Abstract

The present study aimed to evaluate the levels of chlorophyll and carotenoids, as well as the antioxidant potential of various seaweed species collected from the waters of Kartini Beach, Jepara, Central Java, Indonesia. Nine seaweed samples were extracted using methanol, and the pigment content was determined using spectrophotometric analysis. The antioxidant activity of the extracts was assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. The findings indicated that the green seaweed Caulerpa serrulata exhibited the highest concentrations of chlorophyll a (489.42 µg/g) and carotenoids (248.83 µg/g). Conversely, the brown seaweed Sargassum echinocarpum demonstrated superior antioxidant properties, with DPPH radical scavenging activities of 92.59%, followed by C. serrulata extracts at 76.43%.  The antioxidant activity of these extracts exceeded that of ascorbic acid at a concentration of 20 µg/mL (71.21%). A moderate positive correlation was observed between pigment content and antioxidant activity. The variations in pigment composition among different seaweed species significantly influenced their antioxidant potential, with carotenoid-rich brown seaweeds exhibiting higher antioxidant activity compared to green seaweeds. The study's findings reveal the potential of C. serrulata and S. echinocarpum extracts as natural pigments with antioxidant properties. These extracts offer promising applications in the food, nutraceutical, and cosmetic industries as safe and sustainable alternatives to synthetic antioxidants and colorants.

Abstract

Coastal ecotourism development requires an integrated understanding of environmental suitability, ecological carrying capacity, and management strategies to ensure long-term sustainability. This study evaluates the suitability of tourism, carrying capacity, and development strategies for sustainable coastal ecotourism at Karangsong Beach, Indramayu Regency, Indonesia. Tourism suitability and carrying capacity were assessed based on biophysical parameters, field observations, and stakeholder surveys. Tourism suitability was analyzed using the Tourism Suitability Index (TSI), while the Area Carrying Capacity (ACC) was calculated to determine the maximum number of visitors that can be accommodated without compromising environmental quality or visitor comfort. Strategic priorities for ecotourism development were identified using an integrated SWOT analysis and Quantitative Strategic Planning Matrix (QSPM). The results indicate spatial variation in tourism suitability across the study area, with several coastal segments classified as highly suitable for tourism activities, while others require management interventions. The carrying capacity analysis suggests that Karangsong Beach can sustainably accommodate an average of 184 visitors per day under current environmental conditions. Priority development strategies include improving and maintaining tourism facilities, enhancing accessibility, strengthening community participation in tourism management, and conserving mangrove ecosystems as key ecotourism attractions. This study demonstrates that integrating tourism suitability assessment and carrying capacity analysis provides a robust scientific framework for sustainable coastal ecotourism planning. The findings offer practical insights for balancing tourism development, coastal ecosystem conservation, and community-based economic benefits and contribute to sustainable marine ecotourism management in tropical coastal regions.

Abstract

Astaxanthin, a carotenoid with pronounced antioxidant properties, represents a promising candidate for development as a functional food ingredient grounded in local wisdom. Traditional rebon shrimp derivatives including dried rebon, terasi, and cincalok are recognized sources of this pigment; however, systematic data on extraction efficiency, astaxanthin concentration, and antioxidant activity remain scarce. This study evaluated three extraction techniques and characterized antioxidant activity using 2,2-difenil-1-pikrilhidrazil (DPPH), azinobis(3-ethylbenzothiazoline)-6-sulfonic acid (ABTS), and 4,6-tris(2-pyridyl)-s-triazine (FRAP) assays. Samples were vacuum-dried, ground, and subjected to oil extraction, followed by qualitative thin-layer chromatography and quantitative UV-Vis spectrophotometry. Soxhlet extraction yielded the greatest oil output, whereas maceration preserved astaxanthin more effectively due to the absence of thermal degradation. Cincalok exhibited the highest astaxanthin content (10.943 mg/g), followed by terasi and dried rebon. DPPH analysis indicated that cincalok oil possessed the strongest antioxidant activity (IC₅₀ 54.58 mg/L, Soxhlet), while dried rebon demonstrated the weakest. In contrast, ABTS and FRAP assays revealed lower responses for cincalok oil, suggesting that its primary antioxidant mechanism involves free-radical scavenging. These findings highlight cincalok as the most promising natural source of astaxanthin and local antioxidants, underscoring its potential for the development of coastal-based functional foods and health-promoting products.

Abstract

Intensive Pennaeus vannamei aquaculture is a critical reservoir for the emergence of antimicrobial resistance (AMR), driven by the extensive use of antibiotics under high-density production conditions. This study aimed to isolate, characterize, and identify antibiotic-resistant bacteria from intensive shrimp pond environments in the Rowosari and Kaliwungu Districts of Kendal Regency, Central Java, Indonesia. Water, sediment, and shrimp samples were collected via purposive sampling and cultured on thiosulfate-citrate-bile salts sucrose (TCBS) and Zobell Marine Agar. Antimicrobial susceptibility testing was performed using the Kirby-Bauer disk diffusion method, following CLSI guidelines, against erythromycin (ERY), oxytetracycline (OXY), enrofloxacin (ENR), and amoxicillin (AMX). Molecular identification was conducted via 16S rRNA gene sequencing with BLAST homology analysis and neighbor-joining phylogenetic reconstruction using MEGA-X. Four isolates were recovered and characterized in this study. Oxytetracycline resistance was detected universally, and isolate A23 exhibited complete resistance to all tested antibiotics. Molecular analysis identified A23 as V. injensis strain VS1, B15 as V. metschnikovii strain Vm-4, and B12 and B17 as Staphylococcus arlettae, each with 100% sequence homology with reference strains. These findings confirm the presence of diverse AMR bacterial assemblages in intensive shrimp pond environments in Kendal Regency and underscore the critical need for integrated AMR surveillance and prudent antibiotic stewardship in Indonesian shrimp aquaculture.

Abstract

Accurate and rapid species identification of snappers (Lutjanus spp.) is essential for biodiversity assessment and fisheries resource management, particularly in regions with high biodiversity such as the waters of Kaimana and Fakfak, West Papua. This study aimed to identify the economically important checkered snapper (Lutjanus decussatus) using a molecular approach based on DNA barcoding of the cytochrome c oxidase subunit I (COI) gene marker. Six fish tissue samples were collected from the waters of Kaimana and Fakfak, followed by molecular analyses including DNA extraction, PCR amplification, and sequencing. The obtained sequences had a length of 644 base pairs. Sequence data were analyzed by comparison with reference databases using BLAST and the Barcode of Life Data System (BOLD). The results showed that all sample sequences exhibited 100% similarity to reference sequences of L. decussatus in the databases and clustered into a single clade consistently supported by high bootstrap values and low intraspecific genetic distances. The Interspecific and outgroup sequence was clearly separated from the main clade, indicating distinct interspecific divergence. These findings demonstrate the effectiveness of COI-based DNA barcoding for species identification of L. decussatus, and the obtained genetic information can support further studies on genetic diversity and resource management in West Papua.

Abstract

Fenomena upwelling adalah salah satu fenomena kenaikan massa air yang terjadi di perairan Banggai, Sulawesi Tengah, yang mengakibatkan tingginya kandungan nutrien yang memengaruhi keberadaan fitoplankton dan zooplankton, salah satunya zooplankton famili Paracalanidae. Penelitian ini bertujuan untuk mengetahui keberadaan zooplankton famili Paracalanidae di perairan Banggai, Sulawesi Tengah. Pengambilan sampel zooplankton dilakukan di 35 stasiun menggunakan plankton net berukuran 200 μm secara aktif secara vertikal.  Dari kedalaman 150 m hingga permukaan perairan. Berdasarkan hasil pengamatan famili Paracalanidae, ditemukan spesies-spesies berikut: Acrocalanus gracilis, Acrocalanus longicornis, Bestiolina similis, Paracalanus aculeatus, Paracalanus denudatus, Paracalanus nanus, Paracalanus parvus, Parvocalanus crassirostris, dan Parvocalanus elegans. Kelimpahan tertinggi ditemukan pada stasiun 11 dengan nilai kelimpahan 247 ind/m³, lalu diikuti oleh stasiun 20 dengan nilai 236 ind/m³, dan stasiun 23 dengan nilai 232 ind/m³.

Abstract

Green mussels (Perna viridis) represent a significant aquaculture commodity in tropical coastal waters, including Tambak Lorok, Semarang, which is subject to substantial anthropogenic influences from urban settlements, port activities, and land-based waste discharges. Understanding the length-weight relationship (LWR) is essential for elucidating growth patterns, physiological conditions, and population dynamics of this species. This study aimed to analyze the spatial-temporal variation of LWR in P. viridis from Tambak Lorok waters while considering water-quality parameters. Sampling was conducted monthly from May to September 2025 across three cultivation stations using random sampling, yielding a total of 750 individuals. Concurrent measurements of environmental parameters, including temperature, salinity, pH, and dissolved oxygen, were recorded during sampling. The logarithmic equation W = aL^b was applied to determine the length-weight relationship, where W represents weight (g) and L denotes shell length (mm). The results revealed a very strong correlation coefficient (R² = 0.908). The allometric coefficient (b) ranged temporally from 2.294 to 2.580 and spatially from 2.144 to 2.562. All b values were consistently below 3, indicating a negative allometric growth pattern, wherein P. viridis in Tambak Lorok tends to elongate faster than it gains weight. These findings contribute valuable baseline data on the population condition and growth characteristics of green mussels in urban coastal ecosystems, supporting sustainable aquaculture management and environmental monitoring programs in the region.

Abstract

Mangrove merupakan ekosistem pesisir yang tumbuh di zona pasang surut (intertidal) daerah tropis dan subtropis serta menjadi habitat berbagai jenis kepiting. Penelitian ini bertujuan untuk mengetahui struktur komunitas kepiting pada ekosistem mangrove di Desa Pasar Banggi, Kabupaten Rembang. Pengambilan data dilakukan pada empat stasiun pengamatan, masing-masing terdiri atas tiga plot, menggunakan metode deskriptif eksploratif. Analisis meliputi komposisi spesies, kelimpahan, indeks keanekaragaman, keseragaman, dominansi, dan pola sebaran. Hasil penelitian menunjukkan bahwa ekosistem mangrove Desa Pasar Banggi dihuni oleh tujuh spesies kepiting. Nilai rata-rata kelimpahan kepiting berkisar antara 0,02–0,06 ind/m². Indeks keanekaragaman (H') berada pada rentang 1,60–1,77 yang menunjukkan kategori sedang. Indeks keseragaman (E) berkisar antara 0,96–1,00 dan termasuk kategori tinggi, menandakan distribusi individu antarspesies relatif merata. Sementara itu, indeks dominansi menunjukkan tidak adanya spesies yang mendominasi komunitas. Analisis pola sebaran memperlihatkan bahwa sebagian besar spesies kepiting memiliki pola sebaran seragam (uniform). Berdasarkan hasil tersebut, dapat disimpulkan bahwa komunitas kepiting pada ekosistem mangrove Desa Pasar Banggi memiliki tingkat keanekaragaman sedang, keseragaman tinggi, tidak terdapat spesies dominan, serta didominasi oleh pola sebaran seragam. Temuan ini menunjukkan kondisi komunitas kepiting yang relatif stabil dalam mendukung keseimbangan ekosistem mangrove.

Abstract

Karakteristik ukuran butir sedimen merupakan variabel sedimentologi penting untuk memahami proses pengendapan di Laut Maluku. Namun, penelitian mengenai hal ini belum didukung oleh ketersediaan data granulometri vertikal yang memadai. Penelitian ini bertujuan untuk merekonstruksi dinamika lingkungan pengendapan di Laut Maluku berdasarkan variasi vertikal karakteristik ukuran butir sedimen. Sampel inti sedimen (29BC dan 57BC) diambil pada kedalaman 2324 m dan 2286 m.  Analisis ukuran butir dilakukan dengan menggunakan instrumen laser diffraction particle size analyzer (Mastersizer 2000), dengan parameter statistik seperti nilai rata-rata, sortasi, kemencengan, dan kurtosis dihitung dengan perangkat lunak Gradistat versi 8.0. Hasil penelitian menunjukkan bahwa kedua inti sedimen didominasi oleh fraksi lanau kasar hingga lanau sedang dengan tekstur lumpur berpasir dan pola distribusi unimodal. Mekanisme transportasi sedimen berdasarkan diagram C-M didominasi oleh suspensi seragam pada lingkungan semi tertutup berenergi relatif rendah. Meskipun demikian, profil vertikal kedua lokasi merekam stabilitas hidrodinamika yang berbeda. Inti sedimen 29BC lebih dinamis dan fluktuatif (rata-rata 5,534-6,071 ϕ, sortasi 1,439-1,678 ϕ, kemencengan simetris hingga condong berbutir kasar) dengan tren peningkatan ukuran butir ke lapisan atas, merekam penguatan energi hidrodinamika secara temporal yang berkaitan dengan intensitas upwelling aktif selama fase El Niño. Sebaliknya, lokasi 57BC menunjukkan karakteristik vertikal yang homogen dan stabil (rata-rata 5,747-5,935 ϕ, sortasi 1,403-1,537 ϕ, kemencengan konsisten simetris), merefleksikan kondisi lingkungan pengendapan berenergi relatif rendah yang konstan di luar jangkauan fluktuasi arus kuat (di luar area upwelling), serta dipengaruhi suplai material terestrial heterogen karena posisi geografisnya yang lebih dekat dengan daratan.

Abstract

Ekosistem mangrove memberikan jasa ekosistem penting yang mendukung ketahanan lingkungan dan ekonomi masyarakat. Tumpahan minyak di Perairan Karawang tahun 2019 dan 2021 telah menyebabkan kerusakan ekosistem mangrove. Penelitian telah dilakukan di Pantai Sedari, Karawang pada bulan Maret 2023 yang bertujuan untuk mengevaluasi kondisi mangrove pasca tumpahan minyak. Penelitian dilakukan dengan metode random terstratifikasi di 3 lokasi. Pengambilan data meliputi analisis vegetasi, pertumbuhan dan tutupan mangrove, makrozoobenthos, dan pengukuran konsentrasi total petroleum hidrokarbon (TPH) di air laut dan sedimen. Hasil penelitian menunjukkan kondisi ekosistem mangrove terdiri dari 3 spesies mangrove yaitu Rhizophora mucronata, Rhizophora stylosa, Avicennia alba dan Avicennia officinalis. Kondisi ekologis mangrove menunjukkan kategori kerapatan sedang – rapat (1.433 – 1.766 ind/ha), kategori tutupan mangrove sedang – rapat (62,06 ± 5,15% - 78,38 ± 1,47%), Hasil analisis vegetasi menunjukkan Rhizophora memegang peran ekologis yang paling besar di stasiun 1, sedangkan di stasiun 2 dan 3 dipegang oleh Avicenia. Kelimpahan makrozoobenthos (22 – 75 ind/m2). Kondisi ekosistem mangrove berdasarkan residu minyak menunjukkan bahwa lokasi penelitian masih terdampak oleh minyak yang ditunjukkan dengan konsentrasi petroleum hidrokarbon (TPH) pada air laut dan sedimen dalam kategori tinggi (0,887 ± 0,04 - 1,067 ± 0,12   ppm) dan (0,44 ± 0,2 - 4,55 ± 3,85 ppm). Selain itu juga masih terdeteksinya sisa bercak hitam memudar pada akar dan batang mangrove. Oleh karena itu diperlukan upaya untuk membersihkan residu minyak yang masih ada di ekosistem mangrove ini serta tindakan pengawasan terhadap kegiatan-kegiatan yang potensial menyebabkan pencemaran minyak.
