Abstract

Consumption of black soybean tempe can provide rapid improvement in children with poor nutritional status and/or suffering from diarrhea. The antioxidant activity of black soybean tempe is higher than yellow soybean tempe. This study aimed to examine the effect of the amount of black soybean tempe in the feed on hematological status, IgG levels, and IgA levels of rat blood. There was a total of 6 groups, each containing 5 rats. The rats were kept in animal laboratories at room temperature (25–27°C) for 25 days. The six groups were induced Enteropathogenic Escherichia coli (EPEC) suspension with a concentration of 107 CFU / mL of 1 mL per day for seven days, orally using a feeding tube. Parameters analyzed included hematology status, IgG levels and IgA levels. The administration of black soybean tempe had an effect on increasing hemoglobin levels, the number of erythrocytes and the percentage of hematocrit, and it gave the effect of decreasing the number of leukocytes and platelets of rat blood induced with EPEC bacteria. The group of rats that consumed black soybean tempe had no influence on IgG and IgA levels of blood plasma.

Abstract

Tapai is a cassava food product fermented by yeast starter (“Ragi”). Among the amylolitic yeasts that had been isolated from the tapai were Candida guilliermondii, C. tropicalis, Trichosporon mucoides, and Saccharomycopsis fibuligera. The aim of this research was to characterize the physicochemical properties of fermented cassava starch by amylolytic yeasts of indigenus tapai i.e. C. guilliermondii, C. tropicalis, T. mucoides, and S. fibuligera. The process of fermentation of cassava starch was done by inoculating each pure isolate by 5% v / v (105 CFU / ml) into cassava starch suspension 50% b/v (100g starch into 200ml sterile distilled water). The incubation was carried out at 28 °C for 24 h, 48 h, and 72 h. The fermented cassava starch was dried at 50 °C for 24 h. Characterization of physicochemical properties of cassava starch included chemical properties (amylose content), paste properties (RVA), thermal properties (DSC), physical properties (SEM granule macrostructure) and functional groups (FT-IR) of unfermented (native) or fermented cassava starch. The results showed that during the fermentation process, the yeast was able to grow up to 7 log10 CFU/ml and the acidity decreased to pH 3,9. The amylose content of fermented cassava starch decreased to 11,22% from native starch amylose content (15,85%). The paste properties of fermented cassava starch increased at the value of trough viscosity, breakdown, final viscosity, and swelling power. The thermal properties of fermented cassava starch increased to 64,7-67 °C for the initial temperature value of gelatinization (To), 69,1-71,2 °C for gelatinization peak temperature (Tp), 73,9-75 °C for the final temperature of gelatinization (Tc), but decreased gelatinization enthalpy in the range of ΔH 4,84-6,38 J/g. The macrostructure of the longest fermented starch granules (72 hours)occurred liberation into irregular granular shapes. The profile of fermented cassava starch functional groups had a similarity with native cassava starch that had the highest absorption spectra at 3291,2 cm-1 peak (vibration of the O-H bonding group) and 2920 cm-1 peak (C-H group vibration). ABSTRAKTapai merupakan produk pangan ubi kayu yang terfermentasi oleh starter khamir (ragi). Di antara khamir amilolitik yang sudah diisolasi dari tapai yaitu Candida guilliermondii, C. tropicalis, Trichosporon mucoides, dan Saccharomycopsis fibuligera. Tujuan penelitian ini adalah melakukan karakterisasi sifat-sifat fisikokimia pati ubi kayu yang difermentasi oleh khamir amilolitik indigenus tapai yaitu C. guilliermondii, C. tropicalis, T. mucoides, dan S. fibuligera. Proses fermentasi pati ubi kayu dilakukan dengan menginokulasikan masing-masing isolat murni sebanyak 5% v/v (105 CFU/ml) ke dalam suspensi pati ubi kayu 50% b/v (100g pati ke dalam 200ml akuades steril). Selanjutnya dilakukan inkubasi pada suhu 28 °C selama 24, 48, dan 72 jam. Pati ubi kayu terfermentasi dikeringkan pada suhu 50 oC selama 24 jam. Karakterisasi sifat-sifat fisikokimia pati ubi kayu meliputi sifat kimia (kadar amilosa), sifat pasta (RVA), sifat termal (DSC), sifat fisik (makrostruktur granula SEM) dan gugus fungsi (FT-IR) pati ubi kayu alami maupun pati ubi kayu terfermentasi. Hasil penelitian menunjukkan bahwa selama proses fermentasi, khamir mampu tumbuh hingga mencapai 7 log10 CFU/ml dan derajat keasaman mengalami penurunan mencapai pH 3,9. Kadar amilosa pati ubi kayu terfermentasi mengalami penurunan menjadi 11,22% dari kadar amilosa pati alami (15,85%). Sifat pasta pati ubi kayu terfermentasi khamir mengalami peningkatan pada nilai viskositas trough, breakdown, viskositas akhir, dan daya kembang. Sifat termal pati ubi kayu terfermentasi menglamai peningkatan menjadi 64,7-67 °C untuk nilai suhu awal gelatinisasi (To), 69,1-71,2 °C untuk suhu puncak gelatinisasi (Tp), 73,9-75 °C untuk suhu akhir gelatinisasi (Tc), tetapi mengalami penurunan entalpi gelatinisasi pada kisaran ∆H 4,84-6,38 J/g. Makrostruktur granula pati yang terfermentasi paling lama (72 jam) mengalami liberasi dengan bentuk granula yang tidak beraturan. Profil gugus fungsi pati ubi kayu terfermentasi oleh khamir memiliki kemiripan dengan pati ubi kayu alami yaitu memiliki spektra penyerapan tertinggi pada puncak 3291,2 cm-1 (getaran gugus ikatan O-H) dan puncak 2920 cm-1 (getaran gugus C-H).

Abstract

This study was aimed to examine the correlation between temperature and duration of steaming on caffeine and chlorogenic acid reduction as well as to determine energy activation (Ea) according to Arrhenius equation. About 750 g of Robusta coffee was steamed by autoclaving at 100, 110, 120 °C for 1, 2, 3, 4, 5, 6 and 7 hours. The study was conducted in 3 replications. Analyses of caffeine and chlorogenic acid were performed using HPLC.  Steaming coffee beans at 100, 110 and 120 °C resulted in the highest decrease of caffeine and chlorogenic acids content for 7 hours by 13%, 18% and 25% for caffeine; and by 37%, 50% and 59% for chlorogenic acids. At all temperatures investigated, decaffeination and chlorogenic acid reduction followed first order reaction. The decaffeination equation at 100 °C, 110 °C and  120 °C followed equation of  y = -0.019x + 0.862, y = -0.023x + 0.820, and y = -0.033x + 0.759, respectively. Meanwhile, the chlorogenic acid reduction at 100 °C, 110 °C,  and 120 °C followed equation of y = -0.071x + 1.421, y = -0.089x + 1.271, and y = -0.120x + 1.201. Activation energies of decaffeination and chlorogenic acid reduction were 33,543.66 kJ/mol K and 31,934.91 kJ/mol K, respectively. ABSTRAKPenelitian ini bertujuan untuk mengkaji hubungan antara suhu dan lama pengukusan terhadap penurunan kandungan kafein dan asam klorogenat serta menentukan energi aktivasi (Ea) dengan pendekatan persamaan Arrhenius. Biji kopi Robusta masing-masing seberat 750 g dikukus dalam autoklaf (sistem tertutup) pada suhu 100, 110, dan 120 °C masing-masing selama 1, 2, 3, 4, 5, 6, dan 7 jam. Penelitian dilakukan dengan 3 kali ulangan. Analisis kafein dan asam klorogenat dilakukan menggunakan HPLC. Pengukusan biji kopi pada suhu 100, 110, dan 120 °C, menghasilkan penurunan kandungan kafein dan asam klorogenat tertinggi pada pengukusan selama 7 jam. Penurunan kandungan kafein selama 7 jam pada suhu pengukusan 100, 110, dan 120 °C berturut-turut 13%, 18%, dan 25%. Kandungan asam klorogenat mengalami penurunan 37%, 50%, dan 59% berturut-turut pada suhu 100, 110, dan 120 °C.  Pada semua suhu yang diuji, penurunan kafein dan asam klorogenat mengikuti reaksi orde satu. Penurunan kafein mengikuti persamaan y = -0,019x + 0,862,  y = -0,023x + 0,820, dan y = -0,033x + 0,759, sedangkan untuk asam klorogenat mengikuti persamaan y = -0,071x + 1,421,  y = -0,089x + 1,271, dan y = -0,120x + 1,201. Besarnya energi aktivasi penurunan kafein dan asam klorogenat berturut turut adalah 33.543,66 kJ/molK dan 31.934,91 kJ/Mok.

Abstract

Daging ayam merupakan salah satu bahan pangan yang memegang peranan penting sebagai sumber protein hewani dalam pemenuhan kebutuhan gizi masyarakat. Penelitian ini bertujuan untuk (1) mengetahui adanya daya hambat ekstrak kulit dan jantung pisang muli sebagai antimikroba alami dalam menurunkan cemaran Escherichia coli, (2) menentukan konsentrasi terbaik ekstrak kulit dan jantung pisang muli sebagai antimikroba alami untuk menurunkan cemaranEscherichia coli, (3) mengetahui pengaruh penggunaan ekstrak kulit dan jantung pisang muli sebagai antimikroba alami dalam penurunan cemaranEscherichia coli pada daging ayam. Hasil penelitian menunjukkan bahwa ekstrak kulit dan jantung pisang muli memiliki daya hambat sebagai antimikroba alami dalam menurunkan cemaran bakteri Escherichia coli. Hasil penelitian menunjukkan ekstrak kulit pisang muli memiliki daya hambat pertumbuhan bakteri E. coli dengan diameter sebesar 6.45 mm dengan aktivitas antibakteri sedang. Penghambatan ekstrak jantung pisang muli menunjukkan mampu menghambat pertumbuhan bakteri E. coli dengan diameter hambat sebesar 5.63 mm dengan kategori aktivitas antimikroba sedang. Konsentrasi terbaik ekstrak kulit dan jantung pisang muli sebagai antimikroba alami untuk menurunkan cemaran Escherichia coli yaitu masing-masing konsentrasi ekstrak 100% pada taraf nyata 5%. Ekstrak kulit dan jantung pisang muli mampu menurunkan cemaran bakteri Escherichia coli pada daging ayam dengan kategori rendah, dengan angka penurunana dari 8log1.8 menjadi 7log3.0 koloni/g (ekstrak kulit pisang) dan 8log1.6 menjadi 7log4,4 (ekstrak jantung pisang). Total penurunan mikrobia oleh ekstrak kulit dan jantung pisang hanya 1log cycle dengan kategori kativitas anti bakterinya adalah rendah.

Abstract

Maize is one of the important staple foods for people in Timor, East Nusa Tenggara Province, Indonesia. Subsistent farmers store the maize for their own consumption until the next harvest season, for seed and feed.  However, high initial water content of the kernel due to improper drying prior storage initiate serious damage and losses during the maize storage.  High water content promotes the growth of fungi and insects, and increase respiration rate, resulting in rapid deterioration of maize. The purpose of this study was to determine the initial moisture content that might minimize damage and losses of maize in the farmers’ storage, and to study the effects of some plant materials that are used to smoke corns before storage. The experiment was initiated by sun-drying the harvested corncobs for 0, 2, 4, 6, 8, and 10 days (6 hours a day). This experiment was designed using Completely Randomized Design with 6 treatments and 3 replications. Dried corncobs were stored in the farmer’s storage for 4 months. The effects of maize kernels’ initial water content on the development of water content in kernels; the percentage of damaged kernels; and the species of pathogen and insects were investigated during storage with 2-week intervals.  The results demonstrated that drying the corncobs prior storage for 10 days, resulting in 12.96% of water content, significantly decreased the percentage of seed damage to 6.5%, as compared to without drying process which resulted  in 63%.  Aspergillus flavus, Fusarium sp., and Penicillium sp were found to be the main pathogen during storage.  There are no insect pests found during the storage.

Abstract

Angkak, commonly used for food colorant and flavor enhancers in oriental cuisine, is the result of fermentation by Monascus purpureus on steamed rice. In addition to producing pigments Monascus purpureus, Angkak also produces mycotoxins, citrinin, which is hepato-nephrotoxic. Biosynthesis of pigment and citrinin is following a polyketide synthase pathway and then subdivides to form pigment or citrinin. Fatty acids and amino acids are known to be the precursors of red pigment formers in their biosynthetic pathways. The purpose of this research was to know the effect of addition of lauric fatty acid and amino acid glycine on steamed rice substrate to the color characteristic and citrinin level by M. purpureus JK9A. The amount of fatty acid and glycine was respectively 0.5% and 1% (w/w). Fermentation was carried out for 14 days and every two days the solids content of fermented products, pH, number of cells, colors, pigments dissolved in water were measured. The level of citrinin was tested at the end of the fermentation period (14th day). There was no significant difference between treatments for the solids content of about 23  ̶  29% and the number of cells 6.32  ̶  6.56 logCFU/g. While the pH value, color and water-soluble pigments were significantly different between treatment and control. The ˚hue values of glycine and combination of lauric-glycine were 16.11 and 15.33, respectively, lower than controls (22.76). The highest A500nm/A400nm ratio was in the combination treatment of lauric-glycine and the lowest levels of citrinin also in the treatment of lauric-glycine combination. This study noticed that the addition of lauric or glycine and its combination in rice media for Monascus purpureus JK9A fermentation proved to increase the biosynthesis of red pigment (46.34%) and decrease citrinin level up to 49.97%.

Abstract

The corn cob is one of the food waste-material having phytochemical component that of health benefit. The objectives of the research were to isolate phenolic fractions of the corn cob extract and to determine antioxidant and antiphotooxidant activities. The corn cob was extracted with reflux method using ethanol 80% for 2 hours. After that the extracts were filtered and the filtrates were combined and concentrated on rotary evaporator. This crude ethanolic extract was suspended in water and extracted with petroleum ether, ethyl acetate, n-butanol and water, respectively. The best fractions of partition fractionated with colomn chromatography using silica gel 60 and n-hexane: ethyl acetate (4:6). Corncob extract and fraction were evaluated for total phenolic content free radical scavenging activity (1,1-diphenyl-2-picrylhydrazyl radical assay) and total antioxidant capacity (ferric reducing ability of plasma assay). The results of total phenolic content showed that ethyl acetate fraction (163.57 μg/mL) has higher phenolic content than butanol fraction (83.30 μg/mL), ethanol extract (81.53 μg/mL), water fraction (23.71 μg/mL) and petroleum ether fraction (23.57 μg/mL). The results also showed that ethyl acetate fraction has highest free radical scavenging activity and total antioxidant capacity than butanol fraction, ethanol extract, water fraction and petroleum ether fraction. The free radical scavenging activity of fractions II and III exhibited strongest activity than fraction V, I, IV and VI. The results also showed that fraction II has highest total antioxidant capacity than I, III, IV, VI, and V. The results suggest that the ethyl acetate fraction and fraction II contains phenolic compounds which possess antioxidant activities. ABSTRAKTongkol jagung merupakan salah satu limbah tanaman pangan yang mempunyai kandungan fitokimia yang bermanfaat untuk kesehatan. Penelitian ini bertujuan untuk mengisolasi fraksi fenolik dari ekstrak tongkol jagung dan menentukan aktivitas antioksidan. Tongkol jagung diekstraksi dengan cara refluk menggunakan etanol 80% selama 2 jam. Setelah itu disaring dan filtratnya diuapkan dengan rotary evaporator. Ekstrak etanol disuspensikan dalam air dan diekstraksi berturut turut dengan petroleum eter, etil asetat, butanol, dan air. Fraksi pelarut terbaik difraksinasi dengan kromatografi kolom menggunakan silika gel 60 dan eluen n-heksana : etil asetat (4:6). Ekstrak tongkol jagung dan fraksi dievaluasi kandungan total fenolik, aktivitas penangkal radikal bebas (uji 1,1-difenil-2-pikrilhidrazil) dan kapasitas total antioksidan (uji ferric reducing ability of plasma). Hasil uji kandungan total fenolik menunjukkan bahwa fraksi etil asetat (163,57 μg/mL) memiliki kandungan total fenolik yang paling tinggi daripada fraksi butanol (83,30 μg/mL), ekstrak etanol (81,53 μg/mL), fraksi air (23,71 μg/mL) dan fraksi petroleum eter (23,57 μg/mL). Hasil ini juga menunjukkan bahwa fraksi etil asetat mempunyai aktivitas penangkal radikal dan kapasitas total antioksidan paling tinggi daripada fraksi butanol, ekstrak etanol, fraksi air, dan fraksi petroleum eter. Aktivitas penangkal radikal bebas fraksi II dan III memperlihatkan paling kuat daripada fraksi V, I, IV dan VI. Hasil penelitian ini menyarankan bahwa fraksi etil asetat dan fraksi II mengandung senyawa fenolik yang mempunyai aktivitas antioksidan kuat.

Abstract

This experiment aims to the observed morphology, reduction of fineness and distribution particle deterioration of W/O/W double emulsion in instant noodle seasonings which is kept in 3 weeks with different storage temperature and NaCl level treatments. Emulsion structure has an important role to hamper salt release rate from internal to external phase. Structure breakdown shows system inability to maintain continuous salty taste perception during consumption because of the increasing salt release rate in storage period of instant noodle seasoning. Samples are treated with 3 variations of storage temperatures which are low (4 °C), room (25 °C), high temperature (40 °C) and 6 variations of NaCl level which are 0; 0,2; 0,4; 0,6; 0,8; 1%. Samples are categorized into 2 groups, double emulsions, and instant noodle seasonings. The double emulsion is made by 2 phases emulsification to get primary W/O emulsion and final W/O/W emulsion. This experiment showed that low and high-temperature storage affected emulsion and seasoning particle morphology, fineness and distribution throughout several instability phenomena.

Abstract

Fluctuation of the availability and prices of chili happen because the poor of the value chains. This research intended to know the mechanism related to the product, financial and information flow and development strategy to solve the problem of the chili’s value chain on the slopes of Mount Merapi. This research was conducted in Pakem District, Sleman Regency from May-August 2017. The method to determine the farmer respondents is random sampling while the merchants chosen by snowball sampling. The research used descriptive and SWOT analysis. The results showed that the most efficient marketing channels when farmers sell chili to the consumers through the retailers. Overall, the chili’s value chain included in the current category. Unfortunately, there are still some problems such as weak of the farmer cultivation ability and fluctuation of production and price. To solve this problem, the local government need make intensive counseling and training of chili cultivation, post-harvest handling in a professional and strengthen the auction market in Sleman Regency.

Abstract

Seaweed is among fishery commodities with great potential economy prospect. Southeast Maluku District is one of the main producers in Eastern region of Indonesia. Despite the high production since 2012, the number of farmers and the product has declined due to inadequate supply chain coordination and information dissemination among members. Therefore, this study aimed to mitigate the assymetric information in the region using the house of risk (HOR) to identify the risks to be addressed, and also provide response on the source of supply chain risk. Furthermore, Analytic hierarchy process (AHP) with fuzzy approach was used to determine the major factor, and then choose the best alternative to mitigate asymmetric information in the supply chain. Results showed there were five factors that contributed 70% risks. The results also indicated that dependence on local distributor was a factor that had to be prioritized and addressed. In addition, to mitigate the operational risks, findings showed it is necessary to establish seaweed farmers forum, which is the best approach based on the cost and effectiveness. This study also stated that local ,government of Southeast Maluku District was the main actor that helps to overcome the risks and asymmetric information problem. Therefore, the best alternative was to form an information center for seaweed cultivation, which will provide the knowledge of prices and potential buyer outside the region.
