Abstract

The effects of plant growth regulators on somatic embryo genesis in Lilium longiflorum Thunb.leaf cultures were studied. The aim of the research is to study the regeneration ofi. longiflorum via somatic embryogenesis of leaf explant.IAAat5 level of concentrations (0-1 mg/1) combined with BAP at 4 level of concentrations (0-2 mg/1) were tested to induction of somatic embryogenesis in the first stage. Combination of two cytokinins, namely BAP and kinetin, combined with either of four auxins, namely IAA, IBA, NAA or 2,4D were tested to induce the somatic embryo genesis in the second stage. Combination of two concentrations of ABA, namely 0 and 1 mg/1 combined with 5 level concentrations of GA3, namely 0, 0.25, 0.5, 0.75 and 1 mg/1 were tested to improve the somatic embryos maturation. The results showed that somatic embryos were promoted by addition of IAA and BAP. The study of auxin and cytokinin combinations on somatic embryogenesis showed that somatic embryogenesis of kerk lily induced by addition of auxin and cytokinin in the culture medium. The maximum somatic embryo induction was obtained on medium contained of 0.25 mg/1 IAA combined with 1 mg/1 kinetin. Somatic embryos maturation were improved by transferring the somatic embryo from induction medium to maturation medium that contained 0.5 mg/I GA3 +1 mg/1 ABA.

Abstract

A research to study the effect of intercropping on rehabilitated of physic nut (Jatropha curcas L.) was conducted in Muktiharjo Research Station, Pati, Central of Java, from January to December 2010. Plant materials were physic nut rehabilitated by grafting of main branch with IP-2A (1 year old) and intercrops (peanut, soybean, mungbean and sesame). The experiment arranged in randomized block design with 6 replications. The treatments were: 1) intercropping peanut + physic nut, 2) intercropping soybean + physic nut, 3) intercropping mungbean + physic nut, 4) intercropping sesame + physic nut, and 5) physic nut monoculture. Plot size was 8 m x 8 m, plant distance of physic nut 2 m x 2 m, and plant distance for peanut, soybean and mungbean were 25 cm x 25 cm and sesame was 50 cm x 25 cm. The result showed that jatropha seed yield on intercropping physic nut with soybean (980.05 kg/ha) and mungbean (974.22 kg/ha) were not significantly different from physic nut monoculture (1,094.84 kg/ha). Crop that appropriate for intercropped with rehabilitated physic nut (second year) were soybean and mungbean.

Abstract

The Bonokeling society is a part of the Javanese indigenous groups who still maintain and practise the local wisdom in their daily lives. Socio-cultural research concerning the Bonokeling religion has been reported and provided useful information to the society, while the ethnoecological studies about Bonokeling’s local knowledge related to the environment have not been reported. The research aimed to provide a comprehensive study about Bonokeling society on the management and utilization of natural resource and environment. This study used qualitative and quantitative approaches through participatory survey, open-ended interviews, and structured interviews with questionnaires. The results showed that the community recognized five environmental categorizations based on functions, namely settlements, home gardens, agricultural land, sacred sites, and irrigation sources. The plant diversity found were consisted of 288 species with 12 categories of uses. Pari is a species of cultivated plant that has the highest cultural importance value. The Bonokeling local knowledge can be scientifically proven has an ecological value. The management and utilization of agricultural land by the Bonokeling society have been carried out not only to optimize the production, but also provide the benefits to the environment. Besides, the management of sacred site has shown that traditional conservation practices have been maintained well for sustainability.

Abstract

DNA isolation is an important issue in molecular fields especially for specific plant group such as Pinanga (Palmae). Typically, leaflets structure of Pinanga consists of shiny leaflets as common species of palms and mottled leaflets. Pinanga javana and P.coronata form commonly the two types of leaflets. The high quality of DNA of Pinanga with the shiny leaflet is readily purified and that with the mottled leaflet needed DNA purification due to the presence of high polyphenolic compound. Application of the silica binding method was recommended to purify the template DNA. Modified DNA-isolation protocol is relatively quick, simple, least expensive, minimum equipments and chemicals required, and suitable for PCR and endonucleases digestion reaction.
