Abstract

Degree of electrolyte leakage (EL) from leaf tissue after exposure to high temperature has been used as an indicator of heat tolerance. Inthe present study, EL was measured in an attempt to estimate heat tolerance of Viana umbellata (Thumb.) genotypes at three differentgrowth phases. The degree of heat injury is significantly different among the three growth phases and among the genotypes as well as their interaction. However, the degree of heat injury tends to increase over the plant growth. The 18 genotypes of V. umbellata shows moderate genetic variability as shown by its value of genetic variance (46,25 ±16,35), phenotypic variance (85,37 ±27,60), and coefficientof genetic variation (26,36%).

Abstract

The study was designed to explore soil biomass content and soil enzymatic activities that involved in carbon organic soil mineralization. Samples of soil were collected from two locations in Bangka Island. Bulk samples of top soil (TP) and excavated (TG)  were collected from garden soil in Pangkalpinang. Other soils were gathered from forest floor (TU), beneath pelawan (Tristaniopsis merguensis Griff.) trees which is endemic plant to North Bangka forest. Soil biological character was evaluated by measuring soil microbial population, respiration rate, and cellulase and amylase activities. Experiments were carried out in Research Center for Biology, Indonesian Institute of Sciences, on dried soil treatment (Control), soil moistened with 5 mL H2O in 100g soil (Gluc.0), and soil moistened with 5 mL 1% glucose solution (Gluc.1). Bacterial population obtained was higher than fungal population in all soil samples. Soil respiration activity in TU after treatment was 4.51±0.49; 5.26±0,79; and 8.28±1.41 ppm CO2/100g-soil/5-minutes, respectively. Meanwhile , respiration in TP  were 3.65 ± 2.12; 3.57 ± 1.18; and 7.94 ± 1.05 ppm CO2/100g-soil/5-minutes; and in the TG are 2.61 ± 0.70; 3.34 ± 0.94; and 5.46 ± 2.30 ppm CO2/100gsoil/5-minutes. Cellulase activity of all samples were not significantly different. Forest soil cellulase activity was significantly different compared to garden soil. Glucose induction was positively increased amylase activity compared to cellulase. Soil enzyme, soil respiration activities and microbial population of forest soil could be used as a reference to recover degraded land in Bangka Island.

Abstract

Lovastatin is a bioactive material of statin groups and has been used to reduce cholesterol through inhibiting HMG Co-A reductase enzyme activities. Three indigenous strains of Monascus purpureus and three mutans were used in this study produced lovastatin at the range of 0,1 - 1,42%. The objectives of this study were to increase lovastatin productions by co-cultured with several concentrations of Endomycopsis burtonii. M. purpureus (10 cfu/ml) was co-cultured with various concentrations of E. burtonii (10M0 ) cfu/ral at three different feeding times (day 2, 4, and 6). Feeding times and concentrations of E. burtonii significantly increased production of lovastatin by four strains of M. purpureus (JMBA5K, AID, JMBA and TOS). The highest production of lovastatin was achieved by M. purpureus TOS co-cultured by E. burtonii at 10 cfu/ml added at day 6. Expression of the genes that responsible for lovastatin production were analyzed by PCR and RT-PCR method. The phenotypic character of M. purpureus TOS with high lovastatin production was conformed by the high intensity of its gene expression.

Abstract

Mount Cakrabuana in West Java possesses high plant diversity. Administratively, it is belong to Sumedang, Tasikmalaya, Garut,Majalengka, and Ciamis Regencies, The largest area is Sumedang (1.284,62 ha). There are 3 species of rattans found in the mountain Plectocomia elongate Mart, ex Bl., Calamus heteroideus Bl., and C. javensis Bl. Description, habitat, and the usage of the rattans is discussed.

Abstract

Aquilaria malaccensis Lam. is a plant species producing fragrant woody material that contains some resin. The compounds can be used as medicine and perfume. Sesquiterpenoid, one group of compounds has been found being synthesized and subsequently extracted from callus and cell suspension culture of Aquilaria species. The aim of this research was to find a method of producing friable calli and cell suspension cultures from leaves or internodes of A. malaccensis in vitro by using suitable plant growth regulators; cell suspension that will suitably serve as material to produce sesquiterpenoid afterwards. Calli were established in almost all treatments of auxin-cytokinin on both leaves and internod explants. The treatment of 10 mg/L IBA induced the highest percentage of callus coverage from leaves with a rather compact structure. The combined treatment of 1–2 mg/L 2.4-D and 0.2–0.3 mg/L BA induced friable callus formation in more than 80% of cultures with 27–32% callus coverage percentage.  The use of 2,4-D induced a better formation of cell suspension than Picloram, with maximum volume up to 7 mL. Cell suspension culture with fine and homogenous aggregate could be established in the medium supplemented with 0.5 –1 mg/L 2,4-D.

Abstract

Krisis ekonomi yang terjadi di Indonesia terutama bersumber dari dalam negeri kita sendiri.Pemerintah orde baru, terutama sejak era konglomerasi (1987-1994), terlalu mengabaikan berkembangnya kesenjangan.Pertumbuhan ekonomi ternyata hanya sebuah fatamorgana (Mubyarto,2000). Krisis ekonomi 1997-1998 dan keadaan politik yang tidak menentu, mengakibatkan nilai mata uang rupiah dalam tempo yang amat singkat, menurun drastis. Indonesia pun langsung terpuruk. Kegiatan ekonomi tersendat karena sebagian besarnya sangat tergantung kepada bahan baku impor. Banyak perusahaan yang gulung tikar dan banyak buruh terpaksa di PHK. Jumlah penganggur dan jumlah penduduk miskin meningkat dengan tajam.

Abstract

Galunggung Mountain forest area in West Java of post eruption 1982 may served as a natural laboratory for studying plant development and plant growth, including pine trees. Pine trees that were planted by Perhutani at Galunggung had high prospect although they were planted at degraded areas and poor nutrient. Experimental plots were set up in two different locations at Mount Galunggung. Trunk diameter, tree height, total height, soil humidity and soil pH was measured for all pine trees inside the plot at each subplot. All measured parameters resulted in varied values with varied deviation standard. Measurement of trunk diameter, branches height and total height of pine trees in Mount Galunggung suggested that their growth rate was slow and no significant correlation among them.

Abstract

Fermentation of rice by Monascus purpureus could produce red yeast rice (angkak) that can be used as drugs to decrease cholesterol and triglycerides increase thrombocyt in patients of dengue fever. To increase the immune system is needed the compounds that can be produced by microorganisms. The aim of study is to know that several strains of Monascus purpureus could produce the immunomodulatory effects or not. The study is about the invitro of phagocytosic activities of macrophage cell and polymorphonuclear of mice. This study had been used the fermented rice extract of five concentration such as 1 mg/ml, 0.1 mg/ml, 0.01 mg/ml, 0.001 mg/ml and 0.0001 mg/ml. As positive control was used 5 mg/ml extracts of meniran herbal, suspension of Staphylococcus aureus and the liquid of mice peritoneum. The results showed that the extract of fermented rice of Monascus purpureus with 1 mg/ml concentration was the same effectiveness as positive control. The conclusion was the extract of fermented rice could increase the fagocyticyt activities of can be concluded that administration of extract some kind of Monascus purpureus to increase the phagocytosis activities of macrophages and polymorphonuclear cells of mice.

Abstract

This research aims to see the land use change within ethnobotanical perspective. According to observation, interview, and Participatory Rural Appraisal (PRA) it was known that land in Enggano Island have been changed, even the sacred forest transformed, into kebun/garden and ladang/field. The changes of land use was related with demography, economic, and socio-cultural changes in society. Social change affected the species that were planted by the community. The species with a high economic value and a quick harvest and did not need extra treatment planted by local people to replace the previous plants. The other impact that arises is the exixtence of local initiative to restore areas that should functioned as a forest.

Abstract

Zingiberaceae species Zingiber zerumbet var. aromaticum Val. usually propagated by vegetative part of the plant. Study on the proliferation of rhizome explants through in-vitro method were conducted on this species for propagation.The explants were cultured in MS (Murashige & Skoog) composition medium in normal and half strength concentration of macro elements, supplemented with micro elements and vitamins. Plant growth regulator (PGR) cytokinin (kinetin, adenine sulphate) were added in initiation stage.Benzyl adenine (BA), thidiazuron and 2,4D (dichlorophenoxy acetic acid) added in medium as treatments to induce shoot multiplication and adventitious shoot development or rooting plants.The media for induced multiple shoot and roots (plantlets) development, supplemented with PGR BA, thidiazuron, Indole butyric acid (D3A) as treatments and activated charcoal were added as antioxidant.In general, most of cultures were indicated have positively responds to PGR treatments. The ability of culture to produce multiple shoots and roots such as in medium with BAP (8 mg/1) + thidiazuron (0.1 mg/1) + IBA (1 mg/l)-indole butyric acid would give a better opportunity of rhizome explants proliferation and plantlets development of Z. zerumbet var. aromaticum.
