Abstract

Study on the differentiation of several explants by in vitro culture were conducted on Philodendron goeldii for propagationpurposes.Activated charcoal added to the initiating medium did not improve the development of the cultures.In general all type of explants were successfully differentiated into plantlets in all media tested. The most potential explants for propagation of this species are the shoot tip and the stem.Addition of 0.3 ppm NAA to the basal medium with different cytokinins induced roots development on shoot tips,petioles and young leaves cultures.The ability of culture to produce multiple shoots such as in medium with NAA + 2-iP or NAA + BA would give a better opportunity for in-vitro propagation of P. goeldii.

Abstract

Studies on the therapeutic use of insects and insect products have been neglected compared to the use of other animals or plants.This study aims to determine the antibacterial and antioxidant potential of ants extracts. Preliminary study related to antibacterial and antioxidant screeningof 17 extracts of ant colonies that belongs to 8 species were performed by Thin Layer Chromatography (TLC)-Bioautography. Antioxidant activity was measured by DPPH free radical scavenging method. The antibacterial activity was done against three pathogenic bacteria, i.e Bacillus subtilis InaCC B-1, Staphylococcus aureus InaCC B-4 and Escherichia coli InaCC B-5 were performed by non-eluted TLC-autobiography assay. The minimum inhibitory concentration (MIC) and IC50 values of DPPH radical scavenging activity of active extracts were determined by microdilution in 96-well microplate. The results showed 6 extracts active against B.subtilis, 5 extracts active against S.aureus. The lowest MIC value was 512 µg/ ml.  Ten extracts had the antioxidant activity with various IC50 values. The extracts of ants might be used as bioactive resources for antibacterial and antioxidant.

Abstract

The antioxidant properties of methanolic extract of T. copelandii Elmer bark have been evaluated by using free radical (DPPH) scavenging assay, reduction power and ß-caroten bleaching assay. Ascorbic acid (vitamin C) and “butylated hydroxytoluene” (BHT) were used as positive control or antioxidant standard. Total phenolic content was estimated by using Folin-Ciocalteu reagent and expressed in gallic acid (Gallic Acid Equivalent), while total flavonoid content was estimated by using aluminium chloride reagent and expressed in routine (Routine Equivalent). The result shows antiradical activity of extract on concentration 40 µg/ml was 89.96%, lower than vitamin C (95.61%) but higher than BHT ((52.25%). The reducing power of extract on concentration 50 µg/ml has absorbance value 0.644 lower than vitamin C (0.769), but higher than BHT (0.475). During 2 hours incubation, extract on concentration 50 µg/ml inhibited ß-caroten bleaching 69.46% higher than control negative (35.81%), but lower than BHT (85.99%). Estimation of total phenolic and total flavonoid content showed that extract contained 350.40 µg (GAE)/g extract and 277.50 µg (RE)/g extract respectively. The antioxidant properties may be attributed to the presence of phenolic and flavonoid compounds present in the extract.

Abstract

The study is directed t investigate the antimicrobial activity f six ascorbyl fatty acids. The minimum concentrations were observed in ascorbyl caprate and ascorbyl laurate. Ascorbyl caprate was the strongest ester with the minimum inhibitory concentration (1,25-5 nM). Among microorganisms tested,S. cerevisiae was more senseitive than others. The sensitivity of S. cerevisiae was depending on the strains and S.cerevisiae OUT7054 was the most sensitive strain to the ascorbyl caprate which was the sterilizing agent rather than microbiostic agent.The antimicrobial activity of ascorbyl caprate was infuenced remarkably by temperature and pH.The most effective conditbns for sierilizatin of yeast were higher temperature and low pH.

Abstract

Phosphorous insufficiency is a limiting factor for rice production. P deficiency in acid soil of tropical regions causing fixation of Al. Because of it, the giving P fertilizers in acid soil become not efficient. To exploit tolerance varieties to P deficiency and Al toxicity are one of the solutions to decrease P fertilizers. The objective of this experiment was to screen twenty upland rice genotypes for P deficiency and Al toxicity, and double stress Al toxicity and P deficiency in culture solution. The experiment was arranged in a split plot design with three replications. The main plots were aluminium treatment (without and 45 ppm Al), the sub plots were P treatments (0 ppm P, 0,5 ppm P, 5 and 10 ppm P) and twenty genotypes of upland rice were sub-sub plot. The root length and root dry weight characters were used to identify genotypes tolerant to P deficiency and Al toxicity. The results indicated that based on the root length character, Sentani and K36-5-1-1-1 were tolerant to Al toxicity and double stress P deficiency and Al toxicity; while based on the root dry weight character, K36-5-1 -1-1 and NIL-C443 were tolerant to P deficiency, Al toxicity and double stress P deficiency.

Abstract

Entering globalization market, Indonesian government could not reject any import of food products from overseas. To anticipate the possibility of porcine contaminants into processed meat products of imported food such as meat or chicken ball, sausage, tin meat etc., it is important to apply laboratory research on such particular matter in regard to ethical and certain religious concern. This study was intended to identify the possibility of porcine contaminants into either processed meat products or fresh meat.A technique of polymerase chain reaction (PCR) was applied and PCR optimizing was conducted in advanced to obtain the right annealing temperature.Positive control of fresh pork meat was amplified to get porcine Leptin size which is 152bp fragment. Five samples of 4 meat balls and one fresh beef meat were individually collected for their DNA by either from minced or mashed after liquid nitrogen exposure then followed with a series of DNA extraction steps. PCR was assigned by using a specific primer of Leptin gene for porcine identification.Visualization of Leptin fragment was applied either on 1%, 2% of agarose gel or 10-20% gradient acrylamide gel.The result showed that all sample applied were not identified for containing porcine contaminants while positive control was on the right size of 152bp of Leptin gene. Specific primer used in this study was proved that there was not identified porcine Leptin gene on the negative control (fresh beef meat). This study suggests that a method of PCR is a simple analytical method for identification of porcine contaminants and visualization on 2% agarose gel is a cheaper and quicker method while by gradient acrylamide gel showing more clear band however this method is time consuming and expensive.

Abstract

Sejak semula Kebun Raya Bogor - LBN selalu aktif mendatangkan dan tnemperkenalkan jenis-jenis tanaman baru, baik dari luar negeri maupun dari hutan belantara Indonesia sendiri.Kegiatan ini telah mempengaruhi keanekaragaman jenis tanaman yang dipakai penduduk kota madya Bogor dalam menghias halaman dan pekarangan rumahnya,karena mereka mendapat kesempatan pertama untuk mencobakan hasil jerih payah Kebun Raya Bogor itu. Selain itu baik keadaan flsik maupun iklim kota Bogor (ketinggian 190 - 280 m dpi.,rata-rata suhu bulanan 24-26°C, kelembaban udara 76 - 86%,penyinaran matahari 62%,kecepatan angin 1.3 m/detik,curah hujan 4122 mm/ tahun) sangatlah ideal untuk pertumbuhan tanaman.Karenanya tidaklah mengherankan jika lebih dari 625 jenis tanaman telah,dipakai oleh warga kota Bogor sebagai tanaman hias (Widiawati & Rifai 1975).Ini menunjukkan suatu jumlah yang besar mengingat bahwa Bruggeman (1948) hanya mencatat lebih kurang 750 jenis tanaman hias untuk seluruh Indonesia.

Abstract

Two 1-ha (100m by 100m) plots were established at a peat swamp forest of Kelampangan, Central Kalimantan on May 2002. One plot was established at a forest that has been started to regenerate after forest fire December 1997 (PK) and another one at the unburnt natural forest (HAG). Both plots were separated by artificial canal, about 300m away to southwest (HAG) wards and to northeast wards (PK) of the canal. All trees with stem girth at 130cm height of more than 15cm (or about 4.8 cm in diameter) within the plots were enumerated and measured in May 2002 and re-measured again in May 2003. HAG plot was consisted of 3074 tree individuals grouped into 80 species (Fishers'a = 15.02) with total basal areas of 33.19 mTha, dominated by (top five in BA, from higher to lower): Callophyllum canum, Combretocarpus rotundatus, Campnosperma squamatum, Ctenolophon parvifolius and Cratoxylum glaucum. Species with higher number of individual were: C. canum (515 individuals), Cp. squamatum (355), Ct. parvifolius (350), Elaeocarpus petiolatus (183) and Cr. glaucum (125). PK plot consisted of 1158 individuals, 103 species (Fishers'a = 27.3) and total basal areas were 7.43 mVha, dominated by: Co. rotundatus, Cratoxylum arborescens, Palaquium gutta, Shorea teysmaniana and Syzygium ochneocarpum. Species with higher number of individuals were C. arborescens (256 individuals), S. teysmaniana (104), Sy.ochneocarpum (50), Horsjieldia crassifolia (47) and Cp. squamatum (46). Based on its tree diameter and growth rate, most of the trees (1102 individuals) within PK plot were grown after forest fire December 1997, while the rest 56 trees with higher stem diameter were escaped from forest fire December 1997, mostly belong to: C. canum, Co. rotundatus, Dyera lowii, P. gutta. Based on the species number and total basal areas, the recovery rate of peat swamp forest at PK plot after first forest fire December 1997 were categorized as very high. In September 2002, PK plot was burnt again while HAG remain unburnt, and in May 2003, only 2 individuals of Dyera lowii were found to produce new leaves after escaped from the fire, one laid stem of Co.rotundatus produced new shoot and one standing dead tree of Cr. arborescens produced sprouts from the base of the stem. Growth rate relative,mortality rate and recruitment rate of trees at natural un-burnt forest of the HAG plot were also discussed.

Abstract

Pichia pastoris is a group of methylotropic yeast known as a host of expression and protein production which is widely used for biopharmaceutical-based drug production. This yeast can grow fast with a high cell density. Its genetic stability, high cell density, and stress resistance make the development process and scale-up of P. pastoris can increase to a scale of 200,000 liters of culture. In contrast to the expensive and complex development of recombinant protein production in mammalian cells, the development of production in P. pastoris is relatively simple and cheaper. The advantage of P. pastoris as an expression system is that it is able to use methanol as a carbon source by inducing the expression of alcohol oxidase oxidase (AOX) enzyme. Promoter used by this enzyme is also used as a strong promoter for the expression of proteins that we want. Unlike in bacterial and mammalian systems, recombinant protein production in Pichia cells is not contaminated with endotoxins or viruses so it is safer and simplifies the downstream processes in bioproduction. The level of endogenous protein in the low supernatant allows Pichia to cultivate with a high volumetric productivity, therefore the process of protein production becomes very economical. This review provides an overview of several things that must be considered in utilizing P. pastoris as an expression system including the selection of vectors, strains, vector integration mechanisms into the genome, glycosylation processes, and applications in industry.

Abstract

Pathogenic fungus is one of the constraints to increase crop production. Chemical control using fungicides caused negative effects either to the environment or increased pathogen resistance to fungicide. Biological control using microbial-producing ß glucanase is an alternative method to inhibit the growth of pathogenic fungus. The aim of this study was to characterize ß-1,3-1,4-glucanase produced by rice endophytic bacterium, B. cepacia E76. Purification was carried out by ammonium sulphate precipitation, dialysis, and ion exchange chromatography using DEAE sepharose Fast Flow. A further characteristic of the enzyme activity was studied using oatmeal-glucan substrate.Results showed that precipitation using saturated 80% ammonium sulphate generated a good yield with the purity increased by 11 fold and yield of 66%.After chromatography step, the ß-1,3-1,4-glucanase of B. cepacia was successfully purified with an increasedof purity up to 33 fold and yield of 4%. Based on 10% SDS-PAGE, the enzyme profiles had the molecular weight of 15, 48 and 55 kDa.Of the three isozymes, only the 48 kDa isozyme showed the strongest glucanase activity when grown on media containing glucan as substrate.
