Abstract

ICU defined as an intensive monitoring place and life support activities as well as definite therapy in life-threatens disease patients. In this Unit, patients generally receive treatment from various doctors that a patient can receive a variety of drugs from different doctors (polypharmacy). This unit also has higher frequency of drug demand than the other units in the hospital so the potential or actual drug interactions can occur. This study begins with a retrospective pilot study in ICU, concurrent studies in GICU (General Intensive Care Unit), data analysis and conclusions. Detection of drug interactions concurrently on 185 patients obtained 78 drug interactions that consists of 46 (58.97%) pharmacodynamic interactions and 27 (34.61%) pharmacokinetic interactions.

Abstract

Telah diuji efek kemopreventif dari ekstrak etanol temu kunci pada mencit betina yang diinduksi kanker dengan DMBA. Mencit dibagi menjadi 4 kelompok yaitu kelompok normal, kontrol, pembanding, dan kelompok temu kunci dengan dosis 140mg/kg bb. Pengamatan dilakukan setiap hari meliputi munculnya kanker, jenis dan jumlah kanker, serta jumlah kematian mencit. Pada akhir minggu ke-10, seluruh mencit dibedah untuk diamati jumlah dan jenis kanker yang muncul. Hasil menunjukkan bahwa tidak terjadi kematian kelompok temu kunci (0%), sedangkan pada kelompok kontrol dan kelompok pembanding jumlah kematian pada hewan uji mencapai 60%. Pada akhir minggu ke-10, tidak ditemukan kanker pada kelompok normal, ekstrak temu kunci dan obat pembanding, sementara pada kelompok kontrol ditemukan sebanyak 33 kanker. Dapat disimpulkan bahwa ekstrak etanol temu kunci memiliki aktivitas kemopreventif kanker yang ditandai oleh penurunan kejadian kanker, jumlah kanker, dan kematian.

Abstract

Tujuan penelitian ini adalah untuk mengembangkan dan mengevaluasi sediaanmikrosfer mukoadhesif dari ekstrak etanol Centella asiatica yang diketahui memiliki aktivitas antitukak lambung. Simplisia C. asiatica diekstraksi dengan metode refluks menggunakan etanol. Formulasi mikrosfer dilakukan dengan metode gelasi ionotropik menggunakan natrium alginat yang diinkorporasi dengan ekstrak etanol kental serta kitosan dalam larutan kalsium klorida. Variasi dilakukan terhadap konsentrasi natrium alginat, rasio bobot ekstrak dan natrium alginat, konsentrasi kitosan, dan konsentrasi kalsium klorida. Evaluasi meliputi ukuran dan distribusinya, karakteristik permukaan, efisiensi penjeratan, profil kapasitas mengembang, serta uji mukoadhesi in vitro. Formula optimum diperoleh dengan komposisi 3% natrium alginat, rasio ekstrak-natrium alginat 1:2, 0,5% kitosan, dan 0,5 M kalsium klorida. Mikrosfer yang dihasilkan memiliki distribusi ukuran terbanyak antara 630-710 ?m, efisiensi penjeratan 25,48 ± 1,88%, pertambahan bobot pada uji kapasitas mengembang 40,76 ± 1,51% (t=15 menit), dan kekuatan adhesi 78,67 ± 2,89%.

Abstract

Caffeine and nicotine are widely used addictive substances in the world. Several studies have confirmed that nicotine could increase the caffeine intake. Animal studies also indicated that nicotine could enhance caffeine dependence behavior especially with low doses of caffeine. However, study about the behavioral interaction of caffeine and nicotine with its effect on reinstatement is still limited. This experiment was conducted for studying the interaction between nicotine and caffeine in terms of their dependence behaviors. Conditioned place preference (CPP) paradigm was used for establishing the dependence model. Forced swim test (FST) was carried out to observe any effect of caffeine and its combination with nicotine on depressive signs. A lower dose of caffeine (5 mg/kg) induced preference behavior. However, high dose of caffeine (50 mg/kg) stimulated aversive behavior indicated by decreasing preference score. Nicotine injection had no significant effect on the lower dose of caffeine. However at the high dose of caffeine, 0.7 mg/kg of nicotine i.p reduced the aversive behavior and changed the extinction-relapse behavioral pattern resulted from 50 mg/kg of caffeine. Moreover, high dose of caffeine (50 mg/kg i.p) resulted in anxiety behavior and also hyperkinesia shown by lower immobility time in FST. Nicotine injection prior to the high dose of caffeine reduced the anxiety-hyperkinesia manifestation. The result from the current study suggests that nicotine could alter the expression of behavioral manifestation of caffeine, especially with the higher dose of caffeine.

Abstract

Tujuan dari penelitian ini adalah mempelajari interaksi fisika antara alginat dan metformin HCl dalam bentuk mikrosfer. Interaksi fisika diamati dengan menggunakan differential scanning calorimetry (DSC) dan scanning electron microscope (SEM). Mikrosfer dengan efisiensi penjeratan optimum ditentukan jenis interaksinya dengan DSC yang didukung dengan analisis morfologi mikrosfer yang dihasilkan oleh SEM. Termogram DSC menunjukkan interaksi fisika metformin HCl-matriks alginat yang ditunjukkan dengan perubahan profil kurva endotermik pada suhu 228-230°C dan 265-35°C. Hasil SEM mikrosfer menunjukan bahwa metformin berada dalam bentuk kristal sedangkan alginat hasil pautan silang dalam bentuk amorf. Dapat disimpulkan bahwa interaksi fisika yang terjadi adalah ikatan hidrogen yang kuat antara metformin HCl dengan alginat yang berefek pada meningkatnya efisiensi penjeratan.

Abstract

The research was carried out to isolate and characterize the methyl β-orsellinate of lichen Dirinaria sp. collected from the coastal areas of Teluk Nipah, Pulau Pangkor, Perak Malaysia. The sample was extracted in methanol for three times at room temperature. Subsequently, the crude extracts and further analysed using thin-layer chromatography (TLC) and vacuum liquid chromatography (VLC). To derive the pure compounds, the isolation step was performed using radial chromatography (RC). The chemical structure of the isolated compound was determined by several spectroscopies i.e. infra-red (IR), nuclear magnetic resonances (NMR), and mass spectrometric (MS). The compound was identified as methyl β-orsellinate.

Abstract

Ascorbic acid or vitamin C is one of the most widely consumed supplement and available at the market in several preparations such as tablet, sweetlet, effervescent tablet, and ready to drink product. Spectrofluorometric method has been developed in this study for quantitative determination of ascorbic acid in various forms of products based on the reaction between ascorbic acid and methylene blue. Methylene blue concentration decreased along with the addition of ascorbic acid due to a redox reaction which caused the formation of colorless leuco-methylene blue and dehydroascorbic acid. The fluorescence intensity of methylene blue was measured at excitation and emission wavelengths of 664 nm and 686 nm respectively. The proposed method was found to have a good selectivity, linearity, accuracy, and precision. Validation studies demonstrated a good linearity for the method with a correlation coefficient > 0.999 and Vx0 < 2%. The accuracy test also met the requirements with the recoveries were not less than 90.0% and not more than 110.0%. Precision test gave a relative standard deviation (% RSD) not more than 2%. The ascorbic acid content in several marketed products was evaluated by this new spectrofluorometric method and the result was in good agreement for effervescent tablet, isotonic drink and lozenges with the label claim and specification. Due to its simplicity, the method can be used to analyze vitamin C in different product forms.

Abstract

Withdrawal symptom in alcohol dependence is one the main problems in treating alcohol addiction. This study was aimed to clarify the role of the arachidonic acid cascade in the expression of withdrawal signs in animal model of alcohol dependence. Dependence was induced by repeated administration of 15% ethanol in 0.2% sucrose solution for 17 days. On the 18th day, the ethanol was not given, and one hour after the supposed time of ethanol administration, diclofenac at 10 mg/kg was injected i.p. to induce withdrawal. Observation of behavioral withdrawal signs was performed for 30 minutes, starting 25 minutes after diclofenac injection. Following behavioral observation, forced swimming test ensued to assess depression during withdrawal. In different group of mice, ibuprofen at 7.5 mg/kg was given every other day during ethanol regimen, 30 minutes prior to ethanol. Results showed that diclofenac induced the expression of withdrawal signs in mice receiving repeated alcohol. Incidences of facial preening (P<0.01), forepaw licking (P<0.01), grooming (P<0.01) and forepaw tremor (P<0.001) increased significantly by at least three times the normal value. In mice pretreated with ibuprofen significant decreases in body weight reduction (300%, P<0.01) and immobility time (50%, P<0.05) were observed compared to that of ethanol group. Taken together, the data indicates that suppressed arachidonic acid cascade signaling is involved in the expression of alcohol withdrawal.

Abstract

This study was conducted to know the effect of honey from Sumbawa on healthy and alloxan-induced diabetic mice. It consisted of honey quality test, glucose tolerance test (GTT) and alloxan-induced diabetes. On GTT, healthy animals were given pure honey, diluted honey 20% and 50%,  blood glucose levels were measured every 30 minutes for 3 hours. Diabetic mice obtained by inducing alloxan at a dose of 50 mg/kg bw. Blood glucose levels of diabetic mice in range of 400-500 mg/dL. Honey and combination of glibenclamide with honey were given for 21 days. Glibenclamide dose of 0.65 mg/kg bw was used as standard drug. Blood glucose levels were measured on days 10, 17 and 24. On the next day, the animals were sacrificed and pancreas were isolated. Pure honey, 20%, and 50% honey showed to raise blood glucose levels. Blood glucose level in mice group that given pure honey stayed in the normal value of 140 mg/dL until 180 minute observation, significantly different from the group of glucose 20% (p<0.05). Honey and combination of glibenclamide with honey in alloxan diabetic mice did not caused a decrease of blood glucose levels that significantly different compared to the sore group (p>0.05). It can be concluded honey maintains blood glucose levels of healthy mice on a normal value of 140 mg/dL until 180 minutes. In alloxan diabetic test, neither honey nor the combination of glibenclamide and honey did not show a decrease in blood glucose levels that significantly different to the sore group.
