Abstract

Research on the effect of weight and composition of media, temperature and chemically stimulation on fruiting body formation of shiitake (Lentinus edodes) had been done in Wamena, Jayawijaya district, Irian Jay a. The research consisted of three experiments. The first experiment consisted of 3 kinds of media such as media A, B and C(A- sawdust and 100% standard nutrition; B = sawdust and 50% standard nutrition; and C = sawdust without nutrition) and three kinds of temperatures were 20°C (room temperature),4°C (refrigerator temperature) and -10°C(freezer temperature).The second experiment used 4 kinds of media. They were media D, E, F and G which were stimulated by SnCh, CoCh, salicilic acid and the extract of bark of "Seno" (Castanopsis acuminatissima).The third experiment used medium B, weight 500 g and 1000 g (medium H and I).The result showed that medium H& I produced the highest fruiting body. However medium I produced the higher fruiting body compare to medium H).The added of the extract of the bark of C. acuminatissima.SnCh, CoCh and salicilic acid stimulated the fruiting body formation.

Abstract

Research on the diversity of snails in Enggano, Central Bengkulu was held on 16 April 2015 to 5 May 2015 by taking three sampling sites that are north-western part of Enggano (Kampung Bendung at Desa Banjarsari and Desa Meok), the centralpart of Enggano (Desa  Malakoni) and the south-easternpart of Enggano (Desa Kaana). Sampling was conducted using purposive sampling while the analysis using graphical method and statistical cluster and t-test. This study found 722 specimens of snails which consists of 24 species from 17 families. Most of the snails found are in small size. Land snails in the forest dominates the north-easternpart as much as 88.2%. compared to  freshwater snails (11.8%). Terrestrial and freshwater snails share amount 50% each in the forest of the central, while terrestrial snails still dominate in the forest of south-eastern part (land snails: snail freshwater = 71.4%: 28.6% ). The existence of land snails and freshwater can not be separated from the state forests themselves besides the condition of the soil and surface water as a result of the topography of the island

Abstract

A study has been conducted for survival testing of 7 and 7-16 years-preserved fungi in water and liquid-paraffin respectively.Survival testing was done through replanting the preserved fungi in new PDA slant media.The growth observation of fungi was carried out for two weeks long.Each collection number was tested with two replications. Thirteen collection numbers of fungi (Alternaria spp.,Aspergillus spp.,Fusarium spp.,Penicillium spp.,and Trichoderma spp.,)which were preserved for 7 and 8 years long in water and liquid-paraffin respectively were growing again, and the rate of survival were 100%. The survival of the fungi(species and sum of sample were different) which were preserved in liquid-paraffin for 7, 8, 9 and 16 years long were 38,5, 81,5,53,6, and 64,7% respectively.

Abstract

Bonti-bonti (Paratherina striata) is an endemiz and vulnerable fish in Lake Towuti and L. Mahalona. The fish population tend to decreasing due to intensive fishing and habitat quality decline. This study was aimed re reveal the age pyramids and the grouping of fish populations as a basis for describing condition and diversty of fish populations. The study was conducted in L.Towuti at five stations. Samples were collected using experimental gillnet with mesh sized ], ¾, 1, dan 1¼ inches. Meansurement of morphometric characters of  fish consist of 22 characters. Results shows that fish population at each station features a different age pyramid. at station inlent of L. Towuti and Loeha Island, the age pyramid type which is dominated by juveniles a reletively rapid population growth. At station Cape Bakara, outlet of lake Towuti and Beau, the age pyramid type tend to be mederate. This shows the fish population is dominated by medium-size, fish populations in stable condition. Obtained 12 characters that have a strong correlation values. ANCOVA test results showed no difference in morphometric characters between the stations in male and female. Based on ANCOVA test assumed that the Bonti-Bonti at fish station is a population group.

Abstract

The high cost of fish feed is a main problem in aquaculture industries, and the effort to improve the cost efficiency of feed is the main focus for nutritional research program all over the world. Research is needed to identify less expensive and more sustainable ingredients within fish feeds, while maintaining nutritional quality equal to those based mainly on fish meal and soybean meal. Leucaena leaf meal (LLM) is a potential alternative plant protein source because of its high protein content and low cost compared to soybean meal. The Leucaena leaf was soaked in freshwater for 3 days prior to use. This study was a 60-day trial and aimed to examine the replacement of soybean meal with LLM at various level in practical feed for Nile tilapia. This trial used a Completely Randomized Design which was carried out in 15 aquarias sized of 60 x 60 x 50 cm. Triplicate group of nile tilapia (initial body weight 12,04 ± 0,38 g fish-1 ) were fed five isonitrogenous and isocaloric feeds. The control feed was containing soybean meal, whereas in other four feeds, LLM directly replaced soybean meal with the inclusion level of 8, 16, 24 dan 31 % within the feed formulation. Result showed that the growth performance and feed efficiencies of feed containing 8, 16, 24 and 31 % LLM on the formulation were significantly lower compared to control feed. LLM was able to consumed by nile tilapia although there was a tendency of decrease of the feed consumption with the increase of LLM inclusion. It suggested that the additional treatment is needed to improve the quality of LLM, and to increase the inclusion level of LLM in feed.

Abstract

The  aim of study were to determine the chemical compounds and peroxide values (POV) of methanol extract of three plants species belongs to Moraceae family (Artocarpus (Artocarpus sp., A.elasticus, dan Ficus sp.). Their chemical compounds were identified by Cuiley method, while peroxide value was done by iodometri titration. Methanol extract percentage of Artocarpus sp., A.elasticus, dan Ficus sp. were 20.80%, 3.10% and 4.10% respectively. All of the extract tested contained essential oil, sterol and triterpenoid, aglicon flavonoid, tannin, flavonoid glicoside and saponin. POV of Artocarpus sp., A.elasticus, dan Ficus sp. were 325.24, 306.25 and 1580.07 respectively and POV of q tocopherol was 363.96. So, it can be concluded taht all of three methanol extract act as reductor, because their POV less that POV of q tocopherol and might be act antioxidant. These correalted with their chemical compounds such as flavonoid, tannin and peroxide sugar.

Abstract

A study on the influences of alcohol extract of Meniran or Phyllanthus niruri L. (Euphorbiaceae) to blood glucose concentration in rabbits (Oxytolagus cuniculus) had been carried out using glucose tolerance method.The extract was administered orally at 10, 20, 30, 40 and 50 mg/kg BW.Prior to tfie extract administration, all rabbits were put on hyperglycemic condition by injecting glucose solution intravenally at dose of 0.5 g/kg BW.The blood glucose concentration was measured every one hour starting at 2 hours before and until 7 hours after administration. Antidiabetic drug,tolbutamid 250 mg/kgBW.was used as control.The results indicated that extract with dose at 30 mg/kg BW gave a hypoglycemic effect similar to control (Tolbutamid). The blood glucose decreased regularly as the extract dose being higher.Using statistical analyses showed significant differences among the groups, especially after the 2 and 3 hours of treatment (P<0.05).

Abstract

Pisang raja bulu is one of the most important bananas in Indonesia. However, this plant has low toleranee to wilt disease, caused by Fusarium oxysporum f. eubense. Its mass cultivation is inhibited by the absence of variety tolerant to the disease.A wide range of genetic variability will be needed if selection for novel characters is to be conducted, especially when there is no source of resistance gene available for breeding materials. This research consisted of callus induction from primary explant, induction of somaclonal variation using gamma iradiation, and in vitro selection using fusaric acid, followed by regeneration and acclimatization of selected plantlets. The media applied for callus induction was MS (Murashige and Skoog. 1962) + 2,4-D I and 3 mg/l + NAA 0 and 0.1 mg/l and 2,4-D 5 mg/l + BA 0.5 mg/l + Casein hidrolysate (CH) 500 mg/l. The applied gamma irradiation dosage were 0, 5.0. 7.5. 10 and 15 Gy. The irradiated calli was subsequently subcultures on selection media i.e.. MS containing fusaric acid at 30 and 45 mg/l. The living calli was then regenerated on media containing BA, TDZ. with or without proline and arginine. In addition. MS+ kinetin 5 mg/l + 1AA 0,2 mg/l was applied for shoot development. The result showed that the most suitable callus induction media for pisang raja bulu was MS +2.4-D 5 mg/l +BA 0.5 mg/l +CH 500 mg/l. The gamma irradiation of 10 Gy produced somaclone lines which were able to proliferate bud nodules on selection media containing fusaric acid at 30 and 45 mg/l. The media used for shoot development was MS + kinetin 5 mg/l + IAA 0,2 mg/l. Planllet obtained from the in vitro were then successfully acclimatized in the green house.

Abstract

Acanthaceae published in Flora of Java has had significant change especially for nomenclature, distribution and potential uses; thus it affected species names of collections of Acanthaceae in Herbarium Bogoriense of The Indonesian Institute of Sciences. It is therefore necessary to update information of Acanthaceae by conducting herbarium and field surveys as well as reviewing related publications. Validation of the species name, synonymous and distribution were carried out by using some online websites such as IPNI, Tropicos and The plantlist. Meanwhile, the potential uses were obtained from several publications. Examinations on herbarium specimens was conducted in the Herbarium Bogoriense, while field survey was carried out in several locations in Java. This study recorded a total of 164 species of Achantaceeae to which 53 were native in Java (Indonesia), 24 species become synonymous, and five other species have not been published. Some of them have potential chemical substances and uses.

Abstract

Study on the productivity of two species of local crickets; Gryllus testaceus Walk and Gryllus mitratus Burn which are generally found in the local market have been conducted with the main aim at obtaining information on the capability of those species in producing eggs including the length of production cycles.Three levels of box square density (50 female/box,100 female/box and 150 female/box) and two levels of additional feed (green master and papay leaves) were used as treatments.The results showed that the number of eggs produced by G.mitratus were larger with longer production cycles compared to those of G.testaceus. Species and density has highly significant effects on egg production and feed consumption, whereas,additional feed significantly influencing egg production and feed consumption of the two species.The use of green master as additional feed resulted in a higher egg production than those given papay leaves.
