Abstract

Trichinellosis is a zoonotic disease caused by parasitic roundworms of the nematode class Trichinella spp, which is distributed worldwide. Indonesia is one of the countries with trichinellosis cases. This study aims to determine the seroprevalence of Trichinellosis in pigs in Tangerang Village, Banten Province. The sample size is determined with 95% confidence level, the expected prevalence is 5%, and the error rate is 5%. A total of 80 sera were collected from farm (27 sera) and from the sample banks of Disease Investigation Centre Subang (DIC Subang(53 sera)). The sample collection was examined serologically by the ELISA method for the examination of anti-Trichinella antibodies. The results showed that seroprevalence of trichinellosis in this study was 1.25% (confidence interval (CI) 95%; 0,22-6,75%). This case can be a threat to human health. Pigs that show seropositive results were the pigs with traditional husbandry practices with ground flooring and without fences. The results of the questionnaires that distributed to the farmers indicated that all farmers did not know about trichinellosis, this may affect the presence of Trichinella in pigs.

Abstract

Ketamine is anesthesia that commonly used in the rabbit’s surgery as animal model often combined with transquilizer. The aim of this study is to evaluate the rabbit’s cardiac performance after administration of ketamine anesthesia combined with transquilizer. A total of 24 rabbits of New Zealand White strain were divided into four treatment groups, namely ketamine 40 mg/kg BW, combination of ketamine 10 mg/ kg BW and xylazin 3 mg/kg BW, ketamine 10 mg/kg BW and medetomidin 0.125 mg/kg BW and ketamine 10 mg/kg BW and acepromazin 1 mg/kg BW group. Evaluation were performed at 15, 30, 45 and 60 minutes after administration of anesthesia. Evaluation of cardiac performance using echocardiography on heart rate, stroke volume, ejection fraction, fractional shortening and cardiac output. The results showed that the heart rate in all treatment groups decreased along with observation time, except the ketamine group increased after 45 minutes. The stroke volume, cardiac output and fractional shortening in all treatment groups was stable, and the value was not significantly different among time observation (P> 0.05). The ejection fraction of the ketamine combined with transquilizer showed had the same pattern, decreasing at the 30 minutes observation followed by increasing at 45 and 60 minutes observation, while the ketamine group increased at 45 minutes but decreased again at minute 60. The lowest ejection fraction score was seen in the ketamine group. The research suggest that administration of ketamine with combined transquilizer medetomidin showed the most stable cardiac performance during observation.

Abstract

The aim of this study was to compare the quality of Bali bull fresh semen in productive group (below 10 years old) with over productive group and to evaluate the frozen semen quality of bali bull after cryopreservation in Tris egg yolk extender using Dimethyl formamide (DMF) and glycerol in three different concentrations (5%, 6%, and 7%). Semen was collected using artificial vagina. Semen then evaluated macro and microscopically. Macroscopic evaluation conducted visually and microscopic evaluation with the aid of Computer Assisted Sperm Analysis CASA. Immediatelly after evaluation, semen were divided into six aliquots and diluted with Tris egg yolk (TEY) glycerol 5% (TEYG5), TEY-glycerol 6% (TEYG6), TEY glycerol 7% (TEYG7), TEY-DMF 5% (TKTD5), TEY-DMF 6% (TEYD6) and TEY-DMF 7% (TKTD7). Results of experiment showed that there was no difference (P>0.05) among the all parameters, except for semen volume. Semen volume of productive group was higher than over productive age group. No difference was found in post thawing motility of the spermatozoa in TEY extender with 5% glycerol, and TEY with 5%, 6% and 7% DMF. The higher recovery rate of bali bull spermatozoa demonstrated by spermatozoa in TEY extender with glycerol 5% and DMF 6% (63.33±2.40 dan 68.67±2.33). It was concluded that bali bull semen quality did not differ between productive and over productive ages and post thawing semen quality of bali bull in TEY with 5-7% DMF or 5% glycerol was not also different.

Abstract

Insuline-like growth factor binding protein 2 (IGFBP2) is one of the principal binding proteins that has biological functions involved in growth, development, and differentiation. Selection for rapid growth based on molecular Marker Assisted Selection is required to increase production performance. The present study was designed to analyze the associations of IGFBP2 gene polymorphisms with chicken growth and body composition traits. Kampong chicken, a native chicken in Indonesia, is slow-growing chicken. A total of 59 males were used in the current study. Growth and body composition were measured in 24 wk of age. Primers for intron 2 region were designed from genomic chicken sequence. A c.1032C>T SNP of the IGFBP2 gene intron 2 region was detected and PCR-RFLP method was then used to genotype Kampong chicken population. The result showed that IGFBP2 polymorphism was significantly associated with body, carcass, breast, breast muscle, pectoralis minor, leg, and wings weight in Kampong chicken population (P < 0.05). The research suggests that the IGFBP2 gene could be a candidate gene that affects growth and body composition traits in chicken.

Abstract

Sukabumi District is one of the districts in West Java Province that has not been rabies-free since there are still reported rabies dog bites and rabies-positive cases in humans. The aim of the study was to measure the knowledge, attitude and practice of dog’s owner towards rabies which will then be useful for designing the appropriate rabies control programs. A total of 141 dog owners were selected as samples/ respondents. Respondents were taken from two sub-districts in Sukabumi District, which were 66 people from Jampang Tengah Sub-district and 75 people from Cisolok Sub-district. The data were taken using structured questionnaire. The result showed that most of respondent’s level of knowledge (51.1%) was in moderate category, meanwhile the respondent’s level of attitude related to rabies control program mostly (69.5%) was in positive category. But the positive attitude was not reflected with their practice towards rabies control program which many of them (63.1%) was in bad category of rabies control practices. From analytical result showed that only one characteristic variablewhich was the information access about rabies that had significant correlation (c2=9.959, p=0.006) towards respondent’s level of knowledge. Furthermore, another result showed that there was a significant relationship between knowledge variable with respondent’s attitude level (p= 0.000, r= 0.275).

Abstract

Chronic discomfort potentially occurred during a hemodynamic measurements in a rat-tail blood pressure chamber due to the animal is restrained in restricted space for a long period – it took 10 repetitions each at approximately 1.5 minutes for a session of hemodynamic measure. Procedures related to the animal welfare methods are required to improve the accuracy of the measurement which would altered from the discomfort to comfort state of the animal. Welfare procedures are represented by humane handling and habituation of the subject (rats) to the operator and the hemodynamic apparatus. Seven rats were subjected in this study and measured for hemodynamic value in three times point: before (T0), after humane handling (T1) and after human handling and habituation to the measurement apparatus (T2), respectively. Six out of seven subjects showed a significant lowering deviation for all variables of hemodynamic measurements, and that deviation followed by a declining trend from T0 to T2, i.e T0>T1>T2. The results indicated that welfare-based approach would benefit the animals from feeling discomfort and yet improving the accuracy and quality of the outcomes.

Abstract

Dangke whey is one type of protein and can be made edible film. The quality of the edible film made from dangke whey can be improved with the addition of carrageenan. The purpose of this research was to determine the quality of the edible film made from dangke whey with the addition of different concentrations of carrageenan. The qualities of the edible film observed were the microstructure, the color values L *, a * and * b, water vapor transmission rate (WVTR) and thickness. Data analyzed using ANOVA with a completely randomized design. The treatment used of carrageenan with different concentrations: 2%; 2.5%; 3% and 3.5%. Each treatment was repeated 3 times. Microstructure of edible film showed the improvement of interaction between the molecules of constituent materials. Values of color* L were 86.1088.37, the value of color * a were -0.55-0.70 and a value of the color * b were 2.03-3.35. Water vapor transmission rate (WVTR) were 5.08-5.35 g/mm2.jam. The thickness were 0.034-0.036 mm. The use of carrageenan concentrations up to 4% for manufacturing of edible film have not changed for the color * L, * a and * b, the rate of water vapor transmissions and thickness. Based on the microstructure showed that the molecules of edible film properly distributed beginning on the use of carrageenan concentration of 2.5%.

Abstract

Ringworm is an zoonotic infectious skin disease that can infect many types of animals. This disease is caused by dermatophytes fungi. Ringworm cases in cattle were quite widely reported in various countries but a report about the dermatophytes fungi that caused ringworm in bali cattle have never been published. It is very important in the efforts to give more effective therapies. This study aimed to identify the species of dermatophytes fungi that caused ringworm in bali cattle. Eight bali cattles suspected ringworm samples were taken using superficial skin scraping and trichogram (hair pluck) technique. The samples of skin scrapings dan hair on the area of the lesion were taken for direct microscopic examination to find the presence of fungal elements (hyphae or arthrospora). They were dropped with 10% KOH, were allowed for 10-15 minutes, then were observed using microscope. Skin scrapings dan hair samples that showed positive results were cultured on Sabauroud’s Dextrose Agar (SDA) medium for 1-3 weeks dan were identified using Lactophenol Cotton Blue with microscope. The data were analyzed descriptively. Dermatophytes fungi that isolated dan identified were Microsporum gypseum (75% or 6/8) and Microsporum nanum (25% or 2/8). Both of these fungi have ectothrix invasion/infection tipe, which is the forming of arthrospores/arthroconidia is only on the surface/superficial hair shaft therefore the topical therapy is sufficient to resolve the infection.
