Abstract

Grammatophyllum scriptum is a beautiful wild orchid that has an interesting economic value. However, the existence of this ornamental plant species is at risk because of irresponsible exploitation of the natural population and insufficient cultivation efforts. Currently, the Bogor Botanic Garden is developing a program of orchid propagation for enhancing the conservation of all potential orchid collections. The propagation of G. scriptum was basically carried out by culturing the seeds in a modified Hyponex or Vacin & Went medium, transplanting the plantlets into a more suitable medium and finally acclimatizing the resulting seedlings. A study was done to observe the effect of fertilizer application on the growth of acclimatized G. scriptum seedlings, as a means to improve the propagation technique of the species. The result showed that the application of 1 and 2 m1/I organic fertilizer (Sugih) as well as 1 and 2 g/I inorganic fertilizer (Hyponex) increased the seedling mortality, in which the application of 2 g/ml Hyponex fertilizer was being the most damaging treatment, leaving 12,5 % seedlings to survive. However, in terms of seedling growth, the application of 1 m1/I Sugih fertilizer was slightly beneficial to the formation of root and the elongation of stem of the acclimatized seedlings. It may be suggested, therefore, that G. scriptum seedlings require no (or very little amount of) supplementary fertilizer to boost their vegetative growth.

Abstract

Mature seeds of a pitcher plant (Nepenthes grocilis Korth.) were cultured in seven different media, namely half strength MS medium, half strength MS medium supplemented with BAP and NAA, 0.8 KC medium supplemented with coconut water + bean sprout extract, 0.8 KC + H3B03 + CuSO3 + ZnSO4 + coconut water + bean sprout extract, 0.25 KC + Vitamin (MS), HYPONeX + coconut water + sprout bean extract, and HYPONeX + potato extract. Each culture was incubated under light conditions. Young seedlings were subsequently transferred onto fresh media of 0.5 MS and 0.25 KC supplemented with 0, 5, 10, and 15 mg/I GA3 after 15 weeks of sowing. Seed germination and seedling growth were observed weekly. It was noted that the seeds started to germinate in ten weeks after incubation and the highest seed germination was obtained in HYPONeX medium + potato extract. However, the appearance of the seedlings was better on MS media and KC media without organic compound than on the other media. Seedlings in all level of GA3 in half strength MS media and 0.25 KC media grew faster than those in media without GA3.

Abstract

The Family of Magnoliaceae is one of the most primitive taxa in the world.  Knowledge of this family is essential for studies on the origin, evolution and systematics of Angiosperms.  There are 223 species belongs to this family in the world and 25 of them are found in Indonesia. This paper explains taxonomy, distribution, and conservation status of the family Magnoliaceae in Indonesia.

Abstract

Bamboo occurs from lowland to highland. It is commonly utilized for construction, handicrafts and households, but its use as medicine is less known. This inventory revealed that there are six bamboo species that are potential to be used as medicine, i.e. bambu tali (Gigantochloa opus (J.A. & J.H. Schult.) Kurz), bambu ampel gading (Bambusa vulgaris Schrad. ex Wendl. var stricta), bambu pingit (Gigantochloa hasskarliana (Kurz) Backer ex Heyne), bambu tamblang gading (Schizostachyum brachycladum Kurz), bambu buluh lengis (Schizostachyum lima (Blanco) Merr.) and bambu petung (Dendrocalamus asper (Schult.) Backer ex Heyne). Further research is needed to determine the content of active compounds of those bamboo.

Abstract

Dringo (Acorus calamus L.) used as medicinal plant in Indonesian ethnic groups. Those information based on Ristoja 2012 research. The objective of Ristoja was to provide a database of local ethnomedicine knowledge, herbal formula and medicinal plant in Indonesia. Inter Simple Sequence Repeats (ISSR) markers were used to assess the genetic diversity of Dringo from 20 selected ethnic groups in Indonesia. Ten selected ISSR primers generated 82 amplified fragments with 51,2% were polymorphic. Dice coefficient was used to calculate similarity index and UPGMA was used to construct a dendogram. The genetic similarity index among accessions ranged from 76,7-100% thus indicating that low level of genetic diversity in dringo. Genetic diversity database can be useful for medicinal plant mapping and conservation especially for in situ conservation.

Abstract

Leaf anatomy of Piperaceae from Gunung Slamet, Central Java has made with standard  paraffin and paradermal method. The results showed that Piperaceae has dorsiventral leaf, hypostomatatic or amphistomatatic, tetracytic and cyclocytic stomata type. Hypoderm tissue exist in upper and/or lower part of leaf, mesophyll was arranged by 1-2 layers of palisade and 2-7 layers of sponge tissue. There were 3 types of trichome i.e. glandular trichome which had globose cell with short stalk, bicellular glandular trichome and multicellular & uniseriate non-glandular trichome. The shape of the crystal of Piperaceae from gunung Slamet are sand, needle, drusse or prismatic which distributed in hypodermis, mesophyll and midrib. Piperaceae petiole was arranged by open or sometimes closed circle vascular bundles. These open vascular bundles were crescent shape or resemble to U and V shapes.

Abstract

Hubungan kekerabatan (filogenetik) Litsea dan marga-marga yang berdekatan (Actinodaphne, Lindera and Neolitsea) dari suku Lauraceae telah diamati dengan menggunakan data molekular. Analisis dilakukan pada sekuens nukleotida dari gen-gen kloroplas matK dan ndhF. Walaupun gen-gen ini dikenal sebagai untaian pengkode yang berevolusi dengan cepat, tetapi untuk taksa tumbuhan yang diamati pada penelitian ini menunjukkan variasi yang rendah. Pohon-pohon filogenetik hasil analisis gabungan dari sekuens nukleotida matK dan ndhF menunjukkan bahwa marga Litsea tidak monofiletik. Di antara empat seksi dari marga Litsea (seksi Tomingodaphne, seksi Litsea, seksi Conodaphne dan seksi Cylicodaphne), hanya seksi Litsea yang monofiletik.

Abstract

A research was carried out in Orchid Laboratory of Bogor Botanical Gardens to observe the effect of gamma ray irradiation on the growth variation of Brachypezo indusiata (Reichb.f) Garray and suitable culture medium for propagating the plant. The research was arranged in a factorial experiment with a Completely Randomized Design. The first factor was gamma ray irradiation on protocorm-like­bodies (plb) of germinated seeds of B. indusiata in which four levels of irradiation was applied, i.e. 0, 10, 20, and 30 Gy. The second factor was the use of two different culture media, i.e. Vacin & Went medium supplemented with either banana pulp or a mixture of tomato and sprout bean as the essential organic compound of the medium. The result showed that the irradiation treatment improved the survival of B. indusiata explants, most significantly at 20 Gy level of irradiation, and could modify the growth of the resulted plantlets by reducing the height of the plant and the length and width of the leaves. The higher the level of irradiation the smaller the plantlets produced. Meanwhile, the addition of organic compound derived from banana fruit into the basal medium of Vacin & Went enhanced the growth of the plantlets better than that derived from a mixture of tomato fruit and bean sprout. However, there was no significant interaction effect detected between the irradiation treatments and the use of different culture media.

Abstract

Conservation areas with the objective for collection and exchange plant materials have been speculated as weed bank for surrounding areas. Objective of this study was to identify and characterize ruderal invasive weeds in the Bogor Botanic Gardens (BBG). Observations were conducted in all vak (collection blocks) in the BBG in order to identify the weeds species, determine their invasiveness, dominance and distribution. Weeds associations with host plants were observed. Current weed control program and data of dead trees collection were analyzed in relevant to weed. Distribution of weeds outside BBG was observed by transects method following river and road directions. Results showed that there were seven invasive weeds, i.e., Cecropia adenopus (Cecropiaceae), Cissus nodosa Blume (Vitaceae), Cissus sicyoides Blume (Vitaceae), Dioscorea bulbifera L. (Dioscoreaceae), Ficus elastica Roxb. (Moraceae), Mikania micrantha H.B.K. (Asteraceae) and Paraserianthes falcataria (L.) Nielsen (Fabaceae). These seven weeds species invaded 41 out of 215 plant families in BBG. Six species of weeds, i.e., C. adenopus, C. nodosa Blume., C. sicyoides Blume., D. bulbifera L., M. micrantha H.B.K. and P. falcataria (L.) Nielsen, were introduced as BBG collections for the first time while the F. elastica Roxb was considered as native. It is most likely that the weeds dispersal agents are the wind, birds, bats, visitors, and waters. All of these weeds existed in surrounding areas outside BBG. Given the detrimental impact of invasive weeds on the plant collection in BBG, it is necessary to develop long–term comprehensive control measures both inside and neighboring areas by involving other government authorities beyond BBG.
