Abstract

ABSTRACTEffectivity of EDTA (EthylenediaminetetraceticAcid) in Increasing Lead Accumulationof Saccharum spontaneum Grown Under Gold Mine Waste. Lead (Pb) isknown as one of a major metal contaminant in mine tailing. This metal has low solubility,and in many cases, is not readily available. In most soils capable of supporting plantgrow, the soluble Pb2+ levels are relatively low and will not promote substantial uptake bythe plant. In addition, many plants retain Pb2+ in their roots with only minimal trabsportto the aboveground plant portions. Therefore, it is important to find ways to enhance thebioavailability of Pb2+ or to find specific plants that can better translocate the Pb2+ intoharvestable portions for phytoremediation. The success of Pb phytoremediation is tohave significant Pb availability as well as a large quantity of plant biomass with high rateof growth. This experiment was conducted to obtain both purposes. Saccharumspontaneum which proven tolerant and dominant in contaminated area as well as potentialin producing high biomass was used in this research.The plants were grown in wastemedia added by 100 ppm, 200 ppm and 300 ppm of Pb. Ethylenediaminetetracetine Acid(EDTA) was applied to increase Pb availability and plant uptake as well as translocationto the aboveground portions. The results showed that the plant still capable of growingunder the highest level of Pb. EDTA increase Pb availability and plant uptake. Pb accumulationin the aboveground biomass of EDTA treated plants were relatively higher thanuntreated plants.Key words: Timbal, EDTA, phytoremediation

Abstract

Most artemisinin is extracted from an annual herb Artemisia annua L. but the cultivation in Indonesia is limited bythe low yield of the artemisin incontent that is not economically beneficial to industry. Improvement on cultivatedvarieties and cultivation techniques is therefore needed. This work aims to improve the cultivation techniques byevaluating the application effects of NPK, manure, and VAM on the artemisinin yield of A. annua. The experimentwas set using a Split Split Plot Design involving three factors. First factor was the application of four dosages ofNPK (0 kg ha-1 as a control; 40:40:40 kg ha-1; 80:80:80 kg ha-1; and 120:120:120 kg ha-1). Second factor wasmanure addition (using 0 ton ha-1 and 150 tons ha-1). Third factor wasthe application of mycorrhiza (0 g plant-1and15 g plant-1). The experiment was divided into three different groups based on the seed sources (Bandung, Cibodas,and Tawangmangu plants) using similar treatments. The results showed that the plant growth (Relative GrowthRates/RGR of plant heights and number of branches), leaf yield, and total plant biomass were much affected byNPK. The addition of manure has less significant effect on those parameters. Only VAM seems to influence theartemisinin content. The highest artemisinin yield (5 kg ha-1) was relatively low when compared to the productionin USA, China, and Brazil. However the result shows that a low fertilizer input of 40:40:40 kg NPK ha-1 and aninoculation of VAM were recommended for cultivating A. annua resulting a significantly high yield of artemisinin.Keywords: Artemisia annua L., Artemisinin content, NPK, Manure, VAM, Plant Growth, Biomass

Abstract

ABSTRACTEffect of Exploitation Rate on Reproductive Performance in Goldstripe Sardinella (Sardinellagibbosa) in West Java Coastal Waters. The research objective was to explore the effect ofexploitation rate on reproductive performance of goldstripe sardinella (Sardinella gibbosa).Three sites located in West Java coasts were selected representing coastal areas adjacent tothe Indian Ocean (Palabuhan Ratu), Java Sea (Blanakan), and Sunda Strait (Labuan), for fishcollection during May-July 2009. Fish samples were collected and the length was measuredprior to sex determination, observation on gonad morphology as well as gonad maturitydetermination, examination on fecundity and eggs diameter, and protein content analysis.Estimation on exploitation rate (E) was calculated based on length data performance fromwhich the result was correlated with reproductive parameters including the length at firstmaturity, fecundity, eggs distribution and diameter, and protein content analysis. By sitesbasis, variation in E was consistently shown only by eggs protein content in which the lowerthe E estimation the higher the protein contents. However, in response to E, there wereinconsistencies shown by the length at first maturity, fecundity, and eggs diameter. Suchinconsistencies are thought to be associated with population structure of matured female, andvariability in the habitat conditions which determines the magnitude of fish stock.Key words: Sardinella gibbosa, exploitation rate, reproductive performance

Abstract

ABSTRACTThe area of reclamation and revegetation in farmer coal mine has one role as bird habitat. The purpose of this study was to determine indicators of the types of bird that can progress successed reclamation and revegetation program in habitat function of fauna. The method used was bird species and bottom vegetation identification I in each habitat type used transect and mistnet. The data was analyzed using multivariate analysis of hDCCA. The results using hDCCA preedicted that the key indicator of bird characteristics that marks the development of succession in the fifth year is the presence of “insectivore bird” type. Bird species were B15 (Lanius schach), B20 (Prinia flaviventris), B23 (Picoides moluccensis), B28 (Pachychepala hypoxantha), B29 (Rhipidura javanica), B30 (Orthotomus ruficeps), B31 (Orthotomus sericeus).Keywords: hDCCA, insectivore bird, key indicator, reclamation and revegetation,

Abstract

ABSTRACTAnabaena azollae is an heterocyst Cyanophyceaean as symbiont of Azolla sp. Anabaena azollae is able to fix N2 from atmosphere and transform it into ammonium by its heterocyst cell. This research was conducted to study the influence of different concentration level of nitrogen to the dynamic of Anabaena azollae heterocyst cell. Research was conducted by observing heterocyst cell and variation nitrogen concentration in growing media in 21 days. In each sampling of Azolla sp., heterocyst observation was conducted for every 1000 cells in Anabaena azollae filament colonies. Result showed that media with 0 mg/L nitrogen has the highest number of heterocyst cell. In early observation, there was increasing of ammonium concentration in media 0 mg/L and 5 mg/L. Media without nitrogen addition has generated the highest number of heterocyst cell.Keywords: Anabaena azollae, Azolla sp., heterocyst

Abstract

ABSTRACTThe Effect of Depth to Deposition Process on Round Nucleus of Fresh Water Mussel (Anodontawoodiana). One of the affecting factors to the quality of pearl culture is the thickness of pearldepositions (nacre). The objective of this study was to obtain information on best level ofdepth to culture of pearl, to get fast nacre deposition and high quality of pearl. The researchwas conducted for 9 months, in the freshwater pond, was 300 m2 wide and 1 m deep. Freshwaterpearl Anodonta woodiana, sized ranging from 12 – 15 cm were studied. Completely randomizeddesign was used with levels of deep treatment (A) 30 cm; (B) 60 cm and (C) 90 cm. The resultshowed that best thickness of pearl deposition by 90 cm deep (1.30 mm) but hasn’t biggestsignificant (P>0.05) to the deep of 60 cm (1.10 mm) and biggest significant (P< 0.05) to the deepof 30 cm (0.70 mm). The result of implantation was followed that 30, 60 and 90 cm deep were 11.9%; 12.2 %; 12.0 %, whereas survival rate was followed 79.2 %; 79 % and 78.7 %.Keywords: Freshwater mussel; Anodonta woodiana; effect; level of deepKata kunci: Kerang air tawar, Anodonta, woodiana, kedalaman laut

Abstract

An indirect enzyme-linked immunosorbent assay (ELISA) was developed for screening of antibody to avian infectiousbronchitis (IBV). Antigen was prepared from whole virus of infectious bronchitis local isolate PTS-III serotype.Optimum dilution with minimum background for antigen concentration, rabbit anti-chicken conjugate and sera indeveloped ELISA were determined 0.4μg/well, 1:2000 and 1:100, respectively. Correlation optical densities (OD)were compared with a standard commercial ELISA (R2=0.933). The developed ELISA has a better sensitivity to hemagglutinationinhibition (HI) test. The developed local isolate ELISA can be used to detect antibody against infectiousbronchitis virus and it is suitable for sample screening at the diagnostic laboratories.Keywords: ELISA, antibody, chicken, IB PTS-III local isolate

Abstract

Mass propagation of leafy Anthurium (Anthurium spp.) from seed by in vitro technology. We have developed an efficient in vitro propagation of A. plowmanii from seed explants. The protocols include inoculation of seed in MS medium supplemented with 2 g/l BA. This medium is used to proliferate cluster of buds that developed from seeds. To induce the growth of the shoots from the bud clusters, the clusters were devided and transffered to MS medium enriched with 340 mg/l KH2PO4 and 0,1 mg/l BA and 0,05 mg/l NAA. To get high survival rate, the clusterof shoots were acclimatized as a whole cluster that in turn the shoots become easily separatable from each other during the growth. The acclimatized plantlets grew into mature plants through serial transfer to bigger pots and regular fertilizer application. Abstract

ABSTRACTNipah Virus Seroepidemiology in Flying Fox and Pig Husbandry in Several Areasof Indonesia. Nipah is a dangerous zoonotic disease which was carried by flying fox.The disease had been occurred in Malaysia in 1999 and infect pigs and caused humandeath. Indonesia is adjacent country to Malaysia, hence, a serological study had beenconducted on 156 flying fox (P. vampyrus) sera from North Sumatera, West Java, CentralJava and East Java. Besides that, 2740 pig sera was randomly collected in differentprovinces to detect Nipah infection. Both flying fox and pig sera were tested usingELISA test to detect the presence of Nipah antibody. The results indicated that 37 from156 flying fox sera (23.7%) has antibodies against Nipah virus. Infections were occuredin all sampling sites with the prevalence varied from 18% to 33 %. Meanwhile, no pigsera tested (2740) had antibody against Nipah virus. Based on these results it can beconcluded that Nipah virus infections were occurred in flying fox in some parts inIndonesia, but not in pigs. It was suggested that the presence of Nipah virus in Indonesiashould be anticipated. Hence the distribution of its infection in pigs and human must beanticipated. Monitoring of Nipah infection in areas adjacent to Malaysia must be increasedto detect the entering of the disease in Indonesia.Keywords: Nipah, pigs, flying fox, serology
