at vegetative stage, except for phytophagous arthropod. The Shannon-Wiener diversity index (H') was significantly higher at vegetative stage than the remaining stages, while Simpson Dominance index (C) and Species Evenness (E) indices were not significantly different among the three observed stages.

Abstract

In Indonesia, amount of biodiversity could be found including soil microorganism which were useful for sustainable agriculture. Everything lives in agricultural land were interacts and influences each others. Many factors of biotic and a-biotic environment such as land and climate fluctuation influences microbial expansion within the soils. For example arbuscular mycorrhiza (AM) which was formed by myches (fungus) and root of higher plant maybe fluctuated in the expansion due to the soil tillage. Land management and the present higher plant affected its growth and expansion. Mycorrhizae spores as one of the important generative form of various species in the difference host plant. The implication of different host plant often close related to the soil management and properties. Soil sample that was collected from various host plant in the fields or bare land have been found varied spores. Andisols and its properties conserve higher organic matter relatively and developed in the cold temperature, therefore very suitable for AM expansion and also as natural resources stock of AM potentially.Keywords: arbuscular mycorrhiza, andisols, soil tillage, AM colonization

Abstract

In this study the effect of essential oil taken from medicinal plant as antibacterial components in preservative solution of Lisianthus var. Echo (Eustoma grandiflorum) was investigated. The test was done with application of preservative solution.  Cut flowers were treated with different concentrations of Thyme (Thymus vulgaris), Spearmint (Mentha spicata) and Lavender (Lavandula officinalis) essential oil in addition to Sucrose 2.5%. The results showed that there was the longest time in vase life with Thyme in 50 ppm (15.6 days) and the control treatment showed the shortest vase life (11.6 days). Moreover, Thyme with 50 ppm had the highest effect on relative fresh weight and solution uptake. In addition, bacteria-induced stem xylem blockage, extracted from the end of stem, was cultured in NA medium culture with several concentrations of essential oil. The result showed that in pure concentration (100%) inhibition was completed and in various concentrations of essential oil the bacterial population was reduced.Keywords: essential oil, lavender, preservative solution, spearmint, thyme

Abstract

The low efficiency of Phosphate fertilization on wetland rice may be overcame by using quickly dissolve P fertilizer. However, for soils containing high dissolve Fe, such as Alfisol and Oxisol, will induce the formation of Fe-P bonding. Therefore, to use the quickly dissolve P fertilizer  in such soils should be done by split application. The research aimed to assess better P fertilization method for wetland rice which used quickly dissolve P fertilizer given in split application. The research concluded that the application of quickly dissolve P fertilizer to Alfisol and Oxisol which was applied in split application by twice (0 and 30 days after planting) and three times (0, 15 and 30 days after planting) increased the eficiency of P fertilization in wetland rice.  The split application of 75 kgs perhectare (one half of recommended dose) in Alfisol yielded dry seed as high as 6,374 kgs per hectare (at the twice application), 6,280 kgs per hectare (the three times application)c while the control treatment yielded as high as 6,027 kgs per hectare.  In Oxisol the yield of rice was 8,200 kgs per hectare fo the control treatment, 8,027 kgs per hectare for the twice application and 8,440 kgs per hectare for the three time applications.

Abstract

Corn is an important food crop in Indonesia. Plant expansion has been hampered by soilacidity problem and the protein content of many corn varieties was low. This research initiates development of soil-acid-tolerant corn with highquality- protein content. Research was done on 12 factorial treatments and 3 replications as blocks in RCBD. The first factor was corn populations: Toray-1(G1), Toray-2(G2), GS- 5(G3) and GS-10(G4). The second factor was fertilizations: P1(69 kg N+36 kg P2O5+15 kg K2O per ha)); P2(115 kg N+54 kg P2O5+30 kg K2O per ha); and P3(161 kg N+72 kg P2O5+45 kg K2O per ha). The observed variables consisted of several agronomic traits, including the protein content. Results indicated that the corn populations, in general, showed good agronomic traits. The differences were mostly between populations, not between fertilizations, and no interaction was observed. The yield potential ranged from 4.25 to 6.47 ton dry seeds per ha. The protein content of seed resulted from cross ranged from 9.84% to 11.30%, as compared to the parents of 9.11% and 12.62%. This research concludes that genetic factors play an important role as confirmed by heritability estimate (h2)=0.75.Keywords : Corn, protein, acid-soil, tolerance

Abstract

Biodiversity of Rottboellia cochinchinensis from different areas were studied by morphological traits and amplified fragment length polymorphism (AFLP) analysis for classify of an allelopathic ability. The correlation of the similarity/distance between AFLP markers (Jaccard coefficient) and morphological traits (Euclidean distance) was significant with r = − 0.84**. Itchgrass could be divided into two groups from both UPGMA and STRUCTURE analyses: the group A consisted of itchgrass from Chaehom-Lampang, Si Thep-Phetchabun, Phrom Phiram-Phitsanulok, Amphur Muang-Nakhon Sawan, Kamalasai-Kalasin, Amphur Muang-Chachoengsao and Bang Yai-Nonthaburi, whereas itchgrass from Amphur Muang-Chiang Mai, Pak Chong-Nakhon Ratchasima and Kamphaeng Saen-Nakhon Pathom constituted the group B. Allelopathic properties of itchgrass as representative from different group were determined in bioassay test, the result showed that the aqueous extract of itchgrass from Chaehom-Lampang area has a strong allelopathic ability on growth of Echinochloa crus-galli L., Bidens pilosa L. and Lactuca sativa L. than the other group. The molecular analysis was strongly supported in morphological analysis clustering with bioassay test for allelopathic ability, specific morphological traits were soft trichomes, and the dark purple stems and roots which can be used for the preliminary classification of allelopathic ability. Our findings suggest that classification of itchgrass by morphological traits is related to the analysis of the genetic relationship of itchgrass with AFLP analysis, that allowing the assessment of the bio-diversity of itchgrass and their allelopathic potentials.

Abstract

Recently, a new bacterial disease of maize (Zea mays) was observed in Kediri, East Java, Indonesia. Infected plants showed wilt symptoms occasionally accompanied by leaf blight. This study aims to characterize the causal agent of bacterial wilt and leaf blight of maize observed in Kediri. Gram-negative, facultative anaerobic bacteria were isolated from the diseased tissues. All bacterial strains (KD1A, KB2A, KD1, KD4, KB1) gave positive result both in the hypersensitive response and pathogen-nicity assays. However, only KD1 and KB1 strains could be re-isolated from the diseased tissues. Based on several physiological and biochemical assays, the bacteria resembled Pantoea agglomerans. Moreover, the strains showed negative result on PCR amplification using HRP1d and HRP3r, a primer pair specific for detection of P. stewartii subsp. stewartii. Analysis of 16S rRNA gene sequence of KD1 and KB1 showed highest homology at 88% and 90%, respectively to P. agglomerans strain DSM 3493 (NR 0419781).  The homology values were too low to conclude that the bacteria were similar to P. agglomerans. These results suggest that bacterial pathogens isolated from maize in Kediri were strains of Pantoea sp. Keywords: maize, Pantoea sp., wilt, leaf blight

Abstract

Sea level rise is a growing threat for the coastal regions of Bangladesh. It is one of the most densely populated countries of the world where 28% of the population are living in the coastal area. Bangladesh has already been affected by sea level rise through land erosion, salinity intrusion and loss in biodiversity. Saline soil has a detrimental effect upon soil physical and chemical properties. The dominant soil textural classes that occur in the saline areas of these regions are silty clay. In both of the soils pH value of the surface horizon is slightly lower than those of the subsoil and sub stratums. Cation Exchange Capacity (CEC) of all these soils varies from 12.0 to 27.6 meq/100 g soil expressing medium to high status. The organic matter content is medium to high at Asasuni and pretty low at Kalapara. In both areas, nutrient deficiencies of total nitrogen, phosphorous and potassium were quite dominant but sulphur was high. Exchangeable sodium, potassium, calcium and magnesium were in high level. The dominant water soluble cations were Na+, Ca2+, Mg2+ and K+ and anions Cl- and SO4 2-.The amount of accumulated salt was found higher at the surface and decreases with depth.
