Abstract

The study was aimed to analyze the immunogenicity of GRA1 protein derived from clone bearing GRA1 genes from local isolate of Toxoplasma gondii. Analysis of GRA1 protein translated from cDNA of GRA1 is very essential in prior to expressed the gene. Analysis of GRA1 protein derived from clone bearing GRA1 genes was performed using several bioinformatics software which are available as standalone or online software such as CLC Bio Workbench series, BioEdit, BESTORF, GENSCAN, FGENES, BepiPred 1.0, CTL Epitope Finder and SignalP. Translation coding sequences of GRA1 gene into GRA1 peptide sequences revealed 190 amino acids with molecular mass of GRA1 approximately 20.159 kD and isoelectric point at 4.43. GRA1 protein also identified several antigenic domains with six domains were known as epitopes for CD8+/cytotoxic lymphocyte and seven domain as epitopes for B lymphocyte. However, GRA1 protein was considered as good antigen but less immunogenic.

Abstract

Wetlands in Peatland Development Area has potential for the development of paddy, vegetables and palawija. Development of the commodities are limited or restricted by many limiting production factors such as nutrients, water and plant varieties. The purpose of this study is to know the influence of amelioration and fertilization packages on the growth and yield of peanuts in the swampland peat land development area. The treatments consisted of four amelioration and fertilization package i.e.: (1) Farmer’s package I: 250 kg.ha dolomite, (2) -1 -1 -1 -1 Farmer’s package II: 2.5 t.ha straw compost+18 kg.ha P2O5+300 kg.ha fly ash, (3) Recommendation package: 1.0 t.ha dolomite+22.5 -1 -1 -1 -1 -1 -1 kg.ha N + 36 kg.ha P2O5, and (4) Research package: 2.0 t.ha dolomite+22.5 kg.ha N+22.5 kg.ha P2O5 +30 kg.ha K2O. The result showed that the growth of plants (plant height and number of branches) does not differ between treatments, and the obtained peanut pod yield improvement is 50.45% in the research package treatment compared to Farmer’s package I.

Abstract

Gamma ray mutative induction for increasing genetic variation has been applied for plant prime variety engineering. The materials are derrived from seed organ, shoot and calli. Calli is a group of actively dividing cell and have not been organized to form plant. The benefit of using calli explant is that the gamma ray could directly shot to DNA in the nuclear cell in such a way that there is higher opportunity for genetic change to occur. The problema of using calli explant are the difficulties in regenerating the calli into shoots, due to the deformation as a result of radiation process. Therefore, this research is aimed at obtaining the appropriate media formulation for shoot regeneration from Fatmawati-rice calli which has been irradiated with gamma ray. The reseach was conducted in BB-Biogen laboratory consisting of three experiments, such as : (1) calli iradiation with the dosage of 0; 5; 10; 15; 20; 25; 30; 35; 40; 45; 50; 55 and 60 Gy, (2) shoot regeneration at the MS + BA (0, 1, dan 3 mg/l) + IAA (0 dan 0,8 mg/l) media, and BA (0, 1, and 3 mg/l) + zeatin ( 0; 0,1; 0,2 and 0,3 mg/l) + IAA 0,8 mg/l and (3) Shoot induction at MS + IBA (0, 1, 2 and 3 mg/l) media. The result shows that the range of LD50 was obtained at the dosage of 30 Gy, the most apropriate media for shoot regeneration is MS + BA 3 mg/l + IAA 0,8 + zeatin 0,1 mg/l and media for root induction is IBA 1 mg/l.

Abstract

An experiment to study the effect of ellicitor derived from Phytium aphanidermatum On ajmalicine content of Catharanthus roseus root- callus culture has been conducted. Root-callus was induced  from left segment, and grew optimally on Zenk medium with the addition of  10-7 M BAP and 10-5 M NAA. Root-callus was subcultured three times, and followed with elicitation with elicitor derived from autoclaved  P. aphanidermatum. The concentration tested were 0.50, 0.5, 1.0 and 5.0 mg DW/ml, and harcested at 0,18,36 and 72 hours. The ajmalicine was analyzed qualitatively and quantitatively by using High Performance Liquid Chromatography (HPLC). The result showed that ajmalicine content was influenced significantly by concentration of ellicitor and harvesting time. A significant increase of ajamalicine content (153.29 ± 1.61 mg/g DW) was achieved by additon of 1 mg Dw/ml ellicitor after 36 hours incubation.

Abstract

The study of genomic diversity in molecular level on two sub-species i.e indica andjavanica rice types were represented by cv.Pelita 1/1 and Rojolele respectively.Javanica called padi bulu due to long aunt on the grain. Polymerase Chain Reaction (PCR)with 17 primers have been used for detecting genomic variation between these two sub-species.Results of experiment indicated that 12 out of 17 primers showed polymorphic DNA.This study would assist plant breeder to decide,whether to improve rice quality through crossing between javanica and indica is necessary.

Abstract

Menurut International Union for Conservation of Nature and Natural Resources (IUCN )Species Asing Invasif merupakan jenis-jenis tumbuhan asing yang berkembang dan menyebar di luar habitat aslinya, sehingga mengancam ekosistem, habitat, atau jenis yang lain. Species Asing Invasif dapat berasal dari habitat yang berada di luar pulau, kepulauan, benua ataupun negara aslinya. Lebih lanjut The Invasive Sepscies Advisory Committee (ISAC) menegaskan bahwa gangguan terhadap suatu ekosistem akibat masuknya Species Asing Invasif dapat menimbulkan kerugian-kerugian ekonomi, kerusakan lingkungan dan juga gangguan kesehatan manusia.

Abstract

Development of high-yielding varieties depends on choice of parents, include per se performance, morphological and agronomic traits, and genetic diversity as determined through geographic origin. Indicators of success can be expected from the value of genetic progress and some other important genetic parameters. The purpose of this study was to calculate the genetic heterogenity, heritability and genetic advances of some quantitative characters of rice genotypes. Experiments were conducted in the district of Kulon Progo, namely: Wates and Panjatan (irrigated condition, dry season of 2012), in Giripeni (rainfed, medium altitude, wet season of 2012/2013), Samigaluh and Kalibawang (rainfed, high altitude, wet season of 2012/2013). Each experiment was designed using a randomized complete block with three replications.Data indicated that not all environmental conditions appropriate for the selection and development of genotype due to low value heritability and expectation genetic advances of each environment was lower than the value heritability and expectation genetic advances for the combined average of the environment. Wates was suitable location for grain yield selection and development in terms of the high heritability values. Kalibawang, Giripeni and Wates have considerable heritabilty value for 1000 grain weight character, hence they can be used as suitable selection locations for these characters.

Abstract

Antipyretic effect of alcohol extract ofilalang flmperata cylindrica L) root on white rat males had been studied using fever tolerance method.Experimental animals were induced to fever by injecting 0.9 ml/kg BW vaccine kotipa orally.Alcohol extract 5, 10, 30, 50,100 and 150 mg/kg BW was given one hour after injection.The body temperature was measured before injection and every half an hour until the fifth hour after injection.Paracetamol 200 mg/kg BW was used as control.Data were analyzed using ANOVA with the body temperature (T), the time interval (P) and extract doses being given (C) as parameters.The results showed that body temperature increased TC from the basal temperature at the third hour after injection of vaccine.The body temperature did not change significantly at the lower extract doses(5-30 mg/kg BW)(P>0.05),while higher doses (30-150mg/kg BW) linearly lowered the body temperature significantly (P<0.05).The extract dose at 50 mg/kg BW lowered the body temperature similar to paracetamol 200 mg/kg BW.

Abstract

Nitrile and amide bioconversion have received increasing attention due to their ability to provide a range of commercially important chemicals. The experiment was conducted to investigate the potential of bacterial isolate GLB5 to convert methyl cyanide and phenyl cyanide. The samples were collected from various industrial waste. Selection of isolates to utilize  these substrates as a sole source of energy, carbon and nitrogen was conducted on 96 whell microtitter plates, based on the growth ability using INT (Iodo nitrotetrazolium chloride) reagent. Based on the growth  pattern, it showed that the bacterial isolate GLB5 grew well and it was capable of utilizing  methyl and phenyl cyanide compound as the sole source of carbon and nitrogen.  The isolate GLB5 was isolated from industrial waste of Batik factory in Cirebon, and  identified as Rhodococcus pyridinovorans. Bioconversion of methyl cyanide using whole cells of R. pyridinovorans GLB5 showed that ethanamide (C2H5NO) and ethanoic acid (C2H4O2) were detected. Formation of ethanamide and ethanoic acid as the product of bioconversion, indicated that the nitrile hydratase and amidase enzymes  involved in the bioconversion process. Phenyl carboxamide (C7H7NO) as the product of phenyl cyanide bioconversion was also detected,  although  in  low  concentration. In this study, R. pyridinovorans GLB5 was capable of completely converting 300 mM methyl cyanide to ±  140 mM ethanoic acid in relatively short times (<60 minutes).

Abstract

An experiment to study the effect of soft fraction derived from Saccharomyces cerevisiae (Hansen) to elicit ajmalicine production by Catharanthus roseus (L.) G. Don. callus cultures has been conducted. Callus were induced from leaf segment and grew on Zenk medium supplemented with 2,5 x l0M NAA dan 10 M BAP respectively. Callus on the third subculture level were elicited with soft fraction derived from S. cerevisiae. The following concentrations of elicitor tested were 0, 0,5, 1,0 and 2,5 % (g/v), and the harvesting times were 0, 18, 36 and 72 hour respectively. Influence of elicitor to production of ajmalicine by the callus were analyzed qualitatively and quantitatively by using high performance liquid chromatography (HPLC). A significant increasing of ajmalicine content in the callus (303,475 ± 5,602 ug/gDW) was achieved by addition of elicitor of 0,5 % (g/v) after 36 hour. This study showed a significant increase of ajmalicine production in C. roseus callus cultures after being challenged with elicitor derived from S. cerevisiae i.e. 69,334%.
