Abstract

Rabies has been spread to all over part of Bali, caused 160 people and thousands of dogs’ death. Theeffort for preventing rabies have been done, many villages after six months of eradicating rabies programmedhave been free, but a few villages still infected with the rabies. The aim of the study was to find the factorsthat make a number of villages are still infected with rabies. The study was conducted by doing a fieldsurvey to the nine districts of Bali. The survey was aimed at villages that remain infected despite effortsto control rabies has been done in the village. The device of survey is equipped with open ended and closedended questionnaire to be asked to the respondents. Characteristic villages infected with rabies will befigured out after the data were analyzed descriptively. The risk factors that prominent that make Baliinfected rabies include: dogs kept by releasing freely; presence of rabies carrier animals other than dogs;dog free contact with other dogs; the dogs were kept without having fed; puppies obtained from others; theflow of the dogs in and out to the village; And many communities have not obtained adequate counselingregarding the rabies. The conclusion that may be drawn is that there are some of factors that make Baliremain infected rabies.

Abstract

House flies (Musca domestica L.) are one of the major pests found in poultry farms resulting in not onlyannoyance and indirect damage to the poultry production but also transmitting many infectious organismsmechanically and biologically. A highly pathogenic avian influenza virus H5N1 (HPAIV H5N1) collectedfrom field house flies in Java Island, have fully succeeded identified and isolated in 2008. The AIV H5N1isolates were further used in the present study to determine the presence and persistence of the AIV H5N1in the laboratory infected house flies. One hundred house flies from a free AIV poultry farm in Yogyakarta,Indonesia were used in this study. The collected house flies were fasted for 12 hours then divided equallyin control and treated groups. The treated group was allowed to imbibe Dulbecco’s modified eagle medium(DMEM) containing AIV H5N1 while the control group imbibed medium without virus for one hour. Theflies from each group were collected at 12 and 24 hours post-exposure, respectively. All flies were immobilizedat 4°C, immersed in absolute ethanol for a few seconds and dissected under the stereomicroscope to collectthe gastrointestinal (GI) tracts. Based on the RT-PCR results, it is concluded that AIV H5N1 remainedexist in GI tracts of house flies for at least 24 hours post-exposure.

Abstract

Pelung Chicken is classified as dual-purpose chicken (meat and singer). Physical abnormalities affectthe selling price. One of the common physical abnormalities is crooked toes. The aims of this study were todeterminate the crooked toes inheritance pattern and the value of inbreeding coefficient (IC) in Pelungchicken. The method that used in this study is descriptive-explorative and samples were taken usingpurposive sampling analysis. The research was conducted at Cibeber, Cianjur district, West Java.inbreeding coeficient was determined using F = “ (1/2)n+1 (1+Fc) formula and inheritance pattern wasdetermined by the pedigree resulted and chi-square test goodness of fit test. The results show that aCreeper abnormality is inherited as autosomal recessive with the IC value reaches 0.6. The highest valueof IC shows highest risk probability of crooked toes in pelung chicken.

Abstract

Newcastle disease (ND) and Avian Influenza (AI) are infectious diseases and still endemic in Indonesia.Prevention of the disease is conducted by vaccination of birds as the source of the infection. The use ofcombined ND-AI vaccine is expected to be able to prevent both diseases simultaneously. This study aimwas to determine the potency of combined ND-AI vaccine in field condition. Field trial vaccination wasconducted in commercial layer chickens in Tabanan Bali, and the HI test was conducted at the Faculty ofVeterinary Medicine Udayana University, Denpasar. Field trial in commercial layer chickens showed thatthe average HI titer of ND sera from pre-vaccinated chickens was 22.7HI units and AI titer was 21.27 HIunits. The ND titers increased to 25.47 HI Unit, 27.0 HI units, and to 28.73 HI units, whereas AI titersincreased to 27.93 HI Unit, 28.53 HI units, and 28.47 HI units in two, three and four weeks post-vaccinationwith the ND-AI combined vaccine, respectively. Statistically, based on ND and AI antibody pre and postvaccination,it is indicated that the combined ND-AI vaccine was able to induce immune response higherthan the protective titer level (>24). Period of collecting the sera samples also affected the titer of NDVand AI antibodies (P<0.01). Therefore it is recommended that vaccination should be conducted at antibodytiter of < 4 HI Unit.

Abstract

The research was aimed to evaluate the combination effect of inulin dahlia tuber and Lactobacillus spdevelopment micro flora on intestinal and productivity of crossbred local chicken. The present researchwas assigned in a completely randomized design with factorial pattern, 2x3. The first factor was two levelsof dahlia tuber powder as inulin source 0.8% (A1) and 1.2% (A2), and, the second factor was three levels ofLactobacillus sp namely, without Lactobacillus sp (B0), 1.2 mL (108 cfu/mL) (B1) and 2.4 mL (108 cfu/mL)(B2). Each treatment combination was replicated four times with seven birds each. Parameters observedwere total of lactic acid bacteria (BAL), Escherichia coli, pH and body weight gain. The results showed thatfeeding inulin dahlia tubers at 1.2% and Lactobacillus sp at 1.2 mL (108 cfu/mL) sp either in partial orinteraction significantly (P<0.05) affected the total LAB, E. coli, pH and body weight gain. In conclusion,combination of inulin from dahlia tubers and Lactobacillus can increase total LAB and body weight gain,decrease total E. coli and pH.

Abstract

Kinetic study on Anti CPV IgY has been performed on six dogs aged 5-10 months. The IgY was injectedintravenously at dose of 21.4mg /10kg body weight. IgY levels in the blood were determined by ELISA. Aresearch was conducted to find out the kinetics of Anti CPV IgY in dogs blood. The kinetics of IgY wascalculated by using regression analysis to determine the association on the levels of IgY in serum againsttime at injection. The results showed that kinetic parameters were calculated based on first order kinetics.The constant elimination rate of IgY was at the range between 0.007 to 0.015 / h. IgY concentration in thedogs blood was from 0.746 to 0.992 mg / mL. The half-life of IgY was from 1.65 to 4.01 / d. Volumedistribution of IgY was between 21.47 to 28,55 / mL. Total IgY in the dog bodies (AUC) was from 42,60 to142,00 mg / mL.h. The duration of the IgY in the dog’s body was 3.08 to 8.51 days. Clearance time of IgY was0.15 to 0.50 mL / h. In conclusion the kinetics of anti CPV IgY in dog’s body follow one compartment andfirst order model, which are only distributed in the blood with the half-life at 2.5 days, and IgY has lesspossibility to accumulate in the body compared to the IgG.

Abstract

This study aims to determine the genetic variation of gene encoding fusion (F)-protein of avianparamyxovirus (APMV) type-1 isolated from chickens suffering of Newcastle disease (ND) found in achicken farm in the province of Bali throughout the year 2014. There are five isolates got from sick chickenscases/death suspected of being infected by APMV -1, but negative for avian influenza virus (AIV) infectioni.e: D5/AK/2014, B1/AK/2014, T1/ARP/2014, G1/AK/2014, and K1/ARP/2014. Sequence analysis of thefive isolates showed that three isolates of D5, B1 and G1 have a genetic distance of 11,1%, 10,2% and 8,2%when compared to the nucleotide sequence of Bali-1/07 isolated previously from Karangasem regency in2007. Meanwhile, the genetic distance of T1 and K1 isolates are 20,1% and 18% respectively as compareto the sequence of the Bali-1/07 isolate. The composition of amino acids at the cleavage sites enzyme of Fproteinfor 3 isolates of D5, B1 and G1 are R-R-Q- K-R-F, which is the typical characteristic of virulentvirus strain. Meanwhile, two isolates i.e T1 and K1 got from layer chicken had the amino acid sequence ofG- R-Q-G -R-L, which is group into avirulent virus strains. It was concluded that APMV-1 isolates foundin ND cases in the year of 2014 were caused by virulent isolate and were grouped into VII-genotype.Furthermore, It was found that there were two avirulent isolates

Abstract

This study was conducted to determine the seroprevalence of Brucella canis of imported dog for breederthrough Soekarno Hatta International Airport, Cengkareng Tanggerang, Indonesia. A total of 64 serumsamples were obtained from breeder dogs imported to Indonesia between Mei–September 2014. Theserums were examined by Immunochromatographic Assay. None of the serum samples in this studytested positive. Result of this study indicated that antibodies of B. canis were not detected in 64 dogsimported to Indonesia during the research period. A Knowledge, Attitude and Practises (KAP) Survey wasalso performed. This study used 32 respondents and data were analyzed by using path analysis. Therespondents were breeding kennel owners (breeder) who imported dogs through Soekarno HattaInternational Airport during the research period. The study concluded that the knowledge significantlyinfluenced the attitude of the breeder, and also the attitude significantly influenced the practises ofpreventing and controlling brucellosis. The annually income was also identified as a variable thatsignificantly influenced the practises. Practice prevention and control of canine brucellosis can be improvedby increasing the knowledge of the breeding kennel owner.

Abstract

Bovine viral diarrhea virus (BVDV) is a common pathogen that causes major economic losses to dairyindustry worldwide. Amplification of 5’-UTR regions of BVDV genome from blood samples usingreversetranscriptase polymerase chain reaction (RT-PCR) is one of sensitive and accurate techniques forBVDV detection. A common limitation for molecular detection of virus is the ability to obtain high qualityof nucleic acid in the samples. Inappropriate storage conditions will result in a false negative test. FiveEDTA-blood samples collected from BVDV persistently infected cattle were used in the present study.Ten repetitions were done for each sample. The blood were then dripped in the middle part of the FlindersTechnology AssociatesTM (FTA) cards and let it dried for approximately 2 hours. The effectiveness of theFTATMcards for storage and retrieval of BVDV RNA from blood samples molecularly using RT-PCRtechnique were tested and evaluated. The results proved that FTATM cards are effective and safe for thestorage of BVDV genome for a long period at room temperature. Based on the result of the present study,it can be concluded that in veterinary medicine field, the application of FTATM technology needs to bedeveloped to various type of samples and should be used routinely for collecting samples containinginfectious agents.
