Abstract

This study was designed to optimalize the use of epididymal or ejaculate sperm and plasma for in vitro fertilization, that sperm agglutination was found at preparation. The rate of sperm agglutination was calculated the head-to-head sperm agglutination that were incubated in Krebs Ringer-(N-(2hydroxyethyl)piperazine-N’-(2-ethenesulfonic acid) or KR-HEPES medium in 38.50C with 5% CO2 at 1, 3, 5 and 7 hours culture in vitro. The rate of head-to-head sperm agglutination were decreased with time treatments. The cauda of sperm agglutination was lower than that caput, corpus epididymal and ejaculate sperm with statistically significant (P<0.01). These result reflected that distribution of anti-agglutinin might be higher in cauda epididymal than that other areas. Number of protein were characterize with SDS-PAGE as follow 11 bands in caput epididymal, 9 bands in corpus epididymal, 2 bands in cauda epididymal and 4 bands in seminal plasma. The higher distribution of protein was found at range 25-40 kDa in epididymal plasma of ram. However, further investigation should be conducted to determine presumptive anti-agglutinin by advance method.

Abstract

The study was carried out to determine the response of PGF2 ? result extraction of Bali cattle seminal vesicle fluid on the level of progesterone. The seminal vesicle fluid was aspirated and then were. extracted with methanol. This reseach was conducted by devided the mare in the luteal phase into two groups. The first group were treated with PGF2 ? result extraction of Bali cattle seminal vesicle fluid and the second group were treated with dinoprost as a patent product of PGF2 ? were administered intra uterine. The level of progesterone was measured before (0 hours) treatment and at 24, 48, 72 hours after treatment. The level of Progesterone were determined by radioimmunoassay (RIA) technique. The result showed that PGF2 ? result extraction of Bali cattle seminal vesicle liquid decreased the progesterone level 73.03% at 24 hours and 92.79% at 48 Hours. However, there was no significant different between PGF2 ? result extraction of Bali cattle seminal vesicle liquid with PGF2 ? of paten product on the progesterone level decreased. In conclusion, PGF2 ? extraction of Bali cattle seminal vesicle liquid can decreas the level of progesterone in the mare with luteal phase.

Abstract

The objectives of the research was to described habitat charateristics and reproduction aspects of giant frog (Limnonectes cf. grunniens). Some physico-chemical parameters that characteristic for giant frog habitat i.e. air and water temperatur, relative humidity, dissolved oxygen, free CO2 and NH3 in water were measured. Moreover, the reproduction aspects being observed were gonad characteristics, egg size and form, and sperm density. The results indicated that the giant frog habitat was river stream with physical characteristics as following air temperature : 27o–28oC, water temperature : 24o-26 oC, relative humidity : 83%–87%, water turbidity : 1,2–11,6 nephelometric turbidity unit (NTU), and altitude : 20-40 m above sea level. Furthermore, the suitable water characteristics for the habitat were dissolved oxygen : 7.78.2 ppm, free CO2 concentration = 6.6–8.1 ppm, and free NH3 concentration : 0.5–0.6 ppm. The adult giant frog female generally carried 1461.82 eggs in average per individu with diameter ranging from 1.70–3.10 mm. On the other hand, the sperm density of adult male giant frog was found to be 1,614,898 per mm3 with motility rate of 81.23%.

Abstract

The aim of this research was study of Insulin Like Growth -I Complex to level of malondialdehide (MDA) and percentage of intact membrane plasm. Report of several species suggest that seminal plasma contains factors that may influence sperm viability. Seminal plasma was reported to be important for maintaining spermatozoa motility in bull and ram. For improving ram sperm viability and for increasing the resistance of boar spermatozoa to cold shock damage. IGF-I has been identified in the testis, where secreted by Leydig and Sertoli cells. Collection of semen by using artificial vagina from male goats and then characterized of motility and viability of spermatozoa. Identification of Insulin Like Growth Factor –I Complex were done by Native-PAGE 12% techniques, after several bands were identified and was done isolate of first band protein Insulin Like Growth Factor-I Complex. Insulin Like Growth Factor – I (IGF-I) Complex seminal plasma have molecular weight of 150,288 kDa, respectively. Sperm preparation for this research by centrifugation of semen with Bracket and Oliphan’s medium. These sperm preparation were characterized of motility, viability. Then after sperm preparation divided into three groups. The group I, added with Bracket and Oliphan’t medium, group II added with Bracket Oliphan’t and IGF – I Complex medium, group III added with IGF – I Complex medium and then were exposed 45 minutes for incubation and then percentage of MPU and level of MDA was evaluated. The result of this research showed that percentage of level MDA has significant different between group I,group II and group III, (p<0,05). The percentage of MPU sperm has significant different between groups I, group II and group III (p<0,05). The highest of percentage MPU was found in group III and the lower of level MDA was found in group III. Regresion test showed that negative correlation between level of MDA and percentage of MPU sperm.

Abstract

Former finding that avian influenza (AI) virus of H5N1 subtype from Indonesia shows reduced sensitivity against oseltamivir is critically reviewed trough molecular observation of the amino-acid sequence motive of neuraminidase protein (NA) of all H5N1 virus from human and animal in Indonesia available in GeneBank. Amino acid sequence of oseltamivir binding pocket of NA protein on all Indonesian viruses is typical for sensitive virus with a concerved motive of H274, E276, R292 dan N294. Resistance issue could not be explained based on available data.

Abstract

Fracture healing is a complex process that requires the ordered synthesis of matrix proteins and deposition of mineral. Complications such as delayed union or non-union may arise from excessive flexibility bone because low mineral content of bone or apposition of both fracture fragments is not complete yet. The aim of this study is to determine the effect of excessive calcium carbonate (CaCO3) on the healing femoral fracture in rat by examining histopathological change of femoral fracture. Twelve male Sprague Dawley rats, 12 weeks old were subjected to this investigation. The entire rats model at 13 weeks old were treated reposition of femoral dexter fractures with intramedullary pin fixation. The rats model were evenly randomly divided into three groups of four at 15 weeks old ; group I treated with normal diet, group II with normal diet + CaCO3 supplement 75 mg/day, group III with normal diet + CaCO3 supplement 225 mg/day per oral. At the end of the experiment (19 weeks old), the rats were killed, subsequently dexter femur were taken out for determining the histopathological examination. The results showed that excessive suplementation CaCO3 to description of histopathological fracture healing on group III had even better, callus was able to bridge the fracture fragment with completely. The proportion woven bone and trabeculae were higher, while group II were cartilage dominate in callus, and group I were higher fibrous tissue. From the results can be concluded that given CaCO3 225 mg/ day accelerate the healing process of femoral fractures.

Abstract

The purpose of the present study was to investigate the viability of mice and hamster embryos developed in Kalium Simplex Optimized Medium amino acid (KSOMaa) and Hamster Embryo Culture Medium-6 (HECM-6) medium. Female DDY mice were superovulated by injection i.p. of 5 IU Pregnant Mare Serum Gonadotropine (PMSG) and 5 IU Human Chorionic Gonadotropine (hCG) in 48 h interval, hamster (Phodopus campbelli) injected by 2.5 IU PMSG and 2.5 IU hCG 48 h later. Then females were mated with fertile males. Eight-cell embryos were recovered at day 3 after natural mating. The mice embryos were cultured in KSOMaa+5% NBCS (New Born Calf Serum) (T1) and HECM-6+5% NBCS (T2), the hamster embryos were cultured in KSOMaa+5% NBCS (T3) and HECM-6 + 5% NBCS (T4) for further development at 37oC in a humidified atmosphere of 5% CO2 in air for 48 h. The examinations were replicated five times. The T1 embryos developed to compact morulla and early blastocyst 100% (140/140), 92.1% (129/140) to blastocyst and expanded blastocyst, and 22.9% (32/140) became hatching/hatched. The T3 reached 100% (60/60) to compact morulla and early blastocyst, 85.0% (51/60) blastocyst, and 48.3% (29/60) expanded blastocyst, no embryo observed hatching/hatced. The T2 embryos had more expanded blastocyst than T3 (P<0.05), hatching/hatched rate higher than T1 and T3 but lower than T4 (P<0.05). Shortly, KSOMaa enable to support 8-cell stage mice and hamster embryo, but the hamster embryo developed lower at expanded blastocyst stage. HECM-6 is more appropriate than KSOMaa to support 8-cell mice embryos development and suitable to develop 8-cell stage hamster embryos.

Abstract

Research to confirm the abnormalities of blood corpuscles in Bali ducks based on blood smear examination has been conducted. The research samples consisted of 105 ducks from various regencies in Bali. The blood smear examination was conducted by the method described in this literature and the data was collected and tabulated by means of the descriptive method. Erythrocyte abnormalities were: polychromasia (61.9%), anisocytosis (17.1%), poikilocytosis (21.0%). Abnormalities associated with leukocyte composition were: Lymphopenic (46.7%); Heterophylic (44.8%); Heteropenic (3.8%), also Thrombocyto penia (2.9%) was present in 105 ducks. In conclusion 61,9% ducks examined deal with regenerative anaemia. They also have some blood abnormalities such as basophylia, eosinophylia, heterophylia, heteropenia, lymphopenia, and monocytosis.
